Abstract

Description
본 발명은 생물학적 활성 거대분자를 포함하는, 분자의 창자 내강으로부터 장벽을 적절하게 통과하여 체내로의 흡수를 증강하기 위한 방향족 알콜의 사용 방법에 관한 것이다. 특히 본 발명은 바람직하게 장 벽(intestinal wall)을 통과하여 체내로 흡수되는 활성 거대분자 성분을 함유하는 새로운 제약 조성물에 관한 것이다.
친수성 방향족 알콜, 특히 하이드록시기가 방향족 핵에 직접 결합하지 아니한 페녹시에탄올, 페닐에탄올 및 벤질알콜과 같은 방향족 알콜은 수년간 용매 및 가소화제로서 제약의 실시에 사용되어왔으며, 구강 경로를 포함한 다향한 경로를 통하여 투약하는 경우에도 낮은 독성 분포를 갖는다. 그러한 화합물은 상온에서 모두 액체상태이며, 수성 매체에 용이하게 용해된다.
페녹시에탄올과 같은 친수성 방향족 알콜 및 페닐에탄올 및 벤질알콜을 포함한 관련 화합물은 비교적 높은 국부 농도로 존재할 경우, 그러한 방향족 알콜이 장 세포의 장벽층(barrier layer)에 대한 투과성을 일시적으로 증가시키는 장 세포에 대한 특정 활성 범위를 갖는다.
이는 큰 분자(거대분자)도 확산에 의해 통과할 수 있는 공극을 만드는 이러한 세포 사이에 견고한 접합이 개방되는 것에 기인한 것으로 추측된다.

갈레이트 에스테르 또는 특히 프로필 갈레이트의 용도는 US 6,180,666 및 US 5,962,522에 프로필 갈레이트가 시토크롬 P450(특히 세포질세망에 위치하는 CY3PA)의 활성을 저해하여, 장 세포를 통한 작은 분자들의 경로(세포횡단 경로로 알려짐)에서 그들의 대사 분해를 감소시키는 기전을 통한 작은 분자의 생체이용률 증강제로서 각각 기술되어 있다. 프로필 갈레이트 및 다른 갈산의 에스테르가 시토크롬 P450의 유력한 억제제로 나타나며, 가용화(solubilisation) 보조제 없이도, 충분한 프로필 갈레이트가 현저한 영향을 발휘하기 위해 제형에 삽입될 수 있다고 주장된다. 그러나, 거대분자가 도움없이 삽입될 수 없어서 효소가 위치하는 세포질세망과 접촉하지 못하게 되기 때문에 상기 기술된 프로필 갈레이트의 효소 억제제 작용 기전이 거대분자의 생체이용률을 증강시키리라고 기대할 수는 없다. 더욱이, 펩타이드 및 단백질과 같은 거대분자는 작은 약물 분자보다 시토크롬 P450의 작용에 덜 예민하므로, 이러한 효소에 의한 분해는 창자 또는 다른 점막 조직에서 거대분자의 낮은 생체이용률의 주된 원인이 아니다. 보다 큰 장벽은 단순하게도 분자 자체의 크기인데, 이러한 분자의 크기는 분자가 줄지어진 점막조직에 침투하거나 투과하는 것을 방해하며, 이는 이러한 조직을 배열하는 세포가 연속 불투과성 벽을 형성하기 때문이다.

