Description
The present invention relates to a functional composition for whitening, wrinkle improvement and anti-pollution. It is not yet known exactly how much cellular aging is applied to the aging of individuals (humans and animals). It is a well-known theory that only the telomeres at the ends of chromosomes shorten each time a cell divides, and when the limit is reached, the cells die. Recently, there is a study that collagen, the main component of the cell interstitium, decreases to 65% of those in their 20s at the age of 80, thereby contributing to aging. There is also a study that abnormal protein accumulation due to intracellular ER stress is involved in aging.
(EL-Domyati M et al: Exp Dermatol 11, 2002)In skin aging, collagen and proteoglycan, three major components of connective tissue, harden by intermolecular cross-linking or side chain modification, resulting in changes in cell-extracellular matrix interaction. In addition, the reduction of pilose cells and the turnover of epidermal cells (turn over, usually 28 days) decrease to about 40% of those in their 70s to 30s, causing skin blemishes. And the decrease in collagen and ceramide is the main cause of skin wrinkles due to aging. It has been reported that administration of estrogen not only increases collagen in the skin but also improves hyaluronic acid synthesis to improve moisturizing properties (EL-Domyati M et al: Exp Dermatol 11, 2002).
Oxidative stress is known to be an important cause of aging. Cell stress-inducing factors include reactive oxygen species called reactive oxygen species (ROS), such as peroxides, hydroxy radicals, and hydrogen peroxide. In addition, reactive nitrogen species (RNS) such as nitric oxide and peroxynitrite are known, and lipid peroxides are also known as oxidative stress inducers. These oxides are known to cause skin aging or cancer by binding to the DNA base guanine and causing mutations. In addition, there is a report that it is involved in the arachidonic acid cascade and causes vasoconstriction and platelet aggregation, which develops as a risk factor for arteriosclerosis or Alzheimer's disease. As active substances that can inhibit oxidative damage, vitamins C and B12 are known for their water solubility, and natural ingredients such as lycopene and beta-carotene are well known for their fat solubility.
Accordingly, studies using natural products that do not have side effects due to long-term use are being actively conducted. However, research reports on aging regulators using natural products are rarely made, and it is necessary to discover various materials for this.
On the other hand, as a thin film surrounding the outside of the body, the skin functions to protect the human body from physical and chemical stimuli from the external environment, and prevents excessive penetration of contaminants from the outside while suppressing the evaporation of moisture in the body. Recently, as the use of chemicals increases, the skin is exposed to environmental pollutants such as polycyclic aromatic hydrocarbons, aldehydes, nitrogen oxides, nitrates, and fine dust generated in the process of manufacturing, use, and disposal thereof, resulting in erythema, edema, The incidence of mild skin problems such as itchiness, hives, and inflammation and skin diseases such as atopic dermatitis, psoriasis, and skin cancer is increasing.
Therefore, in order to protect skin cells from external stress and prevent aging, it is important to suppress the oxidative stress caused by free radicals and to strengthen the antioxidant defense that can effectively remove free radicals, and recently it is harmless to the human body and has more powerful Research on natural substances with antioxidant power is being actively conducted.
또한, 미용, 식품의 소재로 사용되고 있는 성분들은 히아루론산, 포도씨 추출물, 이소플라본, 콜라겐, 세라마이드, 코엔자임 Q10, 콘드로이틴, 리코펜, 알파 리포산 등이며, 이들의 피부미용효과는 주로 항산화 기능으로부터 기인하고 있다(박장서, 식품과학과 산업, 40, 19 (2007)).Currently, it is reported that superoxide dismutase (SOD) and catalase, which are antioxidant enzymes that remove reactive oxygen species, and vitamins C and E, which are antioxidants, inhibit skin aging and skin cancer. In addition, the ingredients used as materials for beauty and food are hyaluronic acid, grape seed extract, isoflavones, collagen, ceramide, coenzyme Q10, chondroitin, lycopene, alpha lipoic acid, etc., and their skin-beautifying effects are mainly due to their antioxidant function ( Park Jang-seo, Food Science and Industry, 40, 19 (2007)).
On the other hand, along with exogenous skin aging caused by ultraviolet rays, etc., there are problems such as skin aging caused by intrinsic factors such as genetic factors or cell target imbalance. As the concentration increases, it is emerging as a social problem.
That is, as well as problems such as natural cell destruction and skin aging, skin aging and cell destruction are accelerated by the inflow of the above-mentioned pollutants into the body, and even it is pointed out as a problem that causes various diseases.
Specifically, the above-described pollutants not only penetrate into the body but also adversely affect the skin when it comes into direct contact with the skin, and adversely affects the skin, such as dermatitis, skin rash, and skin necrosis. Accordingly, there is an increasing demand for cosmetics capable of protecting the skin from pollutants as well as ultraviolet rays.
