Abstract
본 발명은 내열성 박테리오파지 TS3, TS6, TS13 및 이를 포함하는 살모넬라 억제용 조성물에 관한 것이다. 본 발명의 박테리오파지 TS3, TS6, TS13 및 이를 포함하는 조성물은 내열성 및 pH 안정성이 우수하면서 여러 살모넬라균에 대하여 뛰어난 항균활성을 발휘할 수 있다. 또한, 본 발명의 박테리오파지 TS3, TS6 및 TS13의 조합물은 단일 박테리오파지를 처리할 때와 비교하여 항균력이 훨씬 향상되며, 특히 액체 및 고체 제형의 식품 모델에 적용할 시에도 항균활성이 탁월하여 식품, 사료, 의약 제품에 다양하게 사용가능할 수 있다.
Description
본 발명은 내열성 박테리오파지 TS3, TS6, TS13 및 이를 포함하는 살모넬라 억제용 조성물에 관한 것이다.
살모넬라(Samonella)는 그람 음성의 간균으로, 포자를 형성하지는 않으나 편모를 통해 운동성을 가지며, 감염 시 질환에 따라 급성패혈증을 유발하는 Typhoidal serotype과 만성 복통을 유발하는 Non-Typhoidal serotype으로 나뉜다. 특히, Non-Typhoidal serotype은 감염 시 심한 복통 및 설사를 유발하는 원인균으로, 국내에서 발생하는 식중독 사고의 주요 원인 중 하나이다. 또한, 사료에 오염되어 가금류, 돼지 등에서도 질병을 일으킬 수 있으며, 식육을 통해 사람에게 전염될 수 있기 때문에 중요한 오염인자로서 관리가 필요하다.
항생제는 페니실린이 도입된 1940년 이후로, 의료, 식품, 축산 등 다양한 산업군에서 이용되어 수많은 감염 질환에 대한 치료 및 예방을 가능케 했다. 그러나 불과 60여년 만에 항생제 내성균이 광범위하게 출현하여 각종 감염질환의 치료 실패와 치명적인 결과를 일으키고 있으며, 다제내성균까지 출현하여 미래의 심각한 위협이 되고 있다. 이를 해결하기 위해 WHO(World Health Organization)에서는 항생제를 대체할 수 있는 항균 물질의 개발을 촉구하고 있으며, 이를 통해 내성균 문제를 적극적으로 해결하고자 한다.
한편, 박테리오파지(Bacteriophage)는 박테리아를 숙주세포로 감염하고 사멸하는 바이러스를 말하며, 간단하게 파지(phage)라고도 한다. 일반적으로 박테리아보다 훨씬 작은 입자로서, 원형 혹은 선형의 유전물질과 이를 둘러싼 단백질 캡시드로 이루어져 있으며, 생물권에서 가장 넓게 퍼져 있는 존재로 추측되고 있다. 특히, 특정 균주만을 감염할 수 있어 병원균 외에 미생물 생태계에는 영향을 끼치지 않으며, 사람과 동물을 포함하는 진핵생물에는 작용하지 않아 인체에 무해하다고 볼 수 있다. 또한, 항생제 내성 유전자의 유무와 관계없이 작용할 수 있기 때문에 항생제 대체제로서 가능성이 높은 것으로 판단된다.
박테리오파지의 생활사는 크게 세균의 DNA에 삽입되는 용원성 생활사와 세균을 용해하는 용균성 생활사로 구분되며, 용원성 생활사의 유무에 따라 용원 파지(Temperate phage)와 용균 파지(Virulent phage)로 구분된다. 박테리오파지는 식품, 사료 등에서 첨가제로 사용되고 있으며, 임상 실험을 통해 의약 산업에까지 진출을 노리고 있다. 그러나 파지는 온도, pH, 염 농도 등 외부조건에 대한 저항성이 매우 다양하며, 개별 조건에 따라 그 용균 활성이 달라지므로 이를 보완하기 위한 기술이 필요하다. 특히 살모넬라를 제어하는 기술과 관련하여 식품 및 사료의 가공 공정 시 처리되는 열처리 공정을 고려한 내열성 파지의 적용 방식에 대해서는 개시된 바가 없어 이에 대한 연구가 필요한 실정이다.
