본원 염들의 제약적 용도는 노인성 질환, 특히 대뇌대사 변화에 관련된 질환들 및 혈류감소에 관련된 질환들의 치료용이다.

본원은 신규의 알카로이드 유도체들, 이들의 생산방법과 이들을 함유한 신규의 제약적 조성물들 및 투약형태들에 관한다. 보다 구체적으로, 본원은 노인성 질병 (구체적으로, 대뇌대사 변화 및 혈류감소에 관계된 질환들) 치료에 효과적인 인돌 알카로이드 신규염의 치료적 용도에 관한다.
많은 천연 알카로이드, 합성 알카로이드 유사물 및 천연 알카로이드와 알카로이드 유사물의 유도체들은 여러가지 병리학적 질환들의 치료용 및 또한 건강해 보이는 환자들을 바람직한 생리적 상태들로 유도하기 위한 제약적으로 확대된 용도로 사용된다. 예를들면, 알카로이드들은 심장 혈관분야, 산과학 및 신경학에 사용되어 왔다.

단계 (i) 에 의해 생산된 에스테르 유도체들은 C1-10카복실산과 반응하여 에스테르 그룹들로 전환되는 하나이상의 -OH그룹들을 가질 것이거나 또는 -COOR그룹들 (R 이 C1-6알킬인) 로 전환되는 -COOH 그룹들을 가질 것이다. 이와 유사하게 단계 (iv) 에서는 H - N ≡ 그룹이 RN =그룹 (R 은 상기에서 정의된 바와같은) 으로 전환될 것이고 단계 (x) 에서는 OH그룹이 -OR그룹(R은 상기에서 정의된 바와같은) 으로 전환될 것이다.
"합성 알카로이드 유사물, 및 천연 알카로이드 유도체 " 란 상기의 (i) 내지 (x) 수정 단계들중 하나이상을 거치게되는 천연 알카로이드를 포함하고자 하는 의도에서 사용하였다.
광범위하게 사용되는 일반적인 알카로이드들로는 빈카민 및 아포빈카민과 이의 유도체들, 라우바신, 아포쿠안진, 니세르골린 및 히데르긴이 있다.
유용한 알카로이드들 및 알카로이드 유도체들의 결점은 이들이 일반적으로 매우 높은 친수성을 지니며 따라서 지질상으로 분배되기에 적합한 투약 형태로 즉시 전환되지 않는다는 것이다. 또찬 유용한 투약 형태들은 경피성 투여용 또는 편리한 서방형 조제용으로도 그리 적합하지는 않다.
본원은 이제 상기 결점들을 피한 신규 부류의 알카로이드 유도체들을 개발하였다.
따라서 본원에 따르면, 본원은 포스파티드 산들의 염들 및 제약적으로 활성인, 천연 또는 합성 알카로이드들, 또는 알카로이드 유도체들을 제시하고 있다.
본원 염들은 하기 식에 의해 표현될 수 있을 것이다.

