Abstract
Description
도 2는 본 발명의 히스티딘 태그(his-tag)를 포함하는 청색 형광단백질 his-mBFP의 염기서열(A) 및 아미노산 서열(B)을 나타낸 것이며,
도 3은 기존의 NADP(H) 분석방법(A)과 본 발명에 따른 NADP(H) 분석방법(B)을 비교하여 보여주는 도면이고,
도 4는 본 발명의 히스티딘 태그(his-tag)를 포함하는 청색 형광단백질 mBFP 발현벡터의 모식도이며,
(A: pQE30-mBFP, B: pTrc99A-his-mBFP)
도 5는 SDS-PAGE를 이용하여 친화 크로마토그래프로 분리된 본 발명의 히스티딘 태그(his-tag)를 포함하는 청색 형광단백질 his-mBFP의 순도를 분석한 도면이고,
(Lane 1, size marker; lane 2, cibacron blue 수지로 분리한 his-mBFP; lane 3, Ni-NTA 로 분리한 his-mBFP; lane 4, cibacron Blue 수지와 Ni-NTA 수지로 연속 분리한 his-mBFP)
도 6은 야생형의 청색 형광단백질 mBFP과 본 발명에 따른 히스티딘 태그(his-tag)를 포함하는 청색 형광단백질 his-mBFP의 형광 변화를 비교한 도면이며,
도 7은 온도 변화에 따른 본 발명의 히스티딘 태그(his-tag)를 포함하는 청색 형광단백질 his-mBFP의 형광 변화를 측정한 결과이고,
도 8은 완충액의 pH 변화에 따른 본 발명의 히스티딘 태그(his-tag)를 포함하는 청색 형광단백질 his-mBFP의 형광 변화를 측정한 결과이며,
도 9는 반응 시간에 따른 본 발명의 히스티딘 태그(his-tag)를 포함하는 청색 형광단백질 his-mBFP의 형광 변화를 측정한 결과이고,
도 10은 본 발명의 히스티딘 태그(his-tag)를 포함하는 청색 형광단백질 his-mBFP의 농도에 따른 형광 변화를 측정한 결과이며,
도 11은 계면활성제(SDS) 첨가에 따른 본 발명의 히스티딘 태그(his-tag)를 포함하는 청색 형광단백질 his-mBFP의 형광 변화를 측정한 결과이고,
도 12는 보관에 따른 본 발명의 히스티딘 태그(his-tag)를 포함하는 청색 형광단백질 his-mBFP의 형광활성 유지 정도를 확인한 결과이며,
도 13은 본 발명의 히스티딘 태그(his-tag)를 포함하는 청색 형광단백질 his-mBFP을 이용한 NAD(P)H 의존적 효소의 활성을 확인한 결과이고,
도 14는 잠재적 오염물질(유기/염류 등)로 인해 성장이 유도되어 과발현된 본 발명의 히스티딘 태그(his-tag)를 포함하는 청색 형광단백질 his-mBFP를 포함하는 세포의 형광 이미지이다.
Claims (12)
- 서열번호 3에 기재된 아미노산 서열로 이루어지는 메타게놈 유래의 청색 형광단백질 mBFP(metagenome-derived blue fluorescent protein)와 NADP(H)의 결합으로 mBFP의 발색에 의한 형광의 변화를 측정하는,
생체내외 시료의 NADP(H) 정량법. - 제 1항에 있어서,
상기 형광의 변화는 시료 내 존재하는 NADPH에 상기 mBFP가 직접 결합하여 형광을 증가 또는 감소시키고, 상기 증가 또는 감소된 형광의 세기로부터 NADP(H)의 농도 또는 NADP(H) 의존적 효소의 활성을 측정하는 것을 특징으로 하는 정량법. - 삭제

상기 효소는 NADP(H) 의존적 탈수소효소(dehydrogenase) 또는 산화환원효소(oxidoreductase)인 것을 특징으로 하는 정량법. - 제 1항에 있어서,
상기 정량법은 시료와 mBFP의 반응시 계면활성제를 더 추가하여 혼합하는 것을 특징으로 하는 정량법. - 제 7항에 있어서,
상기 계면활성제는 SDS, Na-deoxycholate, CTAB, DEDAB, 및 CHAPS로터 선택되는 1종 이상인 것을 특징으로 하는 정량법. - 히스티딘 태그(his-tag)가 융합된 메타게놈 유래의 청색 형광단백질 mBFP(metagenome-derived blue fluorescent protein) 포함하는 효소 측정용 단백질을 포함하는 NADPH 회수용 키트.
