Abstract

Description
증식성 질환의 치료에 사용되는, 시클린 의존성 키나제 4/6 (CDK4/6) 억제제, B-Raf 키나제 억제제 및 임의로 미토겐-활성화 단백질 키나제 키나제 (MEK 1/2 또는 MEK) 억제제의 조합물. 본 발명은 또한 증식성 질환의 치료에서의 이러한 조합물의 용도; 작용제 조합물의 제약 조성물, 및 치료 유효량의 이러한 조합물을 증식성 질환을 앓는 대상체에게 투여하는 것을 포함하는, 증식성 질환을 앓는 대상체를 치료하는 방법에 관한 것이다.
종양 발생은 시클린 의존성 키나제 (CDK) 및 그 조절인자의 유전자 변경 및 탈조절과 밀접하게 연관되어 있으며, 이는 CDK의 억제제가 유용한 항암 치료제일 수 있음을 시사한다. CDK의 기능은 특정 단백질을 인산화시키고 이에 따라 활성화 또는 탈활성화시키는 것이다. CDK에 의해 매개되는 촉매 단계는 ATP로부터 거대분자 효소 기질로의 인산-전달 반응을 포함한다. 여러 군의 화합물 (예를 들어 문헌 [Fischer, P. M. Curr. Opin. Drug Discovery Dev. 2001, 4, 623-634]에서 검토됨)이 CDK-특이적 ATP 길항작용에 의해 항증식성 특성을 갖는 것으로 발견되었다.
분자 수준에서의 CDK/시클린 복합체 활성의 매개는 일련의 자극 및 억제 인산화 또는 탈인산화 사건을 필요로 한다. CDK 인산화는 CDK 활성화 키나제 (CAK) 및/또는 wee1, Myt1 및 Mik1과 같은 키나제의 군에 의해 수행된다. 탈인산화는 포스파타제, 예컨대 cdc25 (a & c), pp2a 또는 KAP에 의해 수행된다.
CDK/시클린 복합체 활성은 하기 2종 패밀리의 내인성 세포 단백질성 억제제에 의해 추가로 조절될 수 있다: Kip/Cip 패밀리 또는 INK 패밀리. INK 단백질은 CDK4 및 CDK6에 특이적으로 결합한다. p16ink4 (MTS1로도 공지됨)는 다수의 원발성 암에서 돌연변이되거나 또는 결실되어 있는 잠재적 종양 억제인자 유전자이다. Kip/Cip 패밀리는 p21Cip1, Waf1, p27Kip1 및 p57kip2와 같은 단백질을 포함하며, 여기서 p21은 p53에 의해 유도되고 CDK2/시클린(E/A) 복합체를 불활성화시킬 수 있다. 비전형적으로 낮은 수준의 p27 발현이 유방암, 결장암 및 전립선암에서 관찰된 바 있다. 반대로 고형 종양에서의 시클린 E의 과다 발현이 불량한 환자 예후와 상관관계가 있는 것으로 나타났다. 시클린 D1의 과다 발현은 식도, 유방, 편평세포 및 비-소세포 폐 암종과 연관되었다.
증식 세포에서 세포 주기를 조정하고 구동하는데 있어서의 CDK 및 그의 연관 단백질의 중추적 역할이 상기 요약되어 있다. CDK가 주요 역할을 하는 일부 생화학적 경로가 또한 기재되어 있다. 따라서, 일반적으로 CDK 또는 특이적 CDK를 표적화하는 치료제를 사용하는 증식성 장애, 예컨대 암의 치료를 위한 단일요법의 개발이 잠재적으로 매우 바람직하다. 따라서, 인간 질환을 치료하기 위한 신규 치료제를 발견하는 것이 계속 요구되고 있다. 본 조합물에서 유용한 CDK4/6 억제제는 공개된 PCT 특허 출원 WO2010/020675 (본원에 참조로 포함됨)에 일반적으로 및 구체적으로 기재되어 있다.
단백질 키나제는 매우 다양한 세포 과정의 조절 및 세포 기능에 대한 제어 유지에서 중심 역할을 하는 단백질의 거대 패밀리를 나타낸다. 이상 키나제 활성은 양성 및 악성 증식성 장애, 뿐만 아니라 면역계 및 신경계의 부적절한 활성화로부터 발병하는 질환을 비롯한 다수의 질환 상태에서 관찰된 바 있다.