PA 는 포스파티드산에서 유도된 음이온 또는 여러 포스파티드산들의 혼합물에서 유도된 음이온을 뜻하며,
x : y 는 2 : 1 내지 1 : 2 임.
x : y 의 비는 바람직하게는 1.2 : 1 내지 1 : 1.2, 가장 바람직하게는 1.1 : 1 내지 1 : 1.1 이다.
본원의 신규염들에 있어서, 제약적으로 활성인 천연 또는 합성 알카로이드, 또는 알카로이드 유도체는 인돌 알카로이드, 특히 앞에서 언급한 부류들중 하나이다.
여기서 사용한 "포스파티드산"이란 하기 식을 갖는 화합물로 표현할수 있다.
식중,
R1및 R2(각기 같거나 다를 수 있는)는 각기 C10-24알킬그룹, C10-24알케닐 그룹, 또는 C10-24알카디에닐 그룹임 (상기 그룹들은 일반식 CnH2+1, CnH2-1또는 CnH2-3으로 표현가능함).
바람직하게는, n은 15내지 24, 더 바람직하케는 15 또는 17이다.
보다 구체적으로, 본원은 경구, 좌약 또는 경피성 투여시 생물학적 효능성이 증대됨을 보여주는 염기성 인돌 알카로이드들과 천연 또는 합성 포스타티드 산들의 신규염들에 관한다. 하기에서 설명된 이유들 때문에, 본원에 따른 신규 물질들에 의해 선택된 투여방법으로는 경피적 방법들이 쓰인다.
본원에 따른 특히 유용한 부류의 알카로이드 염들은 주로 말초혈류에 작용하는 알카로이드들 (대뇌 대사를 개선시키거나 또는 대뇌 혈류를 증가시키는 빈카민, 에틸아포빈카미네이트 및 이의 유사물들, 라우바신, 니세르골린 및 에르고트 알카로이드들)에서 제조된 것들이다.
본원의 또다른 양상에 따르면, 본원은 본원에 따른 염 (바람직하게는 상기에서 언급한 바람직한 염들중 하나) 및 제약적으로 용인할만한 부형제로 이루어지는 제약적 조성물들을 제시한다.
본원에 따른 염들은 자신들의 제약적으로 활성인 물질 (즉 제약적으로 활성인, 천연 또는 합성 알카로이드, 또는 알카로이드 유도체)을 일정시간에 걸쳐 방출시킬 수 있는 (특히, 제약적으로 활성인 물질을 경피적으로 방출시킬 수 있는) 서방향의 제약적 투약형태로 조제하기에 특히 적합하다.
이러한 목적을 위하여, 서방형 투약 형태들은 피부에 대는 패취 (Patch), 깁스 또는 붕대 형태가 바람직하다.
본원에 따른 신규염들은 매우 친유성이 크며 경구적 또는 경피적 투여시 탁월한 생물학적 효능성을 나타낸다. 신규염들은 깁스 또는 적당한 겔 또는 적당간 제약적 형태들로 합체되어 뇌의 말초 순환장애 및 (증가된 혈류가 기관작용을 개선시켜주는) 몸의 다른 부위들 치료에 특히 유용하다. 그 제품들 또한 표면 및 신체심부 양자의 미소순환 장애 치료용으로 피부의학 및 화장품분야에서 사용하기에 적합하다.
본원의 염들은 (a) 제약적으로 활성인, 천연 또는 합성 알카로이드, 또는 알카로이드 유도체 및 (b) 하나이상의 포스파티드 산들 (상기 반응물들 (a) 및 (b) 각각은 유리형태 또는 염형성 유도체 형태로 존재함) 의 염화반응으로 이루어지는 방법으로 제조할수 있을 것이다.
염화반응은 바람직하게는 양 반응물들에 적합한 용액내에서 수행된다.
용제는 바람직하게는 할로겐화된 하이드로카본들, 케톤들, 에테르들 및 이들의 혼합물들에서 선택된 것이다.
염화제들로 사용하기에 적합한 포스파티드 산들로는 천연 또는 합성 포스파티드 산들이 있다. 일반적으로 이들은 에스테르 결합을 경유하여 글리세롤의 산소원자에 결합되는 아실쇄들 (같거나 다를수 있고 포화 또는 불포와일수 있는) 을 지닌다. 자신들이 얻어지는 식물 또는 동물 세포조직 화합물들의 천연비에 해당하는 비로 존재하는 여러 지방산들을 갖는 화합물들로 이루어지는 천연 포스파티드산들이 포함된다.
염화반응은 통상 비양성자성 용제들내에서 수행되며 일반적으로 출발물질은 유리염기 형태 또는 약산으로 염화된 알카로이드들 및 유리 포스파티드 산 (들) 이다. 그와는 달리, 염화반응은 음이온 X를 가진 알카로이드 염이 양이온 Y를 갖는 포스파티드산 염 (Y. X염은 본질적으로 반응매질에 불용성임)과 반응하는 염교환 반응 (또는 이중분해 반응) 단계를 거쳐 제조될수 있을 것이다. 예를들면, 알카로이드의 하이드로클로라이드와 포스파티드 산 (들) 의 소듐 또는 포타슘염의 반응이다.
그렇게 얻어진 제약적으로 활성인, 천연 또는 합성 알카로이드들, 또는 알카로이드 유도체들은 비양성자성 용제들에 매우 잘 용해될수 있고, 이들은 농축 및 필요하다면 증발시켜 건조시키거나, 또는 핵산 또는 페트롤륨 에테르와 같은 비용제내에서 불용화시켜 분리시킬수 있다.
염들은 통상 시약들을 몰비 (1M : 1M) 로 반응시켜 수득할수 있다. 염화반응은 일반적으로 시약들이 선택된 용제내에 완전히 용해될때 완료된다.
물로 처리한 경우, 본원의 신규염들은 마이셀 형태를 형성하여 비혼화성의 친유성 유기 용제들내에 자신들이 양적으로 분포될수 있다. 본원의 염들은 가장 극성인 중심핵들(즉, 제약적으로 활성인, 천연 또는 합성 알카로이드, 또는 알카로이드 유도체로 구성되는 분자부위) 주위를 감싸는 아실쇄들의 능력에서 자신들의 친유성을 얻게되며, 그 결과로서 수성매질내에서 리포좀의 미소분산을 형성할수 있게된다고 사료되어진다.
본원의 신규염들을 겔화된 수성 미소분산물들 (리포겔들) 의 형태로, 또는 도포된 조절 방출형 깁스에 합체된 형태로 몸의 여러 부분에 투여할때 이들은 알카로이드의 종래 투약 형태들에 비해 상당한 제약적 이점들을 나타냄을 놀랍게도 알게되었다. 이러한 투약형태에서 본원에 따른 신규염들은 자신들의 활성에 양성적인 효과들을 수반하는, 다른 수준의 생물학적 효능성을 가짐을 보여준다.

