Abstract
본 발명은 종양 세포의 성장 인자-의존성을 저해하기 위한, 프로게스테론 수용체 저해제의 용도에 관한 것이다.
Description
에스트라디올 및 프로게스테론은 흉부암의 발생과 관련있다. 그러나, 진단시 종양의 약 1/3만이 스테로이드 호르몬 의존성을 나타낸다. 대부분의 스테로이드 호르몬-저항성 종양에서, 국소-작용성 오토크린 또는 파라크린 펩티드계 성장 인자에 대한 증식 억제가 일어나는 것으로 생각된다. 이 경우, 성장 인자-수용체-양성 및 스테로이드 호르몬-저항성인 극히 불량한 예후를 갖는 침윤성 종양이 생긴다(엘리지(Elledge) 등의 문헌[Semin. Onkol. 19(1992), 244-253]).
성장 인자는 세포 표면의 매우 유사한 티로신 키나제 수용체에 결합한 후 세포내 신호 전달 경로의 활성화에 의한 세포 성장을 조절한다. 보다 최근의 연구 결과 흉부 암종 세포가 EGF의 유사분열 유발 작용에 대하여 프로게스틴에 의해 감작될 수 있음을 시사한다(그로숑(Groshong) 등의 문헌[Mol. Endocrinol. 11(1997), 1593-1607]). 따라서, 예를 들어 인간 흉부 암종 세포주 T47D에서 프로게스테론이 시클린 1D 및 시클린-의존성 키나제 4의 활성의 일시적 증가가 수반되는 S기에서세포의 개시를 유도하는 것은 불가능하였다. 그러나, 성장 촉진은 하나의 주기에 제한되지 않으며, 제2 주기의 G1/S-전이에서 성장 정지가 따른다(그로숑 등의 상기 문헌(1997); 무스그로브(Musgrove) 등의 문헌[Mol. Cell. Biol. 13(1993), 3577-3587]). 프로게스테론에 의해 중지된 상태에서, 세포는 EGF의 증식 작용에 민감하다. 또한, 프로게스테론은 EGFR, Erb2 및 Erb3을 증가시킴으로써 T47D 세포에 대한 EGF의 작용을 증진시키고, 신호 분자의 티로신 포스포릴화를 증가시키는 것으로 나타났다(레인지(Lange) 등의 문헌[J. Biol. Chem. 273(1998), 31308-31316]; 리쳐(Richer) 등의 문헌[J. Biol. Chem. 273(1998), 31317-31326]). 대조적으로, 프로게스테론 수용체에 영향을 줌으로써 종양 세포에 대한 EGF의 작용의 저해를 나타내는 것이 가능하지 않았다.
본 발명은 종양 세포의 성장 인자 의존성을 저해하기 위한 프로게스테론 수용체 저해제의 용도에 관한 것이다.

본 발명에 따른 시험의 범주내에서, 매우 놀랍게도 프로게스테론 수용체, 예컨대 17α-플루오로알킬 스테로이드의 저해제는 종양 세포, 특히 프로게스테론 수용체의 다량 및(또는) 항상 발현을 나타내는 종양 세포에 대한 성장 인자(예: EGF)의 결합을 적어도 부분적으로 저해할 수 있음이 밝혀졌다.
