Abstract
비경구 투여제제는 생리활성의 펩티드 또는 단백질과 포화 지방산의 폴리글리세롤 디에스테르를 함유하는 매트릭스로 구성되어 있고, 이 매트릭스는 실온에서 고체이다. 생리활성의 펩티드 또는 단백질의 분자량은 2,000 달톤 이상이다. 포화 지방산은 팔미트산, 스테아르산 등과 같이 16 ~ 30 개 정도의 탄소원자를 가지는 지방산을 포함한다. 매트릭스는 주상 또는 입상일 수 있다. 비경구 투여제제는 피하 또는 정맥내 투여된 고형주사제 (예 : 이식용 펠렛 또는 정제), 좌약 등으로서사용될 수 있으며, 1 주간 이상의 장기간 동안 생리활성 펩티드 또는 단백질을 지속적으로 방출할 수 있다.
Description
본 발명은 생리 활성의 펩티드 또는 단백질을 지속적으로 또는 장기적으로 방출시키는데 유용한 서방성 (徐放性) 비경구 투여제제 및 그의 제조방법에 관한 것이다.
치료용 약물의 투여에는 경구투여가 통상적으로 이용된다. 그러나, 생리활성의 펩티드 또는 단백질의 경구투여는 소화 효소에 의한 펩티드 또는 단백질의 가수분해를 야기시켜 소화관으로 부터의 흡수율을 저하시키는 문제점이 있다. 따라서, 이와 같은 생리활성의 펩티드 또는 단백질은 통상 근육내 또는 피하에 반복적으조 주사하거나 정맥내에 점적 주입하는 방법에 의해 투여된다. 그러나, 이 방법들도 매우 제한된 회수의 주사를 필요로 하는 경우에만 허용되고, 만성 투여시에는 바람직하지 못하다. 예컨대, 바이러스에 의한 C 형 간염의 치료를 위해서는 인터페론 - α가 4 주간 이상 계속적으로 매일 투여된다 (참조 : "Journal of Clinical and Experimental Medicine (IGAKU NO AYUMI)",161, 5, 359 - 363 (1992)). 그러나, 이와 같은 만성 또는 빈번한 투여 방법에서, 환자는 크게 구속을 받지 않을 수 없게 된다. 따라서, 생리 활성의 펩티드 또는 단백질의 유효하고 경제적인 투여 시스템의 개발이 요망되어 왔다.
일본국 특허출원 공개 제 2930/1988 호 (JP-A-63-2930) 에서는 폴리락티드에 폴리펩티드를 분산시키는 계를 개시하고 있다. 일본국 특허출원 공고 제 502117/1988 호 (JP-B-63-502117) 및 일본국 특허출원 공개 제 234820/1992 호 (JP-A-4-234820) 에서는 리포솜을 사용한 제제(pharmaceutical preparations)를 개시하고 있고, 일본국 특허출원 공고 제 502574/1991호 (JP-B-3-502574) 에서는 생리활성의 폴리펩티드를 함유하는 리포솜이 겔에 분산된 제제를 제안하고 있다.
그러나, 이들 제제를 투여할 때, 예상외로 약물이 투여 초기에 대규모 방출된다. 따라서, 혈액중 약물농도는 증가하고, 약물의 방출 속도는 일정 범위로 유지될 수 없다. 또한, 제조과정에서 유기용매가 사용되기 때문에, 폴리펩티드가 변성되어 생리활성이 저하된다.
일본국 특허출원 공개 제 2930/1988 호 (JP-A-63-2930) 에서는 아테로콜라겐 - 매트릭스를 포함하고 있고, 이 매트릭스 내에 분산된 생리활성의 폴리펩티드를 지속적으로 방출하는 계를 개시하고 있다. 그러나, 기제로서 사용된 아테로콜라겐은 인간과 다른 이종 또는 별종 동물로 부터 유래되었으며, 항원성을 나타낼 수도 있다.
일본국 특허출원 공개 제 22012/1988 호 (JP-A-63-22012) 에서는 폴리펩티드의 비경구 투여용의 서방성 제제로서, 수불용성 매트릭스 중에 생리활성의 폴리펠티드를 분산시키고 분산액을 압축 성형함으로써 제조한 계를 개시하고 있다. 이 제제는 매트릭스가 생체내에서 침식되는 것을 이용함으로써 생리활성의 폴리펩티드의방출을 조절한다. 따라서, 폴리펩티드는 효소분해되어 생물학적 이용도를 보다 낮추게 된다.
일본국 특허출원 공개 제 85328/1986 호 (JP-A-61-85328) 에서는 생리활성의 폴리펩티드와 폴리글리세롤 지방산 에스테르의 조성물을 함유하며, 이때 폴리글리세롤 지방산 에스테르가 물에 분산되는 제제를 개시하고 있다. 그러나, 이 제제는 용액이기 때문에 제형에 관하여 제한을 받는다. 더군다나, 폴리글리세롤 지방산 에스테르는 생리활성 폴리펩티드의 경피흡수를 촉진시키는데 이용되기 때문에, 약물을 장시간, 즉 24 시간 이상동안 도저히 방출할 수가 없다.
