Abstract
본 발명의 목적은 점안시에 불쾌한 자극이 없고, CPB-I의 장기 보존 안정성이 양호한 점안제를 제공하는 것이며, CPB-I과 함께 다가 알코올, 바람직하게는 카르보닐가가 5 μmol/g 이하인 다가 알코올을 배합한 점안제를 제공하는 것이다.
Description
CPB-I(별명 "아넥신 V")은 인간 태반을 비롯한 생체 내의 조직 및 분비액에 넓게 분포하며[Chem. Pharma. Bull, 38, 1957-1960, 1990], 세포 내에서는 세포질에 존재하는 항혈액 응고 작용 등의 생리 활성을 갖는 물질이다. CPB-I을 인간 또는 동물의 장기로부터 추출할 수 있고(일본 특허 공개 (소)62-174023호 공보), 또한 유전자 재조합 기술에 의해 발현시킬 수 있다는 것(일본 특허 공개 (소)64-20095호 공보)이 보고되어 있다. 또한, CPB-I을 각막 질환 치료제로서 사용하는 것이 보고되어 있다(WO92/08475호).
그런데, 점안제에는 점안시의 눈에 대한 불쾌한 자극을 없애기 위해, 등장화제로서 염화나트륨 또는 염화칼륨 등의 염류를 배합하는 것이 일반적이다. 그러나 상기 염류를 CPB-I을 함유한 점안제에 배합시키면 CPB-I의 장기 보존 안정성이 문제가 되어 바람직하지 않다. 또한, CPB-I의 안정화를 위해 항산화제인 에틸렌디아민테트라아세트산(EDTA) 또는 그의 염, 아스코르브산의 배합을 고려할 수 있는데, EDTA 또는 그의 염은 CPB-I의 창상 치유 작용을 저해할 우려가 있는 한편, 아스코르브산은 안정한 pH역이 CPB-I과 상이하기 때문에 바람직하지 않다.
지금까지, 염기성 아미노산이나 당류를 배합함으로써 CPB-I을 분리 정제, 동결 건조 등을 행할 때 안정화시키는 방법(일본 특허 공개 (평)4-198195호 공보, 일본 특허 공개 (평)4-198196호 공보)이 알려져 있지만, 점안제의 장기 보존 안정성에 대해서는 검토되어 있지 않다. 즉, 점안시에 눈에 대한 불쾌한 자극이 없고, CPB-I의 장기 보존 안정성이 양호한 점안제를 제조하는 것이 곤란하여, 아직 만족할 만한 것을 얻지 못한 것이 현실이다.
본 발명은 점안시에 눈에 대한 불쾌한 자극이 없고, CPB-I의 장기 보존 안정성이 양호한 점안제 및 그의 제조 방법에 관한 것이다.
도 1은 다가 알코올의 카르보닐가가 CPB-I의 안정성에 미치는 영향을 나타낸다.
본 발명은 상기 관점으로부터 이루어진 것이며, CPB-I 함유의 점안제로서, 점안시에 눈에 대한 불쾌한 자극이 없고, CPB-I의 장기 보존 안정성이 양호한 점안제 및 그의 제조 방법을 제공하는 것을 과제로 한다.
본 발명자들은 이상의 사항을 고려하여 예의 연구를 거듭한 결과, 점안시에 불쾌한 자극이 없고, CPB-I의 장기 보존 안정성이 양호한 점안제의 처방을 발견하였다. 즉, CPB-I을 함유한 점안제에 있어서, 등장화제로서 다가 알코올을 함유시킴으로써 점안시에 불쾌한 자극이 없고, CPB-I의 장기 보존 안정성이 양호한 점안제를 제공할 수 있다는 것을 발견하고, 본 발명을 완성하였다. 또한, 다가 알코올을 카르보닐가가 낮은 다가 알코올로 함으로써 CPB-I의 장기 보존 안정성이 더욱 우수한 점안제를 제공할 수 있다는 것을 발견하고, 본 발명을 완성하였다.
