Abstract
본 발명은 유해물질 및 광과 같은 외부환경으로 인한 스트레스로부터 피부를 보호하기 위한 조성물에 관한 것으로, 유해물질 및 광 등의 외부 스트레스 자극에 의한 세포의 비정상적인 반응을 저해하여 외부 환경으로부터 피부를 보호하고, 세포 독성이 없으므로, 약품, 또는 화장품 등의 다양한 분야에 유용하게 적용할 수 있다.
Description
본 발명은 유해물질 및 광(光)과 같은 외부환경으로 인한 스트레스로부터 피부를 보호하기 위한 조성물에 관한 것이다.
인간의 피부는 크게 표피와 진피, 피하지방으로 나누어지고 다시 표피는 각질층, 투명층, 과립층, 유극층, 기저층으로 세분화되어 있다. 피부의 최외각층인 각질층은 죽은 케라틴세포 덩어리로 이루어져 있으며, 주로 단백질로 이루어져 있는 각질 세포로 구성되고, 각질 세포 간의 사이를 세라마이드, 콜레스테롤, 유리지방산 등의 지질로 이루어져 있다. 이 지질층은 체내 수분이 증발되는 것을 방지하며, 외부 침입에 대해 신체를 보호하는 건강학적 기능과 동시에, 피부를 윤택하고 매끄럽게 보이게 하는 심미적 기능을 가지고 있다.
피부는 외부 환경에 직접적으로 노출되는 부위이며, 외부 환경에 존재하는 유해물질에 의하여 홍반, 부종, 가려움 등의 피부 자극 및 염증 반응이 유발되기 쉽다. 유해물질의 접촉에 의하여 독소로 인한 자극 및 염증 반응 과정이 유도되고, 이로부터 생성되는 물질들이 피부의 색소 침착, 피부 탄력 저하, 피부 장벽 손상 촉진을 일으킬 수 있다.
자외선은 피부의 손상 및 노화를 촉진할 수 있음이 널리 알려져 있다. 특히, 피부가 자외선에 과하게 노출되는 경우 자유 라디칼이 축적되어 활성산소의 생성을 유도하여 산화적 스트레스를 유발하고 항산화 체계를 파괴하여 피부의 손상이 발생한다.
또한, TV, 컴퓨터 등 스마트 기기의 디스플레이와 LED 조명기기에서 많이 방출되는 청색광은 가시광선 중에서도 가장 파장이 짧고 강한 380~500 나노미터(nm) 사이의 파장에 존재하는 파란색 계열의 빛으로, 피부에 청색광을 노출시키면 색소침착이 유발된다는 연구 결과가 있다(Luc Duteil et al. Pigment Cell Melanoma Res 2014;27:822-6).
이러한 외부 환경으로부터의 신체 자극은 피부뿐만 아니라 세포의 DNA 손상으로 인하여 암을 유발하거나 중추 및 말초 신경계의 손상, 면역 시스템 파괴, 생식기관 장애, 염증 질환, 만성 알러지 등을 일으킬 수 있다.
그러므로, 유해물질, 자외선 등과 같은 외부 스트레스로부터 피부를 보호하면서도 안전하고 부작용이 없는 조성물에 대한 연구가 필요하다.
본 발명의 목적은 유해물질 및 광과 같은 외부 환경으로 인한 스트레스로부터 피부를 보호하기 위한 조성물을 제공하는 것이다.
상기 과제를 해결하기 위하여 본 발명은,
펩타이드류; 및 아미노산류를 포함하고,
상기 펩타이드류가 니코티노일트리 펩타이드, 펜타펩타이드, 니코티노일헥사 펩타이드, 에스에이치-옥타펩타이드, 테트라카복시메틸 헥사노일디펩타이드, 헥사카복시메틸 디펩타이드 및 바이오티노일 헥사펩타이드-2 아마이드를 포함하고,
상기 아미노산류가 아르기닌, 아스파라긴, 아스파르트산, 글리신, 히스티딘, 히드록시프롤린, 이소류신, 류신, 메티오닌, 페닐알라닌, 프롤린, 세린 및 발린을 포함하는, 유해물질 및 광에 대한 피부보호용 조성물을 제공한다.
일구현예에 따르면, 본 발명에 따른 조성물은 히알루론산을 더 포함할 수 있고, 상기 히알루론산 분자의 질량은 500KDa 내지 2MDa일 수 있다.
일구현예에 따르면, 상기 유해물질은 황사, 미세먼지, 담배연기, 차량 배기가스, 중금속 및 대기 오염물질을 포함하는 군으로부터 선택되는 하나 이상을 포함할 수 있다.
일구현예에 따르면, 상기 조성물은 유해물질 및 광에 의한 스트레스로 인한 피부세포 사멸을 억제할 수 있다.
또한, 상기 조성물은 항산화 활성을 가질 수 있다.
또한, 상기 조성물은 사이클로부탄 피리미딘 다이머(cyclobutane pyrimidine dimers, CPDs)의 형성을 저해할 수 있다.
또한, 상기 조성물은 유해물질을 흡착 또는 배출시킬 수 있다.
