Claims (10)
- 서열번호 1의 아미노산 서열을 가지는 셀룰로모나스 피미(Cellulomonas fimi) 유래의 섬유소결합도메인(cellulose binding domain, CBD) 유전자 및 활성형 단백질의 유전자를 포함하고, 상기 두 유전자가 융합되어 활성을 보유하는 활성형 단백질 응집체 입자 형태로 발현되도록 배치된 재조합벡터.
- 제1항에 있어서, 상기 섬유소결합도메인은 엑소글루카나제(exoglucanase, CBDcex)인 것을 특징으로 하는 재조합벡터.
- 제1항에 있어서, 상기 활성형 단백질은 효소인 것을 특징으로 하는 재조합벡터.
- 제3항에 있어서, 상기 효소는 β-글루쿠로니다아제(β-glucuronidase, gusA) 또는 β-글리코시다아제(β-glycosidase, bglA)인 것을 특징으로 하는 재조합벡터.
- 제1항 내지 제4항 중 어느 한 항의 벡터를 박테리아, 효모 및 곰팡이로 구성된 군에서 선택되는 숙주세포에 도입하여 수득된 재조합 미생물.
- 제5항에 있어서, 상기 박테리아는 대장균(E. coli)인 것을 특징으로 하는 재조합 미생물.
- 제5항의 재조합 미생물을 배양하는 것을 특징으로 하는 섬유소결합도메인과 활성형 단백질이 융합된 활성형 단백질 응집체 입자의 제조방법.
- 제7항의 방법으로 제조되고, 서열번호 1의 아미노산 서열을 가지는 셀룰로모나스 피미(Cellulomonas fimi) 유래의 섬유소결합도메인과 활성형 단백질이 융합되어 활성형 단백질의 활성을 보유하는 것을 특징으로 하는 활성형 단백질 응집체 입자.
- 제8항의 활성형 단백질 응집체 입자에 제니핀(genipin) 또는 글루타알데히드(glutaraldehyde)를 첨가하여 가교시키는 것을 특징으로 하는 가교화에 의한 활성형 단백질 응집체 입자의 고정화 방법.
