Abstract
본 발명은 하기 일반식(1)의 (2R-5S) 또는 (2S-5R) 시스(cis)구조를 가지는 5-(시토시닐-1) 1, 3-옥사티올란에 관한 것으로서,
상기 식에서, R은 질소원을 함유한 단일환 또는 2환식의 헤태로 고리로부터 유래되는 아실 라디칼 또는 아르알킬 라디칼이고, 제2위치의 히드록시메틸기는 2개의 위치 2 및 5에 의해 결정된 평면을 기준으로 시스-위치에 있다. 상기 본 발명의 화합물은 항-바이러스 작용을 가지는 제약 조성물의 활성성분으로서 유용하다.
Description
본 발명은 유기화학 분야에 관한 것으로서, 보다 상세하게는 의화학분야에 관한 것이다. 보다 구체적으로 본 발명은 제2위치가 옥사티올란 고리로 치환된 2', 3'-디데스옥시누클레오티드를 기재로 하는 의화학 분야에 관한 것이다.
구체적으로, 본 발명은 하기 일반식(1)의 (2R-5S) 또는 (2S-5R) 시스(cis) 구조를 가지는 5-(시토시닐-1)1, 3-옥사티올란에 관한 것으로서,
상기 화합물은 하기 두개의 공간 구조식중 하나로 표현될 수 있다 :
상기 식들에서, R은 질소원을 함유하는 모노시클릭 또는 비시클릭 헤테로고리로부터 유도된 아실 라디칼 또는 아르알킬 라디칼이고, 제2위치의 히드록시메틸기는 위치 2 및 5의 두 위치에 의해 결정되는 평면을 기준으로 시스 위치에 있다.
치환체 R에 의해 부여되는 여러 정의 중에서도, 하기 사항이 두드러질 수 있다 :
a) 헤테로고리가 피리딘 구조이고 아실 부분이 2, 3 또는 4 위치에 있는 하기 일반식(1a)의 니코틴 유도체, 또는 이것과 무기 또는 유기산가의 산부가염 :

g) 하기 일반식(1g)의 (디히드로피리딜)알킬 유도체, 또는 이것과 무기 또는 유기산과의 산부가염 :
상기 식에서, R1, X 및 n은 상기 정의한 바와 같고, m은 1 내지 6의 정수이다.
식(1)의 화합물은 자체의 광학적 활성 이성질체로 분리될 수 있다. 따라서, 상기 화합물은 (+)이성질체와 (-)이성질체로 얻어질 수 있다. 레트로바이러스 감염은 심각한 감염의 원인이 되기도 하는데, 후천성 면역결핍증(AIDS)의 감염은 치사에 이르는 것이고, 이보다 덜한 정도가 간염이다. 누클레오시드 또는 헤테로-누클레오시드 유형의 몇가지 화합물은 상기 레트로바이러스의 감염을 치료하는데 현재 임상적으로 사용되고 있다. 누클레오시드 유형의 화합물은 유럽 특허출원 90301335.7호에 개시된 바, AZT(3'-아지도-2', 3'-디데스옥시티미딘)(Proc. Natl. Acad. Sci. 82, 7096-7100, 1985), ddC(2', 3'-디데스옥시시티딘)(Proc. Natl. Acad. Sci. 86, 1911-15, 1986), d4T(2', 3'-디데히드로-3'-데스옥시티미딘)(Biochem. Biophys. Res. Comm. 142, 128-34, 1987), ddl(2', 3'-디데스옥시이노신)(Antiviral Chem. Chemother 2, 221, 1991), BHC-189 또는 3TC(2', 3'-디데옥시-3'-티아시티딘)의 유도체가 있다. 당업자에게 자명한 항레트로바이러스 화합물의 제한된 수 및 제한된 효능이 치료적 측면에서 효능이 증가되고 부작용이 감소된 신규한 화합물의 개발을 제기하였다.
