바람직한 천연 고분자의 예로는, 알부민 및 프롤라민(prolamines)(예를 들어, 제인(zein))과 같은 단백질과 알기네이트, 셀룰로오스 및 폴리히드록시알카노에이트(예를 들어, 폴리히드록시부티레이트)와 같은 다당류가 포함된다.
특히 소화관과 같은 점막 표면의 영상화에 사용하기에 좋은 생체 점착성 고분자로는, 폴리언히드라이드, 폴리아크릴산, 폴리(메틸 메타크릴레이트), 폴리(에틸 메타크릴레이트), 폴리(부틸 메타크릴레이트), 폴리(이소부틸 메타크릴레이트), 폴리(헥실 메타크릴레이트), 폴리(이소데실 메타크릴레이트), 폴리(라우릴 메타크릴레이트), 폴리(페닐 메타크릴레이트), 폴리(메틸 아크릴레이트), 폴리(이소프로필 아크릴레이트), 폴리(이소부틸 아크릴레이트), 폴리(옥타데실 아크릴레이트)가 포함된다.
용매

DOTMA, N-[1-(2,3-디올레오일옥시)프로필-N,n,N-트리에틸암모니움클로라이드; DOTAP, 1,2-디올레오일옥시-3-(트리메틸암모니오)프로판; 및 DOTB, 1,2-디올레오일-3-(4'-트리메틸-암모니오)부타노일-sn 글리세롤과 같은 다양한 캐티오닉 지질이 사용될 수 있다.
가장 바람직한 지질은 인지질이고, 바람직하게는 DPPC, DDSPC, DAPC, DSPC, DTPC, DBPC, DLPC 이고, 가장 바람직하게는 DPPC, DAPC 및 DBPC 이다.
지질의 함량범위는 0.1-30(지질중량/고분자중량)이고; 가장 바람직하게는 0.1-10(지질중량/고분자중량) 사이이다.
다른 소수성 화합물
다른 바람직한 소수성 화합물로는 트립토판, 트리오신, 이소류신, 류신 및 발린과 같은 아미노산, 알킬파라벤(예를 들어, 메틸파라벤) 및 안식향산과 방향족 화합물이 포함된다.
영상화제
가스
생체적합성 또는 제약적으로 수용가능한 어떠한 가스도 마이크로파티클 내에 혼입될 수 있다. 가스라는 용어는 기체 또는 영상화가 수행되는 온도에서 기체가 될 수 있는 어떠한 화합물도 포함한다. 가스는 산소, 질소, 크세논, 아르곤, 질소와 같은 단일화합물, 또는 공기와 같은 혼합물일 수 있다. 불소첨가 가스가 보다 바람직하다. 불소첨가 가스로는, 예를 들면, CF4, C2F6, C3F8, C4F8, SF6, C2F4및 C3F6가 포함된다. 과불소화카본이 특히 바람직한데, 이는 사용 온도에서 응축되지 않고 제약적으로 수용가능한 비수용성 가스를 제공하기 때문이다.
다른 영상화제

MRI에서 조영제로 사용되는 적합한 제제의 예로는, 철, 마그네슘, 망간, 구리 및 크롬 뿐만 아니라 디에틸렌 트리아민 펜아세트산(DTPA) 및 가토펜토테이트 디메글루민(gatopentotate dimeglumine)과 같은 최근에 이용가능한 가탈리늄 킬레이트(gatalinium chelates)가 포함된다.
CAT와 x-선에 사용되는 제제의 예로는, 디아트리조에이트(diatrizoate) 및 이오탈라메이트(iothalamate)에 의해 대표되는 이온성 모노머, 이오파미돌, 이소헥솔 및 이오베르솔과 같은 비이온성 모노머, 이오트롤 및 이오딕사놀(iodixanol)과 같은 비이온성 다이머, 예를 들어 이옥사갈트(ioxagalte)와 같은 이온성 다이머 등의 정맥주사용 요오드 제제가 포함된다. 다른 유용한 제제로는, 경구용 바륨이 포함된다.
