Abstract
약제학적 또는 화장품 조제품에 사용할 액체나 겔형 지질 부형제의 제조방법과 그 조성물을 설명한다. 지질 부형제는 리조인지질 예컨대 모노-올레오일-포스파티딜에탄올아민(MOPE)같은 인지질로 구성된다. 겔을 형성할 때, 비교적 저농도의 즉, 약 1-2%의 지질을 사용한다. 본 발명은 무-독성, 무-자극성 담체나 부형제 단독의 혹은 다른 작용제 예컨대 약물 및 화장품과 혼합한 것으로서 비교적 저 지질농도의 지질전달계 사용법을 발표하였다. 예컨대, 분무식 또는 점적형 지질 부형제는 비막(nesal membrane)을 통한 흡수력을 향상시키는 능력 때문에 비점막에 대한 펩티드(즉 칼시토닌과 인슐린)의 무-자극전달에 특별히 사용된다. 화장품에서, 이것을 단독으로 또는 생물학적 활성 작용제와 혼합하여 사용할 수 있다.
Description
제1도는 MOPE를 부형제 전달 매개체로 사용할 때 비점막을 통과 하는 칼시토닌의 증가를 보여주는 그래프이다.

본 발명은 지질담체와 함께 투여하는 약물과 화장품 전달 분야에 관한 것이다. 더 구체적으로, 본 발명은 계면활성체와 부형제 전달매개체인 지질의 용도를 설명한다.
약물같은 생물학적 활성작용제의 국소적인(눈, 코, 귀, 질, 피부 또는 경피의) 전달은 데옥시콜린산 나트륨이나 프로필렌글리콜 같은 음이온, 무이온 양성의(emphoteric) 또는 잠재적 음이온성 계면활성제를 사용하면 달성된다. 이러한 계면활성제를 사용하는 이유는 상기의 경로를 통한 약물이나 생물학적 작용제의 흡수력이 우수하지 못하여 계면활성제를 사용하므로써 흡수력을 경화시키기 위한 것이다. 예컨대 합성 유화제 같은 계면활성제는 어떤농도, 예컨대 약 14% 내지 75%(표1)의 농도에서 겔을 형성하며 따라서 화장품 공업에서의 담체나 부형제로 이용한다. 여기에서의 계면활성제 농도는 비점막을 자극시킨다. 대조적으로 리포좀 형성에 사용하는 물질은 무-자극성이지만, 막을 통한 약물 흡수력을 강화시키는 것으로 증명되지 않았다. 본 발명은 단독의 무-독성, 무-자극성 담체나 부형제 혹은 다른 작용제와의 복합물 형태인 비교적 저 지질함량으로서 약물과 화장품에 모두 사용하기 위한 지질 전달계의 사용법을 발표한다.
약물 흡수력을 강화하는데 사용하는 계면활성제에 관한 선행기술분야는 담즙염과 다른 계면활성제의 이용도 포함한다(Gordon et al., 1985, Proc. Natl, Acad. Sci. 82;7419-7423;Hirai et al., 1981, Int. J. Pharm. 9:165-172; Duchateau et al., 1987, Int. J. pherm., 39:87-92; and Hanson et al., 1986 IN:Delivery Systems for peptide Drug, Davis, ed., Plenum Press, NY, pp, 233-242). 앞서 설명한 것과 같이, 이 염과 계면활성제는 비점막에 독성인 것으로 발견되었다(Hirai et al., 1981, Int. J. Pharm., 9:173-184).
본 발명은 액체 분무형이나 점적형 또는 별개적으로 점성 액체나 겔형으로 사용할 수 있는 새로운 무-자극성 지질 부형제 시스템을 발표한다. 부형제의 물리적 상태는(즉, 액체, 점성액 또는 겔) 사용된 지질과 그농도 시스템의 pH 또한 시스템에 있는 염의농도에 따라 달라진다. 분무형이나 점적형 지질 부형제는 비막을 통한 펩티드 흡수력을 강화시키는 능력 때문에 비점막에 대한 펩티드(즉, 칼시토닌과 인슐린) 또한 호르몬(즉, 성장 호르몬)의 무-자극 전달에 특별하게 사용된다. 막을 통한 흡수력 증대는 또한 포준적인 더 큰 독성의 부형제와(즉, 데옥시콜레이트) 비교할 때, 부형제의 세포 투과성 강화를 특징으로 한다(제3도 참조). 펩티드 전달과 함께 부형제는 다른 생물학적 활성제중에서 호르몬, 항히스티민제와 비 습윤제 등을 전달할 수도 있다. 본 발명의 겔 부형제에 있는 생물학적 활성제의 용해나 현탁액을 응용부위에 바르면 활성제를 충분히 방출할 수 있다.