본 발명은 다양한 살모넬라균에 대한 억제능을 가지면서 뛰어난 내열성, pH 안정성 및 항균효능을 가지는 박테리오파지 TS3, TS6, TS13 및 이를 포함하는 살모넬라 억제용 조성물을 제공하고자 한다.
본 발명은 살모넬라 균에 특이적인 사멸능을 갖는 서열번호 1로 표시되는 핵산서열로 이루어지는 것을 특징으로 하는 박테리오파지 TS3를 제공한다.
또한, 본 발명은 살모넬라 균에 특이적인 사멸능을 갖는 서열번호 2로 표시되는 핵산서열로 이루어지는 것을 특징으로 하는 박테리오파지 TS6를 제공한다.
또한, 본 발명은 살모넬라 균에 특이적인 사멸능을 갖는 서열번호 3으로 표시되는 핵산서열로 이루어지는 것을 특징으로 하는 박테리오파지 TS13을 제공한다.
또한, 본 발명은 상기 박테리오파지 TS3, TS6 및 TS13을 한 개 이상 포함하는 살모넬라 사멸용 조성물을 제공한다.
또한, 본 발명은 상기 박테리오파지 TS3, TS6 및 TS13을 한 개 이상 포함하는 식품 첨가제를 제공한다.

본 발명은 살모넬라 균에 특이적인 사멸능을 갖는 서열번호 1로 표시되는 핵산서열로 이루어지는 것을 특징으로 하는 박테리오파지 TS3를 제공한다. 또한, 본 발명은 살모넬라 균에 특이적인 사멸능을 갖는 서열번호 2로 표시되는 핵산서열로 이루어지는 것을 특징으로 하는 박테리오파지 TS6를 제공한다. 또한, 본 발명은 살모넬라 균에 특이적인 사멸능을 갖는 서열번호 3으로 표시되는 핵산서열로 이루어지는 것을 특징으로 하는 박테리오파지 TS13을 제공한다.
본 발명의 박테리오파지 TS3과 TS13은 경기도 성남시 수질복원센터에서 수득한 하수 시료와 모란시장에서 구매한 오리 내장 시료로부터 분리하였으며, 박테리오파지 TS6은 서울특별시 관악구 봉천중앙시장에서 구매한 상추 시료로부터 분리하였다.
살모넬라(Samonella)는 그람 음성의 간균으로, 대부분 장내에서 증식하나 독소를 생성하지 않으며, 주로 오염된 음식 및 물의 섭취, 분변에서 구강으로의 경로로 감염된다. 제대로 익히지 않은 날고기 및 가금류, 달걀류, 채소류 등의 음식 식재료에 의해 주로 발생하게 된다. 살모넬라균이 감염되어 발생할 수 있는 대표적인 질환으로는 위장염, 패혈증, 장열, 그리고 불현성 감염증이 있다.
본 발명의 박테리오파지 TS3, TS6 및 TS13은 살모넬라균에 대한 숙주 감수성을 가져 이들을 사멸시킬 수 있으며, 상기 살모넬라균은 일 예로 살모넬라 타이피뮤리엄(Salmonella Typhimurium), 살모넬라 파라티푸스균(Salmonella Paratyphi), 살모넬라 엔테리티디스(Salmonella Enteritidis), 살모넬라 듀블린(Salmonella Dublin), 살모넬라 아리조나이(Salmonella Arizonae)일 수 있다.
또한, 본 발명은 상기 박테리오파지 TS3, TS6 및 TS13을 한 개 이상 포함하는 살모넬라 사멸용 조성물을 제공한다. 본 발명의 살모넬라 사멸용 조성물은 통상적인 살모넬라 제제 제조방법으로 제제화할 수 있으며, 제제화의 방법 및 수단에 대해서는 특별히 한정되지 않으나, 인체에 투여하는 경우는 약제학적으로 허용하는 범위 내에서, 가축 또는 어류에 투여하는 경우는 수의학에서 허용하는 범위 내에서, 제제화하고 그에 따라 투여경로를 정할 수 있다.