As one of the above-mentioned pollutants, benzopyrene may be exemplified. The benzopyrene is produced due to incomplete combustion such as automobile exhaust gas or cigarette smoke, and is a toxic substance that causes cell damage through metabolic conversion to epoxide and reaction with DNA. Recently, as news of inducing atopic dermatitis has been reported, adverse effects on the skin have been revealed, and it is one of the dangerous substances that can cause skin deterioration due to problems such as cell mutation and cell damage.
In addition, there is nitrogen oxide as belonging to the above-mentioned pollutants. The nitrogen oxide is a compound consisting of nitrogen and oxygen, and there are ionized products thereof such as nitrogen monoxide, nitrogen dioxide, dinitrogen monoxide, dinitrogen trioxide, dinitrogen tetraoxide, dinitrogen pentoxide, and the like. In general, nitrogen gas is stable and the atoms constituting its composition are difficult to split into two, but when a very high temperature is applied, the bond may be broken. Often, a high temperature is formed inside the engine of a vehicle, so that the exhaust gas of the vehicle contains a large amount of nitrogen oxides.
The above-mentioned nitrogen oxides may include ammonium or a salt thereof. Nitric acid, another form of nitrogen oxide, reacts with water to form nitric acid, which can also cause acid rain. The nitric acid is combined with ammonium, which is an alkaline substance in the atmosphere, to produce ammonium nitrate. The ammonium nitrate is a particulate matter in the atmosphere, which is secondary fine dust and adversely affects the human body. When the ammonium nitrate comes into contact with the human body, it causes inflammation in the eyes, nasal mucosa, and skin.
Also, there is formaldehyde as belonging to the above-mentioned pollutants. The formaldehyde is a colorless gas with a strong irritating odor at room temperature, also called methanal. The formaldehyde is easily made when a material containing carbon is incompletely combusted, and is synthesized in the air when sunlight and oxygen are added to hydrocarbons such as methane. The formaldehyde is a skin allergen and may cause problems such as skin rash, skin irritation, and the like when in contact with the skin.
The above-described contaminants cause direct inflammation or necrosis when in contact with the skin at a high concentration, and even if the residual amount in the air is continuously contacted, it causes continuous stress on the skin and thus acts as a cause of skin aging.
Compared to the effects of the above pollutants on the skin, the public's awareness of the risks is still low, but as issues such as fine dust have recently emerged, the danger is gradually getting attention, and the number of people who are gradually aware of the dangers of the pollutants is increasing. is increasing
In addition, as a compound word meaning a product that protects the skin from external harmful environments such as fine dust according to various pollution sources and fine dust warnings, the term anti-pollution and the demand for these functional products are increasing. In particular, in the cosmetic field, anti-pollution cosmetics are attracting attention. According to a Chinese market research firm QY Research Report, the size of the anti-pollution skin care market for harmful environments including fine dust is expected to spread to $10 billion by 2025. Interest is growing day by day.
On the other hand, compositions that can protect the skin from these contaminants are being studied, but the diversity is still lacking, the effect is not clear, and there are many cases where they are made of materials that are difficult to manufacture or are difficult to obtain. There is a need for the development of novel compositions.
Also, from an aesthetic point of view, the demand for skin whitening effect is increasing day by day. Several factors are involved in determining a person's skin color and the generation of blemishes and blemishes. Several factors are complexly involved, such as the activity of melanocytes that make melanin, the distribution of blood vessels, the thickness of the skin, carotenoids, and bilirubin. has been Among them, the most important factor is melanin pigment, which is produced by the action of various enzymes in melanocytes in the human body. It is known that genetic factors, hormone secretion, stress, physiological factors such as UV rays, and environmental factors influence the formation of this melanin pigment.
Melanin, which determines a person's skin color, is produced in melanocytes. In melanocytes, enzymes such as tyrosinase exist, and they work together to convert an amino acid called tyrosine, which is always present in the body, as a substrate. Through polymerization and oxidation reaction, melanin, a black-brown pigment, is formed. The melanin thus formed moves through the dendrites of the melanocytes to the epidermal cells called keratinocytes. Here, melanin forms a hat-like structure around the nucleus to protect genes from UV rays and to remove free radicals to protect intracellular proteins. There is no enzyme that decomposes melanin in the living body, but when keratinocytes age and function and fall off from the epidermis, they are removed from the skin together with keratinocytes. However, if melanin is produced excessively than necessary, it causes hyperpigmentation such as spots, freckles and spots, resulting in poor cosmetic results. In addition, as the number of people who enjoy outdoor activities increases due to the increase in the leisure population, the phenomenon of melanin pigmentation due to ultraviolet rays is further intensifying. do. In addition, since ancient times, Asian women have preferred white and fair skin, and considering this as an important standard of beauty, the need for prevention and improvement of skin pigmentation abnormalities and hyperpigmentation is increasing. It became necessary to develop a whitening agent that prevents it, and many efforts have been made in the meantime. Specific examples thereof include inhibitors of tyrosinase activity such as kojic acid and arbutin, hydroquinone, vitamin A, and the like, as well as representative antioxidant vitamin C and derivatives thereof. However, their use is limited due to insufficient skin safety, stability in the formulation, and whitening effect.