제약이나 화장품의 국소전달에 있어서, 점성액이나 겔형의 동일한 부형제를 피부표면에 바를 때 화장품 조제물의 우수한 윤활, 작용시간 또한 공명효과를 보여준다. 이와 같이, 본 발명의 지질 부형제로 만들어진 조제물은 예컨대 파라-아미노벤조산(PABA) 같은 햇빛차단제와 습윤제처럼 화장품 제조물에도 사용할 수 있다. 약제물 같은 생물학적 활성제를 위한 겔부형제는 막을 통해 작용제를 전달하는 지속적인 방출 매질 역할을 한다. 이러한 겔을 코, 눈, 귀로, 피부로, 경피속으로, 질내로 혹은 직장속으로 투여한다.
본 발명에서 사용하는 지질은 인지질 특히, 리조인지질이 적절하다. 리조인지질(lysophospholipids)은 세포에 대해 용해 효과 때문에 약제적인 또한 화장품 계면활성제나 부형제로는 불가능한 대상물이다. 본 발명은 리조포스파티드 특히 리조포스파티딜-에탄올마인, 더욱더 모노-올레오일-포스파티딜 에탄올 아민(MOPE)으로된 약제적 및 화장품 부형제의 무-독성, 무-자극성 조성물에 관계한다.
본 발명은 리조인지질, 특히 모노-올레오일 포스파티딜에탄올아민(MOPE)를 포함하는 무-자극성 지질에 대한 것이다. 부형제는 액체나 겔형이고 또한 증류수나 염용액과 함께 만들 수도 있다. 증류수를 넣으면, 모노-올레오일 포스파티딜에탄올아민 농도가 약 0.1% 내지 0.4% 일 때 액체부형제가 만들어지고 이것의 pH는 약 8.5 내지 10.0이고 약 9.0이 가장 바람직하다.
부형제는 또한 예컨대 인슐린이나 칼시토닌 같은 펩티드로서 생물학적 작용제도 포함다. 별도로, 모노-올레오일 포스파티딜에탄올아민의 농도가 약 0.6% 내지 2.0%이면 부형제가 겔형으로 된다. 이때 부형제의 pH값은 약 8.5 내지 10.0이고 pH9.0이 바람직하다. 겔형 부형제는 또한 예컨대 인슐린이나 칼시토닌처럼 펩티드 같은 생물학적 활성제도 포함할 수 있다.
별도로, 부형제는 화장품으로 사용할 수 있으며, 단독으로 또는 생물학적 활성제를 더 포함하기도 한다. 본 발명의 지질부형제를 이용한 화장품은 액체, 점성액, 또는 겔형으로 된다.
본 발명에서는 비교적 낮은 농도의 지질을 계면활성제와 부형제로 사용할 수 있다. 지질종류, 사용량, 또한 부형제의 물지걱상태(즉, 분무형이나 점적형액, 점성액 또는 맑은 겔형물질)등은 용도와 응용에 따라 다르다. 그러나, 생물학적 활성제 예컨대, 약물과 같은 생물학적 활성이 있는 작용제의 투여와 전달에 있어서, 사용한 지질종류와 양은 예컨대 표피, 피부 또는 비점막 같은 점막처럼 상피막을 통한 약물전달 및 침투를 강화하기에 충분하며 동시적으로, 무-독성 및 무-자극성이다.
이러한 조제물은 전형적으로 점적형이나 분무형 액체이다. 하지만, 점성액이나 겔부형제 역시 여기에 함유된 생물학적 활성제나 화장품에 있어서 우수한 지속적 방출특성을 나타낸다. 화장품의 경우, 본 발명에서 사용한 지질의 종류와 양이 충분하여 국소투여용 담체나 겔을 만들 수 있다.