본 발명의 살모넬라 사멸용 조성물은 유효성분인 본 발명의 박테리오 파지 TS3, TS6 및 TS13 외에 필요에 따라 담체, 기타 보조제 등을 포함할 수 있는데, 상기 담체는 생리식염수, 증류수, 사료, 박테리아용 배지 등이 될 수 있으며, 보조제는 pH 교정 등의 목적으로 완충액 등을 첨가하여 사용할 수 있다.
본 발명의 살모넬라 사멸용 조성물은 액상, 반고체상, 분말상 등의 형태로 제조화될 수 있는데, 반드시 상기 3가지 제제에 한정되는 것은 아니고 다양한 형태의 제형으로 제제화될 수 있다.
본 발명의 살모넬라 사멸용 조성물은 경구, 정맥 내, 동맥 내, 복강 내, 근육 내, 경피, 비측 내, 흡입 또는 직장 등의 경로를 통해 통상적인 방식으로 투여할 수 있다. 본 발명의 살모넬라 사멸용 조성물의 투여량은 투여대상에서 살모넬라균을 사멸시키는 효과를 이루는데 요구되는 양을 의미한다. 따라서, 질환의 종류, 질환의 중증도, 제형의 조율 및 투여대상의 연령, 체중, 일반 건강 상태, 성별 및 식이, 투여 경로, 투여 시간 및 기간, 동시 사용되는 약물을 비롯한 다양한 인자에 따라 그 양이 조절될 수 있다.
본 발명의 박테리오파지 TS3, TS6 및 TS13 또는 이를 포함한 살모넬라 사멸용 조성물은 기존 항생제에 대한 내성 여부와 상관없이 살모넬라균을 사멸시킬 수 있으므로, 살모넬라균의 감염에 의한 질환 치료용으로 사용할 수 있고, 항생제 내성을 갖는 살모넬라균에 사용 시, 기존 항생제로 제거할 수 없었던 살모넬라 내성균을 사멸시킬 수 있으므로, 기존 항생제 대체물로 유용하게 활용될 수 있다. 특히, 식품업, 축산업 및 어업에서, 식품, 축산물 및 수산물의 항생제 잔류 문제 등을 해결할 수 있는 항생제 사용의 대안으로써 제공될 수 있다.
또한, 본 발명은 상기 박테리오파지 TS3, TS6 및 TS13을 한 개 이상 포함하는 식품 첨가제 또는 식품을 제공한다. 상기 식품 첨가제 또는 식품은 박테리오파지 H3, H6 및 H13을 한 개 이상 포함하기만 하면 그 제형에 특별히 국한되지 않으나, 대상 식품은 살모넬라균에 오염되기 쉬운 식품이면 더욱 바람직하다.
또한, 본 발명은 상기 박테리오파지 TS3, TS6 및 TS13을 한 개 이상 포함하는 사료 첨가제 또는 사료를 제공한다. 상기 사료 첨가제 또는 사료는 박테리오파지 H3, H6 및 H13을 한 개 이상 포함하기만 하면 그 제형에 특별히 국한되지 않으나, 살모넬라균에 감염되기 쉬운 가축을 대상으로 하는 사료 첨가제 또는 사료이면 더욱 바람직하다. 사료 첨가제의 일 예로 품질 저하를 방지하기 위하여 첨가하는 결착제, 유화제, 보존제 등이나, 효용 증대를 위하여 사료에 첨가하는 아미노산제, 비타민제, 효소제, 생균제, 향미제, 완충제, 착색제, 추출제, 올리고당 등이 포함될 수 있으며, 그 외에도 사료 혼합제 등을 추가로 포함할 수 있다.
한편, 하기 실험에 의하면, 본 발명의 세 개의 박테리오파지 TS3, TS6 및 TS13 모두 70℃까지 100 %에 가까운 PFU가 유지되는 것을 확인하여 내열성이 우수함을 확인하였고, pH 4 ~ 12까지는 활성이 유지되어 안정적이어서 중성 및 염기성을 띄는 조건에서 식품에는 적용 가능할 것으로 판단되었다. 또한, 박테리오파지 TS3, TS6 및 TS13을 조합하여 처리함으로써 숙주 감염범위를 확장함과 동시에 항균력도 증가시킬 수 있으며, 우유 모델 및 닭가슴살 모델에 적용하였을 때도 살모넬라균주를 완전히 저해하여 항균활성이 우수한 것을 확인할 수 있었다. 이를 통해 본 발명의 박테리오파지 TS3, TS6 및 TS13과 이를 포함하는 조성물은 뛰어난 내열성, pH 안정성 및 항균활성을 가져 가열 살균을 동반한 액체 및 고체 제형의 식품, 사료, 의약 제품에 사용가능성이 높을 것으로 판단된다.