따라서, 본 발명의 목적은 생리활성의 펩티드 또는 단백질이 가수 분해되는 것을 억제하고, 이들의 활성이 저하되는 것을 피할 수 있으며, 장시간 동안 펩티드 또는 단백질이 지속적으로 방출될 수 있도록 하는 서방성 비경구 투여제제를 제공하는 것이다.
본 발명의 다른 목적은 투여 초기에 생리활성의 펩티드 또는 단백질이 과다하게 방출되는 것을 방지하고, 장시간동안 펩티드 또는 단백질이 방출될 수 있는 서방성 비경구 투여제제를 제공하는 것이다.
본 발명의 다른 목적은 고차원 구조의 변성을 억제하고, 생리활성의 펩티드 또는 단백질의 활성저하를 저해하여 생리활성의 펩티드 또는 단백질을 장시간동안 지속적으로 방출하는 서방성 비경구 투여제제를 제공하는 것이다.
본 발명의 다른 목적은 항원성을 일으키는 어떠한 소재도 함유하지 않고, 생리활성의 펩티드 또는 단백질을 1주간 이상동안 방출할 수 있는 서방성 비경구 투여제제를 제공하는 것이다.
본 발명의 다른 목적은 상기와 같은 우수한 특성을 가지는 제제를 간단하고 용이한 방법으로 제조할 수 있는 제제의 제조방법을 제공하는 것이다.
본 발명자들은 상기 목적을 달성하기 위하여 철저한 조사 및 연구를 한 결과, 생리활성의 폴리펩티드 또는 단백질과 배합시킨 수 많은 폴리글리세를 지방산 에스테르로 부터 선택된 특정한 폴리글리세롤 지방산 에스테르를 사용함으로써 수득할 수 있는 제제는 생리활성의 폴리펩티드 또는 단백질의 지속적인 방출을 현저하게 개선시킬 수 있으며, 생리활성의 펩티드 또는 단백질을 장시간동안 지속적으로 방출할 수 있음을 발견하였다. 본 발명은 이러한 발견을 토대로 완성한 것이다.
따라서, 본 발명의 서방성 비경구 투여제제는 "생리활성 펩티드 또는 단백질 (이하, 특별히 언급하지 않는 한, 간탄히 "생리활성 폴리펩티드"라 칭함) 과 포화지방산의 폴리글리세롤 디에스테르를 함유하는 매트릭스로 구성되어 있다. 생리활성 폴리펩티드의 평균 분자량은 2,000 달톤 이상인 경우가 많고, 많은 경우에 있어서 디에스테르는 평균 중합도가 4 정도인 폴리글리세롤과 16 ~ 30 개의 탄소원자를 가지는 포화지방산으로 형성된 디에스테르일 수 있다. 매트릭스는 주상 (柱狀), 입상 (粒狀) 또는 다른 형태일 수 있다. 매트릭스는 이식용의 고형 주사제가 될 수도 있다.
서방성 제제는 생리활성 펩티드 또는 단백질을 용융 또는 연화된 포화지방산의 폴리글리세롤 디에스테르와 혼합하고, 용융 혼합물을 성형함으로써 제조할 수 있다.
본 명세서에서 "평균 중합도가 4 인 폴리글리세롤" 이란 용어는 수산기 가 (value)의 말단 분석법에 의해 예측된 주요 화합물로서의 폴리글리세롤의 중합도를 의미하고, 테트라글리세롤은 물론 주성분으로서 테트라글리세롤및 불가피하게 혼재하는 글리세롤 또는 글리세롤 중합체 (예 : 디글리세롤, 트리글리세롤, 펜타글리세롤 등) 의 혼합물이 포함된다. 따라서, 폴리글리세롤의 평균 중합도는 약 3.7 ~ 4.3 일 수 있다.
"디에스테르" 란 용어는 폴리글리세롤 지방산 에스테르의 에스테르가로 부터 예측된 주요 화합물의 에스테르 결합의 평균가를 의미하고, 디에스테르는 물론 주성분으로 디에스테르 및 불가피하게 공존하거나 혼재할 수 있는 모노에스테르, 트리에스테르를 함유하는 혼합물이 포함된다. 따라서, 디에스테르 중 에스테르 결합의 평균가는 1.7 ~ 2.3 정도일 수 있다.
매트릭스 또는 지방산의 폴리글리세롤 디에스테르가 단일 화합물이 아니라 혼합물인 경우에, 이 물질은 명료한 융점을 나타내지 않으며 특정 온도에서 연화한다. 본 명세서에서 사용된 "융점" 이란 용어는 그 의미내에서는 또한 당해 혼합물의 연화점을 포함한다.
본 발명에서는 생리활성의 폴리펩티드로서, 생리활성을 가지는 각종 펩티드 및 단백질을 사용할 수 있다. 생리 활성의 폴리펩티드의 평균 분자량은, 예컨대, 약 2,000 달톤 이상, 바람직하게는 약 5,000 ~ 1,000,000 달톤, 보다 바람직하게는 약 10,000 ~ 500,000 달톤 및 특히 약 10,000 ~ 100,000 달톤이다. 바람직한 생리활성의 폴리렙티드는 생화학의 분야에서 고차원 구조를 가지는 것으로 표현된 단백질로 분류된 분자를 포함한다.