본 발명에서 대부분의 지질을 부형제로 사용할 수 있다. 가장 바람직한 것은 예컨대, 포스파티딜클린, 포스파티딜에탄올아민, 또한 팔미토일-올레오일-포스파티딜-에탄올아민(POPE) 같은 또한 포스파티딜세린과 포스파티딜글리세롤처럼 음이온성 인지질 같은 인지질등이다. 가장 바람직하게는 리조포스파티딜-에탄올아민 등의 리조인지질을 사용하면 예컨대, 모노-올레오릴-포스파티딜에탄올아민(MOPE)와 모노-미리스토일-포스파티딜에탄올아민(MMPE), 그러나 특히 MOPE를 가장 많이 사용한다. 별개적으로, 리조지질(lysolipid) 모노-올레오일-포스파티드산(MOPE), 모노-올레오일-포스파티딜세린(MOPS), 또한 모노-올레오일-포스파티딜글리세롤(MOPG)도 사용할 수 있다. 또한 sn-1-18:2 시스 -PE와 sn-1-18:3 시스 -PE같은 각각의 2 혹은 3개의 2중결합이 있는 리조포스파티딜에탄올아민 역시 본 발명에서 사용할 수 있다. 별도로, 카르디올리핀도 사용할 수 있다. 그외의 다른 지질은 막을 통한 생물학적 활성제의 전달을 강화하고 동시에 예컨대 표준적혈구 용혈시험으로 측정하였을 때 최소의 자극만 나타내는 능력으로 결정할 수 있다.
본 발명의 실행에 있어서, 스테로이드성 지질 알파-토코페롤헤미숙신산(THS)같은 다른 지질도 액체 부형제로 사용할 수 있다. 다양한 토코페롤과 이들의 수용성 유도체를 사용하여 리포좀을 제조할 수 있다(Janoff et al., PCT Publication NO. 87/02219, April 23, 1987, entitled Alpha Tocopherol-Based Vesicles). 출원인은 또한 미국출원에서 THS를 저 pH에서 안정제와 혼합하고 그후 약물을 THS와 혼합한 약제학적 전달계로서 알파 토코페롤헤미숙신산을 발표하였다(Janoff et al., U.S. Patent application Serial No. 259,988, filed October 19, 1988, entitled Tocopherol- Based Pharmaceutical System). 본 발명에서 THS부형제를 통해 생물학적 활성제와 THS를 압출하여 SUV를 만들고 생물학적 활성제를 전달할 수 있다. SUV 형성을 위한 특별히 바람직한 방법은 Culis et al.,이 발표하였다(PCT Publication No. 87/00238, January 16, 1986, entitled Extrusion Technique for Producing Unilamellar Vesicles), 이 방법에 따라 제조한 소포(versicle)를 막필터를 통해 반복적으로 가압 압출하여 SUV를 형성한다.
액체와 겔부형제로 지질 MOPE를 구성할 수 있다. 1-올레오릴 리조포스파티딜에틸렌아민(MOPE)을 다음처럼 표시한다 :

여기서, R은 1-위치([1]표시 즉 1-올레오릴에 있는 탄소에 부착된 올레오릴기이다). (OH)기는 [2]탄소에 위치한다. 지질을 sn-1-18:1 시스 -PE로 표시하고 이것을 올레오일기의 18 탄소 조성물을 뜻하며 그뒤는 이중결합수를 뜻하는 숫자가 기재되고 이경우는 시스(cis) 구성에 있는 이중결합의 수가 1이다. 전문적 용어로 표현하면, R이 1-위치에 있고 2나 3개의 이중결합과 또한 17탄소원자를 갖춘 것으로 불포화도가 큰 Lyso PEs(LOPEs)을 각각 sn-1-18:2시스-PE와 sn-1-18:3시스-PE로 각각 표시한다. 카르복시산염의 탄소원자는 18번째 탄소원자이다.
전술한 것과 같이 1-올레오일 리조포스파티딜 에탄올아민(MOPE, 또는 sn-1-18:1시스-PE)은 리조인지질의 미소세포열이 아닌 생리학적 pH값의 라멜상을 나타낸다(Tilcock et al., U.S. Patent Application No. 821,366, filed January 22, 1986, entitled Solubilization of Hydrophobic Materials Using Lysopholipids). 그러나 MOPE는 이러한 조건에서 소수성 물질의 미소세포 용해를 촉진하는 높은 pH(즉 약 8.5나 그이상)값에서 미소세포형을 나타낸다. 미소세포에서 이중층으로의 다형상 작용은 (a) 미소세포인 리조포스파티딜콜린과 대조적으로 pH7 및 -20℃ 내지 90℃의 온도에서 라멜라 구성에 상응하는31p-NMR 스펙트럼 ; (b) x-선 산란으로 동일간격으로 떨어진 고리를 만들고 라멜라형 유기체임을 지적하는 MOPE의 x-선 회절형태; (c) pH 8.2에서 생긴 리포좀의 다중 라멜라 특성을 보여주는 동결파열 현미경사진; 또한 (d) 평균 pH9.0에서 미소세포 구조임을 지적하는 등방성 이동을 보여주는 P-NMR 스펙트럼등으로 입증할 수 있다. 앞서 발표한 것과 같이, 리조인지질의 2중층 상 전이에 대해 미소세포를 사용하여 소수성 작용제를 용해할 수 있다(Tilcock et al., 1986, Biochemistry, Vol. 25(4):816-822).