2-[(1-히드록시메틸) 1, 3-옥사티올란-5-일]시토신의 일부 유도체, 즉 히드록시메틸 작용기를 가지는 에스테르류(유럽 특허출원 0 382 526) 또는 피리미딘 고리의 5' 위치가 할로겐화된 유도체는 이미 공지되어 있다(R. F. Schinazi 등, Antimicrob Agent and Chemotherapy 36(1992) 2423-2431).
본 발명의 화합물은 사람의 레트로바이러스, 특히 VIH 및 B형 간염바이러스(HBV)의 복제를 억제할 수 있는 능력을 가지고 있다. 상기 화합물은 BHC-189 또는 TC-3으로부터 유도되고, 시티딘의 엑소시클릭 아미노기에 결합된 1, 4-디히드로 1-메틸 3-퀴놀레일카르보닐기를 구조내에 포함하고 있다. 상기의 특별한 구조체는 혈액뇌 격막의 침투를 증가 또는 촉진시키는 능력을 실험 약리학에 제공한다(Pharmacol. Ther. 19, 337-396, 1983-Methods Enzymol 112, 381-396, 1985-Drug. Des. Del, 1, 51-64, (1986), J Med. Chem. 31, 244-249, 1988-J. Med Chem. 32, 1782-1788, 1989-J. Med Chem. 32, 1774-1781, 1989).
상기 화합물의 화학적 합성 방법은 전통적인 실험 프로토콜을 이용하여 수행되어 왔다. 상기 신규의 헤테로누클레오티드를 수행하는데 2', 3'-디데스옥시 3'-티아시티딘을 출발물질로서 필요로 하며, 이에 대해 문헌상에 몇가지 합성방법이 보고되어 있다[참조 : J. Org. Chem. 56, 6503, 1991-J. Org. Chem. 57, 2217, 1992-Tet. Lett. 33, 4625, 1992-Nucleoside and Nucleotide 12, 225, 1993]. 본원에서는 상기 유도체로부터 출발하여, 본 발명의 대상이 되는 화합물 및 이와 관련된 중간 물질의 합성에 관해서도 설명하기로 한다.
본 발명은 또한 5-(시토시닐-1-) 1, 3-옥사티올란(시스)을 일반식 ROH의 카르복실산의 기능적 유도체의 작용하에, 또는 일반식 R'Z의 알킬화 활성 유도체의 작용하에 제공하는 것으로 이루어지는 일반식(1)의 화합물의 제조방법에 관한 것으로서, 상기 식에서 R은 방향족 또는 디히드로방향족의 모노시클릭 또는 비시클릭 질소원 함유 헤테로고리로부터 유도된 아실 부분이며, R'은 방향족 또는 디히드로방향족의 모노시클릭 또는 비시클릭의 질소원 함유 헤테로고리로부터 유도된 아르알킬 라디칼이며, Z는 쉽게 유리될 수 있는 불안정한 라디칼이다.
보다 구체적으로는, 산 ROH의 기능적 유도체는 할로겐화물, 무수화물, 카보디이미드나 예컨대 페놀에스테르와 같은 활성 에스테르로부터 얻어진 혼합된 무수화물이다.
아랄알킬화 유도체의 경우에, Z는 할로겐 원자이거나 알킬- 또는 아릴술포닐 라디칼이다.
본 발명은 또한 하나 이상의 상기 일반식(1)의 화합물 또는 이것에 유기산이나 무기산을 반응시킨 산부가염을 주성분으로 함유하고, 이를 국소적 또는 전신적 사용에 적합한 비활성, 무독성의 제약학적으로 적합한 담체 또는 비히클과 혼합 또는 결합시킨, 항바이러스 작용을 가지는 제약 조성물에 관한 것이다.
상기 본 발명의 조성물에 있어서, 일반식(1)의 활성 성분의 함량은 투여 경로의 기능에 따라 단위 투여량 당 0.1 내지 100mg 범위이다.