따라서, 본 발명의 요지는 종양 세포에 대한 성장 인자의 결합을 저해하기 위한, 특히 성장 인자에 의해 일어나는 종양 세포 또는 종양의 증식을 저해하기 위한 제제의 제조를 위한, 프로게스테론 수용체의 저해제의 용도이다. 본 발명에 있어서 프로게스테론 수용체의 저해제는 바람직하게는 프로게스테론의 그의 수용체에 대한 결합을 경쟁적으로 저해하는 물질이다. 이 경우, 프로게스테론 수용체의 저해제는 바람직하게는 17α-플루오로알킬 스테로이드중에서 선택되며, 이들은 예컨대 WO 98/34947호에 개시되어 있다. 이러한 17α-플루오로알킬 스테로이드는 하기 화학식 I을 나타낸다:

R7은 수소 원자, 직쇄 C1-C4알킬 기 또는 분지쇄 C3-C4알킬 기이거나, 또는St가 스테로이드 ABC-고리 시스템 A 또는 B를 나타내는 경우, R6과 R7은 함께 추가의 결합을 나타내고,
X는 산소 원자, 히드록시미노 기 =N-OH 또는 2개의 수소 원자를 의미하고,
R8은 Y 라디칼 또는 여러 위치에서 Y 기로 임의로 치환된 아릴 라디칼이며, Y는 수소 원자, 할로겐 원자, -OH, NO2, -N3, -CN, -NR9aR9b, -NHSO2R9, -CO2R9, C1-C10알콕시, C1-C10알카노일옥시, 벤조일옥시-C1-C10알카노일, C1-C10히드록시알킬 또는 벤조일 기를 나타내고, R9a및 R9b는 동일하거나 상이하고, R9는 수소 원자 또는 C1-C10알킬 기를 나타내며, Y가 -NR9aR9b라디칼인 경우, 산과 함께 생리적으로 상용가능한 염이 생성될 수 있고, Y가 -CO2R9(식 중, R9는 수소를 뜻함) 라디칼인 경우, 염기와 함께 생리적으로 상용가능한 염이 생성될 수 있다.
프로게스테론 수용체의 이러한 저해제의 특히 바람직한 예는 화합물 11β-(4-아세틸페닐)-17β-히드록시-17α-(1,1,2,2,2-펜타플루오로에틸)-에스트라-4,9-디엔-3-온(하기 화합물 A)이다. 그러나, 다른 항프로게스틴, 예를 들어 오나프리스톤(11β-[p-(디메틸아미노)페닐]-17α-히드록시-17-(3-히드록시프로필)-13α-에스트라-4,9-디엔-3-온)도 또한 적합하다.
프로게스테론 수용체 저해제의 작용은 다량 및(또는) 구성적 프로게스테론수용체 발현을 나타내는 종양 세포, 예를 들어 프로게스테론 수용체-양성 흉부 암종 세포주 T47D에서 발견된다(사르토리우스(Sartorius) 등의 문헌[Cancer Res. 54(1994), 3668-3877]).
프로게스테론 수용체 저해제는 성장 인자, 특히 EGF 수용체류, 예컨대 EGF 수용체의 성장 인자에 결합하는 인자의 프로게스테론-유도성 증진을 저해한다. 저해제는 특히 바람직하게는 인간 흉부암에 대한 EGF의 결합을 저해한다.
그러므로 본 발명에 따라, 프로게스테론 수용체 저해제는 포유동물 및 바람직하게는 인간에서, 특히 종양의 진행, 특히 스테로이드-의존성 성장으로부터 성장 인자-의존성 성장으로의 진행을 차단하는 종양 치료를 위해 사용될 수 있다. 이러한 식으로, 종양의 유효 치료는, 예컨대 항에스트로겐에 의해 스테로이드-의존성 성장의 단계에서 일어날 수 있으며, 종양은 환자의 예후가 상당히 악화됨과 관련된 성장 인자-의존성 성장의 단계에서 진행될 수 없다. 프로게스테론 수용체 저해제의 투여는 또한 성장 인자-의존성 성장의 단계에서 종양 성장을 느리게 할 수 있다.
본 발명에 있어서, 비스테로이드계 항에스트로겐, 예컨대 타목시펜 및 나폭시딘, 및 랄록시펜 및 EM800이 사용될 수 있다. 나중 2개의 항에스트로겐은 언급된 SERM(Selective Estrogen Receptor Modulators, 선택적 에스트로겐 수용체 조절제)의 대표 물질이며, 또한 SERM의 작용 프로파일을 갖는 다른 화합물, 예컨대 PCT/EP99/05093호에 언급된 화합물 및 그중에서도 특히 5-(4-{5-[(RS)-4,4,5,5,5-펜타플루오로펜틸)술피닐]-펜틸옥시}페닐)-6-페닐-8,9-디히드로-7H-벤조시클로헵텐-2-올이 본 발명에 따라 사용될 수 있다.