개인의 수용기 감응에 따라 한번 또는 그이상의 1 일 복용량으로서 5 내지 50mg 사이의 투약량의 신규염을 상기에서 기술한 부위에 도포할 수 있다. 만성적 퇴행성 질환에 대해서도 동일한 결과를 얻고자 한다면, 요구하는 효과를 나타내기 위하여 목표기관에 충분한 양의 약을 일정하게 공급해야 한다는 것은 명백한 것이다. 이것은 본원에 따른 염들을 이용하여 달성되어 질것이다.
조절방출형 경피성의 제약적 형태들은 본원의 신규염들의 투여용으로 적당하며, 이는 주위기관들의 관련을 최소화하면서 목표기관에 직접적으로 약이 적용가능하게 해주기 때문이다. 조절방출형 깁스가 이러한 목적 달성용으로 특히 효과적인데 이는 5내지 50mg 사이의 여러 알카로이드 염들을 보존제와 합체시킬 수 있기때문이다. 종래의 인지질의 존재 또는 부재하에 리포겔의 형태로 도포된 상기와 동일한 염의 리포좀 형태 또한 유효적임이 입증되었다. 또안 깁스형태가 장기 사용용 (본원 경우에 필수적인) 에 극히 실용적임이 입증되었다. 그러나 본원에 따른 염들은 또한 알약, 유액 (oily solutions), 좌약등과같은 다른 종래 형태로 합체 및 투여가능하다.
표면 및 심부 미소순환 장애들을 치료하기 의하여 사지와 같은 몸의 큰 면적 부위에 수성 미소분산물들 또는 리포겔들 또는 종래의 겔들 및 에멀젼들의 형태로 본원에 따른 염들을 도포할수 있다.
본원에 따른 염들은 Raynaud 병, 공피증 (scleroderma), 선단자람중 (acrocyanosis) 및 다른형태의 사지의 아테롬성 동맥경화증 (무거운 다리, 간헐성 파행, 등)을 치료하기 위하여 국소적으로 도포할수 있다. 소동맥성 (arterioles) 및 전소동맥성 (prearterioles) 모세관내의 근세포들에 작용하여 세포조직들내의 모세혈류를 증가시킴으로써 상기 형태의 질병들에 본 제품들이 유효하다.
본 제품들로서 노화에 기인한 아테롬성 동맥경화증에 의해 표제성 미세혈관 혈행이 손상된 질환들에 또 봉와직염 (cellulitis) 치료 목적의 화장품 응용물들이 있다.
본원에 따른 신규염들은 표면 세포조직내에 더 잘 분포되고 더 오랜시간 작용 부위에 현상 유지될수 있기때문에 상기 질환들에 있어서 염기성 알카로이드 (즉, 염 형성에 사용된, 제약적으로 활성인 천연 또는 합성 알카로이드, 또는 알카로이드 유도체) 보다 더 우수한 결과를 가져다준다.
하기 실시예들은 본원을 보다 더 잘 설명하고자 함이며, 본원에 대한 여하한 제한의 의도는 없음을 이해바란다.
[실시예 1] :
에틸 아포빈카미네이트 디팔미토일 글리세릴포스파티데이트 제조
에틸 아포빈카미네이트 염기 (3.50g)를 교반시키면서 메틸렌 클로라이드 (15ml) 에 용해시키고 주위온도에서 디팔미토일 글리세릴 포스파티드 산 (6.48g)을 가하였다.
시약들이 완전히 용해되었을때 메틸렌 클로라이드내 용액을 진공하에, 40℃를 초과하지않는 온도에서 증발 건조시켜 용제를 완전히 제거시켰다. 거기서 얻어진 잔류물을 5℃, n -헥산내에 분산시킨 다음 여과시켰더니 다음과 같은 특성을 지닌 백색고체 (9.95 g) 가 얻어졌다.
융점 : 약 50℃