증류수나 희석 염농축액(즉, 약 0.5M 염까지)로 수화시킨 pH9.0의 MOPE(수산화 나트륨이나 중탄산 나트륨으로 조정함)은 약 0.1% 농도에서 0.4%농도까지 점적형 또는 분무형 투명액이고 또한 맑은 겔상태인 약 0.4% 내지 2.0%까지의 점도로 증가한다. 인슐린의 침투력 증가에서 효과가 큰 농축물은 증류수에 들어있는 pH9.0의 0.2% MOPE이다.
약 0.1% 내지 2.0%의 MOPE농도와 약 0.5M 까지의 염농도와 또한 ph9.0(중탄산나트륨이나 수산화나트륨으로 조정)의 MOPE는 탁한 유체이다.
분무식 또는 점적식 액체 부형제의 제조에 있어서, 지질(즉, 리조인지질 MOPE)을 약 0.5M 이하의 희석염용액이나 증류수 같은 저이온 함량의 용액과 혼합하고 이때의 온도를 약 5℃ 내지 90℃ 특히 약 20-30℃와 더 바람직하게는 약 25℃로 하고, pH값을 약 8.5 내지 14.0, 특히 pH8.5 내지 pH10.0, 더 바람직하게는 약 pH9.0으로 하고 또한 지질의 농도를 약 0.2% 내지 0.3% 특히 약 0.2%로 한다. 수성용액에 지질 혼합물을 넣고 다시 생물학적 활성제를 함께 혼합하여 생물학적 활성제 농도가 시스템내의 활성제 용해도로 되거나 또는 최종 현탁액의 원하는 농도로 제한하게 한다; 예컨대 칼시토닌의 경우 그농도는 약 100U/㎖이다. 더 구체적으로, 수성용액으로는 증류수를 이용하지만 그외에도 예컨대, 완충액이나 염수용액 같은 염이 들어있는 용액을 사용할 수도 있다. 이 경우, 염농도는 약 0.5M 정도로 크다. 염을 지질 부형제와 함께 사용할 경우 부형제는 탁한 유체형태의 것을 택한다.
별개적으로, 지질을 증류수나 또는 임의의 농도의 염함유용액 같은 용액에서 약 pH6.0 내지 pH8.2 바람직하게는 약 pH 7.0으로 현탁할 수 있다. 부형제용액의 pH가 수성 조성물 및 지질농도에 관계없이 약 7.0 일 때 부형제는 탁한 유체형태로 된다. 인슐린 경우, 지질농도가 약 0.8%일 때 약물흡수력이 증가한다.
겔부형제 제조의 경우, 약 5℃ 내지 90℃ 특히 약 25℃과 약 pH 8.2 내지 14.0, 특히 약 pH 8.5 내지 10.0과 더 바람직하게는 약 pH9.0 에서, 또한 지질농도가 약 0.4%(비교적 무른겔) 내지 10% 이상일 때 지질(즉, 예컨대 리조인지질 MOPE)을 증류수와 함께 혼합하면, 약 2%의 지질농도에서 응고겔이 생긴다. 지질과 물의 혼합 후 생물학적 활성제를 첨가하여 이것의 농도를 겔내의 약물 용해도로 만들거나 그렇지 않을 경우 겔내의 약물현탁액 농도까지로 맞춘다. pH9.0 정도이고 지질농도가 약 0.6% 내지 2.0%인 지질 부형제와 함께 염을 사용할 경우, 부형제는 탁한 유제형태의 것이다.