질소 분위기 하의 60℃에서, 피리딘(3ml)중의 화합물 5의 용액(56mg, 0.12mM)에, 2당량의 p. 톨루엔 술폰산 클로라이드(46mg, 0.24mM)를 첨가하였다. 혼합물을 실온에서 20시간 동안 교반한 후 증발시킨 다음, 5% 시트르산 용액(10ml)으로 세척한 후, 에틸 아세테이트(3×10ml)로 추출하였다. 합쳐진 유기상을 Na2SO4상에서 건조시킨 다음, 증발시켰다. 48mg의 목적하는 화합물이 분리되었다.

5ml의 무수 DMF 중의 2', 3'-디데스옥시-3'-티아시티딘(50mg, 0.2mM)에 1.1 당량의 BOP(93mg, 0.23mM), 1당량의 니플룸산(2-a, a, a-트리플루오로-m-톨루이디노 니코틴산) 및 4당량의 DIEA(146㎕, 0.84mM)을 첨가하였다. 혼합물을 밤새 실온에서 교반하고, 5% 시트르산 용액(10ml)로 세척한 다음 5% 중탄산나트륨 수용액(10ml)로 세척하였다. 형성된 혼합물을 에틸 아세테이트(3×10ml)로 추출하고 황산나트륨 상에서 건조시킨 후, 증발시켰다.
잔류물을 실리카겔 칼럼(용리액 : 에틸 아세테이트/톨루엔 1 : 9) 상에서 크로마토그래피에 의해 정제하여 백색 고체를 얻었다(49mg, 수율 48%).
항바이러스 특성 평가 시험
항HIV시험-실험 프로토콜
a) 일반적 사항
장치 : 벡크만형 TL 100 초원심분리기를 사용하였다. 방사능 표지된 입자의 계수는 휴렛 팩커드사 장치 트리 카브 모델(Tri Carb Model) 1600으로 수행하였다. 신시티아(syncithia)는 전화 현미경 라보버트(invert microscope Lavobert)로 관찰하였다.
용해 완충액 NTE의 조성은 다음과 같다 :

b) 세포배양물에 있어서의 항바이러스 특성 평가시험
항바이러스 작용 평가는 세포계 MT4 상의 HIV-1 바이러스의 세포병원성 효과의 연구를 기초로 하였다. 세포계 MT4는 환자로부터 분리된 원래의 T세포로서 TLHV-I 바이러스에 의해 변형되었다. HIV-1 바이러스의 세포병원성 효과는 현미경으로 관찰가능한 "신시티아(syncithia)"로 불리우는 다핵형성 거대 세포의 형성에 의해 나타났다. 이러한 HIV-1 효과는 감염 후 4 내지 5일에 관찰되었다.
세포병원성 효과는 바이러스에 의해 세포의 감염, 이의 세포내 복제 및 세포에 의해 바이러스 항원의 발현에 직접적으로 상관된다. 따라서, 이러한 효과의 억제는 HIV-1 바이러스 증식의 억제에 상응한다.
MT4 세포를 RPMI 1640 배지(30분 동안 56℃에서 유지된 10%의 중에 보체제거된 송아지 태아 혈청(호르몬, 혈청 성장 인자 등), 1% 글루타민, 1% 페니실린 스트렙토마이신 및 바이러스의 세포 유착을 촉진시키는 2㎍/ml의 폴리렌(Polyren) 중에서 3.105세포/ml로 유지시켰다.
항바이러스제에 의한 치료 작용은 지속적이었다. 사실상, 이러한 효과는 바이러스 감염 전, 감염 동안 및 감염 후에도 있었다. 이어서, 10% 송아지 태아 혈청을 희석하여 MT4 세포가 8일 동안 배양되도록 하고, 신시티아의 형성을 관찰하였다.