In addition, the fact that the effect is temporary and does not comprehensively care for skin health did not satisfy the needs of recent customers who want a fundamental whitening effect.

Preparation Example: Preparation of Extract]
1. Preparation of extracts from sorghum odor
The wild odor extract (SW) was prepared according to the hot water extraction method by washing and shredding to a certain size. In addition, in order to check the difference in the solvent, ethanol was used as a solvent to prepare an extract (SE).
2. Preparation of other plant extracts
In order to confirm the synergistic effect on the extract (SW), sea bamboo shoots extract (NW), thorn extract (TW), and algae extract (EW) were prepared according to the hot water extraction method as a material of the complex extract.
3. Preparation of Mixed Extracts

(Unit: parts by weight)
Experimental Example: Activity evaluation for Preparation Example]
1. Cytotoxicity Assessment
In order to confirm the cytotoxicity of SW, SE and M2, M6 and M10, each Example was treated with Raw 264.7 cells and human keratinocyte cell line (HaCaT human keratinocyte cell line) for each concentration, and MTS assay was performed. The results are shown in FIG. 1 .
Referring to FIG. 1 below, it can be confirmed that there is no problem of cytotoxicity in the group treated with SW, SE and M2, M6 and M10 by concentration. Therefore, it can be seen that there is no problem of cytotoxicity according to the composition of the single extract and the mixed extract.
2. Evaluation of whitening activity
In order to evaluate the inhibitory activity of intracellular melanin production according to the preparation example, the inhibition rate was evaluated by comparing the intracellular melanin production amount with the normal group using the B16F10 cell line. To induce melanin production, the medium was treated with α-MSH or 3-isobutyl-1-methylanthine (IBMX) to accelerate the melanin production process. The medium was removed, the cells were washed with PBS, and the precipitated cells were treated with 1N sodium hydroxide, and the supernatant was recovered after centrifugation to measure the absorbance, and the amount of melanin was evaluated using a calibration curve using synthetic melanin.
[멜라닌 생성 저해율(%)=[(A-B)/A]*100][Melanogenesis inhibition rate (%)=[(A-B)/A]*100]
The melanin production rate of the control using purified water was set to 100 (%), and the melanin production rate according to each preparation example was compared and evaluated, and the melanin inhibition rate (%) of each preparation example was calculated and shown in [Fig. 2] below.
Referring to the following [Table 2], it can be confirmed that the extract shows relatively excellent whitening activity in the case of the extract alone. In particular, it can be seen that both SW, which is a hot water extract, and SE, which is an ethanol extract, show excellent whitening activity.
On the other hand, in the case of the complex extract, it can be seen that a synergistic effect appears when mixed in a certain concentration range. In the case of a mixture of SW and NW, it can be seen that a synergistic effect appears according to the mixing range of M2 and M3. In particular, it can be seen that when out of a certain mixing range, the inhibition rate of melanin production is rather reduced compared to each single extract.
In addition, TW and EW alone have relatively low whitening activity as a single extract, but when mixed with SW at a constant mixing ratio like M5 and M6, a synergistic effect due to the interaction results in very excellent whitening activity than a simple combination. Able to know.
On the other hand, when all of SW, NW, TW and EW are mixed, it can be seen that M10 and M11 have relatively excellent effects, but compared to the mixed composition of M2, M3 or M5, M6, the inhibition of melanin synthesis is reduced. Able to know.
3. Anti-aging and anti-wrinkle activity evaluation
As a cytoprotective effect of CCE on UVB-induced apoptosis, the improvement effect on skin wrinkles was evaluated. To this end, MTT assay analysis was performed on the cell viability in HaCaT cells after UVB exposure. UVB was treated at 290-320 nm, 30 min, 3 times, and for objective comparison, the UVB-treated group without the preparation example was used as a control, and the cell viability (100%) of the control was evaluated. The results are shown in [Fig. 3] below.
Referring to [Table 3] below, it can be seen that the anti-aging effect is excellent due to the cytoprotective activity in the case of SW, SE and NW among the single extracts. In addition, in the case of the mixed composition, it can be seen that the cell protective activity is rather reduced according to the mixing except for the mixed composition of M2, M3 or M5, M6. In particular, it can be seen that when all of SW, NW, TW and EW are mixed, the cytoprotective activity is lower than that of the single extract.