본 발명의 지질부형제나 담체를 예컨대 약물, 호르몬, 단백질, 염료, 비타민 또는 상작용제 같은 생물학적 활성제와 혼합하기도 한다. 본 발명에서 생물학적 활성제는 생물학적으로 활성인 화합물을 뜻하며 예컨대 약물과 또한 펩티드, 호르몬, 독소, 효소, 신경전달물질, 지방단백질, 당단백질, 면역조절체, 면역글로부린, 다당류, 세포수용 결합분자, 헥산, 폴리뉴클레오티드 등과 같은 다른 치료제 또한 염료, 방사선 불투과성 작용제 및 형광작용제 같은 생물학적 추적물질등이 있다.
생물학적 활성제는 소수성이나 친수성이다; 소수성일 경우 본 발명의 지질부형제(즉, MOPE)를 사용하여 작용제를 용해한다(U.S. patent Application to Tilcock et al., U.S. Serial No. 821,366, filed January 22, 1986, entitiled Solubilization of Hydrophobic Materials Using Lysophospholipids의 방법을 사용함). 더 구체적으로 소수성 및 친수성 작용제로 제한적인 것은 아니지만 항-진균성 작용제인 피라미신, 칸디시딘, 필리핀 특히 나이스타틴과 암포테리신 B 같은 폴리엔 항생제가 있다. 또한 다른 생물학적 활성제로서 아미노글리코시드 예컨대, 젠타마이신 같은 항박테리아성 화합물과 리팜파신 같은 항바이러스 화합물, 안티몬 유도체 같은 항-기생 화합물 또한 빈불라스틴, 빈크리스틴, 미토마이신C, 독소루비신, 다우노루비신, 메토르렉사이트 또한 시스플라틴 같은 항종양성 화합물과 그외에도 알부민 같은 단백질, 디프테리아 독소 같은 독소, 촉매 같은 효소, 에스트로겐 같은 호르몬, 아세틸콜린 같은 신경전달물질, 알파-지방단백질 같은 지방단백질, IgG 같은 면역글로부린, 인터페론이나 인터레우킨 같은 면역조절체, 아르세나조 Ⅲ 같은 염료,14C 같은 방사선 동위체,99Te 같은 방사선-불투과성 화합물, 카르복시 형광물 같은 형광성 화합물, 글리코겐 같은 다당류, 에스트로겐 수용단백질 같은 세포수용결합분자, 인도메타신 살리실산 아세테이트, 아이부프로펜, 설린데크, 피록시캄 또한 나프록센 등의 비스테로이드성 항-감염물질과 또한 멕사매타손 같은 항-감염물질, 티몰올이나 필로카르핀 같은 항글라우코작용제, 디부카인 같은 마취제, 티민 같은 헥산, RNA 고분자 같은 폴리뉴클레오티드, 알파-차단제, 베타-차단제, 칼슘로 차단제 ACE 억제제 같은 심장혈관 작용제와 춘 작용제 및 프로스타글란딘등도 포함된다.
본 발명의 지질 부형제에 사용할 수 있는 염용액으로 생리학적 염수, 린저스(Ringers) 염용액, 탄산염완충액 같은 완충액, 메틸글루카민, N-2-히드록시에틸 피페라진-N-2-에탄술폰산(HEPES),에틸렌디민테트라아세트산(EDTA), 또한 트리스(히드록시메틸) 아미노메탄(트리스)등이 포함된다. 이런 염이나 완충용액을 0.5M 염농도까지 사용할 수 있다.
국소투여에서, 지질부형제를 단독으로 또는 생물학적 활성제나 불활성 작용제와 함께 피부표면에 투여하거나 또다른 경로 즉 코, 점막, 눈, 구강, 직장 또는 귀 등을 통해 투여한다. 이렇게 사용하는 경우 지질부형제를 분무형 또는 점적형 액체형태로 또는 겔형태로 전달할 수 있다.
눈이나 귀로 투여할 경우, 현탁액은 예컨대 단일 또는 다중식 점적기나 투여기 등의 전달 시스템을 사용하여 전달할 수 있다. 안구용 조성물로서 히알우론산, 콘드로이틴 황산염, 히드록시프로필 메틸셀룰로오스 또는 폴리비닐알코올 같은 점막유사체도 포함한다; 또한 소르브산 EDTA 나 염화벤질코늄 같은 방부제와 또한 적정량의 희석재 또한/또는 담체물질을 함유할 수 있다. 코로 투여할 때, 일정량을 적하할 수 있는 용기나 다른 그릇과 같은 분무 전달장치를 사용할 수 있다.