Ras-Raf-MEK-ERK 신호전달 경로는 세포 표면 수용체로부터 핵으로 신호를 전송하며, 세포 증식 및 생존에 필수적이다. 인간 암의 10-20%가 종양원성 Ras 돌연변이를 보유하고, 다수의 인간 암이 활성화된 성장 인자 수용체를 갖기 때문에, 이 경로는 개입을 위한 이상적인 표적이다.
세린/트레오닌 키나제의 Raf 패밀리는 3종의 구성원: C-Raf (또는 Raf-1), B-Raf 및 A-Raf를 포함한다. B-Raf 대립유전자의 활성화는 흑색종의 ~70%, 유두상 갑상선 암종의 40%, 난소 저-등급 암종의 30% 및 결장직장암의 10%에서 확인되었다. 대부분의 B-Raf 돌연변이는 키나제 도메인 내에서 발견되며, 단일 치환 (V600E)이 80%를 차지한다. 돌연변이된 B-Raf 단백질은 MEK를 향한 상승된 키나제 활성을 통해 또는 C-Raf의 활성화를 통해 Raf-MEK-ERK 경로를 활성화한다. 본 조합 요법에서의 B-Raf 억제제는 B-Raf 키나제를 수반하는 세포 과정을, 이들 암 세포에서 신호 캐스케이드를 차단하고 궁극적으로 세포의 정지 및/또는 사멸을 유도함으로써 억제한다. 본 조합물에 유용한 B-Raf 억제제는 공개된 PCT 특허 출원 WO2011/025927 (본원에 참조로 포함됨)에 일반적으로 및 구체적으로 기재되어 있다.
MEK는 또한 RAS/ RAF/ MEK/ ERK 경로에서의 주요 단백질이며, 이는 세포 증식 및 생존에 신호전달하고, RAS 또는 RAF 종양유전자에서 또는 성장 수용체 티로신 키나제에서 돌연변이를 갖는 종양에서 빈번하게 활성화된다. 암에서 단지 드물게 돌연변이됨에도 불구하고, MEK1 및 MEK2 단백질의 억제제는 또한 RAS/ RAF/ MEK 신호 전달 경로 신호전달 캐스케이드 내 그의 중심 위치 때문에 소분자 억제를 위해 표적화되었다.
본 조합물에 사용하기 위한 적절한 임의적 MEK 1/2 억제제는 관련 기술분야에 공지되어 있다. 본 발명에 유용한 MEK 1/2 억제제는 PD325901, PD-181461, ARRY142886 / AZD6244, ARRY-509, XL518, JTP-74057, AS-701255, AS-701173, AZD8330, ARRY162, ARRY300, RDEA436, E6201, RO4987655/R-7167, GSK1120212 또는 AS703026을 포함한다.
중요한 실시양태에서, MEK 1/2 억제제는 WO03/077914 (그 전문이 본원에 참조로 포함됨)에 기재된 화합물, 특히 하기 화학식 II 또는 III의 화합물 또는 그의 제약상 허용되는 염 (이하, 각각 화합물 C 및 D로 본원에 지칭됨), 및 WO05/051906, WO05/023251, WO03/077855, US20050049419 및 US7235537 (그 전문이 본원에 참조로 포함됨)에 기재된 화합물을 포함하며, 이는 증식성 질환의 치료를 위한 MEK 1/2 억제제로서의 N3-알킬화 벤즈이미다졸 및 다른 유사한 헤테로시클릭 유도체를 포괄한다.

본 발명은 특히

(b) 하기 화학식의 B-Raf 억제제 또는 그의 제약상 허용되는 염 (이하 화합물 B로 지칭됨), 및

이하, 화합물 A 및 화합물 B의 조합물, 및 화합물 A, 화합물 B 및 MEK 1/2 억제제의 삼중 조합물이 본 발명의 조합물로 지칭될 것이다.
본 발명은 특히 증식성 질환의 치료 또는 예방을 위해 그를 필요로 하는 대상체에게의 개별, 동시 또는 순차적 투여에 유용한 본 발명의 조합물에 관한 것이다.
본 발명은 또한 증식성 질환의 치료 또는 예방을 필요로 하는 대상체에서의 증식성 질환의 치료 또는 예방을 위한 제약 조성물 또는 의약의 제조에 사용하기 위한 본 발명의 조합물에 관한 것이다.