Therefore, it can be confirmed that the skin wrinkle improvement effect due to photoaging appears in the case of the present invention.
4. Anti-Pollution Activity Assessment
(1) Evaluation of resistance to hydrogen peroxide
HaCaT cells were treated with no-treated control and each preparation to distinguish each group, and then treated with hydrogen peroxide for each group, and MTT analysis was performed to confirm the effect of oxidative stress on the induced HaCaT cells. The evaluation of resistance activity to hydrogen peroxide was evaluated in comparison with the control for each treatment group of each preparation, and for objective evaluation, the result of the control was fixed to 1, and the result according to each preparation example was evaluated as an index of 1 to 10. In the index, the higher the number, the better the resistance.
The results are shown in [Fig. 4] below.
(2) Evaluation of resistance to formaldehyde
HaCaT cells were treated with formaldehyde. At the same time, the untreated group was used as a control, and the groups treated with each preparation were separately cultured. After that, the MTT solution was added and cultured, and the cell viability of each example was evaluated by measuring the absorbance after treatment with DMSO. For objective comparison, the results of the control were fixed as
The results are shown in [Fig. 4] below.
(3) Evaluation of skin resistance to aromatic compounds
After washing the silicone specimen, each example mixed with purified water (control) and purified water was applied, and naphtalene, acenaphthylene, acenaphthene, fluorene, benzopyrene was applied to the silicone specimen. (Benzopyrene) was immersed in a solution containing 500 ppm and dried at low temperature. Thereafter, the contents of naphthalene, acenatthylene, acenaphthene, fluorene and benzopyrene remaining in each specimen were analyzed and compared with the control. For objective evaluation, the result of the control was fixed at 1, and the results according to each preparation example were evaluated with an index of 1 to 10. In the index, the higher the number, the better the resistance.
The results are shown in [Fig. 4] below.
(4) Analysis of experimental results
Referring to [Fig. 4] below, in the case of a single extract, NW had the best anti-pollution effect. It can be seen that SW and SE have almost the same effect as that of NW. On the other hand, in the case of the mixed extract, it could be confirmed that a synergistic effect appeared according to the mixed composition of M2, M3 or M5, M6. It can be seen that there is no synergistic effect.
On the other hand, it was confirmed that when all of SW, NW, TW and EW were mixed, a synergistic effect on anti-pollution activity was not confirmed.
Through this, it can be understood that, in the case of the extract, when a specific composition is mixed in a specific range, a synergistic effect according to the interaction is exhibited, thereby increasing the resistance activity against each harmful substance.
5. Evaluation of antioxidant and anti-inflammatory activity
(1) Evaluation of intracellular reactive oxygen species (ROS)
In HaCaT cells, the intracellular reactive oxygen species (ROS) level generated by UVB (UVB (100 mJ/cm 2 ) for 130 min) and the protective action of CCE according to the present invention were measured using DCF-DA (dichlorodihydrofluorescein diacetate) method. and evaluated. The image analysis of intracellular ROS was performed, and the results of each Example were compared and evaluated with the control irradiated with UVB without any treatment. The result of Control was fixed to 1, and the results for each Example were compared and evaluated with an index of 1 to 10.
The results are shown in [Fig. 5] below. In the index, the higher the number, the better the removal activity for active oxygen species.
(2) Anti-inflammatory activity evaluation
Inhibitory activity on inflammation induced by fine dust by processing fine particle PM10 in the HaCaT cells divided into the group treated with the above preparation example and the group not treated with anything (control), and analyzing the production of the inflammatory mediator prostaglandin E2 was tested. The evaluation results were evaluated in comparison with the control, and the index of the control was fixed to 1, and the results for each Example were compared and evaluated with an index of 1 to 10.
The results are shown in [Fig. 5] below. In the index, the higher the number, the better the resistance activity against inflammation induced by the microparticles.
(3) Analysis of experimental results
Referring to [Fig. 5] below, it can be confirmed that SW or SE exhibits excellent antioxidant and anti-inflammatory activity. In addition, in the case of the mixed extract, it can be confirmed that a synergistic effect appears when the mixed composition of M2, M3 or M5, M6 is used. On the other hand, even in the case of the mixed extract, it can be seen that the antioxidant and anti-inflammatory activity is lowered when it is outside the mixed composition range of M2, M3 or M5, M6. In addition, it can be seen that when SW, NW, TW and EW are all mixed, antioxidant and anti-inflammatory activity is lower than that of SW or SE alone.
Therefore, it can be confirmed that when SW or SE is used alone or when a mixed composition according to the M2, M3 or M5, M6 range is used, an excellent functional food or external preparation can be provided.
