HaCaT(immortalized keratinocyte) 세포를 96 well plate에 분주하고 24시간 배양한 후, Serum-free 배지로 교체하고, 시료를 농도 별로 처리하여 24시간 동안 37℃, CO2 5% incubator에 배양하였다. 배양 후 HBSS로 세척한 뒤 H2DcFDA solution을 처리 후 차광하여 incubator에서 30분 반응시켰다. 이후 HBSS로 세척한 뒤 100 mM H2O2 solution을 처리하여 incubator에서 10분 반응시킨 다음 fluorescence value을 측정하여 ROS 생성량을 확인하고, 그 결과를 하기 표 6에 나타내었다.Reactive Oxygen Species (ROS) Assay was performed to confirm antioxidant efficacy.