분류별로 예시하면, 생리활성 폴리펩티드는 단백질, 효소, 핵단백질, 당단백질, 리포단백질, 호르몬과 같은 활성을 가지는 폴리펩티드, 이들 분자들의 작동물질, 길항물질을 함유하는 합성 유사물질 등을 포함한다
본 발명은 다양한 생리활성의 폴리펩티드에 적용할 수 있으며, 그의 종류는 특히 한정되지는 않는다. 생리활성 폴리펩티드의 예로서는, 면역조절인자, 림포킨류, 모노킨류, 사이토킨류, 효소류, 항체, 성장촉진인자, 성장억제인자, 호르몬, 백신 (바이러스, 세균, 기생충류 및 리케차류의 항원을 포함), 혈액응고인자, 그의 각종 전구단백질, 돌연변이 단백질 및 그의 다른 유사체를 들 수 있다.
구체적으로는, 생리활성 폴리펩티드의 예로서, 하기의 생리활성 고분자 화합물, 돌연변이 단백질 및 그의 유사체가 있다.
(1) 인터페론류 (α -, β -, γ - 등), 인터류킨류 (IL - 1, IL - 2, IL-3, IL - 4, IL - 5, IL - 6, IL - 7, IL - 8, IL - 9, IL - 10, IL - 11), 알레르기 억제성 인자, 서프레서 인자, 세포 장해성 당단백질, 면역 세포 장해인자, 면역 독소류, 림포톡신류, 종양괴사인자 (TNF - α, TNF - β 등), 카켁틴 (cachectin), 온코스타틴류, 형질전환 증식인자 (TGF - α, TGF - β 등), 조혈인자 (예 : 에리트로포이에틴), 과립구 - 콜로니 자극인자 (G - CSF), 과립구 - 마크로파지 콜로니 자극인자 (GM - CSF), 마크로파지 - 콜로니 자극인자 (M-CSF), 마크로파지 펩티드류, B - 세포인자 (예 : B - 세포증식인자 등), T - 세포인자 등.
(2) 성장 인자류, 예컨대, 신경성장인자 (NGF), 신경영양인자 (NTF), 뇌신경세포상에 작용하는 폴리펩티드, 상피세포 성장인자 (EGF), 인슐린과 같은 성장인자 (IGF), 성장 호르몬 (GH), 섬유아세포 증식인자 (FGF), 골형성원 (osteogen) 성장인자, 심방성 나트륨 배설증가 인자 (ANP), 연골유도인자 등. 이 카테고리에 속하는 생리활성의 폴리펩티드는, 예컨대, 부갑상선 호르몬 (PTH), 엔도세린 등을 추가로 포함하고 있다.
(3) 혈소판 - 유래 증식인자 (PDGF) 등과 같이 혈소판 증식작용을 가지는 생리활성의 폴리펩티드.
(4) 효소작용을 가지는 생리활성의 폴리펩티드, 예컨대, 제 VIII 인자, 제 IX 인자, 피브린 용해인자, 조직 플라스미노겐 활성화 인자 (TPA), 유로키나제, 프로유로키나제, 스트렙토키나제, 리포코르틴, 마크로코르틴, 단백질 C, C - 반응성 단백질, 레닌 - 저해제, 메탈로프로테아제, 메탄로프로테아제의 조직 억제인자 (TIMP), 슈퍼옥시드 디스무타제 (SOD) 등.
(5) 호르몬과 같은 작용을 가지는 생리활성의 폴리펩티드, 예컨대, 인슐린, 세크레틴, 성장호르몬 방출인자 (GRF), 글루카곤, 가스트린, 프로락틴, 부신피질자극 호르몬 (ACTH), 갑상선자극 호르몬 (TSH), 황체형성 호르몬 (LH), 난포자극 호르몬 (FSH), 콜레시스토키닌, 인융모성 성선자극호르몬 (HCG), 류코키닌, 티모신, 모틸린, 칼리크레인 등.
(6) 백신 항원으로서 작용하는 생리활성의 폴리펩티드, 예컨대, HTLV - 1, HTLV - II, AIDS 바이러스군 (예 : HTLV - III/LAV/HIV 및 HIV - 2 등), 사이토메갈로바이러스, A 형 -, B 형 -, C 형 - 간염 바이러스, 단순 포진 - I 바이러스,단순포진 - II 바이러스, 말라리아, 잠재성 광견병 레트로바이러스, 전염성 위장염 바이러스, 파라인플루엔자 바이러스, 인플루엔자 바이러스, 로타바바이러스군, 호흡성 합포체 바이러스, 수두 - 대상포진 바이러스, 엡스타인 - 바르 바이러스, 백일해와 같은 항원 : 및슈도모나스, 내독소, 파상풍 독소 등과 같은 그람 - 음성 세균이 포함된다. 이와같은 생리활성 폴리펩티드는 단독으로, 또는 합텐과의 결합형태로나 보조제 (adjuvant) 와의 배합형태로 투여될 수 있다.