또한, 상기 조성물은 주름 개선용 또는 탄력 증진용으로 사용될 수 있다.
일구현예에 따르면, 상기 펩타이드류는 에스에이치-옥타펩타이드 100중량부에 대하여,
니코티노일트리 펩타이드 1 내지 50중량부;
펜타펩타이드 10 내지 150중량부;
니코티노일헥사 펩타이드 0.05 내지 10중량부;
테트라카복시메틸 헥사노일디펩타이드 5 내지 70중량부;
헥사카복시메틸 디펩타이드 15 내지 80중량부; 및
바이오티노일 헥사펩타이드-2 아마이드 5 내지 70중량부를 포함하고,
상기 아미노산류는 발린 100중량부에 대하여,
아르기닌 80 내지 180중량부;
아스파라긴 10 내지 60중량부;
아스파르트산 20 내지 80중량부;
글리신 30 내지 100중량부;
히스티딘 50 내지 150중량부;
히드록시프롤린 5 내지 40중량부;

일구현예에 따르면, 상기 조성물 총 중량에 대하여 히알루론산을 1 내지 5중량% 더 포함할 수 있다.
본 발명의 다른 구현예에 따르면, 상기한 바와 같은 조성물을 포함하는, 유해물질 및 광에 의해 유발되는 피부 다크닝, 주름 또는 탄력 저하 개선용 화장료 조성물을 제공한다.
일구현예에 따르면, 일구현예에 따르면, 상기 화장료 조성물은 가교되지 않은 구조를 가지는 히알루론산을 포함할 수 있다.
또한, 본 발명의 다른 구현예에 따르면, 상기한 바와 같은 조성물을 포함하는, 유해물질 및 광에 의해 유발되는 질환의 예방 또는 치료용 약학 조성물을 제공한다. 상기 질환은 암, 아토피 피부염, 접촉성 피부염, 지루성 피부염, 홍반, 일광화상, 광과민증, 여드름, 피부 건조증, 건선, 치사, 면역 독성, 말초 신경계 손상, 중추 신경계 손상, 내분비선 이상, 생식기관 장애, 영아 및 유아의 발달 장애, 빈혈, 기관지염, 폐기종, 폐 기능 감소, 천식, 부정맥, 심장마비, 협심증, 및 심근경색증으로 구성된 군으로부터 선택되는 어느 하나 이상을 포함할 수 있다.
일구현예에 따르면, 본 발명에 따른 약학 조성물은 필러용 주사제 제형일 수 있다.
일구현예에 따르면, 상기 약학 조성물은 가교된 구조를 가지는 히알루론산을 포함할 수 있다.

본 발명은 유해물질 및 광 등의 외부 스트레스 자극에 의한 세포의 비정상적인 반응을 저해하여 피부를 보호하고, 세포 독성이 없으므로, 약품, 또는 화장품 등의 다양한 분야에 유용하게 적용할 수 있다.

본 발명은 다양한 변환을 가할 수 있고 여러 가지 실시예를 가질 수 있는 바, 특정 실시예를 도면에 예시하고 상세한 설명에 상세하게 설명하고자 한다. 그러나, 이는 본 발명을 특정한 실시 형태에 대해 한정하려는 것이 아니며, 본 발명의 사상 및 기술 범위에 포함되는 모든 변환, 균등물 내지 대체물을 포함하는 것으로 이해되어야 한다. 본 발명을 설명함에 있어서 관련된 공지 기술에 대한 구체적인 설명이 본 발명의 요지를 흐릴 수 있다고 판단되는 경우 그 상세한 설명을 생략한다.
본 명세서 내에서 특별한 언급이 없는 한, "내지"라는 표현은 해당 수치를 포함하는 표현으로 사용된다. 구체적으로 예를 들면, "1 내지 2"라는 표현은 1 및 2를 포함할 뿐만 아니라 1과 2 사이의 수치를 모두 포함하는 것을 의미한다.
이하, 본 발명의 구현예에 따른 유해물질 및 광에 대한 피부보호용 조성물에 대하여 보다 상세하게 설명한다.
본 명세서 내의 용어 "피부보호"는 외부 환경으로부터 노출될 수 있는 가혹조건에 대하여 피부 손상의 예방, 개선 또는 치료에 도움을 주는 것을 의미한다. 상기 가혹조건은 특히 유해물질 또는 광(光)을 포함한다.
예를 들면 유해물질은 황사, 미세먼지, 담배연기, 차량 배기가스, 중금속 및 대기 오염물질을 포함하는 군으로부터 선택되는 하나 이상을 포함할 수 있다.
또한, 본 발명에서 사용되는 용어 "광"은 특별한 언급이 없는 한, 자외선, 청색광 또는 이들의 조합을 의미한다.