본 발명은 또한 증식성 질환의 치료 또는 예방을 위한 제약 조성물 또는 의약의 제조를 위한 본 발명의 조합물의 용도에 관한 것이다.
본 발명은 증식성 질환을 갖는 대상체에게 본 발명의 조합물을 증식성 질환에 대해 연합 치료상 유효한 양으로 투여하는 것을 포함하는, 상기 대상체를 치료하는 방법에 관한 것이다.
본 발명은 추가로 치료제로서의 본 발명의 조합물을, 증식성 질환의 진행 지연 또는 치료에 사용하기 위한 그의 동시, 개별 또는 순차적 투여를 위한 지침서와 함께 포함하는 상업용 패키지를 제공한다.

따라서, 본 발명은 추가로

본 발명은 또한 (a) 화합물 A 및 (b) 화합물 B, 및 임의로 화합물 D를 포함하는 조합물, 예컨대 조합 제제 또는 제약 조성물에 관한 것이다.

본 발명의 추가 실시양태에서, 증식성 질환은 흑색종, 폐암 (비소세포 폐암 (NSCLC) 포함), 결장직장암 (CRC), 유방암, 신장암, 예컨대 예를 들어 신세포 암종 (RCC), 간암, 자궁내막암, 급성 골수 백혈병 (AML), 골수이형성 증후군 (MDS), 갑상선암, 특히 유두상 갑상선암, 췌장암, 신경섬유종증 또는 간세포성 암종이다.
본 발명의 추가 실시양태에서, 증식성 질환은 고형 종양이다. 용어 "고형 종양"은 특히 흑색종, 유방암, 난소암, 결장직장암, 및 일반적으로 위장관, 자궁경부암, 폐암 (소세포 폐암 및 비소세포 폐암 포함), 두경부암, 방광암, 전립선암 또는 카포시 육종을 의미한다. 본 조합물은 고형 종양 및 또한 액상 종양의 성장을 억제한다. 또한, 종양 유형 및 사용된 특정한 조합물에 따라, 종양 부피의 감소가 수득될 수 있다. 본원에 개시된 본 발명의 조합물은 또한 종양의 전이성 확산 및 미세전이의 성장 또는 발생을 예방하는데 적합하다. 본원에 개시된 본 발명의 조합물은 불량한 예후의 환자, 특히 전이성 흑색종, 결장직장암 또는 췌장암을 갖는 이러한 불량한 예후의 환자의 치료에 적합하다.
추가 실시양태에서, 증식성 질환은 흑색종 또는 결장직장암이다.
본 발명의 조합물은 RAS/ RAF/ MEK 신호 전달 경로에서의 유전자 변경, 예컨대 예를 들어 B-Raf 돌연변이 또는 유전자 증폭을 갖는 암의 치료에 특히 유용하다.
중요한 실시양태에서, 치료할 암은 B-Raf 돌연변이를 특징으로 하며, 예를 들어, B-Raf 돌연변이된 결장직장암 및 B-Raf 돌연변이된 흑색종이다. 특히, B-Raf 돌연변이는 V600 돌연변이, 예를 들어 V600E, V600K 또는 V600G 돌연변이이다.
증식성 질환의 성질은 다인성이다. 특정 상황 하에, 다양한 작용 메카니즘을 갖는 약물이 조합될 수 있다. 그러나, 단순히 다양한 작용 방식을 갖는 치료제의 임의의 조합물을 고려하는 것이 반드시 유리한 효과를 갖는 조합물을 유도하는 것은 아니다.
본 발명의 제약 조합물의 투여는, 본 발명의 조합물에 사용되는 제약 치료제 중 단 하나만을 적용하는 단독요법과 비교하여, 예를 들어 증상의 완화, 증상 진행의 지연 또는 증상의 억제와 관련된 유익한 효과, 예를 들어 상승작용적 치료 효과 뿐만 아니라, 추가의 놀라운 유익한 효과, 예를 들어 보다 적은 부작용, 보다 지속적인 반응, 개선된 삶의 질 또는 감소된 이환율을 생성할 수 있다.