이러한 생리활성의 폴리펩티드는 천연적으로 발생하거나 유전자 재조합에 의해 제조할 수 있다. 생리활성의 폴리펩티드는 글리코실 사슬을 가지고 있을 수 있으며, 글리코실 사슬의 구조는 서로 상이할 수 있다. 또한, 생리활성의 폴리펩티드는, 예컨대, 상술한 펩티드와 단백질의 돌연변이체, 유도체, 유연체나 유사체, 또는 활성 단편을 포함한다.
생리활성 폴리펩티드는 단독으로 또는 배합하여 사용할 수 있다. 생리활성 폴리펩티드를 활성화시킬 수 있는 물질 및/또는 이 물질과 산술적 또는 상승적 효과를 가지는 다른 성분이 생리활성 폴리펩티드와 배합하여 유리하게 사용될 수 있다. 예컨대, 인터페론 - α는 인터류킨, 렌티난, 미노파겐 등과 같은 활성화 물질 또는 성분과 배합시켜 사용할 수 있다. 활성화 물질 또는 성분은 단독으로 또는 생리활성 폴리펩티드와 배합하여 사용할 수 있다.
본 발명의 특색은 다수의 폴리글리세롤 지방산 에스테르 중에서 특정 폴리글리세롤 지방산 에스테르를 배합시킴으로써 장시간동안 생리활성 폴리펩티드를 지속적으로 방출시킬 수 있다는 점이다.
폴리글리세롤 지방산 에스테르에서, "폴리글리세롤" 이란 " 1 분자중에 n 개 (환상일때) ~ n + 2 개 (직쇄 또는 측쇄일때) 의 수산기와 n - 1 개 (직쇄 또는 측쇄일때) ~ n 개 (환상일때)의 에테르 결합을 가지고 있는 다가 알콜 " 을 의미한다 ["폴리글리세롤 에스테르" Sakamoto Yakuhin Kogyo Co., Ltd., Japan 편집 및 발행 : pp 12 (1986.5.2)]. 폴리글리세롤은 글리세롤의 탈수축합 또는 글리세롤 증류 잔사물로 부터의 회수에 의해 수득할 수 있다.
생리활성 폴리펩티드와 배합하여 사용되는 성분으로서, 본 발명에서는 포화지방산의 폴리글리세롤 디에스티르가 사용된다. 지방산이 불포화 지방산일때, 생리활성 폴리펩티드는 초기 단계에서 방출될 수 있으며, 따라서 서방성은 현저하게 감소하거나 저하될 수 있다. 더군다나, 화합물이 폴리글리세롤과 포화 지방산과의 에스테르인 경우에도, 모노에스테르, 트리에스테르, 테트라에스테르, 펜타에스테르 등은 생리활성 폴리펩티드의 서방성을 현저하게 해치거나 저하시킬 수 있다.
포화 지방산의 예로서는, 팔미트산, 다툴산, 스테아르산, 아라크산, 베헨산, 리그노세르산, 세로트산, 몬탄산, 멜리쓰산 등과 같이 16 ~ 30 개의 탄소원자를 가지는 포화 지방산이 포함된다. 포화 지방산의 바람직한 예로는 16 ~ 22 개의 탄소원자를 가지는 지방산 (예 : 팔미트산, 스테아르산, 베헨산 등), 특히 팔미트산, 스테아르산 등이 포함된다.
이 포화 지방산의 디에스테르는 단일의 지방산 또는 2 종 이상의 지방산 혼합물로 형성된 디에스테르일 수 있다.
폴리글리세롤의 중합도는 서방성에 악영향을 일으키지 않는 특정 가로 특히제한되지 아니하며, 지방산의 종류에 의존하는 범위로 부터 선택된다.
바람직한 폴리글리세롤의 평균 중합도는 3 ~ 5정도이고, 특히 4 이다. 평균중합도가 3 미만이거나 5 보다 클때, 포화 지방산의 종류에 따라 생리활성의 폴리펩티드는 쉽게 방출되는 경향이 있으며, 어떤 경우에는 높은 서방성을 부여하기 어려운 경우가 있다. 따라서, 평균 중합도가 4 인 폴리글리세롤의 사용이 추천된다.
포화 지방산의 폴리글리세롤 디에스테르의 융점은, 예컨대, 약 40 ~ 60 ℃, 바람직하게는 약 42 ~ 58 ℃, 보다 바람직하게는 약 45 ~ 55 ℃ 이다.
포화 지방산의 바람직한 폴리글리세롤 디에스테르는 평균 중합도가 4 인 폴리글리세롤과 16 ~ 22 개의 탄소원자를 가지는 포화 지방산으로 형성된다.