마이크로캡슐 및 그 제조방법
최적의 실시예에서, 마이크로파티클은 분무건조법(spray drying)에 의해 만들어진다. 아래에서 설명하는 용매추출법(solvent extraction), 고온용융캡슐화법
(hot melt encapsulation) 및 용매증발법(solvent evaporation) 등과 같은 다른 기술도 사용될 수 있다. 중요한 특징은, 마이크로파티클을 형성하기 전에 먼저 고분자가 지질에 용해되거나 용융되어야 한다는 것이다. 본 명세서에서 비록 지질이 혼입되는 것으로 특정하게 기술된다 해도, 다른 유용한 소수성 화합물이 지질을 대치할 수 있는 것으로 이해되어야 한다.
바람직한 실시예에서, 원하는 가스의 분류를 유입하거나, 또는 캡슐화된 가스를 제거하기 위해 마이크로스피어에서 진공상태로 축출한 다음 원하는 가스로 충진시켜 가스를 치환할 수 있다.
a. 용매증발법(solvent evaporation)
본 방법에서 고분자와 지질은 염화메틸렌과 같은 휘발성 유기용매에 용해된다. 기공 형성제가 고형으로 또는 용액상으로, 마이크로파티클 형성에 사용되는 상기 용액에 첨가될 수 있는데, 이것은 영상화제로서 가스를 혼입하기 위함이다. 만일 혼입되는 것이 다른 영상화제라면, 영상화제는 고형 또는 용액의 어느 한 형태로 고분자 용액에 첨가될 수 있다. 이 혼합물은 음파처리 또는 균질화되고, 분산 또는 유화된 다음에 트윈 20(TWEENTM20), 트윈 80(TWEENTM80), PEG 또는 폴리(비닐 알코올)과 같은 계면활성제를 함유하는 수용성 용액에 첨가되고 균질화되어 에멀젼을 형성하게 된다. 이렇게 만들어진 에멀젼은 대부분의 유기용매가 증발되어 마이크로스피어가 남게 될 때까지 교반된다. 몇 가지 다른 고분자 농도로도 사용될 수 있다 (0.05-0.60 g/㎖). 다른 크기(1-1000 미크론)와 형태를 가진 마이크로스피어가 이 방법에 의해 얻어질 수 있다. 이 방법은 폴리에스테르 및 폴리스티렌과 같은 비교적 안정한 고분자에 대해 유용하다.

그러나, 폴리언히드라이드 같은 불안정한 고분자는 수분의 존재에 의해 제조공정 중에 분해될 수 있다. 이러한 고분자에 대해서는, 완전한 유기용매 내에서 수행되는 다음의 두 가지 방법이 보다 유용하다.
b, 고온-용융 마이크로캡슐화법(Hot Melt Microencapsulation)

고온-용융 마이크로캡슐화법은 E. Mathiowitz, et al., Reactive Polymer, 6, 275 (1987)에 기술되어 있다. 폴리언히드라이드, 예를 들어 20 : 80의 분자비를 갖는 비스-카복시페녹시프로판과 세바신산(sebacic acid) (P(CPP-SA) 20:80)(Mw 20,000)으로 되어 있는 폴리언히드라이드는 고온-용융 마이크로캡슐화법에 의해 제조될 수 있고, 또는 예를 들어, 폴리(푸마릭-코-세바식)(20:80)(Mw 15,000) 마이크로파티클은 고온-용융 마이크로캡슐화법에 의해 제조될 수 있다.