추가의 이익은 보다 낮은 용량의 본 발명의 조합물의 치료제가, 예를 들어 투여량이 종종 보다 소량일 수 있을 뿐만 아니라 덜 빈번하게 적용될 수 있도록 사용될 수 있거나, 또는 조합물 파트너 중 하나를 단독으로 사용하여 관찰되는 부작용의 발생을 감소시키기 위해 사용될 수 있다. 이는 치료할 환자의 필요 및 요건에 따른다.
본 발명의 조합물이 상기 본원에 기재된 유익한 효과를 생성한다는 것은 확립된 시험 모델에 의해 제시될 수 있다. 통상의 기술자는 이러한 유익한 효과를 입증하기 위해 적절한 시험 모델을 충분히 선택할 수 있다. 본 발명의 조합물의 약리학적 활성은, 예를 들어 본질적으로 하기 기재된 바와 같은 임상 연구 또는 동물 모델에서 입증될 수 있다.

신규 제약 조성물은 약 0.1% 내지 약 99.9%, 바람직하게는 약 1% 내지 약 60%의 치료제(들)를 함유할 수 있다.

본 발명에 따르면, 본 발명의 조합물 중 치료 유효량의 각 조합 파트너는 동시에 또는 임의의 순서로 순차적으로 투여될 수 있으며, 성분들은 개별적으로 또는 고정 조합물로서 투여될 수 있다. 예를 들어, 본 발명에 따른 증식성 질환의 치료 방법은 (i) 유리 또는 제약상 허용되는 염 형태의 작용제 (a)의 투여 및 (ii) 유리 또는 제약상 허용되는 염 형태의 작용제 (b) (및 임의로 유리 또는 제약상 허용되는 염 형태의 작용제 (c))의 투여를, 동시에 또는 임의의 순서로 순차적으로 연합 치료 유효량, 바람직하게는 상승작용적 유효량으로, 예를 들어 본원에 기재된 양에 상응하는 1일 또는 간헐적 투여량으로 포함할 수 있다. 본 발명의 조합물 중 개별 조합 파트너는 요법의 과정 동안 상이한 시간에 개별적으로, 또는 분할 또는 단일 조합 형태로 공동으로 투여될 수 있다. 따라서, 본 발명은 동시 또는 교차 치료의 모든 이러한 요법을 포괄하는 것으로 이해되어야 하며, 용어 "투여하는"은 이에 따라 해석되어야 한다.

독성 없이 효능을 산출하는 본 발명의 조합물 중 조합 파트너 (a) 및 (b)의 최적 비율, 개별 및 조합 투여량, 및 농도는 표적 부위에 대한 치료제의 유용성의 동역학을 기초로 하며, 통상의 기술자에게 공지된 방법을 사용하여 결정된다.
각 조합 파트너의 유효 투여량은 조합물 중 다른 화합물(들)과 비교시 화합물(들) 중 하나의 보다 빈번한 투여를 필요로 할 수 있다. 따라서, 적절한 투여를 허용하기 위해, 포장된 제약 제품은 화합물의 조합물을 함유하는 하나 이상의 투여 형태, 및 화합물의 조합물 중 하나를 함유하지만 조합물 중 다른 화합물(들)은 함유하지 않는 하나 이상의 투여 형태를 함유할 수 있다.
본 발명의 조합물에 사용되는 조합 파트너가 단일 약물로서 시판되는 바와 같은 형태로 적용되는 경우에, 본원에 달리 언급되지 않는 경우에, 그의 투여량 및 투여 방식은 각 시판 약물의 포장 삽입물에 제공된 정보에 따를 수 있다.

단일 투여 형태를 제조하기 위해 담체 물질과 조합될 수 있는 각 조합 파트너의 양은 치료할 개체 및 특정한 투여 방식에 따라 달라질 것이다. 일부 실시양태에서, 본원에 기재된 바와 같은 작용제의 조합물을 함유하는 단위 투여 형태는 작용제가 단독으로 투여되는 경우에 전형적으로 투여되는 조합물 중의 각 작용제의 양을 함유할 것이다.
투여 빈도는 사용되는 화합물 및 치료 또는 예방할 특정한 상태에 따라 달라질 수 있다. 환자는 일반적으로 치료 또는 예방할 상태에 적합한 검정을 사용하여 치료 유효성에 대해 모니터링될 수 있으며, 이는 통상의 기술자에게 익숙할 것이다.
