Abstract
Description
[1]物質を収容可能な複数の収容部が、側壁によって互いに隔てられて形成されている基板上に、当該物質を含む第一の溶媒を導入する物質導入工程と、前記物質導入工程の後に、前記第一の溶媒よりも比重が大きい第二の溶媒を前記第一の溶媒上に導入する物質収容工程と、を含む物質封入方法。
[2]前記第一の溶媒及び第二の溶媒の少なくとも一方が界面活性剤を含む[1]の物質封入方法。
[3]前記第一の溶媒中の前記界面活性剤の濃度が、0.01%〜1%である[2]の物質封入方法。
[4]前記界面活性剤が、TWEEN20又はTriton X−100である[2]又は[3]の物質封入方法。
[5]前記第二の溶媒が、飽和炭化水素、不飽和炭化水素、芳香族炭化水素、シリコーンオイル、ヘキサフルオロプロピレンエポキシド系ポリマー、ハイドロフルオロエーテル構造を持つポリマー、パーフルオロポリエーテル、三フッ化塩化エチレンポリマー及びパーフルオロカーボン構造を持つポリマーからなる群より選択される少なくとも1つ又はこれを含む混合物である[1]〜[4]のいずれかの物質封入方法。
[6]前記第一の溶媒が、水、親水性アルコール、親水性エーテル、ケトン、ニトリル系溶媒、ジメチルスルホキシド、及びN,N−ジメチルホルムアミドからなる群より選択される少なくとも1つ又はこれを含む混合物である[1]〜[5]のいずれかの物質封入方法。
[7]前記側壁が、フッ素系高分子樹脂からなる疎水性の上面を有し、前記第二の溶媒がパーフルオロカーボン構造を有するポリマーである[5]又は[6]の物質封入方法。
[8]前記フッ素系高分子樹脂が、アモルファスフッ素樹脂である[7]の物質封入方法。
[9]前記基板の前記収容部を含む領域が、外部に開放されている[1]〜[8]のいずれかに記載の物質封入方法。

[11]ターゲット分子を特異的に捕捉するビーズと、当該ターゲット分子とを反応させる反応工程と、前記反応工程の後に、前記ビーズを用いて[1]〜[9]のいずれかに記載の物質封入方法を行なうビーズ封入工程と、前記ビーズ封入工程の後に、前記収容部の各々に前記ターゲット分子を捕捉したビーズが収容されているか否かを検出する検出工程とを含むターゲット分子を検出する方法。

前記物質を収容可能な複数の収容部が、側壁によって互いに隔てられて形成されている基板上に、前記物質を含む親水性溶媒を導入する物質導入工程と、前記物質導入工程の後に、前記親水性溶媒よりも比重が大きい疎水性溶媒を前記親水性溶媒上に導入する物質収容工程と、を含む物質封入方法。
[14]前記親水性溶媒及び前記疎水性溶媒の少なくとも一方が界面活性剤を含む[13]の物質封入方法。
[15]前記親水性溶媒中の前記界面活性剤の濃度が、0.01%〜1%である[14]の物質封入方法。
[16]前記界面活性剤が、TWEEN20又はTriton X−100である[14]又は[15]の物質封入方法。
[17]前記疎水性溶媒が、飽和炭化水素、不飽和炭化水素、芳香族炭化水素、シリコーンオイル、ヘキサフルオロプロピレンエポキシド系ポリマー、ハイドロフルオロエーテル構造を持つポリマー、パーフルオロポリエーテル、三フッ化塩化エチレンポリマー及びパーフルオロカーボン構造を持つポリマーからなる群より選択される少なくとも1つ又はこれを含む混合物である[13]〜[16]のいずれかの物質封入方法。
[18]前記親水性溶媒が、水、親水性アルコール、親水性エーテル、ケトン、ニトリル系溶媒、ジメチルスルホキシド、及びN,N−ジメチルホルムアミドからなる群より選択される少なくとも1つ又はこれを含む混合物である[13]〜[17]のいずれかの物質封入方法。
[19]前記側壁が、フッ素系高分子樹脂からなる疎水性の上面を有し、前記疎水性溶媒がパーフルオロカーボン構造を有するポリマーである[17]又は[18]の物質封入方法。
[20]前記フッ素系高分子樹脂が、アモルファスフッ素樹脂である[19]の物質封入方法。
[21]前記基板の前記収容部を含む領域が、外部に開放されている[13]〜[20]のいずれかに記載の物質封入方法。
[22]前記物質は、ビーズ、核酸、タンパク質、ウイルス、細胞及び脂質膜複合体から選択される1以上である[13]〜[21]のいずれかに記載の物質封入方法。
[23]ターゲット分子を特異的に捕捉するビーズと、当該ターゲット分子とを反応させる反応工程と、前記反応工程の後に、前記ビーズを用いて[13]〜[21]のいずれかに記載の物質封入方法を行なうビーズ封入工程と、前記ビーズ封入工程の後に、前記収容部の各々に前記ターゲット分子を捕捉したビーズが収容されているか否かを検出する検出工程とを含むターゲット分子を検出する方法。
[22]前記ビーズに、前記ターゲット分子に特異的に結合する分子が結合している[21]に記載のターゲット分子を検出する方法。