즉, 본 발명은 이하와 같다.
(1) CPB-I과 다가 알코올을 함유하는 것을 특징으로 하는 점안제.
(2) 상기 (1)항에 있어서, 상기 다가 알코올이 글리세린, 프로필렌글리콜, 만니톨 및 소르비톨 중에서 선택되는 것을 특징으로 하는 점안제.
(3) 상기 (1) 또는 (2)항에 있어서, 상기 다가 알코올의 카르보닐가가 5 μmol/g 이하인 점안제.
(4) CPB-I과 함께 카르보닐가가 5 μmol/g 이하인 다가 알코올을 점안제에 배합하는 것을 특징으로 하는, CPB-I을 함유하는 점안제의 제조 방법.
이하, 본 발명을 상세하게 설명한다.
본 발명의 점안제는 CPB-I을 유효 성분으로서 함유한다. CPB-I로서는 인간 유래의 CPB-I을 들 수 있다. 인간 CPB-I의 아미노산 서열은 이미 보고되어 있다(일본 특허 공개 (소)64-20095호 공보). 인간 CPB-I은 서열 1로 표시되는 아미노산 서열을 갖는다.
또한, CPB-I은 전형적으로는 이하의 성질을 갖는다.

⑤ 아미노산 분석으로 아스파라긴산, 트레오닌, 세린, 글루탐산, 프롤린, 글리신, 알라닌, 1/2 시스틴, 발린, 메티오닌, 이소류신, 류신, 티로신, 페닐알라닌, 히스티딘, 리신 및 아르기닌의 존재가 확인됨
본 발명에서 사용하는 CPB-I은 점안제의 유효 성분으로서의 효과, 예를 들면 각막 질환 치료 등의 효과를 갖는 한, 그 유래, 당쇄의 유무 등은 특별히 제한되지 않는다. 또한, 상기 효과를 갖는 한, 아미노산 치환 등의 변경, 개질이 행해진 유도체를 사용할 수도 있다.
본 발명에서 사용되는 CPB-I은 바람직하게는 인간 CPB-I이지만, 상기 유효 성분으로서의 효과를 갖는 한, 서열 1의 아미노산 서열에 있어서, 1개 또는 복수개의 아미노산 잔기가 치환, 결실, 삽입 또는 부가된 아미노산 서열, 또는 C 말단 또는 N 말단에 치환기를 갖는 아미노산 서열을 갖는 단백질일 수도 있다.
CPB-I은 CPB-I을 함유하는 인간 또는 동물의 장기로부터 추출, 정제함으로써 취득할 수도 있지만(일본 특허 공개 (소)62-174023호), 대량 생산의 관점에서는 CPB-I을 코드하는 DNA를 이용한 재조합 DNA 기술에 의해 제조하는 것이 바람직하다. CPB-I 단백질을 코드하는 DNA는 이미 클로닝되어, 그의 염기 서열이 밝혀져 있다(일본 특허 공개 (소)64-20095호). 또한, 이 DNA를 이용하여 CPB-I을 생산시키는 방법이 기재되어 있으며, 이 방법에 의해 생산되는 CPB-I은 본 발명에서 바람직하게 사용할 수 있다.
본 발명의 CPB-I을 코드하는 DNA는, 코드되는 아미노산 서열이 상기 조건을 만족하는 것이면 특별히 제한되지 않지만, 구체적으로는 예를 들면 일본 특허 공개 (소)64-20095호 공보에 기재된 염기 서열을 갖는 DNA를 들 수 있다.
상기와 같은 CPB-I 단백질을 코드하는 DNA는, 이미 CPB-I 단백질의 아미노산 서열 및 이들을 코드하는 DNA의 염기 서열이 밝혀져 있기 때문에, 그 서열에 기초하여 제조한 프라이머를 이용한 PCR에 의해 인간 또는 동물의 염색체 DNA 또는 염색체 라이브러리로부터 취득할 수 있다.
