Abstract
Description
도 2는 억새(Miscanthus sinensis)의 미성숙 화기로부터 캘러스 유도에 미치는 미성숙 화기 길이에 따른 효과를 나타낸 것이다.
도 3은 억새(Miscanthus sinensis)의 미성숙 화기로부터 캘러스 유도에 미치는 2,4-D 및 벤질아데닌 조합처리의 효과를 나타낸 것이다.
도 4는 억새(Miscanthus sinensis)의 미성숙 화기로부터 캘러스 유도에 미치는 배지 고형제의 종류 및 농도에 따른 효과를 나타낸 것이다.
도 5는 억새(Miscanthus sinensis)의 미성숙 화기 유래 캘러스로부터 신초의 재생에 미치는 캘러스 유도배지의 영향을 나타낸 것이다.
도 6은 억새(Miscanthus sinensis)의 미성숙 화기 유래 캘러스로부터 신초의 재생에 미치는 벤질아데닌 및 나프탈렌 아세트산 조합처리의 효과를 나타낸 것이다.
도 7은 억새(Miscanthus sinensis)의 미성숙 화기 유래 캘러스로부터 신초의 재생에 미치는 피타겔 농도의 효과를 나타낸 것이다.
도 8은 억새(Miscanthus sinensis)의 미성숙 화기로부터 기관형성을 통한 식물체 재생을 나타낸 것이다. 억새의 미성숙 화기(A)를 절단하여 캘러스 유도배지에 치상하였다(B). 약 2주 후부터 미성숙 화기에서 캘러스가 유도되기 시작하였으며, 4주 까지 유도된 캘러스(C)를 클러스터에서 분리하여 증식시킨 후, 신초 유도배지에 치상하였다. 약 2~4주 후 캘러스가 증식되면서 신초가 발달하였다(D 내지 F). 재생된 신초를 MS 배지에서 신장 및 발근시킨 후, 온실에서 활착하였다(G 내지 I).
Claims (12)
- (1) MS 배지, 2,4-D (dichlorophenoxyacetic acid), 벤질아데닌, 수크로스 및 배지 고형제를 함유하는 배지에서 억새(Miscanthus sinensis)의 미성숙 화기로부터 캘러스를 유도하는 단계;
(2) MS 배지, 2,4-D (dichlorophenoxyacetic acid), 벤질아데닌, 수크로스 및 배지 고형제를 함유하는 배지에서 미성숙 화기로부터 유도된 캘러스를 증식하는 단계;
(3) MS 배지, 나프탈렌 아세트산, 벤질아데닌, 수크로스 및 배지 고형제를 함유하는 배지에서 상기 증식된 캘러스로부터 신초를 재생하는 단계; 및
(4) MS 배지, 수크로스 및 배지 고형제를 함유하는 배지에서 상기 재생된 신초를 신장 및 발근하는 단계를 포함하는 억새의 미성숙 화기를 이용한 억새 식물체의 대량 생산 방법. - 제1항에 있어서, 상기 미성숙 화기의 길이는 1~20cm인 것을 특징으로 하는 대량 생산 방법.
- 제1항에 있어서, 상기 2,4-D 농도는 1~15 mg/L인 것을 특징으로 하는 대량 생산 방법.
- 제1항에 있어서, 상기 나프탈렌 아세트산 농도는 0.5~1.5 mg/L인 것을 특징으로 하는 대량 생산 방법.
- 제1항에 있어서, 상기 (1) 단계와 (2) 단계의 벤질아데닌 농도는 0.05~0.2 mg/L인 것을 특징으로 하는 대량 생산 방법.
- 제1항에 있어서, 상기 (3) 단계의 벤질아데닌 농도는 0.1~10 mg/L인 것을 특징으로 하는 대량 생산 방법.
- 제1항에 있어서, 상기 수크로스 농도는 20~40 g/L인 것을 특징으로 하는 대량 생산 방법.
- 제1항에 있어서, 상기 배지 고형제는 젤란 검(gellan gum) 또는 한천인 것을 특징으로 하는 대량 생산 방법.
- 제8항에 있어서, 상기 젤란 검 및 한천 농도는 각각 2~3 g/L 및 5~10 g/L인 것을 특징으로 하는 대량 생산 방법.
- (1) MS 배지, 2,4-D (dichlorophenoxyacetic acid) 1~15 mg/L, 벤질아데닌 0.05~0.2 mg/L, 수크로스 20~40 g/L 및 젤란 검 2~3 g/L 또는 한천 5~10 g/L를 함유하는 배지에서 억새(Miscanthus sinensis)의 미성숙 화기로부터 캘러스를 유도하는 단계;
(2) MS 배지, 2,4-D (dichlorophenoxyacetic acid) 1~15 mg/L, 벤질아데닌 0.05~0.2 mg/L, 수크로스 20~40 g/L 및 젤란 검 2~3 g/L 또는 한천 5~10 g/L를 함유하는 배지에서 미성숙 화기로부터 유도된 캘러스를 증식하는 단계;
(3) MS 배지, 나프탈렌 아세트산 0.5~1.5 mg/L, 벤질아데닌 0.1~10 mg/L, 수크로스 20~40 g/L 및 젤란 검 2~3 g/L 또는 한천 5~10 g/L를 함유하는 배지에서 상기 증식된 캘러스로부터 신초를 재생하는 단계; 및
(4) MS 배지, 수크로스 20~40 g/L 및 한천 5~10 g/L를 함유하는 배지에서 상기 재생된 신초를 신장 및 발근하는 단계를 포함하는 억새의 미성숙 화기를 이용한 억새 식물체의 대량 생산 방법. - 삭제
