Abstract
Description
도 2는 고추 녹광 품종의 각 기관에서 CaXEGIP1 유전자의 발현 결과를 나타낸 도면이고,
도 3은 고추 녹광 품종에 고추 세균성 점무늬병균(Xanthomonas campestris pv. vesicatoria)의 병원성 및 비병원성 균주를 접종하였을 때 접종된 잎과 접종되지 않은 잎에서의 CaXEGIP1 유전자의 발현 유도 결과를 나타낸 도면이고,
도 4는 살리실산(Salicylic acid)을 처리한 후 시간별로 CaXEGIP1 유전자의 발현 유도 결과를 나타낸 도면이고,
도 5는 토끼에서 제작된 CaXEGIP1 단백질의 항체를 ELISA test를 이용하여 항원-항체반응을 관찰한 결과를 나타낸 도면이고,
도 6은 고추 녹광품종에 고추 세균성 점무늬병균(Xanthomonas campestris pv. vesicatoria)의 병원성 및 비병원성 균주를 접종하였을 때 접종된 잎과 접종되지 않은 잎에서의 CaXEGIP1 단백질의 항체를 이용하여 웨스턴 블롯팅을 수행함으로써 CaXEGIP1 단백질의 유도 결과를 나타낸 도면이고,
도 7은 그린 플루오레슨트 프로테인(Green Fluoresent Protein, GFP)이 태깅(tagging)된 CaXEGIP1 단백질을 양파 (Allium cepa)의 표피세포에서 과발현시킨 후 공초점현미경 [confocal laser scanning microscope (CLSM)]을 이용하여 CaXEGIP1 단백질이 식물 세포벽(cell wall)을 포함한 세포바깥쪽 (extracellular region)에 위치하고 있음 결과를 나타낸 도면이고,
도 8은 대장균 (Escherichia coli)에서 CaXEGIP1 유전자를 과발현하고 단백질을 순화하여 SDS-PAGE상에서 확인한 결과를 나타낸 도면이고 (Lane 1: CaXEGIP1을 발현하지 않는 E. coli의 단백질, Lane 2: CaXEGIP1을 과발현하는 E. coli의 단백질, Lane 3: 최종적으로 순화된 CaXEGIP1 단백질),
도 9는 순화된 CaXEGIP1 단백질을 이용하여 자이로글루칸 분해 효소 (xyloglucanase)를 억제하는 것을 확인한 결과를 나타낸 도면이고,
도 10은 CaXEGIP1 유전자를 이용한 재조합벡터 pBIN35S:CaXEGIP1로 형질전환된 아그로박테리움 튜메파시엔스(Agrobacterium tumefaciens strain GV3101) 균주를 이용하여 고추 식물에서 CaXEGIP1 유전자를 일시적으로 과발현시킨 후 RT-PCR을 이용하여 유전자의 발현을 확인한 결과이고,
도 11은 CaXEGIP1 유전자를 이용한 재조합벡터 pBIN35S:CaXEGIP1로 형질전환된 아그로박테리움 튜메파시엔스(Agrobacterium tumefaciens strain GV3101) 균주를 이용하여 고추 식물에서 CaXEGIP1 유전자를 일시적으로 과발현시킨 후 CaXEGIP1 단백질의 항체를 이용하여 웨스턴 블롯팅을 수행함으로써 CaXEGIP1 단백질의 유도 결과를 나타낸 도면이고,
도 12는 CaXEGIP1 유전자를 이용한 재조합벡터 pBIN35S:CaXEGIP1로 형질전환된 아그로박테리움 튜메파시엔스(Agrobacterium tumefaciens strain GV3101) 균주를 이용하여 고추 식물에서 CaXEGIP1 단백질을 과발현시킨 부위에서 과민성 세포사멸이 나타나는 것을 관찰한 결과를 나타낸 도면이고,
도 13은 고추 식물에서 CaXEGIP1 단백질을 과발현시킨 부위에서 과민성 세포사멸을 일으키는 신호전달 물질인 활성산소가 축적되는 것을 DAB 염색(3,3'-diaminobenzidine staining)을 이용하여 관찰한 결과를 나타낸 도면이고,
도 14는 VIGS를 통해 CaXEGIP1 유전자의 발현이 억제된 고추식물 (TRV:CaXEGIP1)이 대조구(TRV:00)에 비해 생장이 증가되는 것을 관찰한 결과를 나타낸 도면이고,