c. 용매제거법(solvent Removal)

d. 마이크로파티클의 분무건조법
마이크로파티클은 생체적합성 고분자와 지질을 적절한 용매에 용해시키고, 기공형성 제제를 이 고분자 용액에 분산시킨 다음에 고분자 용액을 분무건조하여 마이크로파티클을 형성함으로써 분무건조법에 의해 만들어질 수 있다. 여기서, 고분자와 기공형성 제제의 용액을 "분무건조"하는 공정은, 용액이 좋은 분무상을 만들기 위해 정밀하게 분말화되고 뜨거운 담체 가스와 직접적으로 접촉됨으로써 건조되는 공정으로 정의된다. 이 기술분야에서 이용가능한 분무건조 장치를 사용하여 고분자 용액은 분무건조기의 입구를 통과하고 건조기내의 튜브를 지난 다음 출구를 통과하여 정밀하게 분말화될 수 있다. 온도는 사용된 가스 또는 고분자에 따라 변할 수 있다. 입구와 출구의 온도는 원하는 산물을 만들기 위해 조절될 수 있다.
고분자 용액의 미립자 크기는, 고분자 용액을 분무하기 위해 사용되는 노즐의 기능, 노즐 압력, 유출속도, 사용된 고분자, 고분자의 농축도, 용매의 타입 및 분무온도(입구와 출구온도 모두)와 분자량에 관계된다. 일반적으로, 농축도가 같다고 할 때 보다 큰 분자량이 캡슐의 크기가 보다 크다. 분무건조법의 전형적인 공정 파라미터는 다음과 같다: 고분자 농도 = 0.005-0.20g/㎖, 입구 온도 = 30-1000℃, 출구온도 = 20-100℃, 폴리머 유출속도 = 5-200㎖/min 및 노즐 직경 = 0.2-4㎜ID. 1 내지 10 미크론의 직경 범위를 갖는 마이크로파티클은 고분자, 농도, 분자량 및 분무속도의 선택에 따라 정해지는 형태로 얻어질 수 있다.
만일, 영상화제가 고형이면, 분무에 앞서 고분자용액에 첨가되는 고형입자로 캡슐화되거나, 또는 분무에 앞서 영상화제가 수용액에 용해된 다음 고분자용액과 함께 에멀젼화되거나, 또는 분무에 앞서 상기 고형물이 적절한 용매내의 고분자와 함께 공용해(cosolubilized)될 수 있다.
e. 하이드로겔 마이크로스피어
폴리포스파젠(polyphosphazene) 또는 폴리메틸메타크릴레이트 같은 겔-타입 고분자로 된 마이크로파티클은, 고분자를 수용성 용액에 용해시키고 이 혼합물에 소정의 기공형성 제제와 현탁된 지질을 현탁시키고, 이 혼합물을 균질화한 후, 미소적(微小適)(작은 물방울)을 형성하는 장치를 거쳐 이를 분출시키고, 반대하전 이온 또는 서서히 교반되는 고분자성 전해질로 구성된 경화조로 떨어지는 미소적(微小適)을 형성함으로써 만들어진다. 이러한 처리체계의 장점은, 제조후 폴리리신과 같은 다중 양이온성 고분자로 코팅함으로써 마이크로파티클의 표면을 추가적으로 변형시킬 수 있다는 능력에 있다. 마이크로파티클 입자는 다양한 크기의 분출제를 사용함으로써 조절되게 된다.
마이크로파티클 형성을 촉진하는 첨가제
영상화제-함유 마이크로파티클이 합성되는 동안 다양한 계면활성제가 첨가될 수 있다. 사용될 수 있는(0.1-5 중량%로) 유화제 또는 현탁화제로는, 물리학적으로 수용가능한 대부분의 유화제가 포함된다. 예를 들어, 담즙산염 또는 담즙산의 천연 및 합성형, 아미노산과 콘쥬게이트된 것과, 타우로데옥시콜레이트 및 콜린산과 같이 콘쥬게이트되지 않은 것 양쪽 모두가 포함된다.
