Abstract
高感度に生体試料を検出することができる検出装置及び検出方法を提供する。生体試料を含む親水性溶媒を充填する複数の収容部(8)を有するマイクロチャンバーアレイ(4)と、前記収容部(8)に対応して画素5が設けられているイメージセンサ(2)とを備える検出装置(50)において、前記マイクロチャンバーアレイ(4)は、前記収容部(8)の開口部(10)に連通している流通路(54)と、前記流通路(54)に連設されている疎水性溶媒供給部(56)と、前記流通路(54)に対し前記親水性溶媒(42)が出入り可能な貫通穴(58)とを有し、前記疎水性溶媒供給部(56)は外部から加えられた力によって疎水性溶媒(44)を前記流通路(54)に流入させることを特徴とする。
Description
本発明は、検出装置及び検出方法に関し、特に生体試料を検出することができる装置に関する。
光学顕微鏡により生きた(活性を有する)タンパク質、核酸などの生体試料の個々の運動を直接的に観察できるようになって以来、主として、光学顕微鏡を用いた測定により、生体試料の分子レベルの機能や活性を検査する方法が採用されている。個々の対象物となる分子を識別するためには、それら分子に蛍光色素、金コロイド又は微小粒子(ポリスチレンビーズ、磁気ビーズなど)の試薬が付加され、光学顕微鏡の分解能では見ることのできない大きさの分子の存在を可視化するといったことが行われる。
ところが、上記方法の場合、大型かつ高価な光学顕微鏡が必要であり、さらに生体試料を直接観察し生体試料の存在をカウントする必要があるので、高速かつ容易に検査をすることができない、という問題があった。
これに対し、生体試料を収容する複数の収容部を有するマイクロチャンバアレイと、当該収容部毎に対応して設けられた画素を有するCMOSイメージセンサとを備える生体試料の検出装置が開示されている(例えば、非特許文献1)。

しかしながら、上記非特許文献1では全ての収容部の開口部は流路で連通しているため、収容部内に収容される生体試料や試薬の濃度が低くなってしまい、測定感度が低下してしまう、という問題があった。
そこで本発明は、高感度に生体試料を検出することができる検出装置及び検出方法を提供することを目的とする。
本発明に係る検出装置は、生体試料を含む親水性溶媒を充填する複数の収容部を有するマイクロチャンバーアレイと、前記収容部に対応して画素が設けられているイメージセンサとを備える検出装置において、前記マイクロチャンバーアレイは、前記収容部の開口部に連通している流通路と、前記流通路に連設されている疎水性溶媒供給部と、前記流通路に対し前記親水性溶媒が出入り可能な貫通穴とを有し、前記疎水性溶媒供給部は外部から加えられた力によって疎水性溶媒を前記流通路に流入させることを特徴とする。
本発明に係る検出方法は、複数の収容部に生体試料を含む親水性溶媒を充填する工程と、前記収容部に励起光を照射する工程と、前記試料の蛍光反応を前記収容部毎にイメージセンサで検出する工程とを備え、前記親水性溶媒を充填する工程は、前記収容部の開口部から前記親水性溶媒を流し込む工程と、前記開口部を疎水性溶媒で閉塞する工程とを有することを特徴とする。
本発明によれば、収容部の開口部を疎水性溶媒で封止し収容部に生体試料を含む親水性溶媒を閉じ込めるので、収容部内に収容される生体試料や試薬の濃度の低下を防いで測定感度を向上することができる。

以下、図面を参照して本発明の実施形態について詳細に説明する。

図1に示す検出装置1は、イメージセンサ2と、干渉フィルタ3と、マイクロチャンバーアレイ4とを備える。検出装置1は、マイクロチャンバーアレイ4に生体試料を含む親水性溶媒42を収容し、当該生体試料の蛍光反応をイメージセンサ2で検出する。
本実施形態の場合、生体試料とは例えばタンパク質、抗体、核酸などの生体分子やウイルス粒子等であり、試薬と反応する。試薬としては例えば、FDG(フルオロデオキシグルコース)や、ビーズを用いることができる。