MT4 시험
시험 전 : 3×106세포/100㎕를 96 웰을 가지는 마이크로역가(microtiter) 플레이트에 분배하고 2000rpm에서 30분 동안 원심분리시켰다. 이후, 펠릿을 1시간 동안 37℃에서 예비 인큐베이팅시키고, 다양한 농도의 항바이러스제 100㎕를 사용하여 시험하였다.
감염 : 100 TCIU(감염 단위)의 HIV-1 바이러스(이러한 역가의 HIV-1 바이러스는 4 또는 5일 후에 신시티아의 형성을 유도하도록 결정된 것이다)를 첨가하는 동안에 마이크로웰에서 감염이 이루어졌다. 항바이러스제는 100 TCIU에서의 바이러스 감염 및 희석시에 항상 존재하였다.
감염 후 : 1시간 동안 37℃에서 인큐베이션 후, MT4 세포를 RPMI 1640 배지로 3회 세척하고, 24 웰의 플레이트상에서 시험하려는 화합물의 각각의 농도를 ml당 3.105세포로 계산하여 배양하였다. 3일이 경과하였을 때, MT4 세포를 1/3로 희석하였다. 항바이러스제의 농도를 유지시켰다. 신시티아가 발생한 각각의 날을 현미경 하에서 관찰하여 대조군 HIV-1와 대해 선택적인 지연이 있는지 살펴보았다. 8일째에, 역전사 효소의 측정을 수행하였다. 따라서, 세포가 감염되지 않은 경우는 항바이러스제에 의해 보호되었다는 것이다.
역전사 효소의 측정
역전사 효소는 RNA 의존 DNA 폴리머라이제이다. 이 효소는 레트로바이러스를 복제할 수 있게 한다. 이로 인해, DNA로 전사된 바이러스 RNA는 세포 게놈에 혼입된다. 프로바이러스 DNA는 세포 효소에 의해 바이러스 RNA로 전사된다. 역전사 효소의 활성을 측정하기 위해, 1ml의 배양액의 상청액을 초원심분리기로 5분 동안 95000rpm으로 100배 농축시켰다. 초원심분리후 얻어진 바이러스 펠릿을 10㎕의 완충액 NTE+TRITON 0.1%(폴리옥시에틸렌 에테르)에 재현탁시켰다. 이를 바이러스를 용해시켜 역전사 효소를 방출시키는데 사용하였다. 시험관내에서, 역전사 효소의 활성이 입증되었다. 이 효소는 잔기가 12 내지 18개인 프라이머 올리고 dT를 가지는 합성 매트릭스 폴리 A를 사용한다. 이 반응의 방사능 표지된 기질은 [3H]dTTP(1mCi/ml에서의 방사 활성 티미딘 트리포스페이트)이었다. 새롭게 형성된 삼중수소 표지된 거대분자를 트리클로로아세트산으로 침전시키고, 여과에 의해 유리 [3H]TTP를 분리시켰다. 역전사 효소의 효소 활성은 착물인 폴리-rA/폴리-dT에 있는 방사성에 의해 측정하였다. 이러한 방사성 표지는 증폭기로서 작용하는 섬광 액체를 첨가한 후에 입자 계수로 측정하였다.
항 HBV 시험-실험 프로토콜
B형 간염(HBV)에 대한 화합물을 활성을 평가하기 위해 사용되는 수개의 방법 중에서, 오리의 B형 간염 바이러스(오리 HBV)패턴을 사용하였다[참조 : J. Med. Virology (1990) 31 82-89; Viral Hepatitis and Liver disease(1988) 506-509; Hepatology(1989) 10 186-191]. 화합물의 항 HBV활성을 시험관내 시험하기 위한 배양 물질은 오리 B형 간염 바이러스(DHBV)에 의해 감염된 오리 간세포로 이루어졌다[참조 : Antimicrob. Agents Chemother. (1989) 33 336-339; J. Med. Virology 40, 59-64; Antivir Res. (1993) 21, 155-171; Antimicrob. Agents chemother. (1993) 37 1539-1542]. 실제, 오리의 B형 간염 바이러스는 사람의 B형 간염 바이러스와 매우 유사한 것으로 인지되어 있다[참조; Viral Hepatitis and Liver Disease(1988) 526-529. Antiviral Research(1987) 8 189-199]. 표준화된 조건(Virology(1988) 171 564-572)하에서, 오리 간세포 배양액은 DHBV의 완전한 복제를 가능하게 하였다. 시험 화합물의 항바이러스 활성은 감염된 간세포를 10 동안 배양하여 생성된 바이러스 DNA의 양의 함수로서 측정되었다.
