Abstract
Description
도 2는 형질전환된 벼 캘러스의 인간 트립신(hTypsin) 유전자의 mRNA 발현 수준을 PCR로 확인한 결과이다.
도 3은 형질전환 벼 캘러스 세포주의 재조합 인간 트립신 단백질의 발현을 (A) SDS-PAGE, (B) 웨스턴 블럿 및 (C) ELISA를 통해 확인한 결과이다. M, 마커(PageRuler TM Protein Ladder Fermentas); PC, 양성 대조구(밀 배아 발현 시스템을 이용하여 인공적으로 합성한 인간 트립신); NC, 음성 대조구(형질전환된 벼 캘러스를 포함하지 않은 배지); 숫자 3, 5, 9, 11, 22 및 25, 형질전환된 벼 캘러스 고유 번호.
Claims (7)
- 서열번호 1의 염기서열로 이루어진 벼 유래의 α-아밀라제(RAmy3D) 프로모터 및 서열번호 2의 염기서열로 이루어진 인간 트립신 단백질 코딩 유전자가 작동가능하게 연결된 벼 형질전환용 재조합 벡터.
- 제1항의 재조합 벡터로 형질전환되어 재조합 인간 트립신을 생산하는 벼 캘러스.
- 제2항에 있어서, 상기 벼 캘러스는 pJKS133-22 (기탁번호: KCTC 18276P)인 것을 특징으로 하는 벼 캘러스.
- (a) 서열번호 1의 염기서열로 이루어진 벼 유래의 α-아밀라제(RAmy3D) 프로모터 및 서열번호 2의 염기서열로 이루어진 인간 트립신 단백질 코딩 유전자가 작동가능하게 연결된 벼 형질전환용 재조합 벡터를 벼 캘러스에 형질전환하는 단계; 및
(b) 상기 (a) 단계의 형질전환된 벼 캘러스를 현탁 배양 배지에서 배양하여 인간 트립신을 생산하는 단계를 포함하는 재조합 인간 트립신의 생산방법. - 제4항에 있어서, 상기 (b) 단계의 벼 캘러스의 배양은 당 결핍 상태에서 이루어지는 것을 특징으로 하는 재조합 인간 트립신의 생산방법.
- 제4항에 있어서, 상기 형질전환된 벼 캘러스는 pJKS133-22 (기탁번호:KCTC 18276P)인 것을 특징으로 하는 재조합 인간 트립신의 생산방법.