이하, 본 발명의 구성을 하기 실시예 및 실험예를 통해 구체적으로 설명하고자 한다. 다만, 본 발명의 권리범위가 하기 실시예 및 실험예에만 한정되는 것은 아니고, 그와 등가의 기술적 사상의 변형까지를 포함한다.
[실시예 1: 본 발명의 박테리오파지 TS3, TS6 및 TS13의 분리 및 동정]
본 실시예에서는 신규한 박테리오파지 TS3, TS6 및 TS13을 분리 및 동정하고자 하였다.
경기도 성남시 수질복원센터에서 수득한 하수 시료와 모란시장에서 구매한 오리 내장 시료로부터 박테리오파지 TS3와 TS13을 분리하였으며, 서울특별시 관악구 봉천중앙시장에서 구매한 상추 시료로부터 박테리오파지 TS6을 분리하였다.
구체적으로, 병원성 살모넬라 타이피뮤리엄(Samonella Typhimurium LT2(c)) 진탕 배양액 50 ㎕, LB 배지 (트립톤(Tryptone) 10 g/L; 효모 추출물 5 g/L; NaCl 10 g/L) 5 mL에 상기 시료 여과액 45 mL을 혼합하고 37℃에서 6시간 동안 배양한 다음, 상기 배양액을 10,000 gDPTJ 15분간 원심분리하여 그 상등액을 0.22 ㎛ 필터로 여과하였다.
시료 배양액을 70℃의 항온수조에 30분간 침지시킨 후, LB 평판 배지의 위에 0.4 %(w/v) 한천 배지 5 mL, 살모넬라 타이피뮤리엄(Samonella Typhimurium LT2(c)) 진탕 배양액 50 ㎕, 시료 배양액 100 ㎕를 혼합한 후, 1.5 % 한천 고체 배지에 굳혀 37℃에서 하룻밤 동안 정치 배양하였다. 그 후, 플레이트 상에 투명한 단일 플라그(plaque)가 관찰되었으며(도 1), 이를 파이펫 팁을 이용하여 분리해낸 다음, 살모넬라 배양액에 넣어 37℃에서 배양하였을 때, 광학 밀도(optical density) 측정을 통해 배양액이 투명해지는 것을 확인할 수 있었다. 그 후, 10,000 g, 15분 조건에서 원심분리하고 그 상등액을 0.22 ㎛ 필터로 여과하여 박테리오파지만 남아있는 용액을 취하였다. 상기와 같은 방법을 통해 TS3, TS6 및 TS13의 파지를 분리하였다.
[실시예 2: 본 발명의 박테리오파지 TS3, TS6 및 TS13의 형태학적 분석 및 유전체 분석]
본 실시예에서는 상기에서 분리한 박테리오파지 TS3, TS6 및 TS13의 형태학적 분석 및 유전체 분석을 수행하였다.
1) 본 발명의 박테리오파지 TS3, TS6 및 TS13의 형태학적 분석
박테리오파지 TS3, TS6 및 TS13에 대해 에너지 여과 투과 전자현미경(Energy-Filtering Transmission Electron Microscope)을 이용하여 그 형태를 관찰하였다. 먼저 박테리오파지 희석액 1010 PFU/mL을 탄소 필름의 구리 그리드(Carbon films on Copper grid)에 올리고, 2 % 우라닐 아세테이트(uranyl acetate)로 음성 염색(negative-staining)한 후 30분 동안 건조하였다. 120 kV 전압의 에너지 투과 전자현미경(LIBRA 120; Carl Zeiss)을 이용하여 염색된 시료를 분석하였다. 그 결과, 도 2와 같이 TS6 및 TS13은 공통적으로 수축성이 없는 긴 꼬리 (long non-contractile tail)를 갖고 있는 것을 확인할 수 있었으나, tail-spkie 형태의 꼬리(tail)를 갖는 TS3 및 TS6과는 달리 TS13은 얇고 긴 형태의 tail fiber를 갖는 것으로 관찰되었다. 따라서 박테리오파지의 분류학적 분류(taxonomic classification)에 의거하여 복합형의 시포비리대(Siphoviridae)에 속하는 것으로 분류하였다.