상기 대기 오염물질은 대기 환경 보전법 제4조(2019년 개정)에 지정된 물질 및 화합물을 포함할 수 있다. 구체적으로, 대기 오염물질은 카드뮴 및 그 화합물, 시안화수소, 납 및 그 화합물, 폴리크로리네이티드 비페닐, 크롬 및 그 화합물, 비소 및 그 화합물, 수은 및 그 화합물, 프로필렌 옥사이드, 염소 및 그 화합물, 불소 화합물, 석면, 니켈 및 그 화합물, 염화비닐, 다이옥신, 페놀 및 그 화합물, 베릴륨 및 그 화합물, 벤젠, 사염화탄소, 이황화메틸, 아닐린, 클로로포름, 포름알데히드, 아세트알데히드, 벤지딘, 1,3-부타디엔, 다환 방향족 탄화수소류, 에틸렌옥사이드, 디클로로메탄, 스틸렌, 테드라클로로에틸렌, 1,2-디클로로에탄, 에틸벤젠, 트리클로로에틸렌, 아크릴로니트릴 및 히드라진으로 구성되는 군으로부터 선택되는 하나 이상의 물질을 포함할 수 있다.
본 발명은 펩타이드류를 포함하는, 유해물질 및 광으로 인한 스트레스에 대한 피부보호용 조성물을 제공한다.
구체적으로, 본 발명은 니코티노일트리 펩타이드(nicotinoyl tripeptide), 펜타펩타이드(pentapeptide-37), 니코티노일헥사 펩타이드(nicotinoyl hexapeptide), 에스에이치-옥타펩타이드(sh-Octapeptide), 테트라카복시메틸 헥사노일디펩타이드(tetracarboxymethyl hexanoyl dipeptide), 헥사카복시메틸 디펩타이드(hexacarboxymethyldipepdite) 및 바이오티노일 헥사펩타이드-2 아마이드(biotinoyl hexapeptide-2 amdie)를 포함한다.
니코티노일트리 펩타이드는 비타민 B3(nicotinic acid, niacin)와 피부 활성화 펩타이드인 트리펩타이드(tripeptide)가 융합된 펩타이드로, 피부 과민 반응을 감소하는 데 도움을 준다.
펜타펩타이드는 피부 미백과 보습에 도움을 준다.
니코티노일헥사 펩타이드는 비타민B3와 외인성 노화를 유발하는 PAR2(신호전달수용체)의 기능을 저하시키는 헥사펩타이드를 연결시킨 것으로, 산화방지 및 피부보호에 도움을 준다.
에스에이치-옥타펩타이드는 글라이신, 히스티딘, 류신, 라이신, 페닐알라닌 및 타이로신으로 구성된 8개의 아미노산으로 이루어진 단일사슬 휴먼펩타이드로 피부의 산화적 손상을 방지한다.
테트라카복시메틸 헥사노일디펩타이드 및 헥사카복시메틸 디펩타이드는 피부컨디셔닝제로 사용된다.
바이오티노일 헥사펩타이드-2 아마이드는 헥사펩타이드-2와 바이오틴의 반응으로 얻은 것으로, C-말단은 아마이드이며, 피부 컨디셔닝제로 사용된다.
일구현예에 따르면, 상기 펩타이드류는 에스에이치-옥타펩타이드 100중량부에 대하여,
니코티노일트리 펩타이드 1 내지 50중량부, 예를 들면 5 내지 30중량부, 예를 들면 10 내지 30중량부;
펜타펩타이드 10 내지 150중량부, 예를 들면 30 내지 100중량부, 예를 들면 50 내지 70중량부;
니코티노일헥사 펩타이드 0.05 내지 10중량부, 예를 들면 0.1 내지 5중량부, 예를 들면 0.5 내지 4중량부;
테트라카복시메틸 헥사노일디펩타이드 5 내지 70중량부, 예를 들면 10 내지 50중량부, 예를 들면 20 내지 40중량부;
헥사카복시메틸 디펩타이드 15 내지 80중량부, 예를 들면 30 내지 60중량부, 예를 들면 40 내지 50중량부; 및
바이오티노일 헥사펩타이드-2 아마이드 5 내지 70중량부, 예를 들면 10 내지 50중량부, 예를 들면 20 내지 40중량부를 포함할 수 있다.
일구현예에 따르면, 본 발명은 아미노산류를 더 포함할 수 있고, 구체적으로 예를 들면 아르기닌, 아스파라긴, 아스파르트산, 글리신, 히스티딘, 히드록시프롤린, 이소류신, 류신, 메티오닌, 페닐알라닌, 프롤린, 세린 및 발린으로 이루어지는 군으로부터 선택되는 하나 이상의 아미노산을 더 포함할 수 있다.
구체적으로 예를 들면 아미노산류는 발린 100중량부에 대하여,
아르기닌 80 내지 180중량부, 예를 들면 50 내지 250중량부, 예를 들면 70 내지 150중량부, 예를 들면 90 내지 130중량부;
아스파라긴 10 내지 60중량부, 예를 들면 20 내지 50중량부, 예를 들면 25 내지 40중량부;
아스파르트산 20 내지 80중량부, 예를 들면 30 내지 70중량부, 예를 들면 35 내지 50중량부;
글리신 30 내지 100중량부, 예를 들면 40 내지 80중량부, 예를 들면 40 내지 60중량부;
히스티딘 50 내지 150중량부, 예를 들면 60 내지 120중량부, 예를 들면 70 내지 100중량부;
히드록시프롤린 5 내지 40중량부, 예를 들면 10 내지 30중량부, 예를 들면 10 내지 20중량부;
이소류신 40 내지 120중량부, 예를 들면 50 내지 100중량부, 예를 들면 60 내지 80중량부;

본 발명에 따른 조성물은 유해물질을 흡착 또는 배출함으로써 피부 세포를 보호할 수 있다.