스테로이드계 항에스트로겐의 예는 EP 0 348 341 A호에 개시된 것, 특히 파슬로덱스, 및 WO 98/07740호에 개시된 것, 특히 11β-플루오로-7α-{5-[N-메틸-N-3-(4,4,5,5,5-펜타플루오로펜틸티오-프로필아미노]-펜틸}-에스트라-1,3,5(10)트리엔-3,17β-디올, 또는 WO 99/33855호에 기술된 것, 특히 11β-플루오로-7α-{5-[메틸-(7,7,8,8,9,9,10,10-노나플루오로-데실)-아미노]-펜틸}-에스트라-1,3,5(10)트리엔-3,17β-디올 또는 그의 약학적으로 상용가능한 유도체 또는 유사체를 포함한다. 항에스트로겐 효과를 갖는 아로마타제 저해제, 예컨대 EP 0 495 825 B1호의 7 내지 8쪽으로부터 알려진 것도 마찬가지로 항에스트로겐으로서 사용될 수 있다.
프로게스테론 수용체 저해제의 투여는 일반적으로 사용되는 방법에 따라, 예를 들어 국부적으로, 국소적으로, 피하내로, 장으로 또는 비경구적으로 수행될 수 있다. 장 투여에 있어서, 특히 정제, 코팅 정제, 캡슐, 환제, 현탁액 또는 용액이 적합하며, 이들은 생약에서 공지된 첨가제 및 부형제를 가지고 통상의 방식으로 제조할 수 있다. 국부 또는 국소 사용에 있어서, 예를 들어 질좌제 또는 경피 시스템(예: 피부 패치)이 적합하다. 피하 투여는 오일상 용액으로 주사함으로써 수행될 수 있다.
투여 단위는, 예를 들어 활성 화합물(들)(=프로게스테론 수용체의 저해제(들)) 0.1 내지 100㎎을 함유할 수 있다. 인간에게 투여하기 위하여, 활성 화합물(들)의 1일 투여량은 약 0.1 내지 400㎎, 바람직하게는 약 10 내지 100㎎, 특히 약 50㎎이다.
또한, 본 발명을 하기 실시예 및 도면에 의해 설명하겠다.
1. 물질 및 방법
물질:
125I-EGF(100mCi/mmol)를 아머샴 버클러(Amersham Buchler)에 의해 얻었다. 화합물 A인 히드로타목시펜(4-OH-Tam), ZM182780 및 에스트라디올은 공지의 방법에 따라 쉐링 아게(Schering AG)의 인스티투트 퓌르 아르즈나이미텔케미(약제 화학 연구소]에서 합성하였다.
세포주:
인간 에스트로겐 수용체(ER)- 및 프로게스테론 수용체(PR)-양성 흉부 암종세포주 T47D(프리이크(Freake) 등의 문헌[BBRC 101(1981), 1131-1138]을 사용하였다.
성장 연구:
RPMI 배지 및 10% 소 태아 혈청, 200nM 인슐린 및 0.1nM 에스트라디올에서, 각 경우에 지시된 화합물의 존재하에, 96웰 평판에서 5000세포/웰로 6일동안 종양 세포를 배양하고, 크리스탈 바이올렛으로 염색하여 성장을 결정하였다.
PR 및 ER 단백질의 양:
세포 용해물내의 PR 및 ER의 양은 푸르만(Fuhrmann) 등의 문헌[Contraception 54 (1996), 243-251]에 기술된 방법에 따라 방사성 표지된 프로게스테론 또는 에스트라디올에 의한 스테로이드 결합 분석을 사용하여 결정하였다.
종양 세포에 대한125I-EGF의 결합:
R5020-전처리된 T47D 세포를 4℃에서125I-EGF와 함께 2시간동안 배양하였다. 비특이성 결합은 항상 전체 결합의 10% 미만이었다.