포화 지방산을 가지는 폴리글리세롤 디에스테르의 예를 들면, 트리글리세롤 디팔미테이트, 트리글리세롤 디스테아레이트 및 트리글리세롤 디베헤네이트와 같은 트리글리세롤 디에스테르 : 테트라글리세롤 디팔미테이트, 테트라글리세롤 디다툴레이트, 테트라글리세롤 디스테아레이트, 테트라글리세롤 디아라케이트, 테트라글리세롤 디베헤네이트, 테트라글리세롤 딜리그노세레이트, 테트라글리세롤 디세로테이트, 테트라글리세롤 디몬탄테, 테트라글리세롤 디멜리스세이트, 테트라글리세롤 모노팔미테이트 모노스테아레이트, 테트라글리세롤 모노팔미테이트 모노베헤네이트 및 테트라리세롤 모노스테아레이트 모노베헤네이트와 같은 테트라글리세롤 디에스테르 : 펜타글리세롤 디팔미테이트, 펜타글리세롤 디스테아레이트, 펜타글리세롤 디베헤네이트 등과 같은 펜타글리세롤 디에스테르를 포함한다. 디에스테르의 바람직한 예로서, 테트라글리세롤 디팔미테이트, 테트라글리세롤 디스테아레이트, 테트라글리세롤 디베헤네이트 등과 같은 테트라글리세롤 디에스테르를 포함한다. 이들 에스테르들은 단독으로 또는 2 종 이상을 배합하여 사용할 수 있다.
폴리글리세롤 디에스테르는 식품 첨가용 유화제로서 유용하고, 생체내에서 그 안정성은 이미 확인되었다. 또한, 디에스테르는 생체 내에서 최종적으로 흡수될 수 있고, 게다가 어떠한 항원성을 나타내지도 않는다
폴리글리세롤 고급 지방산 에스테르 및 펩티드 또는 단백질을 포함하는 매트릭스는 일본국 특허출원 공개 제 223533/1990 호 (JP-A-2-223533), EP-A 443572, 일본국 특허출원 공개 제 237/1993 호 (JP-A-5-2237) 및 제 132416/1993호 (JP-A-5-132416) 에서 개시되고 있다. 그러나, 이들 선행 문헌들은 포화 지방산의 폴리글리세롤 디에스테르와 생리활성 폴리 펩티드의 배합을 함유하는 매트릭스가 생리활성의 폴리펩티드를 극히 장시간동안 방출할 수 있음을 개시하지 않고 있다.
본 발명의 제제는 생리활성의 폴리펩티드와 디에스테르를 함유하는 매트릭스로 구성되어 있다. 통상적으로, 제제는 생리활성 폴리펩티드가 디에스테르 중에 분산되는 매트릭스로 형성되는 것이 바람직하다. 매트릭스는 실온 또는 상온(5 ℃ ~ 35 ℃)에서 고체로 있는 것이 바람직하며, 통상 생리활성의 폴리펩티드는 디에스테르 중에 균일하게 분산되어 있다. 생리활성 폴리펩티드가 제조과정에서 용융된 디에스테르 중에 용해될 때에도, 균일하게 혼합되어 실온에서 폴리펩티드가 고체 형태로 분산된 제제를 제조하는 것이 바람직하다. 이와 같은 제제의 특징은 투여의 초기 단계에 생리 활성의 폴리펩티드가 대규모로 방출되는 것을 억제하고, 그의 고차원 구조를 유지하면서 장시간동안 펩티드를 지속적으로 방출시킬 수 있다는 점이다.
디에스테르에 관한 생리활성 폴리펩티드의 비율은 넓은 범위에서 선택할 수 있으며, 두가지 성분으로 구성된 매트릭스에서 생리활성 폴리펩티드의 비율은, 예컨대, 0.0001 ~ 50 중량%, 바람직하게는 0.001 ~ 20 중량% 및 보다 바람직하게는 0.001 ~ 10 중량% 이고, 잔사물은 디에스테르와 함께 형성된다.
필요한 경우, 매트릭스는 고형제제의 분야에서 통상적으로 사용하는 성분, 예컨대 부형제, 결합제 및 붕해제와 함께 첨가하는 것은 물론, 안정화제, 보존제 등과 같은 각종 첨가제를 첨가할 수도 있다. 안정화제의 예로서, 젤라틴, 알부민, 글로불린, 프로타민, 트레할로스, D - 글루코오스, 덱스트란 등을 들 수 있다. 보존제로서 예를 들 수 있는 것은 파라옥시벤조산 에스테르 (예 : 메틸파라벤, 프로필파라벤 등), 벤질 알콜, 클로로 부탄올, 티메로살 등이 있다.
서방성 비경구 투여제제는 비경구적으로 투여될 수 있는 한, 어떠한 형태라도 좋으며, 통상 환자에게 지나친 고통을 주지 않는 제형의 매트릭스, 예컨대 소형 또는 컴팩트 매트릭스로 구성된다. 본 발명의 특징은 예를 들어 주사바늘을 이용하여 투여할 수 있는 소형 또는 컴팩트 매트릭스 조차도, 생리활성 폴리펩티드를 장시간 동안 지속적으로 방출시킬 수 있다는 점이다. 예컨대, 본 발명 제제의 비경구 투여는 생리활성 폴리펩티드를 단독 또는 분리해서 투여하는 경우와 비교해볼때, 혈액 중 생리활성 폴리펩티드의 지속기간을, 예컨대, 7일 이상으로 연장할 수 있다. 따라서, 제제의 투여횟수 및 환자에게 부여되는 고통을 현저하게 감소시킬 수 있다.
