Abstract
Description
[1]ペリオスチンの発現量増加またはスプライシングバリアントの変化を伴う疾患の治療用医薬組成物であって、ペリオスチン遺伝子の転写段階において、エクソン17をスキップさせる核酸および/またはエクソン21をスキップさせる核酸を有効成分として含有することを特徴とする医薬組成物。
[2]前記核酸がアンチセンス核酸である前記[1]に記載の医薬組成物。
[3]エクソン17をスキップさせるアンチセンス核酸が、配列番号1で示される塩基配列の24143番目〜24323番目の範囲を標的配列とし14〜50塩基からなる1種または2種以上の核酸であることを特徴とする前記[2]に記載の医薬組成物。

(i)ペリオスチンの発現量増加またはスプライシングバリアントの変化を伴う疾患の治療方法であって、当該疾患患者に対して、ペリオスチン遺伝子の転写段階において、エクソン17をスキップさせるアンチセンス核酸および/またはエクソン21をスキップさせるアンチセンス核酸の有効量を投与することを特徴とする治療方法。
(ii)ペリオスチンの発現量増加またはスプライシングバリアントの変化を伴う疾患の治療に使用するための、ペリオスチン遺伝子の転写段階において、エクソン17をスキップさせるアンチセンス核酸および/またはエクソン21をスキップさせるアンチセンス核酸。
(iii)ペリオスチンの発現量増加またはスプライシングバリアントの変化を伴う疾患の治療薬を製造するための、ペリオスチン遺伝子の転写段階において、エクソン17をスキップさせるアンチセンス核酸および/またはエクソン21をスキップさせるアンチセンス核酸の使用。
<実験方法>
C57BL/6J雄マウス8週齢(オリエンタルバイオサービス)を4匹使用した。麻酔下で開腹し、心臓から生理食塩水で臓器灌流を行った後に、各臓器(心臓、大動脈、副腎、肺、脾臓、胃、腎臓、皮膚、脳、肝臓、結腸、精巣)を摘出した。定法に従って臓器をホモジナイズし、RNeasy Plus Mini Kit(Qiagen)を用いてトータルRNAを抽出した。DNase処理したトータルRNAをHigh-Capacity cDNA Reverse Transcriptase Kit(Applied Biosystems)を用いて逆転写し、得られたcDNAを定量PCRに供した。試料中の転写物のコピー数は、既知のコピー数を有する各スプライシングバリアント(Pn1、Pn2、Pn3およびPn4、図1参照)プラスミドを使用して決定した。定量PCRには、ViiA-7 real-time PCR system(Applied Biosystems, Foster City, CA, USA)を用いた。統計処理にはJUMP統計ソフトウェアパッケージを使用し、分散分析およびTukey-Kramer adjustmentを行った。
結果を図2に示した。健常マウスにおいては、測定したすべての臓器においてもPn4(エクソン17およびエクソン21がスキップしたスプライシングバリアント)のコピー数が他のスプライシングバリアント(Pn1、Pn2、Pn3)に比較しておよそ100-10000倍多いことが証明された。
<実験方法>
(1)ノックアウトマウスの作製
エクソン17ノックアウトマウス(Pn 17KO)、エクソン21ノックアウトマウス(Pn 21KO)、全ペリオスチンノックアウトマウス(Pn null)をそれぞれ作製した。具体的には、Pn nullはエクソン2およびエクソン3に対するターゲッティングベクター、Pn 17KOはエクソン17に対するターゲッティングベクター、Pn21 KOはエクソン21に対するターゲッティングベクターをそれぞれ作製し、ES細胞に導入後、キメラマウスを得た。引き続き正常マウスと交配させて誕生したヘテロ接合体マウス同士を交配し、ノックアウトマウス(ホモ接合体)を作製した。これらのノックアウトマウスの作製は理化学研究所に依頼した。