CPB-I을 코드하는 DNA의 제조, 재조합 DNA 기술을 이용한 CPB-I의 제조에 있어서, 염색체 DNA의 제조, 염색체 DNA 라이브러리의 제조, 혼성화, PCR, 플라스미드 DNA의 제조, DNA의 절단 및 연결, 형질 전환 등의 방법은 일본 특허 공개 (소)64-20095호에 기재되어 있다.
본 발명에서 사용하는 CPB-I의 함유량은, 점안제의 전량에 대하여 0.0001 내지 1 중량%, 바람직하게는 0.001 내지 0.1 중량%, 특히 바람직하게는 0.003 내지 0.03 중량%일 수 있다.
본 발명의 점안제에는 상기 CPB-I과 함께 다가 알코올을 함유시킨다. 다가 알코올로서는 바람직하게는 2가 내지 6가의 알코올, 구체적으로는 글리세린, 프로필렌글리콜, 만니톨 및 소르비톨 등을 들 수 있으며, 바람직하게는 글리세린을 들 수 있다. 다가 알코올의 함유량은 점안제 조성물이 등장이 되는 정도의 양, 구체적으로는 점안제의 전량에 대하여 0.1 내지 20 중량%, 바람직하게는 1 내지 10 중량%, 특히 바람직하게는 1.5 내지 5 중량%일 수 있다.
다가 알코올 중에 포함되는 알코올 산화물, 구체적으로는 카르보닐 화합물의 함량이 높으면, 점안제의 보존 중에 CPB-I의 활성이 저하되는 경우가 있다. 따라서, 다가 알코올은 카르보닐화가 낮은 것을 사용하는 것이 바람직하다. 구체적으로는, 카르보닐가는 후술하는 실시예에 나타낸 값보다 낮은 값, 예를 들면 5 μmol/g 이하, 보다 바람직하게는 3 μmol/g 이하, 더욱 바람직하게는 1 μmol/g 이하인 것이 바람직하다.
여기서, 본 발명에서 사용하는 다가 알코올의 카르보닐가란, 예를 들면 시료를 일정한 조건하에서 2,4-디니트로페닐히드라진으로 발색시켜 흡광도법으로 구했을 때의 시료 1 g 중의 카르보닐 화합물의 μmol 수를 말한다[A. S. Heick, M. F. Benca, Jr. Mitchell, JAOCS 31, 88(1954), 미쯔나가 신지, 시마무라 우마지로, 유까가꾸; 7, 275(1958)]. 이하에, 카르보닐가의 일반적인 측정 방법을 나타낸다.
<카르보닐가의 측정 방법>
농글리세린 1 g을 측량하고, 알데히드 무함유 에탄올을 첨가하여 전량을 50 ㎖로 한다. 제조한 액체 5 ㎖를 25 ㎖의 메스 플라스크에 넣어 3.6 %(W/V) 트리클로로아세트산 용액(톨루엔에 용해) 2 ㎖와 0.05 %(W/V) 2,4-디니트로페닐히드라진 용액(톨루엔에 용해) 3 ㎖를 첨가하여 교반하고, 60±1 ℃의 항온조에서 30 분간 반응시킨 후, 실온까지 방냉한다. 또한, 4 %(W/V) 수산화칼륨ㆍ알데히드 무함유 에탄올 용액 5 ㎖를 첨가하고, 톨루엔을 첨가하여 25 ㎖로 한다. 수산화칼륨 용액을 첨가하고 나서 10 분 후에 435 nm에서 흡광도를 측정한다. 농글리세린을첨가하지 않고 공시험을 행하여 보정한다. 알데히드 표준 원액(노나날[와꼬 쥰야꾸] 0.142 g을 톨루엔에 용해하여 100 ㎖로 하고, 이 액체 5 ㎖에 톨루엔을 첨가하여 100 ㎖로 한 것)을 1, 3, 5 및 7 ㎖ 채취하고, 알데히드 무함유 에탄올을 첨가하여 각각 25 ㎖로 한다. 각 용액 5 ㎖를 25 ㎖ 메스 플라스크에 넣고, 상술한 표준 시험법과 동일한 조작을 행하여 검량선을 작성한다.