트랜스활성화 분석:
T47D 세포를 MTV-LUC(카토(Cato) 등의 문헌[EMBO J., 9: 2237-40])로 일시적으로 트랜스고정하고 1nM R5020의 부재하 또는 존재하에 배양하였다. PR-매개성 길항작용에 대한 시험에서, 일시적으로 트랜스고정된 T47D 세포를 R5020으로 처리하고, 또한 증가 농도의 화합물 A 또는 RU486으로 처리하였다. 24시간 후, 루시페라제 시험을 수행하였다.
2. 결과
도 1은 여러 시험 물질의 항증식 작용을 나타낸다. 0.1nM E2및 증가 농도의 화합물 A(▲), 오나프리스톤(■), ZK191703(●) 또는 4-OH-Tam(◆)의 존재하(상부 대각선 부분) 또는 부재하(하부 대각선 부분)에 T47D 세포를 배양하였다. T47D 세포의 경우, 화합물 A는 또한 극히 작은 농도에서 상당한 항증식 작용을 나타낸다.
도 2는 T47D 세포내 PR- 및 ER-단백질의 양을 나타낸다.
도 3은 T47D 세포내 PR의 전사 활성을 나타내며, 여기서 개개의 세포를 MTV-LUC로 일시적으로 트랜스고정하고, (a) (Co)의 부재하에 또는 1nM R5020의 존재하에 배양하였다. PR-매개성 길항물질에 대한 시험에서, 일시적으로 트랜스고정된 T47D 세포를 0.1nM R5020 및 증가 농도의 화합물 A 또는 RU468(b)로 처리하였다.
도 4에서, T47D 세포에 대한125I-EGF 결합의 스캣챠드 분석이 도시되어 있다. 세포를 20nM의 화합물 A를 함유하거나 함유하지 않는 20nM R5020의 존재하에 48시간동안 배양한 다음 세척하였다. 그 다음, 4℃에서 2시간동안 배양하여 EGF 0.25 내지 150ng/㎖의 농도 범위에 걸쳐 EGF-결합을 결정하였다. 삽입 그림은 자유 리간드 농도의 대수에 비교하여 결합된 리간드의 양을 나타낸다. 화합물 A(하부 도면)가 첨가되는 경우 모니터링(상부 도면)에 대하여 R5020(중간 도면)에 의해 일어난 EGF 결합의 증가를 차단하는 것이 가능하였음이 분명하다.
도 5에서, 완전 T47D 세포에 대한125I-EGF의 결합이 도시되어 있다. 이를 위하여, 세포를 2 또는 20nM R5020 및 화합물 A 또는 오나프리스톤, 또는 화합물 A 단독으로 48시간동안 처리하였다. 여기서 화합물 A가 R5020에 의해 일어난 T47D 세포에 대한 EGF-결합의 증가를 차단함을 또한 알 수 있다. 유사한, 그러나 상당히 더 약한 효과가 오나프리스톤에 경우 발견된다.
3. 고찰
상기 결과들은 다량 및 구성적 PR 접촉을 갖는 T47D 세포의 에스트라디올-촉진된 성장이 화합물 A에 의해 효과적으로 차단되었음을 나타낸다.
트랜스활성화 분석에 의해, PR은 T47D 세포에서 전사 활성이고 화합물 1에 의해 차단될 수 있었음을 나타내는 것이 불가능하였다.
T47D 세포를 R5020으로 자극하면 2배 또는 3배 증가된 EGF-수용체 발현이 일어나며, 이는 화합물 A에 의해 차단되었다. 동시에, 세포에 대한 EGF의 결합은 2배 내지 3배 증가되며, 화합물 A에 의해 방지될 수 있고 오나프리스톤에 의해서는 덜 효과적으로 방지될 수 있었다. R5020-처리된 세포에 대한 증가된 EGF-결합은 증진된 EGF-수용체 발현 또는 EGF 수용체와 erbB2 사이의 증가된 헤테로이량체 형성에 의해 일어날 수 있었다.