본 발명의 지질 부형제를 사용하는 생물학적 활성 작용제 제형을 치료학적으로 인간을 포함한 포유동물에게 사용하여 생물학적 활성 약물 전달효과가 있을 감염이나 다른 조건을 치료할 수 있다. 질병상태 치료에서 사라에게 투여할 때 담당의사는 대상환자에 대한 적정의 투여량을 최종 결정하며 이것은 환자 개개인의 나이, 중량, 반응과 또한 질병특성 및 심각성에 따라 다양하다고 예상된다.
화장품의 국소전달에 있어서, 점성액이나 겔형태의 지질 부형제를 피부표면에 바를 때 화장품 조제물의 우수한 윤활, 작용시간 또한 공명 특성을 보여준다. 화장품 조제물로 사용할 때 지질부형제를 단독으로 혹은 생물학적 활성제나 다른 작용제(즉 향수, 파라핀, 오일, 착색제, 글리세린등)와 혼합하여 피부에 바르고 그결과 피부의 조직과 탄력성은 표면특성으 개선하여 건조도나 탈수도가 줄어들고 따라서 주름살 형성과 그외의 육안으로 알수 있는 다른 노화증거를 피할 수 있다.
본 발명의 지질 부형제로 만들 수 있는 다른 조제물은 선스크린(햇빛차단제)(즉, 파라-아미노벤조산)(PABA)과 습윤제 등의 화장품 조제물도 포함한다. 화장품에 이용할 경우 제형을 통상적인 실행에 따라 투여할 수 있다.
다음의 실시예는 보기를 든것이며 본 발명 범위를 제한하지 않는다.
클로로포름 속의 MOPE(8.0㎎0을 건조시켜 37℃의 수조속에 있는 동근바닥 플라스크상에 지질막이 생기도록 한다. 20-50uL의 0.1N 수산화나트륨(NaOH)을 사용하여 pH 9.0으로 조절한 1.0㎖ 증류수 속에 지질막을 재현탁한다. 1㎖의 칼시토닌(Calcimar로 제공 USV, Tuckahoe, N. Y., as a 200U/㎖ soultion)을 재현탁 MOPE에 더하고 용액을 15초간 강하게 취저으면서 혼합한다. pH갓을 0.1N NaOH로 다시 pH 9.0이 되도록 조정한다.
[실시예 2]
실시예1의 방법을 4.0㎎ MOPE를 사용하여 실행하면 0.2% MOPE 용액이 된다.
[실시예 3]
중량이 250-300g인 여덟마리의 Wistar Kyoto 수컷쥐를 1.6g/㎏의 분취량으로 20% 우레탄 용액을 복강내로(IP) 투여하여 마취시킨다.
쥐의 코를 절개하고 하라이법에 따라 시료를 점적한다(Hussain et al., 19800, J. Pharm. Sci., 69(12):1411-1413). 약물을 사전 투여한 피검물을 각 쥐에게서 떼어내고(안와후방(retrocrbital)에서 1.0㎖취함) 영기게하여 통상의 기술로 혈청을 분리한다. 시료가는 주사기를 비공속에 넣어 주사하여 칼시토닌-MOPE용액(쥐 1마리당 50uL 투여)을 투여한다.
칼슘혈청을 Sigma Diagnostics Calcium Kit No. 587로 비색 측정한다(Sigma, St. Louis, MO). 약물을 사전 투여한 대조표본과 비교할 때 혈청의 칼슘농도의 변화를 시간에 대한 함수로서 나타 내었다(제1도).
[실시예 4]
클로로포름 속의 MOPE(40.0㎎)을 건조시켜서 37℃의 수조에 있는 둥근바닥 플라스크상에 지질막을 형성한다. 20-50uL의 0.1N 수산화나트륨(NaOH)을 사용하여 pH 9.0으로 조절한 10.0㎖의 증류수속에 지질막을 재현탁한다. 25U/㎎으로된 4㎎의 포르신(porcine) 인슐린분말(Eli Lilly, Indianapolis, IN)을 MOPE용액과 혼합하고 교반 혼합한다.