野生型マウス(C57BL/6J、雄、n=5)、Pn 17KO、Pn 21KOおよびPn null(雄、各n=10)の8週齢における歯牙を観察し、代表例の写真を撮影した。
野生型マウス(C57BL/6J、雄、n=5)、Pn 17KO、Pn 21KOおよびPn null(雄、各n=10)の6週齢〜16週齢まで2週ごとに体重および尻尾長を測定した。統計処理にはJUMP統計ソフトウェアパッケージを使用し、分散分析およびTukey-Kramer adjustmentを行った。
図3に各群の歯牙の観察結果(各群の代表例の写真)を示した。Pn nullの歯牙は、野生型マウス(WT)、Pn 17KO、Pn 21KOの歯牙と比較して小さく、歯牙の形成障害が生じていると考えられた。
体重測定の結果を図4に、尻尾長の測定結果を図5にそれぞれ示した。Pn nullはWT、Pn 17KO、Pn 21KOより体重が少なく、尻尾長が短かった。
<実験方法>
野生型マウス(C57BL/6J、雄、8週齢)を使用した。麻酔下で開腹し、片側尿細管を4-0silk糸にて結紮して片側尿細管結紮(Unilateral ureteral obstruction: UUO)モデルを作製した。片側尿細管結紮モデルは、急性腎不全から線維化を生じ、慢性腎不全へ移行するモデルであることが知られている(Kidney Int. 2009, Jun;75(11):1145-52)。術前(0日目)、術後3、7、14、21、28、42および49日目に、それぞれ6匹のマウスから結紮側の腎臓を摘出し、参考例1と同じ方法でトータルRNAを抽出してcDNAを取得し、各ペリオスチンスプライシングバリアント(Pn1、Pn2、Pn3およびPn4、図1参照)の発現量を、以下の各スプライシングバリアントに特異的なプライマーセットを用いて定量PCRにより測定した。統計処理にはJUMP統計ソフトウェアパッケージを使用し、分散分析およびTukey-Kramer adjustmentを行った。

野生型マウスの片側尿細管結紮モデルにおけるペリオスチンスプライシングバリアントの発現を測定した結果を図6に示した。図中、*は0日目に対して有意差があることを示す(P<0.05)。尿細管を結紮した腎臓において、いずれのスプライシングバリアントの発現も非常に増加し、術後21日目〜28日目頃にピークに達した。Pn 2およびPn 4のmRNAは、42日目〜49日目にほぼベースラインまで減少したが、Pn 1およびPn 3のmRNAは、42日目以後も依然としてより高いままであった。
<実験方法>
野生型マウス(C57BL/6J)、Pn 17KOおよびPn null(雄、8週齢)を使用し、参考例3と同じ方法で片側尿細管結紮モデルを作製した。術後21日目に結紮側の腎臓を摘出し、参考例3と同様に厚さ4μmのパラフィン包埋切片を作製し、マッソントリクローム染色キット(GeneCopoeia)を用いてマッソントリクローム染色を行った(各n=5)。マッソントリクローム陽性領域(線維化領域)の面積をIMARIS software(Zurich, Switzerland)とBZ-II Analyzer(Keyence, Osaka, Japan)を用いて測定した。

マッソントリクローム陽性領域(線維化領域)の測定結果を、腎臓の面積に対する線維化領域の面積の割合(%)として図9に示した。図中、*はWTに対して有意差があることを示す(P<0.05)。Pn nullおよびPn 17KOのマッソントリクローム陽性領域は野生型マウスに比べて有意に減少していた。
以上の結果から、エクソン17を含むペリオスチンはTGF-βのシグナル伝達に関与し、線維化を促進するが、ペリオスチンのエクソン17を遮断することにより、線維化が抑制されることが示された。
参考例1〜4の知見から、ペリオスチン全体の阻害は片側尿細管結紮モデルの腎線維化を抑制するが、体格の小型化や歯牙の形成不全を引き起こす可能性があるため、ペリオスチンエクソン17の選択的阻害がより安全な治療法につながると考えられた。ペリオスチンエクソン21の選択的阻害についても同様と考えられる。そこで、エクソン17またはエクソン21をスキップしたmRNAを生成させるアンチセンス核酸の使用を試みた。