ビーズは、平均粒子径1μm~4μmであることが好ましい。ビーズの平均粒子径を小さくすることにより、ビーズをマイクロチャンバーアレイ4へ効率的に封入できると共に、マイクロチャンバーアレイ4の高密度化を図ることができる。なお、「平均粒子径」とは、ここでは電子顕微鏡観察又は動的光散乱法を用いて測定した数値をいう。
本実施形態の場合、検出装置1はイメージセンサ2上に干渉フィルタ3を介してマイクロチャンバーアレイ4が搭載されている。イメージセンサ2は、例えばCMOSセンサを用いることができる。イメージセンサ2は、所定の大きさの画素5が複数形成されていると共に、ボンディングパッド部7が設けられている。干渉フィルタ3は、接着剤によってイメージセンサ2の画素5上に固着されており、マイクロチャンバーアレイ4に照射される励起光がイメージセンサ2の画素5に入射することを防止する。
マイクロチャンバーアレイ4は、例えばガラス、シリコン、高分子樹脂等で形成された板状部材からなるチャンバー本体6と、当該チャンバー本体6に形成された複数の収容部8とを有する。チャンバー本体6は、表面が疎水性を有することが好ましい。疎水性とは、親油性と同じ意味であり、疎水性溶媒44との親和性が親水性溶媒42との親和性よりも高いことをいう。この場合チャンバー本体6表面は、例えば撥水性の樹脂、フッ素系高分子樹脂等を用いることができる。フッ素系高分子樹脂としては、高い疎水性を有し、かつ、生体試料に対する毒性が低いアモルファスフッ素樹脂等を好適に用いることができる。アモルファスフッ素樹脂としては、例えば、CYTOP(登録商標)、TEFLON(登録商標)AF2400、及びTEFLON(登録商標)AF1600から選択することができる。
収容部8は、チャンバー本体6表面から厚さ方向に穿設された貫通穴である。収容部8は、試薬と生体試料を効率的に反応させるため、生体試料及び試薬の濃度を十分に高くする必要があり、そのため容積がフェムトリットルクラスであることが好ましい。
図2Aに示すようにイメージセンサ2は、所定の大きさを有する画素5が複数連設されている。本実施形態の場合、イメージセンサ2は1辺の長さが7.5μmの正方形の画素5を複数有する。
図2Bに示すようにマイクロチャンバーアレイ4は、収容部8が所定の間隔を開けて連設されている。当該間隔は、入射した励起光を遮断し得る長さに設定される。本実施形態の場合、マイクロチャンバーアレイ4は15μmの間隔を開けて形成された収容部8を複数有する。
図2Cに示すように、マイクロチャンバーアレイ4は、1個の収容部8が1個の画素5に対応するようにイメージセンサ2上に固定される。
また本図には図示しないが、干渉フィルタ3と、マイクロチャンバーアレイ4及びイメージセンサ2の間の側壁に、黒色の染料を含有する遮蔽材を塗布するのが好ましい。これにより、干渉フィルタ3と、マイクロチャンバーアレイ4及びイメージセンサ2の間の隙間から励起光がイメージセンサ2の画素5に入射することを防ぐことができる。
次に、本実施形態に係る検出装置1の製造方法について説明する。まず、厚さ150μmのイメージセンサ11の画素5上に接着剤(本実施形態の場合、CYTOP(登録商標))14を2μmの厚さに塗布する。次いで、180℃で加熱しながら一面に厚さ3μmの干渉フィルタ3を設けた厚さ60μmのSi基板12を、干渉フィルタ3の表面を上記接着剤14に重ね、干渉フィルタ3を介してSi基板12とイメージセンサ11とを固定する(図3A)。
次いで、Si基板12上に200nmの厚さのAl膜16を蒸着法により形成し、当該Al膜16上にレジスト18を設ける(図3B)。
次いで、レジスト18にパターンを形成し貫通穴20を穿設する(図3C)。ボンディングパッド部7を保護する厚膜レジスト22を形成した後で、ウェットエッチングによりAl膜16に貫通穴24を形成する(図3D)。