또한, 자외선에 의한 피부 상해를 억제할 수 있다. 피부는 자외선으로 인하여 DNA 상해를 입을 수 있고, 사이클로부탄 피리미딘 다이머(cyclobutane pyrimidine dimer, CPD)가 생성된다. CPD는 인접하고 있는 피리미딘 염기의 6각형 고리 구조 내에 이중결합이 단파장(약 254 nm) 자외선 에너지에 의해 변화되어 4각형의 새로운 결합을 형성함으로써 비정상적 결합을 유도하고 DNA에 구조적 결함을 야기하는데, 본 발명에 따른 조성물은 CDP 형성을 억제하여 자외선으로 인한 피부세포 사멸을 억제할 수 있다.
또한, 항산화 활성을 향상시키고, 유해물질 및 광으로 인한 스트레스에 의한 피부 세포 손상을 예방 및 개선하여 피부 다크닝, 주름 또는 탄력 저하를 개선할 수 있다.
일구현예에 따르면, 본 발명의 조성물은 통상적인 화장료 조성물, 피부 외용제 약학 조성물 등 피부학적으로 적용이 가능한 어떠한 제형으로도 제조될 수 있다. 피부학적으로 적용이 가능하다는 것은 적용 대상에게 비교적 비독성이고 무해한 유효작용을 할 수 있는 조성물로서, 피부에 적용할 수 있는 외용제를 포함할 수 있으며, 상기 조성물로부터 기인하는 부작용이 유효 성분의 효능을 저하시키기 않고, 적용 대상에 심각한 자극을 유발하지 않으면서 유효성분들의 활성 및 물성을 손상시키지 않는다는 것을 의미한다.
또한, 본 발명의 피부 외용제 조성물은 당업계에서 통상적으로 제조되는 어떠한 제형으로도 제조될 수 있으며, 예를 들어, 용액, 현탁액, 유액, 유탁액, 페이스트, 겔, 팩, 크림, 로션, 파우더, 비누, 계면활성제-함유 클렌징, 오일, 분말 파운데이션, 유탁액 파운데이션, 왁스 파운데이션, 스프레이 및 모발 화장료 등으로 제형화될 수 있으나, 이에 한정되는 것은 아니다.
구체적으로, 상기 화장료 조성물은 스킨로션, 스킨소프너, 스킨토너, 아스트린젠트, 로션, 밀크로션, 모이스쳐 로션, 영양로션, 맛사지크림, 영양크림, 모이스처크림, 핸드크림, 파운데이션, 에센스, 영양에센스, 팩, 비누, 헤어샴푸, 풋샴푸, 클렌징폼, 클렌징로션, 클렌징크림, 바디로션 및 바디클린저의 제형 등으로 제조될 수 있고, 예를 들면 화장료용 필러로 제조될 수 있다.
화장료용 필러는 피부 표면에 도포하는 방법으로 적용할 수 있고, 예를 들면 향료, 잔탄검, 왁스, 버터, 오일, 계면활성제, 보습제, 알코올 등을 포함할 수 있고, 통상적으로 포함할 수 있는 화장료 조성물의 구성 성분이라면 특별히 제한하지 않고 포함할 수 있다. 또한, 화장료용 필러 조성물이 히알루론산을 포함하는 경우, 상기 히알루론산은 가교되지 않은 구조(non-crosslinked)를 가질 수 있다.
본 발명의 다른 구현예에 따르면, 유해물질 및 광에 의해 유발되는 질환의 예방 또는 치료용 약학 조성물을 제공한다. 구체적으로 예를 들면, 상기 질환은 암, 아토피 피부염, 접촉성 피부염, 지루성 피부염, 홍반, 일광화상, 광과민증, 여드름, 피부 건조증, 건선, 치사, 면역 독성, 말초 신경계 손상, 중추 신경계 손상, 내분비선 이상, 생식기관 장애, 영아 및 유아의 발달 장애, 빈혈, 기관지염, 폐기종, 폐 기능 감소, 천식, 부정맥, 심장마비, 협심증, 및 심근경색증으로 구성된 군으로부터 선택되는 어느 하나 이상일 수 있다.
일구현예에 따르면, 본 발명에 따른 약학 조성물은 약제적으로 허용 가능한 담체를 포함할 수 있다. 약제학적으로 허용되는 담체는 결합제, 활탁제, 붕해제, 부형제, 윤활제, 가용화제, 분산제, 안정화제, 현탁화제, 색소, 향료, 완충제, 보존제, 무통화제, 가용화제, 등장제 등을 혼합하여 사용할 수 있다. 본 발명의 약학 조성물의 제형은 상술한 바와 같은 약제학적으로 허용되는 담체와 혼합하여 다양하게 제조될 수 있다. 본 발명에 따른 약학 조성물의 제형은 의료용 보충재, 용액, 현탁액, 연고, 정제, 캡슐 등으로 제형화 할 수 있으나, 상기한 바에 한정하지는 않는다.