발명의 개요

제약의 실시에서 항산화제로 사용된 상기 개시한 방향족 알콜은 총 제형의 0.1%w/v까지의 농도로 포함되어 있다(문헌[Handbook of Pharmaceutical Excipients, Eds Wade & Weller, The Pharmaceutical Press, London UK, 2nd edition 1994]에 개별 화합물 항목을 참조하라). 일반적으로 화합물의 보다 높은 농도가 추가된 항산화제의 유리한 효과를 나타내지 못하므로, 따라서 표준 제약의 실시에서는 제형에 있어서 항산화제의 농도를 0.1%이하로 제한하는 것으로 생각된다. 그러나, 본 발명에 따라 흡수 증강제로 사용될 경우에는, 이러한 화합물의 효능은 상당히 높은 수준까지 농도 의존적이며, 제약 제형에 있어서 그들의 비율은 이전의 당업계에서 먼저 서술된 것보다 훨씬 높다.
본 출원인의 지식으로는, 이전의 당업계에서 제약 제형에 있어서 항산화제로서 이러한 제제의 용도를 제안한 적이 없다. 이들 제제 중 어느 것도 구강 경로에 의한 거대분자의 흡수 증강에 있어서 어떠한 역할을 하지 못하고 또는 이러한 제제가 제형에 높은 농도로 포함될 수 있는 것은 산화제에 대한 표준 제약의 실시이다.
예를 들어, EP-A-0295941은 BHA, BHT 또는 PG가 제형의 생물학적 효율성에 필수적이지 않다는 것을 명확하게 하기 위하여 그들이 선택적으로 함유될 수 있는 구강 투약형 제형을 개시하였다. 이러한 제제의 어떠한 농도도 특정되지 않았고, 캡슐에서의 빠른 방출을 보장하기 위해 즉시 용해되는 것이 바람직한 본 발명과는 상당히 대조적으로, 제형은 조절-방출 투약 형태로서 의도된 것이다.
WO-A-0222158은 시클로스포린(거대분자가 아님)을 함유하고 일반적으로 BHA, BHT 및 PG를 항산화제로서 함유하는 조성물을 제공한다. 항산화제에 대한 특정 농도가 주어져 있지 않다고 하더라도 항산화제로서 그 화합물들의 사용은 0.1% wt 미만의 농도라는 것을 시사한다.
US-A-5342625는 다시 시클로스포린을 함유하는 조성물을 개시한다. 가용화 보조제가 시클로스포닝의 사전농축(pre-concentrate) 미세에멀션을 형성하는 것을 돕기 위하여 존재할 수도 있다. BHA 또는 BHT는 항산화제로서 낮은 농도로 존재할 수 있다.
BHA 및 BHT는 또한 식물성 유지 운반체의 존재하에서 안정한 약물, 보다 특히 쓴 맛을 가진 물-민감성 약물을 함유하는 US-A-3996355의 조성물에서 항산화제로 존재할 수도 있다.
바람직한 가용화 보조제는 소듐 타우로콜레이트 또는 타우로데옥시콜레이트와 같은 담즙산 또는 담즙산염, 벤질알콜, 페닐에탄올, 페녹시에탈올, 트랜스큐톨(transcutol) 및 이소프로판올을 포함하나, 이에 한정되지는 않는다.
본 발명의 범위 내에서 분류되는 활성 거대분자 성분은 인체 또는 동물체 내부로, 특히 장 벽을 통과하여 흡수될 경우 유리한 효과를 갖는 것이 가능한 모든 분자들을 포함한다. 유리한 효과는 예컨대 치료 효과, 미용 효과 또는 예방 또는 피임과 같은 방지 효과일 수 있다. 활성 거대분자 성분은 천연(생물학적), 합성 및 반합성 기원일 수 있다.

본 발명의 바람직한 구체예에서, 흡수될 활성 거대분자 성분은 칼시토닌, 인슐린, 저분자량 헤파린, 에리스로포에이틴, 인간성장호르몬 및 부갑상샘호르몬 특히 칼시토닌, 인슐린 및 부갑상샘호르몬에서 선택된다.
사용되는 추가적인 부형제의 성질에 따라 본 발명의 제약 조성물은 액체, 고체, 반고체 또는 겔 형태일 수 있다. 본 발명의 제약 조성물은 볼 및 설하 점막, 비측 구개, 폐, 직장, 장 관(대장 및 소장 포함) 및 질의 상이한 점막 세포에 대한 접근을 부여하는 임의의 경로를 통한 투약에 바람직하다. 액체, 반-고체 또는 겔 제형의 경우에는 이들은 무수물 또는 가수물 중 하나일 수 있다.
본 발명의 조성물을 작용시키고자 한 부위가 장인 경우에, 본 제형의 내용물이 소장 또는 대장에 도달하기까지 희석되지 않고 긴밀한 결합으로 함께 잔류할 수 있도록 본 조성물은 위에서 견딜 수 있는 장용코팅제로 피복되어 있는 것이 바람직하다. 이러한 제형은 무수물인 것이 바람직하다. 액체 형태의 조성물은 장도착캡슐로서 적절하게 투약될 것인 반면, 고체 제형은 장도착캡슐 또는 알약 중 하나, 바람직하게는 장도착알약으로 투약될 수 있다.