[실시예 2] :
에틸 아포빈카미네이트 및 수소화된 소야 (soya) 포스파티드산 염의 제조
에틸 아포빈카미네이트 (3.50g)를 교반하면서 메틸렌 클로라이드 (15ml) 내에 용해시키고 산 -염기 적정에 의해 검측된 평균 분자량 698 인 수소화된 소야 포스파티드 산 (지방산들의 천연비로 존재하는) (7 g) 을 가하였다.
시약들이 완전히 용해되었을때 메틸렌 클로라이드내 용액을 진공하에, 40℃를 초과하지않는 온도에서 증발 건조시켜 용제를 완전히 제거시켰다. 거기서 얻어진 잔류물을 5℃, n -헥산내에 분산시킨 다음 여과시켰더니 다음과 같은 특성을 지닌 백색고체 (10.5 g) 가 얻어졌다.
융점 : 52.8℃

[실시예 3] :
니세르골린 및 수소화된 소야 포스파티드 산 염의 제조
에틸 아포빈카미네이트 (4.84g)를 교반하면서 메틸렌 클로라이드 (15ml) 내에 용해시키고 산 -염기 적정에 의해 검측된 평균 분자량 698 인 수소화된 소야 포스파티드 산 (지방산들의 천연비로 존재하는) (7g)을 가하였다.
시약들이 완전히 용해되었을때 메틸렌 클로라이드내 용액을 진공하에, 40℃를 초과하지않는 온도에서 증발 건조시켜 용제를 완전히 제거시켰다. 거기서 얻어진 잔류물을 5℃, n -핵산내에 분산시킨 다음 여과시켰더니 다음과 같은 특성을 지닌 백색고체 (11.7g) 가 얻어겼다.
융점 : 150 - 190℃ (dec.)

[실시예 4] :
빈카민 및 수소화된 소야 포스파티드 산염의 제조
빈카민 염기 (3.54g)를 교반하면서 메틸렌 클로라이드 (15ml) 내에 용해시키고 산-염기 적정에 의해 검측된 평균 분자량 698인 수소화된 소야 포스파티드산(지방산들의 천연비로 존재하는) (7g)을 가하였다.
시약들이 완전히 용해되었을때 메틸렌 클로라이드내 용액을 진공하에, 40℃를 초과하지 않는 온도에서 증발 건조시켜 용제를 완전히 제거시켰다. 거기서 얻어진 잔류물을 5℃, n -헥산내에 분산시킨 다음 여과시켰더니 다음과 같은 특성을 지닌 백색고체 (10.5g) 가 얻어졌다.