의료용 보충재로는 의료용 필러, 창상피복재, 뼈이식재, 연조직 보충재, 치과재료 등을 포함할 수 있고, 인공조직, 인공장기, 세포배양지지체, 약물전달제제, 이식용 바이오센서 등에 적용할 수 있다.
일구현예에 따르면, 본 발명의 조성물은 필러용 주사제의 형태로 제조될 수 있으며, 예를 들면 알긴산(alginic acid), 카르복시메틸셀룰로즈(carboxymethyl cellulose), 키토산(chitosan), 덱스트란(dextran), 콜라겐(collagen), 젤라틴(gelatin), 펙틴(pectin), 아가(agar), 아밀로즈(amylose), 사이클로덱스트린(cyclodextrin) 및 엘라스틴(elastin)으로 이루어진 군으로부터 선택되는 하나 이상의 담체를 포함할 수 있다. 필러용 주사제 조성물이 히알루론산을 포함하는 경우, 상기 히알루론산은 가교되어 있는 구조(cross-linked)를 가질 수 있다.
또한, 예를 들면 생분해성 고분자 지지체(scaffold)로 히알루론산(hyaluronic acid), 폴리글리콜산(PGA), 폴리락트산(PLA), 폴리락트산-글리콜산 공중합체(PLGA), 폴리-ε-카프로락톤(PCL), 폴리아미노산(polyamino acid), 폴리안하이드라이드(polyanhydride), 폴리오르쏘에스테르(polyorthoester) 및 이들의 공중합체를 포함할 수 있다.
일구현예에 따르면, 본 발명에 따른 조성물은 히알루론산을 포함한다. 히알루론산은 이의 염 또는 가교된 히알루론산을 포함할 수 있으며, 예를 들면 히알루론산 나트륨(sodium hyaluronate)을 포함할 수 있다. 또한, 예를 들면 히알루론산 분자의 질량은 500KDa 내지 2Mda, 예를 들면 1 내지 2Mda, 예를 들면 1 내지 1.5Mda일 수 있다. 본 발명에 포함할 수 있는 히알루론산 함량은 조성물 총 중량에 대하여 1 내지 5중량%, 예를 들면 1 내지 3중량%일 수 있다.
일구현예에 따르면, 생분해성 고분자 지지체는 공극이 형성된 것일 수 있고, 예를 들면 염, 발포성 염, 탄수화물, 탄화수소 왁스 등을 사용하여 공극을 형성할 수 있다.
또한, 본 발명에 따른 필러용 주사제는 국소 마취제, 항히스타민제, 비타민 등을 포함할 수 있다.
국소마취제로는 예를 들면, 리도카인(lidocaine), 에티도카인(etidocaine), 부피바카인(bupivacaine), 테트라카인(tetracaine), 메피바카인(mepivacaine), 프로카인(procaine), 프릴로카인(prilocaine), 로피바카인(ropivacaine) 등을 포함할 수 있으나, 주사제용 국소마취제라면 특별히 제한하지 않는다.
항히스타민제로는 예를 들면 plokon(piprinhydrinate), 클로로페니라민(chlorpheniramine), 디페닐피랄린(diphenylpyraline, piprinhydrinate), 디펜히드라민(diphenhydramine), 세티리진(cetirizine) 등을 포함할 수 있으나, 주사제용 항히스타민제라면 특별히 제한하지 않는다.
이하, 본 발명이 속하는 기술 분야에서 통상의 지식을 가진 자가 용이하게 실시할 수 있도록 본 발명의 실시예에 대하여 상세히 설명한다. 그러나 본 발명은 여러 가지 상이한 형태로 구현될 수 있으며 여기에서 설명하는 실시예에 한정되지 않는다.

실험예 1: 청색광(blue light)에 대한 세포 보호
본 발명에 따른 조성물의 청색광에 대한 세포 보호 효과를 확인하기 위하여 사람 섬유아세포(NHDFs)를 사용하여 세포 생존율 시험법인 MTS((3-(4,5-dimethylthiazol-2-yl)-5-(3-carboxymethoxyphenyl)-2-(4-sulfophenyl)-2H-tetrazolium) assay 로 측정하였다.
사람섬유아세포(normal human foreskin-derived Dermal fibroblasts, NHDFs;ATCC,CRL-2522)세포를 24웰 플레이트(24well plate)에 3Х104 cells/well 농도로 5% CO2, 37℃의 조건에서 10% 소태아혈청과 100 U/mL 페니실린-스트렙토마이신(penicillin- streptomycin)을 함유하는 DMEM으로 24시간 배양하였다. 실시예 조성물을 0 ~ 1.0% 농도로 준비하여 배양된 세포에 24시간 처리 후, 청색광(blue light, 453nm,40J/cm2)을 6시간 조사하여 세포생존율을 측정하였다. 측정방법은 배양된 세포의 배지를 제거한 후 MTS solution(0.33 mg/mL MTS)을 1시간 내지 4시간 동안 처리하여 분광광도계(Epoch 2, BioTek)로 490nm에서 흡광도를 측정하였다.