이들 결과는 인간 흉부 암종 세포에서 PR- 및 성장 인자-신호 시스템 사이의 상호작용을 나타낸다. 항프로게스틴의 사용에 의해, 스테로이드-의존성 성장으로부터의 종양 세포의 진행은 성장 인자-의존성 성장이 저해되거나 방지된다.
Claims (10)
- 종양 세포 및(또는) 종양에 대한 성장 인자의 결합을 저해하기 위한 제제의 제조를 위한, 프로게스테론 수용체의 저해제의 용도.
- 제1항에 있어서, 성장 인자에 의해 생성되는 종양 세포 및(또는) 종양의 증식이 저해됨을 특징으로 하는 용도.
- 제1항 또는 제2항에 있어서, 프로게스테론 수용체의 저해제가 하기 화학식 I의 17α-플루오로알킬 스테로이드중에서 선택되는 용도.<화학식 I>상기 식에서,R1은 메틸 또는 에틸 기를 나타내고,R2는 식 CnFmHo의 라디칼을 나타내며, 식중 n은 2, 3, 4, 5 또는 6이고, m은 1보다 크고, m+o는 2n+1이고,R3은 유리, 에테르화 또는 에스테르화 히드록시 기를 나타내고,R4및 R5는 각각 수소 원자를 나타내며, 함께 추가의 결합 또는 메틸렌 기를 나타내고,St는 하기 화학식 A, 화학식 B 또는 화학식 C의 스테로이드 ABC-고리 시스템을 나타내고,<화학식 A><화학식 B><화학식 C>R6은 수소 원자, 직쇄 C1-C4알킬 기 또는 분지쇄 C3-C4알킬 기 또는 할로겐 원자이고,R7은 수소 원자, 직쇄 C1-C4알킬 기 또는 분지쇄 C3-C4알킬 기이거나, 또는 St가 스테로이드 ABC-고리 시스템 A 또는 B를 나타내는 경우, R6과 R7은 함께 추가의 결합을 나타내고,X는 산소 원자, 히드록시미노 기 =N-OH 또는 2개의 수소 원자를 의미하고,R8은 Y 라디칼 또는 여러 위치에서 Y 기로 임의로 치환된 아릴 라디칼이며, Y는 수소 원자, 할로겐 원자, -OH, NO2, -N3, -CN, -NR9aR9b, -NHSO2R9, -CO2R9, C1-C10알콕시, C1-C10알카노일옥시, 벤조일옥시-C1-C10알카노일, C1-C10히드록시알킬 또는 벤조일 기를 나타내고, R9a및 R9b는 동일하거나 상이하고, R9는 수소 원자 또는 C1-C10알킬 기를 나타내며, Y가 -NR9aR9b라디칼인 경우, 산과 함께 생리적으로 상용가능한염이 생성될 수 있고, Y가 -CO2R9(식 중, R9는 수소를 뜻함) 라디칼인 경우, 염기와 함께 생리적으로 상용가능한 염이 생성될 수 있다.
- 제3항에 있어서, 프로게스테론 수용체의 저해제가 화합물 11β-(4-아세틸페닐)-17β-히드록시-17α-(1,1,2,2,2-펜타플루오로에틸)에스트라-4,9-디엔-3-온인 용도.
- 제1항 내지 제4항중 어느 한 항에 있어서, 종양 세포가 다량 및(또는) 구성적 프로게스테론 수용체 발현을 나타내는 용도.
- 제1항 내지 제5항중 어느 한 항에 있어서, 종양 세포가 흉부 암종 세포인 용도.
- 제1항 내지 제6항중 어느 한 항에 있어서, EGF 수용체에 결합하는 EGF 및(또는) 다른 인자의 결합이 종양 세포에서 저해되는 용도.
- 제1항 내지 제7항중 어느 한 항에 있어서, EGF 수용체와 erbB2 사이의 헤테로이량체의 형성이 저해되는 용도.
- 제1항 내지 제8항중 어느 한 항에 있어서, 종양 치료법인 용도.
- 제9항에 있어서, 스테로이드-의존성 성장으로부터 성장 인자-의존성 성장으로의 종양의 진행을 저해하는 용도.