[실시예 5]
실시예4의 인슐린-MOPE 용액을 실시예3의 방법에 따라 쥐의 비공속에 주입한다. Right Sprague-Dawley 수컷 쥐(300g 중량의)를 Weiner et al의 방법에 따라 당뇨병 환자로 만든다(1985, J. Pharm. Sci., 74:922-925). 1시간 후 혈액 시료를 수거하고 실시예3의 표준방법에 따라 혈청을 분리한다. Coat-A-Count 인슐린125I 방사선 면역분석법에 따라 혈청의 인슐린을 분석한다(Diagnostic Products Corp. , Los Angeles, CA).
[실시예 6]
클로로포름 속의 MOPE(80.0㎎)을 건조하여 37℃의 수조에 있는 둥근바닥 플라스크상에 지질막을 형성한다. 20-50uL의 0.1N 수산화나트륨(NaOH)을 사용하여 pH 9.0으로 조절한 10.0㎖의 증류수속에 지질막을 재현탁한다. 25U/㎎으로된 4㎎의 포르신 인슐린분말(Eli Lilly, Indianapolis, IN)을 MOPE 용액과 혼합하고 교반 혼합한다. 그결과로 나온 0.8% MOPE 인슐린겔을 실시예5의 방법에 따라 당뇨병이 생긴쥐에게 투여한다. 인슐린-MOPE 투여후 1시간 후에 혈액을 취하고, 혈청내 인슐린 농도의 분석결과를 MOPE 농도에 대해 그래프로 나타내었다(제2도).
상기의 실시예의 방법을 반복하고 희석하면 각각 0.4%, 0.2% 또한 0.1% MOPE 10㎖이 나오고 이것을 4.0㎎의 인슐린과 혼합하여 혈청중 각 표본의 인슐린 농도에 따른 결과를 표2에서 함께 그래프로 나타낸다.
[실시예 7]
1% 데옥시콜레이트 용액(Sigma, St. Louis, MO)을 증류수와 함께 만들어 총 10㎖이 되게한다. 4㎎을 인슐린을 함께 혼합하고 교반하여 용해시키면 0.5U의 인슐린 용액이 된다.
실시예3의 방법에 따라 인슐린-데옥시콜레이트 용액을 주입한다. Coat-A-Count 방사선 면역분석용 기구를 사용하여 실시예5와 같이 혈청내 인슐린을 분석한다. 그결과는 표3과 같다.

클로로포름 속의 MOPE(80.0㎎)을 건조하여 37℃의 수조에 있는 둥근바닥 플라스크상에 지질막을 형성한다. 20-50uL의 0.1N 수산화나트륨(NaOH)을 사용하여 pH 7.0으로 조절한 10.0㎖의 증류수속에 지질막을 재현탁한다. 25U/㎎으로된 4㎎의 포르신 인슐린 분말(Eli Lilly, Indianapolis, IN)을 MOPE 용액과 혼합하고 교반 혼합한다. 그결과로 나온 0.8% MOPE 인슐린겔을 실시예5의 방법에 따라 당뇨병이 생긴쥐에게 투여한다. 인슐린-MOPE 투여후 1시간 후에 혈액을 취하고, 혈청내 인슐린 농도의 분석결과를 MOPE 농도에 대해 그래프로 나타내었다(제4도).

Claims (7)
- 모노올레일 포스파티딜에탄올아민(MOPE)으로 이루어진 지질성분을 포함하는 지질 부형제에 있어서, 모노올레일 포스파티딜에탄올아민(MOPE)의 농도는 0.1% 내지 0.4% 또는 0.6% 내지 2.0% 로 구성되는 것을 특징으로 하는 지질 부형제.
- 제1항에 있어서, 부형제는 액상형이고, 모노올레일 포스파티딜에탄올아민(MOPE)의 농도는 0.1% 내지 0.4% 로 구성되는 것을 특징으로 하는 지질 부형제.
- 제2항에 있어서, 증류수를 추가로 포함하는 것을 특징으로 하는 지질 부형제.
- 제1항에 있어서, 부형제는 겔(gel)형이고, 모노올레일 포스파티딜에탄올아민(MOPE)의 농도는 0.6% sowl 2.0%로 구성되는 것을 특징으로 하는 지질 부형제.
- 제1항에 있어서, 지질 부형제의 pH는 8.5 내지 10.0인 것을 특징으로 하는 지질 부형제.
- 제5항에 있어서, 지질 부형제의 pH는 9.0인 것을 특징으로 하는 지질 부형제.
- 제1항에 있어서, 지질 부형제의 생물학적 활성제를 포함하는 것을 특징으로 하는 지질 부형제.