MTS 시험을 통한 블루라이트로부터 사람섬유아세포의 보호효과 결과는 도 1에 나타내었다. 그 결과, 블루라이트 조사에 의한 세포생존율에 있어서, 본 발명에 따른 조성물을 처리한 군은 조성물을 처리하지 않은 블루라이트 단독 조사군 대비 1 내지 11% 높은 세포생존율을 나타내었다.
실험예 2: 자외선에 의한 피부손상 방지(ex vivo)
본 발명에 따른 실시예 조성물(product S)의 자외선에 의한 피부손상 방지 효과를 확인하기 위하여 실시예 조성물을 처리한 피부생검(skin biopsies)에 UVA/UVB를 조사하여 피부절편(skin explants)의 형태 (H/E Staining) 및 사이클로부탄 피리미딘 다이머(Cyclobutane Pyrimidine Dimers, CPDs) 양을 측정하였다. 자외선 조사는 DNA손상을 일으키며, 이는 CPDs형성에 의해 발생하는 것이다.
생체 외 모델(ex vivo)인 full-thickness human skin tissues는 윤리적인 승인과 사전 환자 동의서를 확인하였고, 자세한 사항은 표 2와 같다. 채취한 피부조직은 5% CO2, 37℃의 조건에서 24시간 동안 배양 후 시험을 진행하였다.
피부 절편은 12 well plate에 24시간 배양 후 30% 농도의 시료(실시예 조성물)를 24시간 후, 5시간 동안 자외선을 조사(UVA/UVB)하여 Hematoxylin/eosin 염색법(H/E staining)에 의해 피부조직(6um thick section)의 형태를 광학현미경(DM2000, lens20X)으로 관찰하였다(도 2).
그리고, CPDs 시험조건은 상기 방법과 동일하게 시료를 처리 후 자외선 조사(UVA/UVB)를 시간별(30분, 1시간 30분, 5시간)로 조사하여 분석하였다.
측정방법은 paraffin-embedded tissue section으로 immunohistochemistry(IHC)을 사용하여 CPDs 생성량을 측정하였고(도 3), 광학현미경으로 시료조직을 관찰하였다(도 4). 단, 자외선 A 조사는 50J/cm2, 자외선 B 조사는 500mJ/cm2로 하였다.
도 2에서는 자외선에 의한 광노화현상의 지표로 피부 표피층의 두께와 진피층의 밀도를 확인하였으며, 피부조직을 관찰한 결과, 실시예 조성물을 처리하지 않은 자외선 조사군은 표피층의 두께가 두꺼워지고, 진피층의 밀도가 감소하는 것을 확인하였다. 반면, 실시예 조성물(product S)을 처리한 군에서는 표피층이 얇아지고, 진피층의 밀도가 증가한 것을 확인하였다.
그리고 도 3에서, CPDs 생성량은 실시예 조성물을 처리하지 않은 자외선 조사군에 대비하여, 실시예 조성물을 처리한 자외선 조사군에서 평균 3 내지 7% 감소한 결과를 나타내었다.
또한, 도 4에서 CPDs 생성 결과를 피부조직 내에서 관찰한 결과 실시예 조성물을 처리하지 않은 자외선 조사군에서는 CPDs 생성이 뚜렷하게 나타나는 반면, 실시예 조성물을 처리한 군에서는 조성물을 처리하지 않은 자외선 조사군 대비 감소한 경향을 나타내었다.
실험예 3: 대기오염물질에 대한 세포 보호(in vivo)
본 발명에 따른 조성물의 대기오염물질에 대한 세포 보호 효과를 확인하기 위하여 사람 섬유아세포(NHDFs)에 대기오염물질인 도시 먼지(urban dust)와 실시예 조성물을 처리하였다. 측정은 세포 생존율 시험법인 MTS((3-(4,5-dimethylthiazol-2-yl)-5-(3-carboxymethoxyphenyl)-2-(4-sulfophenyl)-2H-tetrazolium)assay로 하였다.
도시 먼지(SRM1649b, 미국 NIST(US National Institute of Standards and Technology)는 1976~1977년도에 워싱턴D.C에서 수집한 대기입자를 사용하였다. 도시먼지에는 다양한 화합물이 포함되어 있으며, 그 종류로는 PAHs(Polycyclic aromatic hydrocarbons), nitrosubstituted polycyclic aromatic hydrocarbons, PCB(polychlorinated biphenyl congeners, chlorinated pesticides, decabromodiphenyl ether(decaBDE), dibenzo-p-dioxin 및 dibenzofuran congeners를 포함한다. 입자사이즈는 대략 10um정도로 혼합되어 있다.
사람섬유아세포(Normal human foreskin-derived Dermal fibroblasts, NHDFs;ATCC,CRL-2522)세포를 24 웰 플레이트(24well plate)에 3Х104 cells/well 농 도로 5% CO2, 37℃의 조건에서 10% 소태아혈청과 100 U/mL 페니실린-스트렙토마이신(penicillin- streptomycin)을 함유하는 DMEM으로 24시간 배양하였다. 실시예 조성물은 0 ~ 10% 농도로 준비하여 배양된 세포에 6시간 처리 후, Urban Dust 200ug/ml을 처리하고 18시간 동안 배양한 뒤 세포생존율을 측정하였다. 측정은 배양된 세포의 배지를 제거한 후 MTS solution(0.33 mg/mL MTS)을 1시간 내지 4시간 동안 처리하여 분광광도계(Epoch 2, BioTek)로 490nm에서 흡광도를 측정하는 방법으로 하였다.