본 발명은 증식성 질환을 갖는 대상체에게 본 발명의 조합물을 증식성 질환에 대한 연합 치료상 유효한 양으로 투여하는 것을 포함하는, 상기 대상체를 치료하는 방법에 관한 것이다. 특히, 본 발명의 조합물로 치료할 증식성 질환은 흑색종 또는 결장직장암, 특히 B-Raf 돌연변이된 흑색종 또는 결장직장암, 예를 들어 V600 B-Raf 돌연변이된 흑색종 또는 결장직장암이다. 또한, 치료는 수술 또는 방사선요법을 포함할 수 있다.
본 발명은 추가로 증식성 질환, 특히 암의 치료에 사용하기 위한 본 발명의 조합물에 관한 것이다.
본 발명은 추가로 치료제로서의 본 발명의 조합물을, 증식성 질환의 진행 지연 또는 치료를 필요로 하는 대상체에서의 증식성 질환의 진행의 지연 또는 치료에 사용하기 위한 그의 동시, 개별 또는 순차적 투여를 위한 지침서와 함께 포함하는 상업용 패키지를 제공한다.
하기 실시예는 상기 기재된 본 발명을 예시하지만; 이들은 어떠한 방식으로도 본 발명의 범위를 제한하고자 하는 것은 아니다. 본 발명의 제약 조합물의 유익한 효과는 통상의 기술자에게 그 자체로 공지된 다른 시험 모델에 의해 또한 결정될 수 있다.
실시예 1
화합물 A의 감수성 및 화합물 A 및 B의 조합 효능을 화합물 B-감수성 원발성 인간 흑색종 HMEX1906 이종이식편에서 평가하였다. 각 작용제가 이전의 연구에서 종양 정체 또는 퇴행을 유발하였기 때문에, 본 연구의 목적은 본 발명의 조합물로의 치료 후 시간-대-재성장을 연구하는 것이다.
종양을 현탁액 같이 세포주 내로 쵸핑/민싱하였다 (종양을 균질화시킴). 매트리겔 7mL 및 HBSS 1.5mL를 첨가하였다. 매트리겔이 두꺼워질 때까지 현탁액을 손바닥에서 가온하고, 18G 바늘을 사용하여 암컷 누드 마우스의 우측 측복부로 s.c 이식하였다. 치료는 이식 16일 후에 시작하였다.
화합물 A는 Braf 돌연변이체 HMEX1906 이종이식편 모델에서 유의한 항종양 활성을 나타내며, 150mg/kg,qd에서 정체 T/C=7% 및 250mg/kg,qd 용량에서 76% 퇴행을 나타내었다. 화합물 B는 3mg/kg,bid 용량에서 57% 퇴행을 나타내었다. 본 발명의 조합물은 연구 제46일 [투여 31일]에 91% 퇴행을 나타내었다.
비히클 군 및 화합물 A [150mg/kg] 단일 작용제를 31일 동안 투여하였고, 마지막 투여 후 종결하였다. 화합물 A [250mg/kg], 화합물 B 단일 작용제 및 본 발명의 조합물을 45일 동안 투여하고, 마지막 투여 [연구 제59일] 후 종양 성장 지연에 대해 모니터링하였다.
치료된 동물에서는 어떠한 유의한 체중 감소도 관찰되지 않았다.
실시예 2
목적: 원발성 인간 흑색종 (BRAF 돌연변이체) HMEX2613 이종이식편에서 화합물 A, 화합물 B, 및 화합물 A 및 B의 조합물에 대한 반응 지속성을 평가하였다. 본 연구의 목적은 단일 작용제로 및 본 발명의 조합물로 치료한 후 시간-대-500 mm3까지의 재성장을 조사하는 것이다.
종양 이종이식편을 실시예 1에 기재된 것과 유사한 방식으로 암컷 누드 마우스에 이식하였다. 치료를 이식 19일 후에 시작하고, 비히클 및 화합물 A 단일 작용제로 투여된 군에서는 11일 동안 또는 화합물 B 단일 작용제 및 본 발명의 조합물로 투여된 군에서는 18일 동안 계속하였다.