바람직한 장용코팅제는 당업계에 공지되어 있고, 그 예에는 셀룰로스, 아세테이트, 프탈레이트, 쉘락(shellac) 및 유드라지트(Eudragit)의 L 및 S 계열, 구체적으로 유드라지트 L12.5P, L12.5, L100, L100-55, L30, D-55, S12.5P, S12.5 및 S100에서 선택되는 것과 같은 폴리메타크릴레이트(polymethacrylate)를 포함된다. 또한, 트리에틸 시트레이트 및 폴리소르베이트 80과 같은 적당한 가소제 또는 습윤제가 코팅제 혼합물에 포함될 수 있다.
캡슐, 바람직하게는 HPMC 또는 젤라틴 캡슐에 적당한 코팅제의 선택은 유드라지트 제품을 지원설명하는 입수용이한 문헌 및 당업자의 경험을 기반으로, 당업자라면 용이하게 결정할 수 있다.
목적한 작용 부위가 비측 점막인 경우에, 제형은 수용액 또는 건조 분말의 형태일 수 있으며, 이는 스프레이로 투약될 수 있다.
목적한 작용 부위가 직장인 경우, 투약 방법은 캡슐 형태의 외피에 담겨있거나 또는 침식성 좌약의 기질에 혼힙되어 있는 무수 액체 또는 고체 형태인 것이 적당하다.
질에 투여용의 경우, 겔 형태의 제형의 투약도 바람직하다.
방향족 알콜 흡수 증강제는 불수용성인 것이 바람직하다. 증강제는 중량의 1 내지 40% 함량, 바람직하게는 중량의 5 내지 35% 함량, 보다 바람직하게는 중량의 10 내지 30% 함량으로 조성물 내에 존재한다.
본 발명의 조성물에서, 방향족 알콜 흡수 증강제는 활성 거대분자 성분의 양과 동등하거나 그 이상의 함량(중량에 기초하여)으로 존재한다. 이는, 활성 거대분자가 흡수되었을 때, 그것의 정상적인 유리한 효과를 발휘하는 적절한 양이 함께 존재하여 흡수의 증강을 야기하기 위한, 장 세포 장벽층(장 벽)에서의 방향족 알콜 흡수 증강제의 효과적인 농도를 제공한다. 본 발명의 실시자는 방향족 알콜 흡수 증강제의 함량 및 관련된 활성 거대분자 성분의 치유 효능에 필수적인 함량(예, 혈 농축 농도)에 기초한 활성 거대분자 성분의 함량을 선택할 수 있다. 캡슐에 들어있는 혼합물에서 방향족 알콜 흡수 증강제 대 활성 거대분자 성분의 중량 비율은 적절하게는 최소 1:1, 바람직하게는 최소 5:1, 예컨대 1:1 내지 100:1, 바람직하게는 3:1 내지 50:1, 가장 바람직하게는 5:1 내지 20:1이다.
가용화 보조제 대 방향족 알콜 흡수 증강제의 비율은 적절하게는 최소 1:1, 바람직하게는 1:1 내지 10:1 그리고 가장 바람직하게는 1.5:1 내지 5:1 이다.
활성 거대분자 성분의 절대 함량은 사용되는 복용법 및 관련된 환자에 대하여 정상적인 유리한 효과를 발휘하기 위한, 성분의 투약량에 기초하여 선택된다. 이러한 함량의 결정은 본 발명의 실시자의 맨틀(mantle) 범주 내에 있다.
구강 투약을 위한 조성물에 있어서, 캡슐의 내용물이 정상적인 치료 효과를 달성하기 위한 적절한 함량의 활성 거대분자 물질을 함유하는 것이 바람직하다. 예를 들어, 이러한 조성물은 캡슐 조성물의 중량(캡슐 자체의 중량은 포함하지 아니함)을 기초로 하여 0.05 내지 50%, 바람직하게는 0.1 내지 25%, 보다 바람직하게는 0.1 내지 10% 중량의 활성 거대분자 성분을 함유할 수 있다.