실험 프로토콜
간세포 생성을 위해 DHBV에 의해 감염된 3주령의 어린 수컷 오리를 사용하였다. 오리를 마취하여 치사시키고, 간세포는 길로우조(Guillouzou)(Research in isolated and cultured hepatocytes, J. Libbey Eurotex Ltd/INSERM, 1986)의 방법을 사용하여 분리시켰다. 상기 세포를 라보위츠(Leibowitz)의 배지와 5㎍/ml 소 인슐린 중에서 배양하고, 여기에 7×10-5몰 코티존 헤미숙시네이트 및 1.5% 디메틸술폭사이드(DMSO)를 첨가하였다. 세포의 농도는 8×106세포이었고, 이를 100×20mm의 배양 상자에 분무하였다. 세포를 분무시킨 후에 시험 화합물을 첨가하였다. 배지를 10일 동안 매일 새롭게 하였다. 세포 상청액중의 오리 B형 간염 바이러스의 바이러스 DNA 생성을 포렐 등에 따른 "도트 블럿(Dot blot)"(Viral Hepatitis and Liver Disease, 1988, 506-509; Hepatology, 1989, 10, 186-191)에 의해 측정하였다. 억제율은 누클레오티드 유도체로 처리되지 않은 DHBV에 의해 감염된 배양액과 관련하여 세포의 상청액에 존재하는 DNA양의 %로서 표현하였다.
바이러스학적 결과
HIV 시험
본 출원에 그의 합성이 기재되어 있으며, 관련 구조는 선행하는 종래 기술에 공지되어 있는 본 발명의 화합물은 감염된 세포가 MT4 형인 경우에 BRU HIV-1 바이러스(BRU는 시험된 바이러스 균주이다)의 복제를 억제하는 효과를 갖는다.
표 1에 이러한 시험 결과를 기재하였다. 칼럼 1은 연구한 가장 대표적인 화합물의 이름을 나타내고 있으며, 칼럼 2에서는 IC50값(감염 후 7일째에 측정된, 감염된 MT4 세포에서 HIV의 복제를 50% 억제하는 성분의 농도)을 기재하고 있다.
DBHV 시험
가장 대표적인 화합물에 대하여 상기 프로토콜 부분에 해당되는 부분의 오리 바이러스 HBV(DHBV)의 복제에 대한 효과를 시험하였다.

Claims (14)
- 하기 일반식(1)의 (2R-5S) 또는 (2S-5R) 시스(cis) 구조를 가지는 5-(시토시닐-1) 1, 3-옥사티올란 :상기 식에서, R은 질소원을 함유한 모노시클릭 또는 비시클릭의 헤테로고리로부터 유도된 아실 라디칼 또는 아르알킬 라디칼이고, 제2위치의 히드록시메틸기는 위치 2 및 5에 의해 결정되는 평면을 기준으로 시스 위치에 있다.
- 제1항에 있어서, 하기 일반식(1)의 (2R-5S) 구조를 가지는 5-(시토시닐-1) 1, 3-옥사티올란 :
- 제1항에 있어서, 하기 일반식(2)의 (2S-5R) 구조를 가지는 5-(시토시닐-1) 1, 3-옥사티올란 :
- 제1항 내지 제3항중의 어느 한 항에 있어서, 하기 일반식(1a)의 5-(시토시닐-1) 1, 3-옥사티올란 :상기 식에서, X는 수소, 할로겐 원자, 니트로기, 저급 알콕시 또는 트리플루오로메틸 라디칼이고, n은 1 내지 3의 정수이며, 아실 부분은 2, 3 또는 4위치에 있다.