[실시예 5] :
니세르골린 디팔미토일 - 글리세릴 포스파티데이트의 제조
니세르골린 염기(4.84g)를 교반시키면서 메틸렌 클로라이드(50ml)에 용해시키고 주위온도에서 디팔미토일 글리세릴 포스파티드 산 (6.48 g) 을 가하였다.
시약들이 완전히 용해되었을때 메틸렌 클로라이드내 용액을 진공하에, 40℃ 를 초과하지않는 온도에서 증발 건조시켜 용제를 완전히 제거시켰다. 거기서 얻어진 잔류물을 51℃, n -헥산내에 분산시킨 다음 여과시겼더니 다음과 같은 특성을 지닌 백색고체 (11.1g) 가 얻어졌다.

[실시예 6] :
라우바신 및 수소화된 소야 포스파티드산 염의 제조
라우바신 염기(3.52g)를 교반하면서 메틸렌 클로라이드(100ml)에 용해시키고 산 -염기 적정에 의해 검측된 평균 분자량 698 인 수소화된 소야 포스파티드 산(지방산들의 천연 비로 존재하는) (7g)을 가하였다.
시약들이 완전히 용해되었을때 메틸렌 클로라이드내 용액을 진공하에, 40℃를 초과하지않는 온도에서 증발 건조시켜 용제를 완전히 제거시켰다. 거기서 얻어진 잔류물을 5℃ n - 헥산내에 분산시킨 다음 여과시켰더니 다음과 같은 특성을 지닌 백색고체 (10.3 g) 가 얻어졌다.

[실시예 7] :
수소화된 소야 포스파티드산의 니세르골린 염 함유 경피적 흡수용 깁스 제조
수소화된 소야 포스파티드산의 니세르골린염을 약 방출을 조절하는 접착물질과 합체시켰다. 이의 조성은 하기와 같다 :
경피적 깁스는 3 개의 층 (즉, 안쪽의 지지층, 몇가지 피복물들이 도포되어 고착된 활성 성분을 함유한 접착막 및 외피층)으로 구성되어 있다. 약은 지속적인 투여를 위해서 24시간 동안에 걸쳐 방출된다.
[실시예 8] :
에틸 아포빈카미네이트 및 수소화된 소야 포스파티드산 염함유 리포겔의 제조
100kg의 리포겔을 제조하기 의하여 사용된 조제물은 하기 시약들로 이루어진다 :
Claims (11)
- 하기 식을 갖는 염.식중,Alk 는 빈카민, 아포빈카민, 라우바신, 아포쿠안진, 니세르골린 및 이들의 염 및 에스테르로 구성된 그룹으로부터 선택된 알카로이드에서 유도된 양이온이고,PA 는 하기식에 의해 표현되는 포스파티드산 또는 포스파티드산의 혼합물이고,[식중, R1및 R2는 서로 같거나 다를수 있고, 각각 C10-24알킬그룹, C10-24알케닐 그룹, 또는 C10-24알카디에닐 그룹임]x : y 는 1.1 : 1 내지 1 : 1.1의 범위 내에서 실질적으로 동일한 몰비임.
- 제1항에 있어서, 알칼로이드가 에틸 아포빈카미네이트인 염.
- 제 1항에 있어서, Alk가 빈카민, 아포빈카미네이트, 라우바신, 아포쿠안진 및 니세르골린으로 구성된 그룹으로부터 선택된 알카로이드에서 유도된 양이온인 염.
- 하기 식을 갖는 염.식중,Alk 는 빈카민, 에틸 아포빈카민, 라우바신, 아포쿠안진 및 니세르골린으로 구성된 그룹으로부터 선택된 알카로이드에서 유도된 양이온이고,PA 는 하기식에 의해 표현되는 포스파티드산 또는 포스파티드산의 혼합물이고,[식중, R1및 R2는 서로 같거나 다를수 있고, 각각 C10-24알킬그룹, C10-24알케닐 그룹, 또는 C10-24알카디에닐 그룹임]x : y 는 1.1 : 1 내지 1 : 1.1의 범위 내에서 실질적으로 동일한 몰비임.