기공형성 제제는, 기공형성능을 증진시키기 위해 0.01 내지 90 중량 대 용량%의 양으로 함유될 수 있다. 예를 들어, 분무건조법, 용매증발법에서, 휘발성 염, 예를 들어 암모늄 비카보네이트, 초산 암모늄, 염화암모늄 또는 안식향산 암모늄 등과 같은 기공형성 제제 또는 다른 냉동건조된 염이 먼저 물에 용해된다. 기공형성 제제를 함유하는 이 용액은 고분자 용액으로 유화되어 고분자내에 기공형성 방울(drop)들을 만든다. 다음에 이 유화제는 분무건조되거나 또는 용매증발/추출 공정을 통해 제거된다. 고분자가 침전된 후 경화된 마이크로파티클이 동결되고 기공형성 제제를 제거하기 위해 동결건조된다.
마이크로파티클의 크기
폐 모세혈관을 통과할 수 있는 주사가능한 마이크로파티클의 제조에 관한 바람직한 실시예에서 마이크로파티클은 약 1 내지 10 미크론의 직경을 갖는다. 보다 큰 마이크로파티클은 폐를 막을 수 있고, 보다 작은 마이크로파티클은 충분한 음파반사성(echogenicity)을 제공할 수 없다. 보다 큰 마이크로파티클은 주사외의 다른 경로, 예를 들어 (소화관의 검진을 위한) 경구, 다른 점막표면에의 적용(직장, 질, 경구, 비강), 또는 흡입 등과 같이 투여될 때 유용하다. 경구투여에 대한 바람직한 입자 크기는 약 0.5 미크론 내지 5㎜이다. 제약적으로 수용가능한 유용한 운반체로는 글리세롤 및 트윈20(TWEENTM20)을 함유하는 식염수와 트윈20(TWEENTM20)을 함유하는 등장성 만니톨이 포함된다. 입자크기 분석은 광학 현미경, 스캔닝 전자현미경, 또는 투과 전자현미경에 의한 계수기 측정으로 수행될 수 있다.
표적화
마이크로파티클은 마이크로파티클을 형성하는 고분자, 마이크로파티클의 크기, 및/또는 마이크로파티클에 대한 리간드의 결합 또는 부착의 선택을 통해 특이적 또는 비특이적으로 표적화될 수 있다. 예를 들어, 생물학적 활성분자, 또는 전하를 띠는 분자, 친유성 또는 친수성 입자가 마이크로파티클의 표면에 부착될 수 있다. 추가로, 조직의 점착을 최소화하고, 또는 생체내에서 마이크로파티클의 특정 표적화를 촉진하는 분자가 마이크로파티클에 부착될 수 있다. 대표적인 표적화 분자에는, 항체, 레시틴 및 특정 타입의 세포 표면에 있는 수용체에 의해 특별히 결합되는 다른 분자가 포함된다.
RES에 의한 흡수 억제
또한, 마이크로파티클의 흡수와 제거는 고분자의 선택 및/또는 점착과 흡수를 최소화하는 분자의 혼입 또는 커플링을 통해 최소화될 수 있다. 예를 들어, 마이크로파티클에 의한 조직의 점착은, 마이크로파티클 표면에 대해 공유적으로 결합하는 폴리(알킬렌 글리콜) 성분에 의해 최소화될 수 있다. 표면 폴리(알킬렌 글리콜) 성분은, 물에 대해 높은 친화성을 가져 입자 표면으로의 단백질의 흡수를 감소시킨다. 그러므로, 세망내피계(RES)에 의한 마이크로파티클의 인식과 흡수가 감소된다. 예를 들어, 폴리(알킬렌 글리콜)의 말단 히드록실 기는 생물학적 활성분자 또는 전하를 나타내는 분자, 친유성 또는 친수성 입자를 마이크로파티클의 표면으로 공유적으로 부착시키는데 사용될 수 있다. 이 기술분야에서 이용가능한 방법이, 생체내에서 마이크로파티클의 송달 성질, 안정성 또는 다른 성질을 향상시키기 위하여 마이크로파티클에 어떠한 종류의 리간드를 부착시키는 데에도 이용될 수 있다.