MTS 시험을 통한 대기오염물질인 Urban Dust에 대한 사람섬유아세포의 보호 효과 결과는 도 5에 나타내었다. 그 결과, 실시예 조성물 처리군은 조성물을 처리하지 않은 Urban Dust 단독 처리군 대비 세포생존율이 8 내지 13% 더 높게 나타났다.
따라서, 본 발명은 대기오염물질로부터 피부 오염방지(antipollution) 효과를 나타내는 것을 확인하였다.
실험예 4: 대기오염물질 및 자외선에 대한 세포 보호(in vivo)
본 발명에 따른 조성물의 대기오염물질 및 자외선에 대한 세포 보호 효과를 확인하기 위하여 사람 섬유아세포(NHDFs)에 대기오염물질인 Urban Dust와 자외선 A를 조사하여 펩타이드조성물에 대한 세포보호효과를 확인하였다. 평가는 세포 생존율 시험법인 MTS((3-(4,5-dimethylthiazol-2-yl)-5-(3-carboxymethoxyphenyl)-2-(4-sulfophenyl)-2H-tetrazolium)assay로 측정하였다.
사람섬유아세포(Normal human foreskin-derived Dermal fibroblasts, NHDFs;ATCC,CRL-2522)세포를 24웰 플레이트(24well plate)에 3Х104 cells/well 농도로 5% CO2, 37℃의 조건에서 10% 소태아혈청과 100 U/mL 페니실린-스트렙토마이신(penicillin- streptomycin)을 함유하는 DMEM으로 24시간 배양하였다. 시료(실시예 조성물)는 1% 농도로 준비하여 배양된 세포에 24시간 처리 후, Urban Dust 80ug/ml를 24시간 처리하였다. 그리고, 자외선 A를 10J/cm2 조사하여 세포생존율을 측정하였다. 측정은 배양된 세포의 배지를 제거한 후 MTS solution(0.33 mg/mL MTS)을 1시간 내지 4시간 동안 처리하여 분광광도계(Epoch 2, BioTek)로 490nm에서 흡광도를 측정하는 방법으로 하였다.
MTS 시험을 통한 대기오염물질인 Urban Dust(UbD)와 자외선 A(UVA)조사에 의해 실시예 조성물의 사람섬유아세포 보호 효과 결과는 도 6에 나타내었다. 그 결과, 실시예 조성물을 처리한 군은 조성물을 처리하지 않은 Urban Dust 및 자외선 A 조사군 대비 세포생존율이 약 16% 이상 높게 나타났다.
따라서, 본 발명은 대기오염물질과 자외선 조사를 동시에 처리하였을 때 세포보호 효과를 나타내는 것을 확인하였다.
실험예 5: 대기오염물질 및 자외선에 대한 세포 산화스트레스
본 발명에 따른 조성물의 세포 산화스트레스 보호 효과를 확인하기 위하여 자유라디칼(ROS)을 측정하였다.
세포 내 자유라디칼(ROS) 생성량을 측정하기 위해 H2DCFDA(2'-7'dichlorodihydrofluorescein diacetate) assay를 이용하였다. H2DCFDA assay는 자유라디칼에 의해 산화되어 형광을 내는 DCF(2'-7'dichlorodihydrofluorescein)로 변환되는 원리로 측정하는 것이다.
시험방법은 사람섬유아세포(Normal human foreskin-derived Dermal fibroblasts, NHDFs;ATCC,CRL-2522)세포를 24 웰 플레이트(24well plate)에 3Х104 cells/well 농도로 5% CO2, 37℃의 조건에서 10% 소태아혈청과 100 U/mL 페니실린-스트렙토마이신(penicillin- streptomycin)을 함유하는 DMEM으로 24시간 배양하였다. 시료(실시예 조성물)는 1% 농도로 준비하여 배양된 세포에 24시간 처리 후, DCFDA(Fisher Scientific, D399)를 10uM 첨가하여 30분간 배양하였다. DCFDA는 HBSS 용액에 희석하여 사용하였다. DCFDA용액을 제거한 후 HBSS 용액으로 2회 세척하고, Urban Dust 80ug/ml를 24시간 처리 및 자외선 A를 10J/cm2 조사하였고, 세포 내 형광 광도를 형광광도계(Victor3,1420 Multilabel counter, Perkin Elmer)를 이용하여 excitation:485nm; emission:520nm에서 측정하였다. 그 결과는 도 7에 나타내었으며, 시료를 처리한 군에서 시료를 처리하지 않은 군 대비 약 73% 이상 자유라디칼 소거능이 향상된 것으로 확인하였다.
