Abstract
Description
도 2는 비브리오 파라해모라이티쿠스 표면 결합 DNA 앱타머의 제작을 위한 SELEX 과정 중 각 라운드에서 회수된 비브리오 파라해모라이티쿠스 표면 결합 DNA 앱타머의 농도를 나노드랍을 이용하여 정량적으로 측정한 결과이다.
도 3은 비브리오 파라해모라이티쿠스 표면 결합 DNA 앱타머 제작을 위한 SELEX 과정 후 선별된 라운드에서 확보한 비브리오 파라해모라이티쿠스 표면 결합 DNA 앱타머 후보군들의 1차 (a), 2차 (b) 용리 액을 나노드랍을 이용하여 정량적으로 측정한 결과이다.
도 4는 m-fold 프로그램을 이용하여 비브리오 파라해모라이티쿠스 표면 결합 DNA 앱타머 VPCA-01과 VPCA-02의 예상 구조를 나타낸 것이다.
도 5는 비브리오 파라해모라이티쿠스 표면 결합 DNA 앱타머를 스트렙트아비딘이 코팅된 Sensor chip SA의 표면에 고정시키고 비브리오 파라해모라이티쿠스 생균을 결합시키는 과정을 나타낸 것이다.

Claims (7)
- 비브리오 파라해모라이티쿠스(Vibrio parahaemolyticus) 생균의 표면에 특이적으로 결합하는 서열번호 11 내지 13 중 하나의 염기서열을 갖는 올리고뉴클레오타이드인 것을 특징으로 하는 DNA 앱타머(aptamer).
- 하기 단계를 포함하는 것을 특징으로 하는 제 1 항에 따른 DNA 앱타머를 선별하는 방법:
(ⅰ) PCR 기법을 이용하여 DNA 앱타머를 증폭하고, ssDNA 앱타머를 제조하는 단계;
(ⅱ) 비브리오 파라해모라이티쿠스를 배양하고 세척하는 단계; 및
(ⅲ) 비브리오 파라해모라이티쿠스 생균을 이용한 Live Cell SELEX 기법을 통해 비브리오 파라해모라이티쿠스 생균 표면에 특이적으로 결합하는 DNA 앱타머를 선별하는 단계.
- 제 1 항에 따른 DNA 앱타머를 유효성분으로 포함하는 비브리오 파라해모라이티쿠스(Vibrio parahaemolyticus) 생균의 검출용 조성물.
- 제 1 항에 따른 DNA 앱타머를 유효성분으로 포함하는 비브리오 파라해모라이티쿠스(Vibrio parahaemolyticus) 생균의 검출용 키트.
- 제 4 항에 있어서,
상기 키트는 DNA 앱타머가 칩상에 고정화된 칩 형태인 것을 특징으로 하는 키트.
- 제 4 항에 있어서,
상기 키트는 DNA 앱타머가 기판 상에 고정화된 마이크로어레이 형태인 것을 특징으로 하는 키트.
- 하기 단계를 포함하는 것을 특징으로 하는 비브리오 파라해모라이티쿠스(Vibrio parahaemolyticus) 생균의 검출 방법:
(a) 비브리오 파라해모라이티쿠스 생균을 함유하는 검사 시료와 제 1 항에 따른 DNA 앱타머를 접촉시키는 단계; 및
(b) 상기 DNA 앱타머에 결합된 비브리오 파라해모라이티쿠스 생균을 확인하는 단계.