2) 본 발명의 박테리오파지 TS3, TS6 및 TS13의 유전체 분석
본 발명의 박테리오파지 TS3, TS6 및 TS13에 대해 EDTA, 프로테나아제(Proteinase) K, 10 % SDS를 사용하여 파지 DNA를 노출시켰으며, 에탄올 침전법에 따라 DNA를 추출하였다. 이 후, 세니젠에서 Illumina Miseq (Illumina, 미국)을 이용하여 차세대염기서열분석(Next generation sequencing)을 수행한 다음, 해당 파지들의 서열 정보를 확보하였으며, GeneMark.hmm. software와 The RAST server software를 활용하여 오픈 리딩 프레임(Open reading frame, ORF)을 분석하였다. 박테리오파지 TS3, TS6의 유전체 분석 결과, 각각 41,568 bp, 41,515 bp의 크기를 가지며, 전체 유전체 서열은 서열번호 1, 2로 제시되어 있다.
[실험예 1: 본 발명의 박테리오파지 TS3, TS6 및 TS13의 내열성 분석]
본 실험예에서는 본 발명의 박테리오파지 TS3, TS6 및 TS13의 내열성 분석을 수행하기 위해 히트 블록(heat block)을 사용하여 60℃, 65℃, 70℃, 75℃에 노출시킨 뒤, 각 온도별로 남아있는 파지에 대해 플라크 형성 유닛(plaque forming unit, PFU)을 비교하였다.
보다 구체적으로, 3차 정제된 물 990 ㎕를 준비한 뒤 각 온도가 되도록 히트 블록(heat block)을 이용하여 조정하였다. 107 PFU/mL이 되도록 박테리오파지를 주입한 뒤, 총 60분 동안 방치하였으며, 10분 단위로 생존한 파지 수를 확인하였다. 각 온도에 노출된 박테리오파지에 대해 씨리얼 딜루션 드로핑 메소드(Serial dilution dropping method)를 이용하여 PFU를 측정하였다. 0.4 % 한천 배지 5 ml에 12시간 배양한 살모넬라 타이피뮤리엄(Samonella Typhimurium LT2(c))을 섞어 15 % 한천 고체 배지에 부은 후, 20분 동안 상온에서 건조하였다. 박테리오파지를 희석배수별로 10 ㎕씩 분주하여 말린 후, 37℃에서 12시간 이상 정치배양하였다. 그 결과, 도 3과 같이 약 107 PFU/mL의 박테리오파지를 각 온도에 노출시켰을 때, 세 파지 모두 70℃의 열을 1시간 동안 처리했을 때, PFU가 유지되는 것을 확인할 수 있었으며, 75℃에서 급격하게 감소되는 것이 확인되었다. 이를 통해 박테리오파지 TS3, TS6 및 TS13은 내열성이 우수한 박테리오파지임을 확인할 수 있었다.
[실험예 2: 본 발명의 박테리오파지 TS3, TS6 및 TS13의 pH 안정성 분석]
본 실험예에서는 본 발명의 박테리오파지 TS3, TS6 및 TS13의 pH 안정성을 분석하기 위해 다양한 pH 범위(pH 2, 3, 4, 5, 6, 7, 8, 9, 10, 11, 12)에서의 안정성을 평가하였다. 수산화나트륨과 염산을 이용하여 각 pH 범위의 LB 배지를 각각 제작하였으며, 각 pH 용액 990 ㎕와 109 PFU/mL의 박테리오파지 10 ㎕를 희석한 후, 1시간 동안 상온(25℃)에서 방치하였다. 그 다음, 살모넬라 타이피뮤리엄(Samonella Typhimurium LT2(c))이 접종된 고체 배지에 희석배수별로 분주하고 37℃에서 12시간 배양하여 생성되는 단일 플라그를 계산하였다. 그 결과, 도 4와 같이 107 PFU/mL의 박테리오파지를 처리하였을 때, pH 2,3에서는 활성을 보이지 않았지만 pH 4 ~ 12까지는 활성이 유지되어 안정적인 것을 확인하였다. 이를 통해 중성 및 염기성을 띄는 조건에서 식품에는 적용 가능할 것으로 판단된다.