본 발명의 조성물은 추가적으로 하나 또는 그 이상의 다른 흡수 증강 물질, 예컨대 중간쇄지방산 및 중간쇄모노사카라이드를 함유할 수 있다.
본 발명의 조성물은 의약물의 조제에 통상적으로 사용되는 임의의 첨가제를 선택적으로 포함할 수 있으며, 그 예에는 항산화제, 항미생물제, 현탁화제, 충전제, 희석제, 흡수제, 활주제(glidant), 결합제, 응결방지제, 윤활제, 붕해제, 팽창제, 점도 조절제, 가소화제 및 산도 조절제(구체적으로, 장 환경을 7 내지 7.5 사이로 조정하는 제제)가 포함된다. 적당한 팽창제에는 소듐 스타치 글리콜레이트(sodium starch glycolate), 사전젤라틴화된 전분, 미세결정형 셀룰로스, 크로스프로비돈(crosprovidone) 및 마그네슘 알루미늄 실리케이트 또는 이의 혼합물이 포함된다. 소듐 스타치 글리콜레이트 및 다른 다당류계 팽창제는 5 내지 10중량%의 양으로 존재할 수 있다. 크로스포비돈은 5 내지 30중량%의 양으로 존재할 수 있다.
본 발명의 조성물은 추가적으로 바람직한 조성물의 활동을 시너지효과로 증강하기 위한 가외의 활성 성분을 선택적으로 함유할 수 있다. 예컨대 활성 거대분자 성분이 인슐린일 경우, 조성물은 또한 흡수된 인슐린에 신체 반응을 향상시킬 수 있는 인슐린 민감제(sensitiser)를 함유할 수 있다. 이러한 방법에 사용될 수 있는 민감제의 예는 트로글리타존(troglitazone), 피오글리타존(pioglitazone), 로시글리타존(rosiglitazone) 및 글리타존계 분자의 다른 구성원들이다.
방향족 알콜 흡수 증강제 및 활성 거대분자 성분을 함유하는 혼합물이 캡슐 또는 알약에 내재된 본 발명의 조성물에 있어서, 제형은 실질적으로 무수물인 것이 바람직하다. 본 발명의 보다 바람직한 구체예에서, 전체 조성물은 실질적으로 무수물인 것이 좋다. 본 발명의 문장 중에서 실질적으로 무수성물 이란, 혼합물 중량을 기준으로 5% 미만, 바람직하게는 1% 미만, 보다 바람직하게는 0.5% 미만의 수분이 존재하는 것을 의미한다.
본 발명에 사용된 활성 거대분자 성분에 따라서, 치료법에 의한 인간 또는 동물 신체의 다양한 증상 및 질환의 치료에 사용되거나, 또는 인간 또는 동물 신체의 질환 및 증상의 진단에 필수적인 거대분자의 도입에 사용될 수 있다. 본 발명의 조성물은 약학 조성물 또는 미용 조성물인 것이 바람직하다.
본 발명의 조성물에서 캡슐에 함유된 혼합물은 액체, 반-고체 또는 겔 형태일 수 있으며, 이는 용액 또는 마이크로미립자 분산액 형태 중 하나로 존재할 수 있다. 즉, 흡수될 위한 활성 거대분자 성분은 용액 또는 마이크로미립자 분산액 형태 중 하나의 제형으로 첨가될 수 있다. 대안적으로, 조성물은 고체 형태일 수 있다.
본 발명의 조성물은 활성 거대분자 성분의 실질적 무수 혼합물 및 방향족 알콜 흡수 증강제를 제조하고, 그 다음 선택적으로 피복되지 않은 캡슐을 혼합물로 채우고 바람직한 침투 성질을 수득하기 위해, 선택적으로 그것을 적절한 중합체 혼합물로 코팅함으로써 바람직하게 생산될 수 있다.