본 발명의 점안제는 일반적으로 사용되는 첨가제, 예를 들면 완충제, 점조제, 안정화제 및 비이온 계면활성제 등을 함유할 수 있다.
완충제로서는 붕산, 붕사, 시트르산, 인산제일염, 아세트산염, 인산제이염, 탄산수소염 및 탄산염 등을 들 수 있다. 완충제의 함유량은, 점안제의 전량에 대하여 0.01 내지 2 중량%, 바람직하게는 0.05 내지 1 중량%, 특히 바람직하게는 0.1 내지 0.5 중량%일 수 있다. 점안제의 pH는 바람직하게는 5 내지 10, 보다 바람직하게는 6 내지 9이다.
점조제로서는 폴리비닐알코올, 폴리비닐피롤리돈 및 카르복시비닐 중합체 등을 들 수 있다. 점조제는 점안액의 눈 밖 유출을 방지하고, 눈 안에서의 체류 시간을 연장하여 약효를 높인다. 또한, CPB-I 점안제를 진탕하면 CPB-I의 석출이 발생할 우려가 있지만, 점조제를 함유함으로써 CPB-I의 석출이 억제된다. 점조제의 함유량은 점안제의 전량에 대하여 0.01 내지 2 중량%, 바람직하게는 0.025 내지 1 중량%, 특히 바람직하게는 0.05 내지 0.5 중량%일 수 있다.
안정화제로서는 카프릴산나트륨, 아르기닌, 아황산나트륨, 아황산수소나트륨 등을 들 수 있다. 안정화제의 함유량은 점안제의 전량에 대하여 0.01 내지 2 중량%, 바람직하게는 0.05 내지 1 중량%, 특히 바람직하게는 0.1 내지 0.5 중량%일 수 있다.
비이온 계면활성제로서는 지방산 다가 알코올, 폴리옥시에틸렌 소르비탄 지방산 에스테르, 폴리옥시에틸렌 경화 피마자유 등의 폴리옥시에틸렌에테르, 알킬폴리옥시에틸렌에테르, 알킬카르보닐폴리옥시에틸렌, N,N'-디(폴리옥시에틸렌)알칸아미드 등의 에틸렌옥시드 부가체, 지방산 자당 에스테르, N,N'-디(알카놀)알칸아미드 및 폴리옥시에틸렌 블럭 공중합체 등을 들 수 있다. 비이온 계면활성제의 함유량은 점안제의 전량에 대하여 0.01 내지 5 중량%, 바람직하게는 0.02 내지 2 중량%, 특히 바람직하게는 0.03 내지 1 중량%일 수 있다.
또한, CPB-I의 장기 보존 안정성의 관점에서, 일반적으로 점안제의 등장제로서 사용되는 염화나트륨 또는 염화칼륨 등의 염류는 사용하지 않거나, 또는 함유량을 안정성을 손상시키지 않을 정도로 낮추는 것이 바람직하다.
본 발명의 점안제는 점안 용액 제제이다. 이 제제는 사용시 용해 제제 또는 희석 제제로 제조할 수도 있다. 사용시 용해 제제로 제조할 경우에는, CPB-I과 다가 알코올을 별도로 포장할 수도 있다.
본 발명을 실시예에 의해 더욱 구체적으로 설명한다.