종양 이종이식편을 실시예 1에 기재된 것과 유사한 방식으로 암컷 누드 마우스에 이식하였다. 치료를 이식 19일 후에 시작하고, 비히클 및 화합물 A 단일 작용제로 투여된 군에서는 11일 동안 또는 화합물 B 단일 작용제 및 본 발명의 조합물로 투여된 군에서는 18일 동안 계속하였다.
결과는 도 1에 그래프로 도시하였다.
화합물 A는 T/C 45%를 갖는 Braf 돌연변이체 HMEX2613 이종이식편 모델에서 중간 정도의 항종양 활성을 나타내었다.
화합물 B 및 본 발명의 조합물은 완전 종양 퇴행을 나타내었다.
비히클 군 및 화합물 A 단일 작용제를 11일 동안 투여하였고, 이어서 인도적으로 안락사시켰다. 화합물 B 단일 작용제 및 화합물 B/화합물 A 조합물을 18일 동안 투여하고, 마지막 투여 후에 종양 재성장에 대해 모니터링하였다.
화합물 B 단일 작용제는 마지막 투여 5일 후 종양 재성장 및 연구 종료시 37% 무종양 동물을 나타내었다 [8개 종양 중 5개가 재성장함].
본 발명의 조합물은 마지막 투여 26일 후 종양 지연 및 연구 종료시 62% 무종양 동물을 나타내었다 [8개 종양 중 3개가 재성장함].
치료된 동물에서는 어떠한 유의한 체중 감소도 관찰되지 않았다.
실시예 3
목적: 5 mg/kg 화합물 B의 존재 하에 성장하고 이에 대해 내성이 있는 HMEX1906 원발성 흑색종 모델 (HMEX1906-R5)에서 본 발명의 조합물의 효능을 평가하는 것
약물 제제: 화합물 A는 0.5%MC/0.5% 트윈80 중에서 제제화하고, 화합물 B는 20%PEG300/3%ETPGS 중에 제제화하였다.
종양 이종이식편을 실시예 1에 기재된 바와 유사한 방식으로 암컷 누드 마우스에 이식하였다. 마우스는 종양 덩어리 이식 후에 화합물 B를 투여받지 않았다.
평균 종양 부피 266 mm3 및 평균 체중 25 그램을 갖는 마우스를 이식 18일 후에 하기 군으로 할당하였다.
군: 10마리 마우스/군, 경로 PO, 투여 부피 0.2 mL
군 1: 비히클, 0 mg/kg bidx14
군 2: 화합물 A, 250 mg/kg qdx21
군 3: 화합물 B, 5 mg/kg bidx21
군 4: 화합물 A 250mg/kg qd x21 + 화합물 B 5 mg/kg bid x21
결과:
실시예 4
물질 및 방법 - 화합물 원액을 DMSO 중에 최종 농도 10mM로 제조하였다. 작업 원액을 적절한 세포 배양 배지 중에 3배 증분으로 연속 희석하여 화합물 B 및 D에 대해 2.7μM 내지 1.2nM, 및 화합물 A에 대해 10μM 내지 4.6nM 범위의 최종 검정 농도를 달성하였다.
세포주, 세포 배양, 세포 생존율 측정 - A-375 및 WM-266-4 세포를 아메리칸 타입 컬쳐 콜렉션(American Type Culture Collection: ATCC)으로부터 구입하였다. A-375 세포는 DMEM 배지 (ATCC) 중에 배양하고, WM-266-4 세포는 EMEM 배지 (ATCC) 중에 배양하였으며, 이들 둘 다 10% 태아 소 혈청 (깁코(Gibco))으로 보충하고, 37℃/5% CO2에서 인큐베이션하였다. 흔히 발생하는 내성 대립유전자 지표를 발현하도록 조작된 세포주를 노파르티스-에머리빌(Novartis-Emeryville)로부터 입수하였다. 이들 내성 모델은 돌연변이체 MEK1P124L, 말단절단된 p61-BRAFV600E 또는 돌연변이체 NRASQ61K를 발현하는 A-375 세포, 및 돌연변이체 MEK1C121S, 말단절단된 p61-BRAFV600E 또는 돌연변이체 NRASQ61K를 발현하는 WM-266-4 세포를 포함한다. 이들 세포를 선택 마커 G418를 함유한 적절한 모 배지 중에 및 5uM LFE158 (MEK 돌연변이체) 또는 LIH720 (말단절단된 p61-BRAFV600E)의 존재 하에 배양하였다.