- 제1항 내지 제3항중의 어느 한 항에 있어서, 하기 일반식(1b)의 5-(시토시닐-1) 1, 3-옥사티올란 :상기 식에서, R1은 탄소 원자가 1개 내지 10개인 알킬 라디칼이고, X 및 n은 제4항에서 정의된 바와 같으며, 아실 부분은 2, 3 또는 4 위치에 결합되어 있다.
- 제1항 내지 제3항중의 어느 한 항에 있어서, 하기 일반식(1c)의 5-(시토시닐-1)-1, 3-옥사티올란의 4차 유도체 :상기 식에서, A는 유기 또는 무기의 음이온이고, R1은 탄소 원자가 1개 내지 10개인 알킬 라디칼이고, X 및 n은 제4항에서 정의된 바와 같으며, 아실 부분은 2, 3 또는 4 위치에 결합되어 있다.
- 제1항 내지 제3항중의 어느 한 항에 있어서, 하기 일반식(1d)의 N-퀴놀레시닐 5-(시토시닐-1) 1, 3-옥사티올란 :상기 식에서, 카르보닐 라디칼은 제2, 3, 4 위치에 결합되어 있고, X는 수소, 할로겐, 트리플루오로메틸 라디칼, 저급 알콕시 또는 니트로기이고, n'은 1 내지 6의 정수이다.
- 제1항 내지 제3항중의 어느 한 항에 있어서, 하기 일반식(1e)의 N-디히드로퀴놀레시닐 5-시토시닐 1, 3-옥사티올란 :상기 식에서, R1및 X는 제4항에서 정의된 바와 같으며, n'은 1 내지 6의 정수이다.
- 제1항 내지 제3항중의 어느 한 항에 있어서, 하기 일반식(1f)의 N-4차 퀴놀레시닐 5-시토시닐 1, 3-옥사티올란 :상기 식에서, R1, X 및 n'은 제4항에서 정의된 바와 같으며, 아실기는 2, 3 또는 4 위치에 결합되어 있으며, A는 유기 또는 무기 음이온이다.
- 제1항 내지 제3항중의 어느 한 항에 있어서, 하기 일반식(1g)의 (N-디히드로피리딜 알킬) 5-시토시닐 1, 3-옥사티올란 :상기 식에서, R1, X 및 n은 제4항에서 정의된 바와 동일하며, m은 1 내지 6의 정수이다.
- 제1항에 따른 화합물의, 유기 또는 무기산과의 산부가염.
- 제1항에 따른 화합물의 광학 활성 이성질체.
- (시스) 2-(히드록시메틸)-5-[n4-(3"-피리딜 카르보닐)시토신 1'-일] 1, 3-옥사티올란, 또는 라세미 형태 또는 광학 활성 형태의 유기 또는 무기산과의 부가염인 제1항에 따른 화합물.
- 5-(시토시닐-1) 1, 3-옥사티올란(시스)을 일반식 ROH(여기에서, R은 방향족 또는 디히드로방향족의 모노시클릭 또는 비시클릭의 질소원 함유 헤테로고리로부터 유도된 아실 부분이다)의 카르복실산의 기능적 유도체의 작용하에, 또는 일반식 R'Z(여기에서, R'은 방향족 또는 디히드로방향족의 모노시클릭 또는 비시클릭 질소원 함유 헤테로고리로부터 유도된 아르알킬 라디칼이며, Z는 쉽게 유리될 수 있는 불안정한 라디칼이다)의 아르알킬화 활성 유도체의 작용하에 제공하는 단계를 포함하는, 제1항에 따른 화합물을 제조하는 방법.