진단 적용
마이크로파티클은 일반적으로 인산완충식염수 또는 식염수 또는 만니톨과 같은 제약적으로 수용가능한 운반체와 결합한 다음, 검진에 유효한 양으로 적절한 경로를 이용하여, 즉 일반적으로 주사에 의해 혈관으로, 또는 경구로 환자에게 투여된다. 캡슐화된 영상화제를 함유하는 마이크로파티클은 혈관 조영 뿐만 아니라 간 및 신장 질환의 검진에 대한 적용, 심장에 대한 적용, 종양의 크기와 조직의 검출 및 특정에, 그리고 말초혈액의 점도측정 등에 이용된다. 또한, 마이크로파티클은 조직 점착을 최소화하는, 또는 전술한 것과 같이 생체내에서 신체의 특정 영역에 마이크로파티클을 표적화하는 리간드와 결합될 수 있다.
상기의 방법과 조성이 다음과 같은 비제한적인 실시예에 의해 설명된다.
실시예 1: 레시틴을 갖는 옥타플루오로프로판 PEG-PLGA/PLGA 마이크로파티클의 제조
3.2g의 PEG-PLGA(75:25)(IV=0.75 dL/g Birmingham Polymers), 6.4g의 PLGA(50:50)(IV=.4 dL/g Henley Chemicals), 23㎎ 레시틴 (Spectrum chemicals), 및 193㎎의 팔미틴산(Spectrum chemicals)이 190㎖의 염화메틸렌에 용해되었다. 0.70g/㎖ 초산 암모늄 10.8㎖가 상기 고분자용액에 가해지고, 고분자/초산 암모늄이 상기 고분자용액에 가해지고, 이 고분자/초산 암모늄 혼합물이 버티스(Virtis) 균질기를 사용하여 1분 동안 10,000 RPM으로 균질화되었다. 이 용액은 20㎖/min의 유속으로 펌프되고, 부치 랩(Bucchi Lab)분무건조기를 이용하여 분무건조되었다. 입구온도는 40℃이었다. 마이크로파티클 분말이 모아지고, 120시간 동안 FTS 트레이 동결건조기로 동결건조되었다. 이 마이크로파티클은 5㎖ 퍼폼 바이알로 분배되어 부틸 스토퍼로 밀봉되었다. 상기 바이알은 10 psig의 압력에서 옥타플루오로프로판으로 충진되고, 가스하에서 3분간 계속적으로 정화되었다. 이 시점이후에 상기 바이알은 사용될 때까지 4℃로 저장되었다. 상기 입자의 직경은 1-10 미크론의 범위를 가졌고, 계수측정기로 측정할 때 개수 평균 2 미크론 정도를 나타내었다. 스캔닝 전자현미경은 입자가 일반적으로는 평활한 표면의 구형이며, 때때로 톱니같은 형상을 가진다는 것을 보여주었다.
실시예 2: 디팔미토일포스파티딜콜린(DPPC)을 가지고 있는 옥타플루오로프로판 PEG-PLGA/PLGA 마이크로파티클의 제조
레시틴 대신에 29.6㎎의 디팔미토일포스파티딜콜린이 사용되는 것을 제외하고는 실시예 1과 같은 방법으로 마이크로파티클을 제조하였다.
실시예 3: 디스테아로일포스파티딜콜린(DSPC)을 가지고 있는 옥타플루오로프로판 PEG-PLGA/PLGA 마이크로파티클의 제조
레시틴 대신에 29.9㎎의 디스테아로일포스파티딜콜린(Avanti, Birmingham A1)이 사용되는 것을 제외하고는 실시예 2와 같은 방법으로 마이크로파티클을 제조하였다.