(1)アンチセンス核酸
ヒトペリオスチン遺伝子のエクソン17またはエクソン21をスキップさせるモルホリノアンチセンスオリゴヌクレオチドをフナコシから購入した。各アンチセンス核酸の塩基配列を以下に示す。
ヒトエクソン17アンチセンス:5'-CCATGTATAACATTGATTTTTACCTTCAGT-3'(配列番号3)
ヒトエクソン21アンチセンス:5'-TTGTTGTCCTTTTACTAACCTCCCT-3'(配列番号4)
コントロールアンチセンス:5'-CCTCTTACCTCAGTTACAATTTATA-3'(配列番号6)
ヒトエクソン17アンチセンスの標的は、配列番号1の24267番目〜24296番目であり、ヒトエクソン21アンチセンスの標的は、配列番号1の29540番目〜29564番目である。
ヒト心臓線維芽細胞(コスモバイオ、Catalog #6300)およびヒト乳がん細胞MDA-MB-231(ATCC番号:HTB-26)を使用した。ヒト心臓線維芽細胞およびヒト乳がん細胞MDA-MB-231は、10%ウシ胎児血清(FBS)および1%ペニシリン/ストレプトマイシンを含有するDMEMで培養した。これらの細胞に、ヒトエクソン17アンチセンスまたはヒトエクソン21アンチセンスを、トランスフェクション試薬「Endo-Porter(Gene Tools)」を用いて導入した。アンチセンス核酸は、培地に最終濃度10μMで添加した。導入後48時間目に細胞からトータルRNAを抽出してcDNAを取得し(参考例1参照)、RT-PCRを行った。PCR産物の有無をアガロースゲル電気泳動で確認した。

結果を図13に示した。図13から明らかなように、アンチセンス核酸の導入により、目的のスプライシングバリアントが抑制されていることが示された。
<実験方法>
ヒト腎臓近位尿細管上皮細胞(Human renal proximal tubular epithelial cells: hRPTEC、ATCC番号:PCS-400-010)を10%ウシ胎児血清(FBS)および1%ペニシリン/ストレプトマイシンを含有する腎臓上皮細胞用培地(REBM: renal epithelial cell basal medium、Cambrex Bio Science Inc.)で培養した。この細胞に、実施例1と同様の方法でヒトエクソン17アンチセンス(Pn exon 17 AS)またはコントロールアンチセンス(Control AS)を導入した。導入後24時間目に血清を含まない培地に交換し、その24時間後に培地にヒトTGF-β1(PeproTech)を添加し、さらに24時間培養した。培養終了後、細胞からトータルRNAを抽出してcDNAを取得し(参考例1参照)、スプライシングバリアントPn 1(エクソン17およびエクソン21を含む、図1参照)の発現量(各n=3)、およびTGF-βにより誘導される下流シグナル(αSMA、Snail 1、TNFα、CTGF、Vimentin)のmRNA発現量(各n=4)を、以下のプライマーを用いて定量PCRで測定した。統計処理にはJUMP統計ソフトウェアパッケージを使用し、分散分析およびTukey-Kramer adjustmentを行った。

スプライシングバリアントPn 1の発現量の結果を図14に示した。また、TGF-βにより誘導される下流シグナルのmRNA発現量の結果を図15に示した。図14、15において、*はTGF-β(-) Control ASに対して有意差があることを示し(P<0.05)、**はTGF-β(+) Control ASに対して有意差があることを示す(P<0.05)。図14に示したように、TGF-β(+) Control AS群では、スプライシングバリアントPn 1の発現量が有意に増加した。図15に示したように、TGF-β(+) Control AS群では、TGF-βにより誘導されるαSMA、Snail 1、TNFα、CTGFおよびVimentinの発現がいずれも有意に増加していたが、TGF-β(+) Pn exon 17 AS群では、いずれも発現が有意に抑制されていた。この結果から、ペリオスチンエクソン17を選択的にスキップさせるアンチセンス核酸が、TGF-β1シグナル伝達を介する線維化の抑制に有効であることが示された。