제제는, 예컨대, 피하 또는 근육내에 투여될 수 있는 고형 주사제 (예 : 작은 환약 또는 이식 등) 또는 좌약과 같은 점막 흡수 조성물로서 사용할 수 있다. 제제의 형상 또는 형태는 제형에 따라 선택될 수 있으며, 예컨대, 산제, 과립제 또는 환제로서 분말상 또는 과립상 ; 이식용으로 투여가능한 작은 환약 또는 정제로서 편평상, 타원상, 간상 또는 주상(pillar) ; 또는 좌약으로서 구상이나 난상 (oval) 일 수 있다. 주사제로서 사용될때, 제제는 주상 또는 분말상인 경우가 빈번하다. 제제의 형태중 바람직한 것으로는, 예컨대, 원주상 또는 컬럼상과 같은 주상 및 구상과같은 과립상이 있다.
또한 본 발명의 비경구 제제의 크기는, 환자에게 과도한 고통을 주지 않는 한, 제형에 따라 선택될 수 있다. 주사제로서는, 제제가 주상 매트릭스일때, 그 크기는 11G 이하의 주사바늘을 사용하여 투여할 수 있는 것으로서, 예컨대, 약 3 mm이하의 직경과 약 30 mm 이하의 길이, 바람직하게는 약 1 mm 이하의 직경과 약 20 mm 이하의 길이, 더욱 바람직하게는 약 0.1 ~ 1 mm의 직경과 약 1 ~ 20 mm 의 길이로, 특히 바람직한 것은 원주상 또는 주상이다. 주사가능한 과립형 매트릭스의 그레인 또는 입자크기는 최대직경이 약 1 mm 이하, 바람직하게는 약 150 μm 이하 및 더욱 바람직하게는 약 1 ~ 100 μm이다. 매트릭스의 중량은 제제의 형태 또는 형상에 따라 선택할 수 있는데, 통상 주사제로서는 매트릭스의 중량은 예컨대, 약 40 mg 이하이고, 바람직하게는 약 1 ~ 25 mg 이다.
본 발명의 제제는 여러가지 방법으로 제조될 수 있는데, 폴리펩티드를 변성시키는 유기용매는 어떠한 것도 사용하지 않는 것이 바람직하다. 이와 같은 방법으로서 예컨대, 포화 지방산의 용융 또는 연화 폴리글리세롤 디에스테르와 생리활성의 폴리펩티드를 혼합하고, 생성된 용융 혼합물을 제제로 성형시키는 방법이 있다. 생리활성의 폴리펩티드가 수용액에서는 열역학적으로 불안정하다 할지라도, 동결건조분말과 같은 고체형태에서는 예상외로 안정하다. 따라서, 균일한 혼합용으로 건조분말과 같은 고체분말 또는 과립상의 생리활성 폴리펩티드를 사용하는 것이 바람직하다.
성형에서는, 제제의 형태 또는 형상에 따라 어떠한 성형방법도 사용될 수 있다. 예컨대, 주사제는 용융 혼합물을 바늘을 가진 주사기에 흡입시키고, 바늘로 부터 충진물을 압출하여 주상 생성물을 수득하는 방법 또는 용융 혼합물을 회전판 또는 디스크상에 적하하고, 적하물을 원심분리나 텀블링하여 구상 생성물을 수득하는 방법에 의해 제조할 수 있다. 또한 용융 혼합물을 분무하고, 분말 생성물을 냉각시켜 미립자상 제제를 제조하거나 펠렛과 같은 성형품을 제트밀과 같은 분쇄수단에 의해 미립자를 수득한다.
하기의 실시예 및 실험예는 단지 본 발명을 보다 상세히 기술하려고 한 것으로 본 발명의 범위를 한정하는 것으로 해석되어서는 아니된다.
실 시 예
실시예 1
테트라글리세롤 디팔미테이트 (300 mg ; 에스테르 결합수 2.0 ; Sakamoto Yakuhin Kogyo Co., Ltd., Japan 제조) 를 48 ℃ 에서 가열하여 용융시키고, 동결건조분말상 인터페론 - α 7.2 mg 을 첨가하였다. 용융 혼합물의 일부를 11G 의 주사바늘속에 흡입시키고, 실온으로 냉각시킨 후, 충진물을 바늘로 부터 압축 성형하여 원주상 매트릭스 제제 (직경 1 mm, 길이 10 mm, 중량 약 10 mg)를 수득하였다.
실시예 2
테트라글리세롤 디스테아레이트 (300 mg ; 에스테르 결합수 2.0 ; Sakamoto Yakuhin Kogyo Co., Ltd., Japan 제조) 를 58 ℃ 에서 가열하여 용융시키고, 이 용융물에 인터페론 - α의 동결건조분말 7.2 mg 을 첨가하였다. 용융 혼합물의 일부를 11G 의 주사바늘속에 흡입시키고, 실온으로 냉각시켰다. 충진물을 압출성형하여 원주상 매트릭스 제제 (직경 1 mm, 길이 10 mm, 중량 약 10 mg)를 수득하였다.