실시예 4: 디아라키도일포스파티딜콜린(DAPC)을 가지고 있는 옥타플루오로프로판 PEG-PLGA/PLGA 마이크로파티클의 제조
레시틴 대신에 29.9㎎의 디아라키도일포스파티딜콜린(Avanti, Birmingham A1)이 사용되는 것을 제외하고는 실시예 2와 같은 방법으로 마이크로파티클을 제조하였다.
실시예 5: 시험관내에서의 마이크로파티클의 후방산란 측정
초음파빔에 촛점이 맞추어진 마이크로파티클 현탁액 10 마이크로리터를 노출시킴으로써 실시예 1-4에서 제조된 옥타플루오로프로판을 함유하는 각각의 고분자성 마이크로파티클의 후방산란을 얻었다. 후방산란된 어쿠스틱 파워는 샘플에 대한 깊이로서 다음과 같이 결정되었다. 파동-수용체(Pulser-receiver, PanametricsModel 5800)가 사용되어 초음파 트랜듀서(2.25㎒)를 쇽 흥분시키고, 생리식염수의 마이크로파티클 현탁액에 초음파의 파동을 보낸다.
37℃로 온도가 조정된 워터 바스에 들어있는 원통의 샘플 챔버(식염수 55㎖)에 상기 현탁액을 포함시켰다. 상기 챔버를 트랜듀서로부터 1.5 인치에 위치시키고, 바로 그 트랜듀서를 챔버의 어쿠스틱 윈도우에 촛점을 맞추었다. 현탁액내에서 마이크로파티클이 유지되도록 챔버를 15rpm으로 회전시켰다. 용해된 산소
(OrionModel 840)로 측정된 바와 같이, 약 90%의 공기 포화상태가 유지되도록 상기 식염수에 가스를 용해시켰다. 어쿠스틱 파워 테스트 시스템의 작동은, LabVIEW(National Instruments)의 프로그램으로 운영되는 PC로 조절된다. 이 컴퓨터는 파동-수용체를 작동시켜 초음파 트랜듀서를 쇽 흥분시킨다.
후방산란된 신호가 상기 동일한 트랜듀서로 수용되고, 되돌아 방출되는 신호는 상기 파동-수용체에 의해 증폭되었다. 이 증폭된 신호는 100MSa/s에서의 디지털화를 위해 디지털 오실로스코프(LeCroymodel 9310AM)를 통과하였다. 디지털화된 신호는 계속적으로 진행되었다. 이 신호는 FFT에 의해 정산되고 분석되었으며, 트랜듀서의 밴드폭 6 dB 이상으로 합성되었다. 아비트라리 유니트(arbitrary units)에 있어서 마이크로파티클 현탁액의 깊이 기능과 같이, 상기 시스템으로 수집된 어쿠스틱 데이터는 합성 후방산란력 (IBP)으로 전환되었다. IBP 대 깊이 데이터는 50 펄스부터 평균화되고, 베스트 피팅 직선은 IBP 데이터를 통해 결정되고, 후방산란 효과를 촉진하는 y-인터셉트가 결정되었다. 각각의 샘플은 총 시간 10분 이상으로 2.5분 간격으로 테스트되었다.
4개의 서로 다른 마이크로파티클에 대한 시간 기능에 따른 후방산란이 도 1에 나타나있다. 레시틴은 사슬 길이가 다른 인지질의 혼합이다. 인지질에 부착된 지방산 사슬길이가 길어짐에 따라 후방산란의 매그니튜드는 보다 오랜시간 지속되는데, 이는 마이크로파티클내의 옥타플루오로프로판의 증가된 안정성을 나타낸다. 또한, 고도로 정제된 인지질을 사용하는 것이, 레시틴이 함유된 인지질의 혼합물과 비교하여 가스의 안정화면에서 보다 효과적이다.
Claims (30)
- 생체적합성 고분자로 형성되고 영상화제가 혼입되어 있는, 진단영상을 위한 마이크로파티클의 제조방법으로서, 상기 마이크로파티클에 대한 수분의 침투율 또는 흡수율을 감소시키는 유효량의 소수성 화합물을 상기 고분자에 혼입시키는 것을 포함하는 방법.