<実験方法>
ルシフェラーゼ発現マウス乳がん細胞(4T1-Luc)を10%FBSおよび1%ペニシリン/ストレプトマイシンを含有するDMEMで培養した。マウスペリオスチン遺伝子のエクソン17をスキップさせるモルホリノアンチセンスオリゴヌクレオチドをフナコシから購入した。塩基配列を以下に示す。
マウスエクソン17アンチセンス:5'-TGCTGAAAACATAGAAAGTGGAGCA-3'(配列番号5)
マウスエクソン17アンチセンスの標的は、配列番号2で示されるマウスペリオスチン遺伝子の21118番目〜21142番目である。
結果を図16に示した。コントロールアンチセンスを導入した4T1-Luc細胞を投与したマウスでは、高いルシフェラーゼ活性が測定され、4T1-Luc細胞が肺に転移していることが示された。一方、エクソン17アンチセンスを導入した4T1-Luc細胞を投与したマウスでは、ルシフェラーゼ活性は非常に低く、4T1-Luc細胞がほとんど肺に転移していないことが示された。この結果から、ペリオスチンエクソン17を選択的にスキップさせるアンチセンス核酸が、乳がん細胞の肺への転移を抑制することが示された。
<実験方法>
ペリオスチンを高発現するヒト乳がん細胞株BT549を10%FBSおよび1%ペニシリン/ストレプトマイシンを含有するRPMI1640で培養した。BT549細胞は、エストロゲン受容体陰性、プロゲステロン受容体陰性、HER2陰性のトリプルネガティブ乳がん細胞株である。
BT549細胞に、実施例1と同様の方法で、ヒトエクソン17アンチセンスまたはヒトエクソン21アンチセンスを導入した。その後細胞を96ウェルプレートに5×103個/ウェルで播種した。コントロールとして、コントロールアンチセンスを導入したBT549細胞を、同様に96ウェルプレートに播種した。播種の翌日に、パクリタキセルを10nMになるように各ウェルに添加した。72時間培養後、細胞増殖(生細胞数)およびATP産生能(ATP定量)を評価した。生細胞数の測定は、CellTiter 96 AQueous One Solution Cell Proliferation Assay(商品名、プロメガ、#G3582)を用いて行った(MTSアッセイ)。ATP量の測定は、CellTiter-Glo Luminescent Cell Viability Assay(商品名、プロメガ、#G7570)を用いて行った(ATPアッセイ)。各群間比較にはTukey-Kramer法を用いた。
MTSアッセイの結果を図17に示した。エクソン21アンチセンスを導入したBT549細胞は、エクソン17アンチセンス導入したBT549細胞およびコントロールアンチセンスを導入したBT549細胞と比較して有意に細胞増殖が抑制された(図17(A))。エクソン17アンチセンスを導入したBT549細胞、エクソン21アンチセンスを導入したBT549細胞およびコントロールアンチセンスを導入したBT549細胞にそれぞれパクリタキセルを処理すると、エクソン17アンチセンスを導入したBT549細胞およびエクソン21アンチセンスを導入したBT549細胞は、どちらもコントロールアンチセンスを導入したBT549細胞より、有意に細胞増殖が抑制された(図17(B))。
<使用動物>
参考例2で作製したエクソン17ノックアウトマウス(Pn 17KO, n=6)、エクソン21ノックアウトマウス(Pn 21KO, n=6)、全ペリオスチンノックアウトマウス(Pn null, n=6)を使用した。コントロールとして野生型マウス(BALB/c, n=6)を使用した。いずれも8週齢の雌を使用した。