실시예 3
테트라글리세롤 디팔미테이트와 동결건조분말상 인터페론 - α 의 용융 혼합물을 실시예 1 과 동일한 방법으로 제조하였다. 용융 혼합물을 1 ml - 주사기 (Terumo Co., Ltd., Japan) 에 흡입시키고, 온도를 50 ℃ 로 유지시킨 상태에서, 27 G 의 주사바늘 (Terumo Co., Ltd., Japan) 로 부터 압출시키면서 공기총 ( Hakuko Co., Ltd., Japan) 으로 매트릭스를 분무하여 마이크로스피어 (microspheres) 를 수득하였다. 마이크로스피어를 체 (16 메쉬)에 통과시켜 직경 1 mm 이상의 과립물을 제거하였다.
실시예 4
48 ℃ 로 가열하여 용융시킨 후, 용융된 테트라글리세롤 디팔미테이트 (300 mg ; Sakamoto Yakuhin Kogyo Co., Ltd., Japan) 에 인터류킨 - 2 의 동결건조분말 1.5 mg 을 가하였다. 용융 혼합물의 일부를 11 G 의 주사 바늘에 흡입시키고, 실온으로 냉각시킨 후, 바늘로 부터 충진물을 압출성형하여 직경 1 mm, 길이 20 mm 및 중량 약 20 mg 을 가지는 원주상 매트릭스 제제를 수득하였다.
실시예 5
테트라글리세롤 디팔미테이트 (300 mg ; Sakamoto Yakuhin Kogyo Co., Ltd., Japan) 를 48 ℃로 가열하여 용융시키고, 이 용융물에 인슐린의 동결건조분말 1.5 mg 을 가하였다. 용융 혼합물을 주사기를 사용하여 11 G 의 주사 바늘에 흡입시키고, 실온으로 냉각하였다. 바늘로 부터 충진물을 압출성형하여 원주상 매트릭스 제제 (직경 1 mm, 길이 20 mm, 중량 약 20 mg) 를 수득하였다.
비교예 1
테트라글리세롤 디팔미테이트 대신에 글리세롤 모노팔미테이트 (에스테르 결합수 1.0 : Riken Vitamin Co., Ltd., Japan) 를 사용한 점을 제외하고는 실시예 1과 동일한 방법을 실시하여 원주상 매트릭스 제제 (직경 1 mm, 길이 10 mm, 중량 약 10 mg) 를 수득하였다.
비교예 2
테트라글리세롤 디팔미테이트 대신에 디글리세롤 모노팔미테이트 (에스테르 결합수 1.0 ; Sakamoto Yakuhin Kogyo Co., Ltd., Japan) 를 사용하고 실시예 1 의 공정을 수행하여 직경 1 mm, 길이 10 mm 및 중량 약 10 mg 인 원주상 매트릭스 제제를 수득하였다.
비교예 3
테트라글리세롤 디스테아레이트 대신에 글리세롤 모노스테아레이트 (에스테르 결합수 1.0 ; Takeda Chemical Industries, Ltd., Japan) 를 사용한 점을 제외하고는 실시예 2 의 방법과 동일하게 실시하여 원주상 제제 (직경 1 mm, 길이 10 mm, 중량 약 10 mg) 를 수득하였다.
비교예 4
테트라글리세롤 디스테아레이트 대신에 디글리세롤 모노스테아레이트 (에스테르 결합수 1.0 : Sakamoto Yakuhin Kogyo Co., Ltd., Japan 제조) 을 사용한 점을 제외하고는 실시예 2 의 공정을 반복 실시하여 직경 1 mm, 길이 10 mm 및 중량 약 10 mg 인 원주상 매트릭스 제제를 수득하였다.
비교예 5
테트라글리세롤 디스테아레이트 대신에 테트라글리세롤 모노 스테아레이트 (에스테르 결합수 1.0 : Sakamoto Yakuhin Kogyo Co., Ltd., Japan 제조) 를 사용한점을 제외하고는 실시예 2 와 동일한 공정을 실시하여 원주상 메트릭스 제제 (직경 1 mm, 길이 10 mm 및 중량 약 10 mg) 를 수득하였다.
비교예 6
테트라글리세롤 디스테아레이트 대신에 테트라글리세롤 트리스테아레이트 (에스테르 결합수 3.0 ; Sakamoto Yakuhin Kogyo Co., Ltd., Japan) 를 사용하고 실시예 2 의 공정을 실시하여 원주상 제제 (직경 1 mm, 길이 10 mm, 중량 약 10 mg) 를 수득하였다.
실시예 6
테트라글리세롤 디팔미테이트 대신에 테트라글리세롤 디미리스테이트 (에스테르 결합수 2.0 ; Sakamoto Yakuhin kogyo Co., Ltd., Japan) 를 사용한 점을 제외하고는 실시예 2 와 동일한 방법을 실시하여 원주상 제제 (직경 1 mm, 길이 10 mm, 중량 약 10 mg)를 제조하였다.
실험예
웅성 JCL-SD 쥐 (6 주생) 의 배부 (back) 피하에 11 G 의 주사바늘을 사용하여 실시예 1 과 2 및 비교예 1 내지 6 에서 수득한 매트릭스 제제를 각각 10 mg 투여하였다. 각각의 원주상 매트릭스 제제는 인터페론 - α 를 4 × 107국제단위 (IU) 함유하고 있다.
