도 2는 자성입자에 의해 핵산추출 공정을 나타낸 개념도이다.
도 3은 TGF-β1 유전자를 SNP 검출하기 위한 일구체예에 따른 공정도이다.
도 4는 본 발명의 SNP 판별 방법에 의해 784명의 혈액 시료에서 TGF-β1의 SNP를 판별한 결과를 나타낸 도이다.
도 5는 본 발명의 일구체예에 따른 단일염기 다형성 판별 방법의 각 단계를 나타낸 개념도이다.

Claims (18)
- 단일염기 다형성(single nucleotide polymorphism, SNP) 판별 자동화 장치에 있어서,
제1반응용기, 반응용기 지지대, 및 반응용기 하부에 ON/OFF 방식으로 자력을 인가하는 수단을 구비하며, (+) 전하를 띠는 자성입자를 사용하는 DNA 분리 공정 및 제1작용기로 수식된 PCR 프라이머를 사용하는 PCR 공정을 수행하는 제1반응부;
제1반응부로부터 제1작용기 수식 폴리뉴클레오티드 함유 PCR 반응 산물을 제2반응부로 이동시키는 수단; 및
제2반응용기, 반응용기 지지대, 및 반응용기 하부에 ON/OFF 방식으로 자력을 인가하는 수단을 구비하며, 제1작용기 수식 폴리뉴클레오티드를 제1작용기에 결합하는 제2작용기로 수식된 자성입자에 고정화하는 공정, 제1작용기 수식 폴리뉴클레오티드의 단일가닥 (single stranded) 화 공정 및 SNP 판별용 프로브로 혼성화(hybridization)하는 공정을 수행하는 제2반응부
를 구비한 것이 특징인 단일염기 다형성 판별 자동화 장치.
- 제1항에 있어서, 제1반응용기에는 (+) 전하를 띠는 자성입자가 수용되고, 제2반응용기에는 제1작용기에 결합하는 제2작용기로 수식된 자성입자가 수용되어 있는 것이 특징인 단일염기 다형성 판별 자동화 장치.
- 제1항에 있어서, (i) (+) 전하를 띠는 자성입자가 구비되어 있는 제1반응용기에서 제1반응용기 내 수용되어 있는 시료로부터 나온 (-) 전하를 띠는 핵산과 상기 자성입자를 결합시킨 후 자력을 인가하여 결합형(B)/유리형(F) 분리로 핵산을 분리하고,
(ii) 제1반응용기에서 제1작용기로 수식된 PCR 프라이머를 사용하여 (-) 전하를 띠는 DNA가 (+) 전하를 띠는 자성입자에 붙어있는 상태에서 PCR을 진행하여, 한쌍의 프라이머로 정의되는 DNA 부분을 증폭하고,
(iii) PCR로 증폭된 DNA를 제1반응용기로부터 제2반응용기로 이동시키고,
(iv) 제2반응용기에서 제1작용기에 결합하는 제2작용기로 수식된 자성입자를 사용하여 제1작용기로 수식된 PCR 프라이머를 사용하여 증폭된 DNA를 상기 자성입자에 고정화시키고,
(v) 제2반응용기에서 DNA를 단일가닥(single stranded)화시키고,
(vi) 제2반응용기에서 1종 이상의 SNP 판별용 프로브와 자성입자에 고정화되어 있는 DNA 단일가닥과 혼성화(hybridization)하도록 프로그램되어 있는 것이 특징인 단일염기 다형성 판별 자동화 장치.
- 제1항에 있어서, 제1반응용기에 자력을 인가하여 (-) 전하를 띠는 핵산과 결합되어 있는 (+) 전하를 띠는 자성입자를 반응용기의 특정부위에 모으고,
DNA 단일가닥에 혼성화된 SNP 프로브를 분리하는 조건하에 제2반응용기에 자력을 인가하여 자성입자를 반응용기의 특정부위에 모으면서 자성입자로부터 SNP 판별용 프로브를 분리시키는 것이 특징인 단일염기 다형성 판별 자동화 장치.
- 제1항에 있어서, 지지대 또는 반응용기의 온도를 조절하는 가온냉각장치 및 선택적으로 온도 센서를 더 구비한 것이 특징인 단일염기 다형성 판별 자동화 장치.
- 제1항에 있어서, SNP 판별용 프로브는 표지물질로 수식된 것이 특징인 단일염기 다형성 판별 자동화 장치.
- 제1항에 있어서, 하나 이상의 파이펫(pipette)이 장착된 이동할 수 있는 헤드부를 추가로 구비한 것이 특징인 단일염기 다형성 판별 자동화 장치.
- 제7항에 있어서, 파이펫(pipette)은 혈액 용혈제, PCR용 혼합액, 표지물질로 수식된 1종 이상의 SNP 프로브로 구성된 군에서 선택된 시약을 제1반응용기 또는 제2반응용기에 운반하는 것이 특징인 단일염기 다형성 판별 자동화 장치.
- 제7항에 있어서, 파이펫팅에 의해 교반을 수행하여 반응용기 내 반응을 촉진하는 것이 특징인 단일염기 다형성 판별 자동화 장치.
- 제7항에 있어서, SNP 프로브 또는 표지물질 검출기가 헤드부에 장착되어 있는 것이 특징인 단일염기 다형성 판별 자동화 장치.