플레이트 레이아웃, 세포 분배 및 화합물 첨가 - 스크리닝을 위해, 세포를 8-채널 스탠다드 카세트가 장착된 멀티드롭 콤비(MultiDrop Combi) (써모-피셔(Thermo-Fisher))를 사용하여 웰당 500 (A-375) 또는 750 (WM-266-4) 세포 밀도로 384-웰 플레이트 (써모 사이언티픽(Thermo Scientific), cat# 4332) 내 배지 80ul 중에 시딩하였다. 전 웰에 걸친 세포의 균등 분배를 촉진하기 위해, 세포를 1000 RPM에서 짧게 원심분리하고, 실온에서 30분 동안 인큐베이션하였다. 모든 플레이트를 37℃, 5% CO2에서 24시간 동안 인큐베이션한 후에 화합물을 첨가하였다. 화합물 원액을 적절한 배양 배지 중에 신선하게 준비하고, 200nl 핀 도구를 갖춘 PAA 로봇을 사용하여 첨가하였다. 최소 3개의 반복 웰에서, 72시간 후의 단일 작용제 및 조합물 효과를 제조업체의 프로토콜에 따라 셀 타이터 글로(Cell Titer Glo) (프로메가(Promega))를 통한 세포 ATP 수준의 정량화에 의해 및 현미경검사 영상화에 의해 둘 다 평가하였다. 영상화를 위해, 세포를 플레이트에 고정시키고, 제어된 분배 속도를 갖는 웰메이트(WellMate) 분배기를 통해 PBS 중 10% PFA, 0.3% TX-100의 용액과 함께 투과시켰다. 세포 핵을 훽스트(Hoechst) 33342 (H3570, 인비트로젠(Invitrogen))로 염색하고, 모든 필요한 세척 단계를 바이오텍(BioTek) 세척액에 의해 수행하였다.
자동화 영상 분석 - 인셀 애널라이저(InCell Analyzer) 2000 (지이 헬스케어(GE Healthcare), 28-9534-63)로부터의 영상은 TIFF 포맷으로 있고, 2048x2048 픽셀의 크기를 가졌으며, 이는 384-웰 플레이트의 전 웰을 캡쳐한 것이다. 자동화 영상 분석 파이프라인은 오픈-소스, 통계 프로그래밍 언어 R에서의 주문-제작 스크립트 및 바이오컨덕터 패키지 이비이미지(BioConductor package EBImage)의 함수를 사용하여 확립하였다. 목적은 세포 생존율에 대한 근사치로서 웰당 생존 핵 (세포)의 수를 정량화하는 것이다. 파이프라인은 하기 7 단계로 구성되었다: (I.) 강도 피크의 수를 감소시키기 위한 이미지의 평활화, (II.) 포그라운드 (신호)를 백그라운드 (잡음)으로부터 분리하기 위한 임계 함수의 적용, (III.) 핵에 대한 시드로서 작용하는, 포그라운드에서의 국부 최대치의 확인, (IV.) 아주 근접한 국부 최대치의 필터링, (V.) 남아있는 국부 최대치로부터 핵의 확대, (VI.) 및 확대된 핵으로부터 목적하는 특징 (핵의 수, 크기 특징 및 강도 특징)의 추출. 마지막 단계 (VII.)로서, 파편 (예를 들어 단편화 핵)을 계수로부터 제외하기 위해, DMSO- 및 스타우로스포린-처리 웰에서 확인된 목적물을 사용하여 각각 생존 및 단편화 핵에 대한 특징 분포를 수득하였다. 이들을 사용하여 생존 및 단편화 핵을 구별하는 컷-오프를 설정하였다. 단편화 핵의 수를 확인된 목적물의 총 수에서 빼고, 결과는 그 웰에 대한 최종 계수로서 보고하였다.
데이터 정규화 - 데이터는 각 처리 (화합물) 조건에 대한 삼중 측정, DMSO-처리 웰의 42회 반복 측정, 스타우로스포린-처리 웰의 이중 측정을 포함하였다. 데이터를 DMSO 측정의 중앙값에 대해 정규화시키고, 삼중의 중앙값을 계산함으로써 요약하였다. 데이터를 챌리스(Chalice) 내로 입력하여 화합물 상승작용을 계산하였다.