실시예 1 세포 배양 단층을 침투성화함에 있어서의 효과
카고-2 세포들(Caco-2 cells)(인간 대장 선암으로부터 유래된 세포열)은 상부 구획이 배지 200㎕ 및 하부 구획이 배지 600㎕를 함유한 두 수성 구획을 분리하는 다공성 세포막(공극도 0.4㎛, 표면적 0.33㎠)의 표면에 융합성 단층으로 성장한다. 단층을 통과하는 전기 저항은 상부 및 하부 구획에서 단층의 양쪽에 배지로 삽입된 전극에 연결되어 있는 상피 볼토미터(voltohmeter)를 사용하여 측정되었다. 이러한 전환-상피 전기 저항(TEER)은 방향족 알콜을 상부구획에 첨가하기 이전, 첨가 후 15분에 즉시 측정되었다(전형적인 결과는 아래의 표에 나타내었다). 네 개의 복제품이 각각의 화합물에 사용되었는데, 그들의 농도는 아래 표에 나타내었다. 초기 값의 50%를 초과하는 농도의 하강은 중요한 것으로 생각된다. 농도를 감소시키는 것은 관찰된 효과를 감소시키는 경향이 있다.

실시예 2 인슐린, 프로필 갈레이트 및 소듐 타우로콜레이트를 함유하는 제형의 제조
소듐 타우로콜레이트를 150mg의 양으로 하여 75mg의 프로필 갈레이트와 유리 바이알(vial)에서 혼합하고 증류수 825㎕를 첨가하였다. 용해는 상온에서 오랜 기간 교반하여도 일어나지 않으나 초음파조(ultrasonic bath)에서 단시간의 초음파 처리로 데운 후에는 무색 투명의 용액이 수득되었다. 8.4mg 양의 소(bovine) 인슐린을 혼합하면서 첨가하고, 이후 인슐린 부유액을 교반하면서 빙초산 10㎕를 첨가하였다. pH 3.15인 투명한 용액이 빠르게 수득되었다. 바이알의 내용물을 교반하면서 빠르게 냉각시켰고 밤새 동결건조 시켰다. 다음날 건조 고형물이 수득되었다. 10mg 양의 고형물을 2ml 바이알에서 무게를 달았고 증류수 50㎕를 첨가하였다. 투명한 용액이 빠르게 형성되었다.
실시예 3 인슐린, 프로필 갈레이트 및 소듐 타우로데옥시콜레이트를 함유하는 제형의 제조
타우로콜레이트 대신 타우로데옥시콜레이트를 사용하여 실시예 2에서 기술한 바와 동일한 조건을 사용하였다. 건조 이전의 최종 용액의 pH는 3.36이었다. 건조 고형물에 증류수를 첨가함과 즉시 투명한 용액이 빠르게 형성되었다.
실시예 4 칼시토닌, 프로필 갈레이트 및 소듐 타우로콜레이트를 함유하는 제형의 제조
연어 칼시토닌 2.3mg을 증류수에 용해시킨 것을 제외하고 실시예 2에서 기술한 바와 동일한 조건을 사용하였고, 전체 용액은 타우로콜레이트 및 프로필 갈레이트의 혼합물에 첨가하였다. 증류수의 첨가 즉시 투명한 용액이 빠르게 형성되었다.
실시예 5 칼시토닌, 프로필 갈레이트 및 소듐 타우로데옥시콜레이트를 함유하는 제형의 제조
타우로콜레이트 대신 타우로데옥시콜레이트를 사용하였다는 것을 제외하고, 실시예 4에서 기술한 바와 같은 동일한 조건을 사용하였다.
실시예 6 부갑상샘호르몬, 프로필 갈레이트 및 소듐 타우로콜레이트를 함유하는 제형의 제조
칼시토닌 대신 부갑상샘호르몬 0.5mg을 사용하였다는 것을 제외하고, 실시예 4에서 기술한 바와 같은 동일한 조건을 사용하였다.
실시예 7 부갑상샘호르몬, 프로필 갈레이트 및 소듐 타우로데옥시콜레이트를 함유하는 제형의 제조
타우로콜레이트 대신 타우로데옥시콜레이트를 사용하였다는 것을 제외하고, 실시예 6에서 기술한 바와 같은 동일한 조건을 사용하였다.
실시예 8 부갑상샘호르몬, 프로필 갈레이트 및 소듐 타우로데옥시콜레이트를 함유하는 제형의 제조
담즙산/PG 혼합물이 단백질의 첨가 없이 건조되었다는 것을 제외하고는 실시 예 7에서 기술한 바와 같은 동일한 조건을 사용하였고, 부갑상샘호르몬을 건조 분말로서 동결건조 후의 건조 잔류물에 첨가하였다.
실시예 9 인간성장호르몬, 프로필 갈레이트 및 소듐 타우로데옥시콜레이트를 함유하는 제형의 제조
부갑상샘호르몬 대신 인간성장호르몬 20mg을 사용하였다는 것을 제외하고, 실시예 8에 기술된 바와 같은 동일한 조건을 사용하였다.