さらにドライエッチング(Deep Reactive Ion Etching : DRIE)によりSi基板12に貫通穴26を形成する(図3E)。次いで、レジスト18及びAl膜16を混酸(酢酸,リン酸,硝酸,純水(体積比4:4:1:1))により除去する(図3F)。
次いで、ガラス基板28上に載置してボンディングパッド部7を保護する厚膜レジスト30を形成した後で、イメージセンサ11の裏面にAlのパターン32を形成し(図4A)、さらにDRIEにより外形加工を施して検出装置1を得ることができる(図4B)。
形成された検出装置1は、ポリイミド基板34上に固定され、ボンディングパッド部7においてワイヤボンディングされる(図4C)。
ボンディングワイヤ36、及びボンディングパッド部7はエポキシ樹脂40で覆われている。また、干渉フィルタ3と、マイクロチャンバーアレイ4及びイメージセンサ2の間に、黒色の染料を含有する遮蔽材38が設けられている(図4D)。
次に、上記のように構成された検出装置1の動作及び効果を説明する。まず、生体試料と試薬とを含む親水性溶媒42をマイクロチャンバーアレイ4の収容部8に導入する(図5A)。親水性溶媒42は例えば、水、親水性アルコール、親水性エーテル、ケトン、ニトリル系溶媒、ジメチルスルホキシド(DMSO)、及びN,N-ジメチルホルムアミド(DMF)からなる群より選択される少なくとも1つ又はこれを含む混合物等を用いることができる。
次いで、検出装置1を減圧環境下に放置することにより、脱気をする。これにより、収容部8内の空気を除去し、生体試料を含む親水性溶媒42を収容部8に効率的に導入することができる(図5B)。
次いで収容部8の開口部10周辺に疎水性溶媒44を導入する(図5C)。疎水性溶媒44は、親水性溶媒42と混ざらない溶媒であればよく、例えば飽和炭化水素、不飽和炭化水素、芳香族炭化水素、シリコーンオイル、パーフルオロカーボン、ハロゲン系溶媒、及び疎水性イオン液体からなる群より選択される少なくとも1つ又はこれを含む混合物等を好適に用いることができる。
疎水性溶媒44を収容部8の開口部10周辺に導入することにより、各収容部8に疎水性溶媒44により覆われた液滴を形成し、収容部8内に生体試料を封入することができる。これにより、生体試料をフェムトリットルクラスの収容部8に封入することができる。
次いで、検出装置1のマイクロチャンバーアレイ4に対し、光源46から励起光を照射する(図6)。生体試料に結合した所定の酵素によって蛍光基質が分解されて蛍光物質が遊離する。この蛍光物質を干渉フィルタ3を介して各収容部8に対応する画素5Aで検出する。一方、親水性溶媒が充填されなかった収容部及び収容部同士の間に対応する画素5B,5Cにおいて蛍光物質は検出されない。
これにより、検出装置1は、収容部8の開口部10を疎水性溶媒44で封止し収容部8に生体試料を含む親水性溶媒42を閉じ込めるので、収容部8内に収容される生体試料や試薬の濃度の低下を防いで測定感度を向上することができる。
また、検出装置1は、試薬が収容されている収容部8の数と、生体試料を捕捉した試薬が収容されている収容部8の数とを検出して、試薬の全数のうちターゲット分子を捕捉した試薬の数の割合を算出することができる。これにより、ターゲット分子の濃度を定量化することが可能となる。
しかも、本実施形態の場合、イメージセンサ2は各収容部8に対応した画素5において各収容部8の試薬の有無及び生体試料を捕捉した試薬が収容されているか否かを検出することができるので、従来のように光学顕微鏡を必要とせず検出システムの小型化を図ることができると共に、検出作業の高速化を図ることができる。
