Abstract
Description
(1)乳タンパク質濃度が1.9質量%以上である、血中アミノ酸濃度上昇促進用発酵乳。
(2)乳タンパク質濃度が3.0質量%以上である、(1)に記載の血中アミノ酸濃度上昇促進用発酵乳。
(3)発酵乳が、ラクトバチルス・デルブルッキー・サブスピーシーズ・ブルガリカス(Lactobacillus delbrueckii subsp. bulgaricus)およびストレプトコッカス・サーモフィルス(Streptococcus thermophilus)により発酵されたものである、(1)または(2)に記載の血中アミノ酸濃度上昇促進用発酵乳。
(4)血中必須アミノ酸濃度上昇促進用発酵乳である、(1)〜(3)のいずれかに記載の血中アミノ酸濃度上昇促進用発酵乳。
(5)血中分岐鎖アミノ酸濃度上昇促進用発酵乳である、(1)〜(3)のいずれかに記載の血中アミノ酸濃度上昇促進用発酵乳。
(6)(1)〜(5)のいずれかに記載の発酵乳を含む、血中アミノ酸濃度上昇促進用食品。
(7)乳タンパク質濃度が1.9質量%以上である発酵乳を、対象に摂取させることを含んでなる、血中アミノ酸濃度上昇促進方法。
(8)血中アミノ酸濃度上昇促進のための、乳タンパク質濃度が1.9質量%以上である発酵乳の使用。
(9)乳タンパク質濃度が1.9質量%以上である発酵乳を含む、血中アミノ酸濃度上昇促進用食品組成物。
(10)血中アミノ酸濃度上昇促進用食品組成物の製造のための、乳タンパク質濃度が1.9質量%以上である発酵乳の使用。
(11)血中アミノ酸濃度上昇を促進するための、乳タンパク質濃度が1.9質量%以上である発酵乳。
Lactobacillus delbrueckii subsp. bulgaricus OLL205013株は、2017年2月3日付け(原寄託日)で独立行政法人 製品評価技術基盤機構 特許微生物寄託センター(日本国千葉県木更津市かずさ鎌足2−5−8 122号室)に、受託番号NITE BP−02411の下でブダペスト条約に基づき国際寄託されている。
本発明の発酵乳の調製
下記表1に記載の配合によりベースミックスを調合した。調合後のベースミックスを95℃で達温殺菌した後、スターターを添加して43℃で発酵し(終了時のpHは4.3)、本発明の発酵乳を調製した。スターターとして、ラクトバチルス・デルブルッキー・サブスピーシーズ・ブルガリカス(Lactobacillus bulgaricus subsp. bulgaricus)2038株とストレプトコッカス・サーモフィルス(Streptococcus thermophilus)1131株(いずれの菌も「明治ブルガリアヨーグルト」(登録商標、株式会社明治製)から入手した)とを組み合わせて用いた。また、ベースミックスの配合に用いた脱脂粉乳は株式会社明治より入手した。本発明の発酵乳の組成を下記表2に表す。
乳中のタンパク質吸収性評価
8週齢のSD系雄ラット(日本エスエルシー株式会社より入手した)を1週間の馴化の後に用いて、実施例1で調製した発酵乳中のタンパク質の吸収性を評価した。
(1)投与試験
実施例1で調製した発酵乳をさらに65℃、30分間殺菌した後、試験飲料として用いた。実施例1で調製した発酵乳と同じ、脂質、無脂乳固形分、固形分組成(%)となるように脱脂乳を水に溶解した試料を対照飲料(発酵を行っていない)として用意した。実施例1で調製した発酵乳と、対照飲料とは同じ乳タンパク質濃度となる。
8週齢のSD系雄ラット13匹について、ネンブタール麻酔下で門脈カテーテルを留置した。
カテーテル留置から1日後に一晩の絶食を行い、体重の平均値が均一になるように脱脂乳(対照飲料)群(6匹)、脱脂発酵乳(実施例1で調製した発酵乳(本発明の発酵乳))群(7匹)に群分けを行った。各群には脱脂乳または脱脂発酵乳(実施例1で調製した発酵乳(本発明の発酵乳))を10mL/kgBWで胃ゾンデを用いて単回経口投与した。単回経口投与前、投与から30、60、90、120、および240分後にカテーテルより血液を200μLずつヘパリン処理済みチューブに採取した。採取した血液から血漿を得た後、−80℃で凍結保存し、アミノ酸濃度の測定に用いた。
血漿に、3.5%スルホサリチル酸二水和物を1:2の割合で添加し、遠心分離を行い除蛋白した後、0.45μmフィルター(Durapore PVDF 0.45μm,
Millipore)で濾過しサンプルとした。サンプル中のアミノ酸濃度は高速アミノ酸分析計(L−8800, Hitachi)で測定した。
測定したアミノ酸、総アミノ酸(TAA)、必須アミノ酸(EAA)、非必須アミノ酸(NEAA)、および分岐鎖アミノ酸(BCAA)を下記表3に示した。

- 乳タンパク質濃度が1.9質量%以上である、血中アミノ酸濃度上昇促進用発酵乳。
- 乳タンパク質濃度が3.0質量%以上である、請求項1に記載の血中アミノ酸濃度上昇促進用発酵乳。
- 発酵乳が、ラクトバチルス・デルブルッキー・サブスピーシーズ・ブルガリカス(Lactobacillus delbrueckii subsp. bulgaricus)およびストレプトコッカス・サーモフィルス(Streptococcus thermophilus)により発酵されたものである、請求項1または2に記載の血中アミノ酸濃度上昇促進用発酵乳。
- 血中必須アミノ酸濃度上昇促進用発酵乳である、請求項1〜3のいずれか一項に記載の血中アミノ酸濃度上昇促進用発酵乳。
- 血中分岐鎖アミノ酸濃度上昇促進用発酵乳である、請求項1〜3のいずれか一項に記載の血中アミノ酸濃度上昇促進用発酵乳。
- 請求項1〜5のいずれか一項に記載の発酵乳を含む、血中アミノ酸濃度上昇促進用食品。