실시예 11 칼시토닌, 프로필 갈레이트 및 트랜스큐톨을 함유하는 제형의 제조
트랜스큐톨 200㎕에 프로필 갈레이트 100mg을 교반하여, 상온에서 1분 후에 투명한 용액을 수득하였다. 그 다음 결과물 용액 200㎕를 고형 칼시토닌 1mg을 포함하는 바이알에 옮겼다. 고형물을 분산시키기 위하여 바이알을 단시간 교반하였 고, 이후 37℃에서 1시간 동안 교반하여, 투명한 용액을 수득하였다. 이 용액 100㎕를 새로운 바이알에 옮기고 여기에 증류수 100㎕을 첨가하였다. 모든 구성 성분은 단일상 투명 액체로서 용액에 잔류하였다.
실시에 12 칼시토닌, 부틸화된 하이드록시 톨루엔 및 트랜스큐톨의 제조
트랜스큐톨 200㎕에 부틸화된 하이드록시 톨루엔 100mg을 교반하여, 상온에서 수 분 후에 투명한 용액을 수득하였다. 그 다음 결과물 용액 200㎕를 고형 칼시토닌 1mg을 함유한 바이알에 옮겼고, 이후 37℃에서 1시간 동안 교반하여 투명한 용액을 수득하였다. 이 용액 100㎕를 새로운 바이알에 옮기고 여기에 증류수 100㎕를 첨가하여, 37℃에서 선명한 유백광 용액을 수득하였다.
Claims (29)
- (a) 활성 거대분자 성분; 및(b) 부틸화된 하이드록시 톨루엔, 부틸화된 하이드록시 아니솔, 및 프로필 갈레이트로부터 선택되는 방향족 알콜 흡수 증강제의 혼합물을 함유하는 구강 투여용 제약 조성물로서,상기 방향족 알콜 흡수 증강제는 활성 거대분자 성분의 중량과 같거나 그 이상의 중량으로 존재하며,상기 방향족 알콜 흡수 증강제가 프로필 갈레이트일 때, 상기 혼합물은 (c) 수성 매체에서 방향족 알콜 흡수 증강제의 용해도를 상승시킬 수 있는 가용화 보조제를 추가로 포함하는 제약 조성물.
- 제1항에 있어서, 상기 조성물이 pH 3 내지 7에서 침투성으로 되는 장용코팅제(enteric coating)로 피복된 것을 특징으로 하는 제약 조성물.
- 제1항 또는 제2항에 있어서, 상기 혼합물이 5중량% 미만으로 수분을 함유하는 것을 특징으로 하는 제약 조성물.
- 제1항 또는 제2항에 있어서, 방향족 알콜 흡수 증강제가 부틸화된 하이드록시 톨루엔 또는 부틸화된 하이드록시 아니솔이고, 조성물이 수성 매체에서 방향족 알콜 흡수 증강제의 용해도를 상승시킬 수 있는 가용화 보조제를 추가로 포함하는 것을 특징으로 하는 제약 조성물
- 제1항 또는 제2항에 있어서, 상기 혼합물이 방향족 알콜 흡수 증강제를 1중량% 이상 함유하는 것을 특징으로 하는 제약 조성물.
- 제1항 또는 제2항에 있어서, 상기 방향족 알콜 흡수 증강제 대 활성 거대분자 성분의 중량 비율이 적어도 5:1인 것을 특징으로 하는 제약 조성물.
- 제1항 또는 제2항에 있어서, 상기 혼합물이 용액 또는 마이크로미립자 분산액 형태인 것을 특징으로 하는 제약 조성물.
- 제1항 또는 제2항에 있어서, 상기 혼합물이 고체 형태인 것을 특징으로 하는 제약 조성물.
- 제1항 또는 제2항에 있어서, 상기 활성 거대분자 성분이 폴리펩타이드 또는 단백질, 폴리뉴클레오티드, 폴리사카라이드 또는 이들의 혼합물인 것을 특징으로 하는 제약 조성물.

- 제1항 또는 제2항에 있어서, 상기 가용화 보조제가 복합(conjugated) 담즙산 또는 복합 담즙산염, 벤질알콜, 페닐에탄올, 페녹시에탄올, 2-(2-에톡시에톡시)에탄올 및 이소프로판올 중에서 선택되는 것을 특징으로 하는 제약 조성물.
- 제1항 또는 제2항에 있어서, 상기 활성 거대분자 성분이 인슐린, 칼시토닌, 성장호르몬, 부갑상샘호르몬, 및 에리스로포에이틴 중에서 선택되거나,상기 활성 거대분자 성분이 GLP-1(글루카곤 유사 펩타이드 1; Glucagon-like peptide 1), GCSF(과립구 콜로니-자극 인자; Granulocyte colony-stimulating factor), 또는 단일-, 이중-, 또는 삼중 나선 RNA인 것을 특징으로 하는 제약 조성물.

- 구강 투여용 제약 조성물에서,장 벽을 통과하는 거대분자의 흡수 증강제로서,부틸화된 하이드록시 톨루엔, 부틸화된 하이드록시 아니솔, 및 프로필 갈레이트로부터 선택되는 방향족 알콜을 사용하는 방법으로서,상기 방향족 알콜이 프로필 갈레이트일 때, 수성 매체에서 방향족 알콜 흡수 증강제의 용해도를 상승시킬 수 있는 가용화 보조제가 추가로 사용되는,방향족 알콜을 사용하는 방법.