マウス乳がん細胞株4T1を10%FBSおよび1%ペニシリン/ストレプトマイシンを含有するDMEMで培養した。細胞懸濁液を調製し、1×103個の細胞をマウスの左足底に移植した。移植3週間後にマウスを安楽死させ、肺を摘出してブラウン液で染色し、転移コロニー数を目視でカウントした。各群の比較にはTukey-Kramer法を用いた。
結果を図19に示した。野生型マウス(WT)では多数の肺転移コロニーが観察された。一方、エクソン17ノックアウトマウス(Pn 17KO)、エクソン21ノックアウトマウス(Pn 21KO)、全ペリオスチンノックアウトマウス(Pn null)では、いずれも野生型マウスと比較して、有意に肺転移コロニー数が減少していた。
Claims (11)
- ペリオスチンの発現量増加またはスプライシングバリアントの変化を伴う疾患の治療用医薬組成物であって、ペリオスチン遺伝子の転写段階において、エクソン17をスキップさせる核酸および/またはエクソン21をスキップさせる核酸を有効成分として含有することを特徴とする医薬組成物。
- 前記核酸がアンチセンス核酸である請求項1に記載の医薬組成物。
- エクソン17をスキップさせるアンチセンス核酸が、配列番号1で示される塩基配列の24143番目〜24323番目の範囲を標的配列とし14〜50塩基からなる1種または2種以上の核酸であることを特徴とする請求項2に記載の医薬組成物。
- エクソン17をスキップさせるアンチセンス核酸が、配列番号1で示される塩基配列の24191番目〜24193番目、24215番目〜24220番目、24247番目〜24254番目、24249番目〜24258番目、24252番目〜24255番目および24273番目〜24275番目の少なくとも1つの範囲を標的配列とする請求項3に記載の医薬組成物。
- エクソン21をスキップさせるアンチセンス核酸が、配列番号1で示される塩基配列の29412番目〜29595番目の範囲を標的配列とし14〜50塩基からなる1種または2種以上の核酸であることを特徴とする請求項2に記載の医薬組成物。
- エクソン21をスキップさせるアンチセンス核酸が、配列番号1で示される塩基配列の29460番目〜29462番目、29468番目〜29474番目、29472番目〜29479番目、29509番目〜29515番目、29525番目〜29531番目、29530番目〜29536番目、29531番目〜29538番目、29534番目〜29539番目、29534番目〜29541番目、29536番目〜29542番目および29545番目〜29547番目の少なくとも1つの範囲を標的配列とする請求項5に記載の医薬組成物。
- エクソン17をスキップさせるアンチセンス核酸および/またはエクソン21をスキップさせるアンチセンス核酸を発現するアデノ随伴ウイルスベクターを含有する請求項2〜6のいずれかに記載の医薬組成物。
- ペリオスチンの発現量増加またはスプライシングバリアントの変化を伴う疾患が、ペリオスチン遺伝子のエクソン17および/またはエクソン21を含むスプライシングバリアントの増加を伴う疾患である請求項1〜7のいずれかに記載の医薬組成物。
- ペリオスチンの発現量増加またはスプライシングバリアントの変化を伴う疾患が、心不全、腎不全、乳がん、胆管細胞がん、膵がん、悪性黒色腫、神経膠芽腫、気管支喘息、糖尿病性網膜症、変形性膝関節症、アトピー性皮膚炎、特発性間質性肺炎、加齢黄斑変性からなる群から選択される少なくとも一種である請求項8に記載の医薬組成物。
- 心不全の治療、腎不全の治療、糖尿病性網膜症の治療、乳がんの転移抑制または悪性黒色腫の転移抑制に用いられる請求項8に記載の医薬組成物。
- ペリオスチンの発現量増加またはスプライシングバリアントの変化を伴う疾患の治療薬と組み合わせて使用される請求項1〜10のいずれかに記載の医薬組成物。