[실험예 3: 본 발명의 박테리오파지 TS3, TS6 및 TS13의 숙주 감수성 분석]
본 실험예에서는 본 발명의 박테리오파지 TS3, TS6 및 TS13의 숙주 감수성 분석을 수행하기 위해 씨리얼 딜루션 드로핑 메소드(Serial dilution dropping method)를 사용하였다. 0.4%의 한천 배지 5 mL에 12시간 배양한 살모넬라를 각각 섞어 15 % 한천 고체 배지에 부은 후, 20분 동안 상온에서 건조하였다. 박테리오파지를 희석배수별로 10 ㎕씩 분주하여 말린 후, 37℃에서 12시간 이상 정치배양하였다.
파지에 대한 감수성을 보이는 숙주의 경우 플레이트 상에서 투명한 플라그를 형성하는데, 이는 숙주가 완전히 사멸된 결과이다. 한편, 약한 감수성을 보이는 박테리오파지의 경우, 드로핑 스팟(dropping spot)이 뿌옇게 되어 불투명한 자국을 형성하게 된다. 박테리오파지 TS3, TS6 및 TS13의 숙주 감수성을 분석한 결과는 하기 표 1과 같았다.

그 결과, 박테리오파지 TS3, TS6 및 TS13은 사람 및 동물에 대해 병원성을 보이는 다양한 살모넬라 아종에 대해 각기 다른 감수성을 보였다. 세 파지 모두 살모넬라 타이피뮤리엄(Salmonella Typhimurium), 살모넬라 파라티푸스균(Salmonella Paratyphi)에 대해 뚜렷한 항균활성을 보였고, TS3은 살모넬라 엔테리티디스(Salmonella Enteritidis), TS6은 살모넬라 듀블린(Salmonella Dublin), TS13은 살모넬라 아리조나이(Salmonella Arizonae)에 대해 추가적으로 사멸할 수 있는 것을 확인하였다. 특히, TS3, TS6 및 TS13을 모두 혼합한 조성물을 처리한 경우 실험한 모든 살모넬라 균주에 대해 항균활성을 나타내는 것을 확인하였다.
[실험예 4: 본 발명의 박테리오파지 TS3, TS6 및 TS13의 조합물 항균효과 분석]
본 실험예에서는 본 발명의 박테리오파지 TS3, TS6 및 TS13의 조합물 항균효과 분석을 수행하고, 이를 식품모델(액체, 고체)로 적용하여 확인하였다.
1) 본 발명의 박테리오파지 TS3, TS6 및 TS13의 조합물 항균효과 분석
본 발명의 박테리오파지 TS3, TS6 및 TS13을 조합하여 살모넬라 타이피뮤리엄(Samonella Typhimurium LT2(c))처리하였을 때, 그 항균력이 향상되는지 여부를 SpectraMax i3x (Molecular Devices, 미국) Plate reader를 이용하여 측정하였다. 37℃ weak shaking 조건에서 108 CFU/mL (OD600nm= 0.15)에 대해 MOI(Multicity of infection=CFU/PFU) 1의 박테리오파지 조성물을 처리하였을 때, OD600nm 값이 감소되는 경향을 관찰하였다(도 5).
구체적으로 도 5의 (A)와 같이 단일 박테리오파지를 처리할 시, TS3과 TS13은 4시간 째, 저항성 균주가 출현하였으며, TS6은 항균력이 비교적 약할뿐더러 6시간 째, 저항성 균주가 출현하는 것을 관찰할 수 있었다. 반면, 도 5의 (B)와 같이 TS3과 TS6을 함께 처리하였을 때는 효과가 없는 것으로 나타났지만, 두 파지를 TS13과 함께 처리하였을 때는 항균력이 증가함과 동시에 파지 저항성 균주의 출현이 8시간까지 증가하는 것으로 나타났다. 이를 통해 박테리오파지 TS3, TS6 및 TS13을 조합하여 처리함으로써 숙주 감염범위를 확장함과 동시에 항균력도 증가시킬 수 있음을 확인할 수 있었다.