本発明に係るビーズ封入方法について、図1を参照して説明する。図1は、本発明に係るビーズ封入方法の手順を説明するための模式図であり、アレイ1の側方断面図を示す。
ビーズ導入工程について、図1Aを参照して説明する。
界面活性剤は、特に限定されないが、例えばTWEEN20(CAS番号:9005-64-5、モノラウリン酸ポリオキシエチレンソルビタン)及びTriton X−100(CAS番号:9002-93-1、一般名ポリエチレングリコールモノ−4−オクチルフェニルエーテル(n≒10))などが挙げられる。第一の溶媒20への界面活性剤の添加濃度は、特に限定されないが、好ましくは0.01〜1%である。
ビーズ収容工程について、図1B〜Hを参照して説明する。
基板10の収容部13を含む領域14は、先に述べた通り、上部開放空間とされていてよいが、領域14を閉鎖空間とする場合には、ビーズ導入工程とビーズ収容工程との間に、領域14内を脱気する脱気工程を行ってもよい。脱気方法としては、例えば、アレイ1を減圧環境下において放置する方法等を好適に用いることができる。具体的には、約0.1気圧の減圧デシケータ内にアレイ1を約30秒放置する方法などである。
次に、本発明に係るターゲット分子を検出する方法について、説明する。本発明に係るターゲット分子を検出する方法は、反応工程と、ビーズ封入工程と、検出工程とを含む。
次に、アレイ1の構成について、図1を参照して詳細に説明する。
アレイ1とビーズ21は、キットとして構成することができる。アレイ1の収容部13の各々は、このキットが備えるビーズ21を1個のみ収容可能であるように構成されている。
次に、本発明に係るターゲット分子検出装置50の構成について、図2を参照して説明する。図2は、本発明に係るターゲット分子検出装置の一実施形態を模式的に示す図である。
本発明に係る物質封入方法において、基板上に形成された収容部に封入する物質は、ビーズ、核酸、タンパク質、ウイルス、細胞及び脂質膜複合体などであってよい。上記の実施形態では、ビーズ例として説明した。
Claims (11)
- 物質を収容可能な複数の収容部が、側壁によって互いに隔てられて形成されている基板上に、当該物質を含む第一の溶媒を導入する物質導入工程と、
前記物質導入工程の後に、前記第一の溶媒よりも比重が大きい第二の溶媒を前記第一の溶媒上に導入する物質収容工程と、を含む物質封入方法。 - 前記第一の溶媒及び第二の溶媒の少なくとも一方が界面活性剤を含む請求項1記載の物質封入方法。
- 前記第一の溶媒中の前記界面活性剤の濃度が、0.01%〜1%である請求項2記載の物質封入方法。
- 前記第二の溶媒が、飽和炭化水素、不飽和炭化水素、芳香族炭化水素、シリコーンオイル、ヘキサフルオロプロピレンエポキシド系ポリマー、ハイドロフルオロエーテル構造を持つポリマー、パーフルオロポリエーテル、三フッ化塩化エチレンポリマー及びパーフルオロカーボン構造を持つポリマーからなる群より選択される少なくとも1つ又はこれを含む混合物である請求項1〜3のいずれか一項に記載の物質封入方法。
- 前記第一の溶媒が、水、親水性アルコール、親水性エーテル、ケトン、ニトリル系溶媒、ジメチルスルホキシド、及びN,N−ジメチルホルムアミドからなる群より選択される少なくとも1つ又はこれを含む混合物である請求項1〜4のいずれか一項に記載の物質封入方法。
- 前記側壁が、フッ素系高分子樹脂からなる疎水性の上面を有し、前記第二の溶媒がパーフルオロカーボン構造を有するポリマーである請求項4又は5に記載の物質封入方法。
- 前記基板の前記収容部を含む領域が、外部に開放されている請求項1〜6のいずれか一項に記載の物質封入方法。
- 前記物質は、ビーズ、核酸、タンパク質、ウイルス、細胞及び脂質膜複合体から選択される1以上である請求項1〜7のいずれか一項に記載の物質封入方法。
- ターゲット分子を特異的に捕捉するビーズと、当該ターゲット分子とを反応させる反応工程と、
前記反応工程の後に、前記ビーズを用いて請求項1〜8のいずれか一項に記載の物質封入方法を行なうビーズ封入工程と、
前記ビーズ封入工程の後に、前記収容部の各々に前記ターゲット分子を捕捉したビーズが収容されているか否かを検出する検出工程とを含む、ターゲット分子を検出する方法。 - 前記ビーズに、前記ターゲット分子に特異的に結合する分子が結合している請求項9に記載のターゲット分子を検出する方法。
- 基板上に形成された収容部に物質を含む親水性溶媒を導入し、前記収容部に導入された前記物質を含む親水性溶媒を疎水性溶媒で被覆することによって前記収容部内に前記物質を封入する方法であって、
前記物質を収容可能な複数の収容部が、側壁によって互いに隔てられて形成されている基板上に、前記物質を含む親水性溶媒を導入する物質導入工程と、前記物質導入工程の後に、前記親水性溶媒よりも比重が大きい疎水性溶媒を前記親水性溶媒上に導入する物質収容工程と、を含む物質封入方法。