콘트롤로서, 인터페론 - α 의 4 × 107IU 를 함유하는 수용액을 사용하였다.
투여후, 시간이 흐름에 따라 꼬리정맥으로 부터 0.6 ml의 혈액을 채취하여 혈청 샘플을 수득하였다. 세마리의 쥐로 부터 각각 혈청 샘플을 채취하고, 각 혈청샘플에서 인터페론- α의 농도를 2 류의 항 인터페론 - α 항체를 사용한 샌드위치 ELISA 에 의해 측정하고, 평균치를 계산하였다. 표준 인터페론 - α의 단위로, 칸페론 - TM (Takeda Chemical Industries, Ltd., Japan) 을 사용하였다. 투여후 시간 경과에 따라 혈청에서 인터페론 - α농도의 평균치는 표 1 과 2에 기재되어 있다.
표 1 과 2 에서, " ND " 는 " 검출 불가능 " 임을 의미한다.
표 1 및 표 2 로 부터 명백히 알 수 있듯이, 폴리글리세롤 고급 지방산 에스테르 중에서 테트라글리세롤 디팔미테이트와 테트라글리세롤 디스테아레이트를 사용하여 수득한 실시예 1 과 실시예 2 의 제제 각각은 1 주간 이상동안 인터페론 - α를 지속적으로 방출할 수 있고, 인터페론 -α의 방출기간은 비교예의 제제보다 2배 이상 길다.
Claims (18)
- 생리활성 펩티드 또는 단백질 및 16 내지 30 개의 탄소 원자를 갖는 포화 지방산의 폴리글리세롤 디에스테르를 함유하는 매트릭스로 구성된 서방성 비경구 투여제제.
- 제 1 항에 있어서, 폴리글리세롤의 평균 중합도가 4 인 서방성 비경구 투여제제.
- 제 1 항에 있어서, 폴리글리세롤의 평균 중합도가 4 이고, 포화 지방산의 탄소원자수가 16 - 30 개인 서방성 비경구 투여제제.
- 제 1 항에 있어서, 생리활성 펩티드 또는 단배질이 상기 디에스테르 중에 분산된 서방성 비경구 투여제제.
- 제 1 항에 있어서, 생리활성 펩티드 또는 단백질의 평균 분자량이 2,000 달톤 이상인 서방성 비경구 투여제제.
- 제 1 항에 있어서, 생리활성 펩티드 또는 단백질이 인터페론, 인터류킨 또는 인슐린인 서방성 비경구 투여제제.
- 제 1 항에 있어서, 상기 포화 지방산의 탄소원자수가 16 ~ 22 개인 서방성 비경구 투여제제.
- 제 1 항에 있어서, 상기 포화 지방산이 팔미트산 또는 스테아르산인 서방성 비경구 투여제제.
- 제 1 항에 있어서, 생리활성 펩티드 또는 단백질의 비율이 매트릭스를 기준으로 0.0001 ~ 50 중량 % 인 서방성 비경구 투여제제.
- 제 1 항에 있어서, 상기 매트릭스가 주상 (pillar) 또는 입상 (granular) 인 서방성 비경구 투여제제.
- 제 1 항에 있어서, 상기 매트릭스가 이식용의 고형 주사제인 서방성 비경구 투여제제.
- 평균 분자량이 5,000 ~ 1,000,000 달톤인 생리활성 펩티드 또는 단백질과, 평균 중합도가 4 인 폴리글리세롤과 16 ~ 22 개의 탄소원자를 가지는 포화 지방산의 디에스테르를 함유하는 매트릭스로 구성된 제제로서, 매트릭스를 기준으로 하여 0.001 ~ 20 중량 % 의 생리활성 펩티드 또는 단백질이 디에스테르 중에 분산되며,피하 또는 근육내에 투여할 수 있는 매트릭스로 구성된 서방성 비경구 투여제제.
- 제 12 항에 있어서, 상기 포화 지방산이 팔미트산, 스테아르산 또는 베헨산인 서방성 비경구 투여제제.
- 제 12 항에 있어서, 상기 디에스테르의 융점이 40 ~ 60℃ 이고, 상기 매트릭스가 실온에서 고체인 서방성 비경구 투여제제.
- 제 12 항에 있어서, 생리활성 펩티드 또는 단백질이 인터페론인 서방성 비경구 투여제제.
- 생리활성 펩티드 또는 단백질과 포화 지방산의 용융 또는 연화된 폴리글리세롤 디에스테르를 혼합하고, 용융 혼합물을 성형시키는 것으로 구성된 서방성 비경구 투여제제의 제조방법.
- 제 16 항에 있어서, 용융 혼합물을 주상 또는 입상으로 성형하는 서방성 비경구 투여제제의 제조방법.
- 제 16 항에 있어서, 건조시킨 분말상의 생리활성 펩티드 또는 단백질을 용융 또는 연화된 디에스테르와 혼합시키는 서방성 비경구 투여제제의 제조방법.