실험예 6: 주름 개선 효과
본 발명에 따른 조성물에 대한 주름 개선 효과를 확인하기 위하여 표 1에 따른 실시예 조성물을 포함하는 주사제를 사용하였다.
건강한 35세에서 55세의 여성을 대상으로 실시하였으며, 피부에 2주 간격으로 상기 주사제를 2ml/3회 주사하여 주름 개선 효과를 확인하였다. 주름의 상태는 사진촬영 및 3D 분석으로 평가하였다.
그 결과는 도 8 내지 10에 나타내었으며, 주름이 있는 부위에 대하여 주사제의 주입 전과 후의 사진을 비교하였다.
도 8 내지 10에 나타난 바와 같이, 본 발명에 따른 조성물을 2주간 3회 주사하였을 때, 주름 개선율이 10~20% 향상되었음을 확인하였다.
상기와 같은 결과로부터 확인할 수 있는 바와 같이, 본 발명은 피부 세포에 독성이 없고, 외부환경의 유해물질 또는 광으로부터 피부 세포를 보호할 수 있음을 확인하였으며, 구체적으로는 유해물질, 광 또는 이들 조합으로
이상의 설명은 본 발명의 기술 사상을 예시적으로 설명한 것에 불과한 것으로서, 본 발명이 속하는 기술 분야에서 통상의 지식을 가진 자라면 본 발명의 본질적인 특성에서 벗어나지 않는 범위에서 다양한 수정 및 변형이 가능하다. 또한, 본 발명에 개시된 실시 예들은 본 발명의 기술 사상을 한정하기 위한 것이 아니라 설명하기 위한 것이고, 이러한 실시 예에 의하여 본 발명의 기술 사상의 범위가 한정되는 것은 아니다. 본 발명의 보호 범위는 아래의 청구범위에 의하여 해석되어야 하며, 그와 동등한 범위 내에 있는 모든 기술 사상은 본 발명의 권리범위에 포함되는 것으로 해석되어야 할 것이다.
Claims (17)
- 펩타이드류; 및 아미노산류를 포함하고,
상기 펩타이드류가 니코티노일트리 펩타이드, 펜타펩타이드, 니코티노일헥사 펩타이드, 에스에이치-옥타펩타이드, 테트라카복시메틸 헥사노일디펩타이드, 헥사카복시메틸 디펩타이드 및 바이오티노일 헥사펩타이드-2 아마이드를 포함하고,
상기 아미노산류가 아르기닌, 아스파라긴, 아스파르트산, 글리신, 히스티딘, 히드록시프롤린, 이소류신, 류신, 메티오닌, 페닐알라닌, 프롤린, 세린 및 발린을 포함하는, 유해물질, 광 또는 이들의 조합에 대한 피부보호용 화장료 조성물. - 제1항에 있어서,
히알루론산을 더 포함하고, 상기 히알루론산 분자의 질량이 500KDa 내지 2Mda인 것인, 화장료 조성물. - 제1항에 있어서,
상기 유해물질이 황사, 미세먼지, 담배연기, 차량 배기가스 및 중금속 중 하나 이상을 포함하는 대기 오염물질인 것인, 화장료 조성물. - 제1항에 있어서,
상기 조성물이 필러용 제형인 것인, 화장료 조성물. - 제1항에 있어서,

헥사카복시메틸 디펩타이드 15 내지 80중량부; 및
바이오티노일 헥사펩타이드-2 아마이드 5 내지 70중량부를 포함하고,
상기 아미노산류가
발린 100중량부에 대하여,
아르기닌 80 내지 180중량부;
아스파라긴 10 내지 60중량부;
아스파르트산 20 내지 80중량부;
글리신 30 내지 100중량부;
히스티딘 50 내지 150중량부;
히드록시프롤린 5 내지 40중량부;
이소류신 40 내지 120중량부;
류신 50 내지 150중량부;
메티오닌 5 내지 40중량부;
페닐알라닌 40 내지 120중량부;
프롤린 10 내지 50중량부; 및
세린 20 내지 80중량부를 포함하는 것인, 화장료 조성물. - 제1항에 있어서,
상기 조성물 총 중량에 대하여 히알루론산을 1 내지 5중량% 더 포함하는 것인, 화장료 조성물. - 펩타이드류; 및 아미노산류를 포함하고,
상기 펩타이드류가 니코티노일트리 펩타이드, 펜타펩타이드, 니코티노일헥사 펩타이드, 에스에이치-옥타펩타이드, 테트라카복시메틸 헥사노일디펩타이드, 헥사카복시메틸 디펩타이드 및 바이오티노일 헥사펩타이드-2 아마이드를 포함하고,
상기 아미노산류가 아르기닌, 아스파라긴, 아스파르트산, 글리신, 히스티딘, 히드록시프롤린, 이소류신, 류신, 메티오닌, 페닐알라닌, 프롤린, 세린 및 발린을 포함하는,
유해물질, 광 또는 이들의 조합에 의해 유발되는 아토피 피부염, 접촉성 피부염, 지루성 피부염, 홍반, 일광화상, 광과민증, 여드름, 피부 건조증 및 건선 중 하나 이상의 질환의 예방 또는 치료용 약학 조성물. - 제11항에 있어서,
