실시예 1:바실러스 서브틸리스(Bacillus subtilis)를 이용한 홍삼박 발효Example 1: Red ginseng gourd fermentation using Bacillus subtilis

홍삼액기스를 추출하고 남은 홍삼박의 발효를 위하여 배양조성물에 포함이 가능한 홍삼박 함량을 조사하였다. 기초 배양조성물로는 건조 중량 대비, 대두박 30%, 소맥피 20%, 전분 5%, 당밀 5%, 그리고 옥수수분말을 10%로 하였고 홍삼박 첨가량은 0%, 1%, 5%, 10%, 20%로 달리하였으며, 나머지 펄라이트(Perlite)로 충당하였으며, 총 수분함량은 70%로 하였다. 상기 홍삼박 배양조성물을 100℃이상의 온도에서 30분간 살균처리하였다. 살균처리가 완료된 배양조성물에 1.0x109 cfu/ml 이상으로 배양된 바실러스 서브틸리스(Bacillus subtilis)를 1 ~ 5% 접종하였다.Red ginseng extract was extracted and the contents of red ginseng gourd were included in the culture composition for fermentation of the remaining red ginseng gourd. 상기 실시예를 통하여 얻어진 발효홍삼박 제조 공정을 기초로 발효홍삼박을 제조하였고, 이를 건조한 후에 락토바실러스 아시도필러스(Lactobacillus acidophilus), 아시퍼질러스 오리제(Aspergillus oryzae), 사카로마이세스 세레비제(Saccharomyces cerevisae), 스트렙토마이세스 또는 스트렙토코커스 써모필러스(Streptococcus thermophilus) 또는 이들의 조합 생균제를 혼합하여 24시간 동안 2차 발효공정을 거쳐 가축 사료첨가용 조성물을 제조하였다. 발효홍삼박을 주성분으로 하는 가축 사료첨가제의 제조 공정은 도 2에서 보는 것과 같다. 발효홍삼박의 급여가 산란계의 생산성에 미치는 영향을 조사하였다. 55주령에 해당하는 산란계를 대상으로 사료에 발효홍삼박을 0.1% 첨가한 후에 12주간 사육하며 발효홍삼박의 효능을 평가하였다. 그 결과는 표 3에서 보는 것과 같다. 발효홍삼박을 급여한 실험군에서 발효홍삼박을 급여하지 않은 대조군에 비하여 유의적을 우수한 난 생산성을 나타내었으며, 난 중량 또한 유의적으로 높게 나타났다. 난 질량과 난 각두께 또한 발효홍삼박의 급여를 통하여 향상됨을 알 수 있었다. 발효홍삼박의 급여가 이유자돈의 성장에 미치는 영향을 알아보기 위하여 4주령의 이유자돈 사료에 발효홍삼박을 0.1% 첨가하여 4주간 사양평가를 실시하였다. 그 결과는 표 4에서 보는 것과 같다. 발효홍삼박의 급여를 통하여 이유자돈의 일당 증체량이 증가하였고, 사료효율 또한 개선됨을 알 수 있었다. 발효홍삼박의 급여를 통하여 이유자돈의 혈액 중에 면역물질이 감소하는 결과를 얻었으며, 이는 이유자돈의 사양상태가 매우 안정되었음을 나타내는 결과였다.