또한, 이하에 기재된 실시예 및 비교예에서 나타낸 점안제는, 모두 pH를 7.5로 조절하기 위해 붕산과 붕사 또는 디소듐 모노히드로겐포스페이트와 모노소듐 디히드로겐포스페이트 디히드레이트를 각각 조정하여 배합하였다. 또한, 모든 제제의 침투압을 286 mOsm으로 조정하도록 농글리세린, 프로필렌글리콜, 소르비톨, 염화나트륨 및 염화칼륨을 조정하여 배합하였다.
<실시예 1>
CPB-I 0.01 g, 농글리세린 2.3 g, 붕산 0.11 g, 붕사 0.085 g, 폴리옥시에틸렌 소르비탄 지방산 에스테르 0.05 g, 폴리비닐피롤리돈 0.1 g을 멸균 정제수에 첨가하여 전량을 100 ㎖로 하고, 교반 용해하였다.
<실시예 2>
농글리세린 2.3 g 대신에 프로필렌글리콜 2.1 g, 붕사 0.085 g 대신에 붕사 0.016 g을 첨가한 것 외에는, 실시예 1과 동일한 방법으로 제조하였다.
<실시예 3>
농글리세린 2.3 g 대신에 소르비톨 4.0 g, 붕산 및 붕사 대신에 디소듐 모노히드로겐포스페이트 0.26 g 및 모노소듐 디히드로겐포스페이트 디히드레이트 0.05 g을 첨가한 것 외에는, 실시예 1과 동일한 방법으로 제조하였다.
<실시예 4 내지 6>
카르보닐가가 상이한 농글리세린을 사용한 것 외에는, 실시예 1과 동일한 방법으로 제조하였다.
<비교예 1>
농글리세린 2.3 g 대신에 염화나트륨 0.8 g, 붕사 0.085 g 대신에 붕사 0.011 g을 첨가한 것 외에는, 실시예 1과 동일한 방법으로 제조하였다.
<비교예 2>
농글리세린 2.3 g 대신에 염화칼륨 1.0 g, 붕사 0.085 g 대신에 붕사 0.011 g을 함유한 것 외에는, 실시예 1과 동일한 방법으로 제조하였다.
<실시예 및 비교예의 점안제의 평가>
실시예 1 내지 6 및 비교예 1 내지 2의 점안제에 대하여 CPB-I의 안정성을 검토하였다. CPB-I의 안정성은 45 ℃에서 2 주일간 보존했을 때의 CPB-I의 잔존율 (제조 직후를 100 %로 함)로서 측정하였다.
결과를 표 1, 표 2 및 도 1에 나타내었다. 또한, 도 1은 표 2의 결과를 그래프로 나타낸 것이다.
이상과 같이 실시예 1 내지 3 및 비교예 1 내지 2의 결과로부터, 점안제에 등장화제로서 다가 알코올을 함유시키면, CPB-I의 장기 안정성이 양호한 점안제를 얻을 수 있음을 확인할 수 있었다.
또한, 실시예 1, 4 내지 6의 결과로부터 다가 알코올의 카르보닐가가 낮을 수록 CPB-I의 장기 안정성이 양호한 점안제를 얻을 수 있음을 확인할 수 있었다.
본 발명에 의해 점안시에 불쾌한 자극이 없고, CPB-I의 장기 보존 안정성이 양호한 점안제를 제공할 수 있다.
Claims (4)
- CPB-I과 다가 알코올을 함유하는 것을 특징으로 하는 점안제.
- 제1항에 있어서, 상기 다가 알코올이 글리세린, 프로필렌글리콜, 만니톨 및 소르비톨로 이루어지는 군에서 선택되는 것을 특징으로 하는 점안제.
- 제1항 또는 제2항에 있어서, 상기 다가 알코올의 카르보닐가가 5 μmol/g 이하인 점안제.
- CPB-I과 함께 카르보닐가가 5 μmol/g 이하인 다가 알코올을 점안제에 배합하는 것을 특징으로 하는, CPB-I을 함유하는 점안제의 제조 방법.