2)
본 발명의 박테리오파지 TS3, TS6 및 TS13의 조합물의 액체 식품 모델 적용
본 발명의 박테리오파지 TS3, TS6 및 TS13의 조합물의 항균효과를 우유 모델에 적용하여 분석하였다. 멸균 우유에 박테리오파지 107 PFU/mL을 처리하고, 항온수조를 이용하여 30분간 63℃의 온도를 처리하였다. 그 후, 104 CFU/mL의 살모넬라 타이피뮤리엄(Samonella Typhimurium LT2(c))을 처리하여 상온 조건(25℃)과 저온 조건(4℃)에서 48시간 배양하여 시간에 따른 균의 감소를 정량적으로 측정하였다. 이를 비교 분석하기 위해, 파지 비처리군(LT2(c))과 비내열성 파지 NH4 처리군을 함께 실험하여 본 발명의 내열성 박테리오파지 TS3, TS6 및 TS13의 조합물 활성을 확인하였다.
그 결과, 도 6과 같이 본 발명의 내열성 박테리오파지 TS3, TS6 및 TS13 조합물 처리군(Cocktail로 기재)은 상온 조건(25℃) 배양조건에서 12시간 째 살모넬라균주가 검출되지 않았으며 24시간에서 6.1 logCFU를 감소시키는 것으로 측정되었고, 저온 조건(4℃) 배양조건에서 살모넬라균주가 완전히 저해되어 검출되지 않았다. 반면 비내열성 파지 NH4 처리군은 살모넬라균주를 전혀 억제하지 못하는 것으로 나타났다. 이를 통해 본 발명의 내열성 박테리오파지 TS3, TS6 및 TS13 조합물은 우유에 적용되어 항균제로 작용할 수 있으며, 더 나아가 제조공정에서 가열 살균을 동반한 액체 제형의 식품, 사료, 의약 제품에도 사용될 수 있을 것으로 판단된다.

본 발명의 박테리오파지 TS3, TS6 및 TS13의 조합물의 고체 식품 모델 적용
본 발명의 박테리오파지 TS3, TS6 및 TS13의 조합물의 항균효과를 닭가슴살 모델에 적용하여 분석하였다. 항온수조를 이용하여 각각의 107 PFU/mL, 108 PFU/mL 박테리오파지 조합물이 담긴 물을 66℃로 조정한 후, 닭가슴살 큐브(cube)(1cm x 1cm x 1cm)을 1분 30초간 침지 상태로 방치하였다. 그 후, 105 CFU/mL의 살모넬라 타이피뮤리엄(Samonella Typhimurium LT2(c))을 오염시킨 후, 상온 조건(25℃)과 저온 조건(4℃)에서 48시간 배양하여 시간에 따른 균의 감소를 정량적으로 측정하였다. 이를 비교 분석하기 위해, 파지 비처리군(PC)과 비내열성 파지 NH7 처리군을 함께 실험하여 본 발명의 내열성 박테리오파지 TS3, TS6 및 TS13의 조합물 활성을 확인하였다.
그 결과, 도 7과 같이 본 발명의 내열성 박테리오파지 TS3, TS6 및 TS13 조합물 처리군(Cocktail로 기재)은 파지 비처리군에 비해 살모넬라 균주를 감소시키는 것을 확인할 수 있었고, 특히 108 PFU/mL을 처리하여 48시간 동안 배양하였을 때, 상온 조건(25℃) 배양조건에서는 5.5 logCFU를 감소시키는 것으로 측정되었고, 저온 조건(4℃) 배양조건에서 3.5 logCFU를 감소시키는 것으로 확인되었다. 반면 비내열성 파지 NH7 처리군은 열에 의해 파괴됨에 따라 살모넬라균주를 전혀 억제하지 못하는 것으로 나타났다. 이를 통해 본 발명의 내열성 박테리오파지 TS3, TS6 및 TS13 조합물은 닭가슴살에 적용되어 항균제로 작용할 수 있으며, 더 나아가 제조공정에서 가열 살균을 동반한 고체 제형의 식품, 사료, 의약 제품에도 사용될 수 있을 것으로 판단된다.