- 제1항에 있어서, 상기 소수성 화합물은, 고분자의 중량에 대한 소수성 화합물의 중량이 0.01 내지 30의 비율로 상기 고분자와 혼합되는 것을 특징으로 하는 방법.
- 제2항에 있어서, 상기 소수성 화합물은 0.01 내지 30의 비율(지질 중량/고분자 중량)로 상기 고분자와 혼합되는 지질인 것을 특징으로 하는 방법.
- 제1항에 있어서, 상기 지질은 지방산 및 그 유도체, 모노-, 디 및 트리글리세라이드, 인지질, 스핑고리피드, 콜레스테롤 및 스테로이드 유도체, 오일, 비타민 및 테르펜으로 구성된 군으로부터 선택되는 것임을 특징으로 하는 방법.
- 제4항에 있어서, 상기 지질은 포스파티드산류, 포화 및 불포화 지질을 모두 갖는 포스파티딜콜린, 포스파티딜에탄올아민, 포스파티딜글리세롤, 포스파티딜세린, 포스파티딜이노시톨, 리조포스파티딜 유도체, 카디오리핀(cardiolipin) 및 β-아실-y-알킬인지질로 구성된 군으로부터 선택된 인지질인 것을 특징으로 하는 방법.
- 제5항에 있어서, 상기 인지질은 디올레오일포스파티딜콜린, 디미리스토일포스파티딜콜린, 디펜타데카노일포스파티딜콜린, 디라우로일포스파티딜콜린, 디팔미토일포스파티딜콜린, 디스테아로일포스파티딜콜린, 디아라키도일포스파티딜콜린, 디베헤노일포스파티딜콜린, 디트리코사노일포스파티딜콜린, 디리그노세로일포스파티딜콜린 및 포스파티딜에탄올아민으로 구성된 군으로부터 선택되는 것임을 특징으로 하는 방법.
- 제1항에 있어서, 상기 영상화제는 불소첨가가스, 산소, 크세논(xenon), 아르곤, 헬륨 및 공기로 구성된 군으로부터 선택된 가스인 것을 특징으로 하는 방법.
- 제7항에 있어서, 상기 가스는 CF4, C2F6, C3F8, C4F8, SF6, C2F4및 C3F6으로 구성된 군으로부터 선택되는 것임을 특징으로 하는 방법.
- 제8항에 있어서, 상기 가스는 옥타플루오로프로판인 것을 특징으로 하는 방법.
- 제1항에 있어서, 상기 영상화제는 자기공명영상화(MRI), 컴퓨터 단층촬영, x-선, 양전자 방사 단층촬영(PET), 단일광자 방사 컴퓨터 단층촬영으로 구성된 군으로부터 선택된 영상화 기술에 의해 검출가능한 것임을 특징으로 하는 방법.
- 제1항에 있어서, 상기 마이크로파티클은 합성 고분자로 형성된 것임을 특징으로 하는 방법.
- 제1항에 있어서, 상기 마이크로파티클은 천연 고분자로 형성된 것임을 특징으로 하는 방법.
- 제1항에 있어서, 상기 마이크로파티클은 생체점착성 고분자로 형성된 것임을 특징으로 하는 방법.
- 제11항에 있어서, 상기 마이크로파티클은 폴리(히드록시산), 폴리언히드라이드, 폴리오르토에스테르, 폴리아미드, 폴리카보네이트, 폴리알킬렌, 폴리알킬렌 글리콜, 폴리알킬렌 옥사이드, 폴리알킬렌 테레프탈레이트, 폴리비닐 알코올, 폴리비닐 에테르, 폴리비닐 에스테르, 폴리비닐 할로겐화물, 폴리비닐피롤리돈, 폴리실록산, 폴리(비닐 알코올), 폴리(비닐 아세테이트), 폴리스티렌, 폴리우레탄 및 그들의 공중합체, 합성 셀룰로오스, 폴리아크릴산, 폴리(부틸산), 폴리(발레르산) 및 폴리(락티드-코-카프로락톤), 에틸렌비닐아세테이트, 그들의 공중합체 및 혼합물로 구성된 군으로부터 선택된 합성 고분자로 형성된 것임을 특징으로 하는 방법.