도 15는 VIGS를 통해 CaXEGIP1 유전자의 발현이 억제된 고추식물 (TRV:CaXEGIP1)이 대조구(TRV:00)에 비해 병원성 및 비병원성 고추 세균성 점무늬병균(Xanthomonas campestris pv. vesicatoria) 접종 후 CaXEGIP1 유전자와 방어관련 CaBPR1 (Capsicum annuum basic pathogenesis-related 1) 및 CaDEF1 (Capsicum annuum defensin 1) 유전자의 발현이 억제되는 것을 관찰한 결과를 나타낸 도면이고,
도 16은 VIGS를 통해 CaXEGIP1 유전자의 발현이 억제된 고추식물 (TRV:CaXEGIP1)이 대조구(TRV:00)에 비해 병원성 및 비병원성 고추 세균성 점무늬병균(Xanthomonas campestris pv. vesicatoria)의 생장이 증가되어 감수성이 증진되는 결과를 나타낸 도면이고,
도 17은 CaXEGIP1 유전자를 이용한 재조합 벡터 pTA7002:CaXEGIP1로 형질전환된 아그로박테리움 튜메파시엔스(Agrobacterium tumefaciens strain GV3101) 균주를 이용하여 형질전환 애기장대(Arabidopsis thaliana ecotype Col-0) 식물을 제작한 뒤 데사메타손(dexamethasone, DEX) 처리에 의해 특이적으로 CaXEGIP1 유전자의 발현이 증가되는 것을 관찰한 결과를 나타낸 도면이고,
도 18은 pTA7002:CaXEGIP1로 형질전환된 애기장대 식물 (DEX:CaXEGIP1)에서 CaXEGIP1 단백질의 항체를 이용하여 웨스턴 블롯팅을 수행함으로써 DEX 처리에 의해 CaXEGIP1 단백질의 유도되는 것을 관찰한 결과를 나타낸 도면이고,

도 21은 pTA7002:CaXEGIP1로 형질전환된 애기장대 식물 (DEX:CaXEGIP1)에서 DEX 처리에 의해 뿌리와 배축(hypocotyl)의 세포생장에 억제가 일어나는 것을 관찰한 결과를 나타낸 도면이고,
도 22는 pTA7002:CaXEGIP1로 형질전환된 애기장대 식물 (DEX:CaXEGIP1)에서 DEX 처리에 의해 CaXEGIP1 단백질이 과량으로 축적될 경우 병원성 (Pst) 및 비병원성 [Pst (avrRpm1)] 슈도모나스 시링게 패소바 토마토 DC3000 [Pseudomonas syringae pv. tomato DC3000 (Pst DC3000)]을 접종한 식물에서 세포사멸이 DEX를 처리하지 않은 대조구 식물에 비해 강하게 일어나는 것을 관찰한 결과를 나타낸 도면이고,
도 23은 CaXEGIP1 유전자를 이용한 재조합 벡터 pBIN35S:CaXEGIP1로 형질전환된 아그로박테리움 튜메파시엔스(Agrobacterium tumefaciens strain GV3101) 균주를 이용하여 담배 식물 (Nicotiana benthamiana)에서 CaXEGIP1 단백질을 과발현시킨 부위에서 과민성 세포사멸이 나타나는 것을 관찰한 결과를 나타낸 도면이다.
Claims (9)
- 식물병 저항성 관련 유전자로서 고추 유래 자일로글루칸 분해효소 억제 단백질을 암호화하는 하기 (a) 또는 (b)의 분리된 유전자(CaXEGIP1):
(a) 서열번호 2의 아미노산 서열을 갖는 단백질을 코딩하는 유전자;
(b) 서열번호 1의 염기서열을 갖는 유전자. - 서열번호 2의 아미노산 서열을 갖는 분리된 단백질(CaXEGIP1).
- 제1항 기재의 유전자를 포함하는 재조합 벡터.
- 제3항에 있어서, 상기 재조합 벡터는 pBIN35S:CaXEGIP1, pTA7002:CaXEGIP1, 또는 pTRV2:CaXEGIP1 인 것을 특징으로 하는 재조합 벡터.
- 제1항 기재의 유전자 또는 제3항 기재의 재조합 벡터에 의해 형질전환된 식물체.
- 식물체로부터 mRNA 분리하여 제1항 기재의 CaXEGIP1 유전자의 차별적 발현을 확인하는 단계를 포함하는 식물체의 병 또는 화학물질 저항성 탐색방법.
- 고추 유래 자일로글루칸 분해효소 억제 단백질을 암호화하는 유전자 CaXEGIP1의 발현을 억제하여 식물체의 생장을 촉진하는 방법.
- 삭제
- 서열번호 2의 아미노산 서열 중 219~231 번째의 아미노산 서열(NIYNPKKIDISKD, 서열번호 3)을 항원(epitope)으로 이용하여 제작된 서열번호 2의 아미노산 서열을 갖는 분리된 단백질(CaXEGIP1)을 특이적으로 검출할 수 있는 항체(α-CaXEGIP1).