- 생체적합성 고분자로 형성되고 영상화제가 혼입되어 있는, 진단영상을 위한 마이크로파티클로서, 상기 마이크로파티클에 대한 수분의 침투율 또는 흡수율을 감소시키는 유효량의 소수성 화합물을 상기 고분자에 혼입시키는 것을 포함하는 마이크로파티클.
- 제15항에 있어서, 상기 소수성 화합물은 고분자의 중량에 대한 소수성 화합물의 중량으로서 0.01 내지 30의 비율로 상기 고분자와 혼합되는 것을 특징으로 하는 마이크로파티클.
- 제16항에 있어서, 상기 소수성 화합물은 0.01 내지 30의 비율(지질 중량/고분자 중량)로 상기 고분자와 혼합되는 지질인 것을 특징으로 하는 마이크로파티클.
- 제17항에 있어서, 상기 지질은 지방산 및 그 유도체, 모노-, 디 및 트리글리세라이드, 인지질, 스핑고리피드, 콜레스테롤 및 스테로이드 유도체, 오일, 비타민 및 테르펜으로 구성된 군으로부터 선택되는 것임을 특징으로 하는 마이크로파티클.
- 제18항에 있어서, 상기 지질은 포스파티드산류, 포화 및 불포화 지질을 모두 갖는 포스파티딜콜린, 포스파티딜에탄올아민, 포스파티딜글리세롤, 포스파티딜세린, 포스파티딜이노시톨, 리조포스파티딜 유도체, 카디오리핀(cardiolipin) 및 β-아실-y-알킬인지질로 구성된 군으로부터 선택된 인지질인 것을 특징으로 하는 마이크로파티클.

- 제25항에 있어서, 상기 고분자는 폴리(히드록시산), 폴리언히드라이드, 폴리오르토에스테르, 폴리아미드, 폴리카보네이트, 폴리알킬렌, 폴리알킬렌 글리콜, 폴리알킬렌 옥사이드, 폴리알킬렌 테레프탈레이트, 폴리비닐 알코올, 폴리비닐 에테르, 폴리비닐 에스테르, 폴리비닐 할로겐화물, 폴리비닐피롤리돈, 폴리실록산, 폴리(비닐 알코올), 폴리(비닐 아세테이트), 폴리스티렌, 폴리우레탄 및 그들의 공중합체, 합성 셀룰로스, 폴리아크릴산, 폴리(부틸산), 폴리(발레르산) 및 폴리(락티드-코-카프로락톤), 에틸렌비닐아세테이트, 그들의 공중합체 및 혼합물로 구성된 군으로부터 선택되는 것임을 특징으로 하는 마이크로파티클.
- 제17항에 있어서, 상기 지질은 상기 고분자와 함께 액화되어 상기 마이크로파티클을 형성하는 것임을 특징으로 하는 마이크로파티클.
- 제27항에 있어서, 상기 지질과 상기 고분자는 둘다 용매에 용해된 다음, 마이크로파티클을 형성하는 것임을 특징으로 하는 마이크로파티클.
- 제27항에 있어서, 상기 가스는 고분자와 지질이 고화된 다음에 상기 마이크로파티클에 혼입되는 것임을 특징으로 하는 마이크로파티클.
- 제15항에 있어서, 상기 고분자는 단백질과 다당류로 구성된 군으로부터 선택되는 천연 고분자인 것을 특징으로 하는 마이크로파티클.