次に、実際に製造した検出装置1を用いて行った評価について説明する。製造した検出装置1のマイクロチャンバーアレイ4は、厚さが60μm、収容部8の有効長さが4μm、収容部8同士の間の間隔が15μmとなるように形成されている。イメージセンサ2は、チップサイズが1.2mm×3.6mmのCMOSセンサを用い、画素5の1辺の長さが7.5μm、アレイサイズが120×268とした。
まず、励起光の適切な入射角度について確認した。励起光には水銀ランプからの白色光を青色干渉フィルタ(透過中心波長469nm,波長幅35nm)により選択した青色光を用いた。マイクロチャンバーアレイ4に対し照射する励起光は、図7に示すように、収容部8の開口部10に対し斜め上方から照射するのが好ましい。すなわち収容部8の開口部10に対し、垂直方向に励起光を照射した場合、イメージセンサ2の画素5に励起光が直接入射してしまう。これに対し、図7Bに示すように、収容部8の開口部10に対し斜め上方から照射した場合、イメージセンサ2は当該励起光を検出しないので、より確実に蛍光物質を検出することができる。
図8は、励起光の入射角度とイメージセンサ2における検出強度の関係を示すグラフであり、●はマイクロチャンバーアレイ4あり、□はマイクロチャンバーアレイ4なしの結果である。入射角度は、鉛直方向を0度とし、当該鉛直方向の伸びる軸と励起光が入射する方向に平行な軸とのなす角をいう。この結果から入射角度を変えることによりマイクロチャンバーアレイ4が励起光を遮断し得ることが確認できた。
図9は、本実施形態に係る検出装置1で検出した結果を示す図である。なお、親水性溶媒42としてリン酸緩衝液、生体試料としてβ-gal(β-galactosidase:10nM)、試薬としてFDG(4mM)を用いた。
図9Aはマイクロチャンバーアレイ4を上方からマイクロスコープで観察した画像である。各収容部8が明るく光っていることから、各収容部8に試薬と反応した生体試料が収容されていることが確認される。
図9B~図9Dは、図9A中のAにおける検出装置1の検出結果を示す。時間経過と共に蛍光反応が進み、それに伴い蛍光反応を検出する画素5が増加していることが確認された。
図10Aは検出装置1における検出時間と検出強度との関係を示すグラフである。この結果から、本実施形態に係る検出装置1は、従来の光学顕微鏡の結果(図10B)と遜色のない検出結果を得られることが確認された。
(第2実施形態)
次に本発明の第2実施形態に係る検出装置1について図11を参照して説明する。上記第1実施形態と同様の構成については同様の符号を付し、簡略のため説明を省略する。
次に本発明の第2実施形態に係る検出装置1について図11を参照して説明する。上記第1実施形態と同様の構成については同様の符号を付し、簡略のため説明を省略する。
本図に示す検出装置50は、イメージセンサ2と、干渉フィルタ3(本図には図示しない)と、本体52と、マイクロチャンバーアレイ4とを備える。
本体52は、底面が開口しており、上面が閉塞している筒状の部材で構成されている。本体52の底面の開口を塞ぐようにイメージセンサ2が設けられている。イメージセンサ2上に設けられたマイクロチャンバーアレイ4上には流通路54が形成されている。流通路54はマイクロチャンバーアレイ4の各収容部8とそれぞれの開口部10を介して連通している。流通路54及び全ての収容部8は親水性溶媒42で満たされている。
流通路54の一端には疎水性溶媒供給部56が設けられている。疎水性溶媒供給部56には疎水性溶媒44が収容されている。疎水性溶媒供給部56の本体52上面には、外力で弾性変形可能な変形部62が設けられている。
本体52の上面には、流通路54と外部とを連通する一対の貫通穴58と、当該貫通穴58を閉塞する蓋部60とが着脱自在に設けられている。本体52は、内部の高さAが疎水性溶媒44の油滴の大きさ以下であることが好ましい。内部の高さAを油滴の大きさ以下とすることにより、疎水性溶媒44と親水性溶媒42が混ざり合わずに本図に示すように分離した状態で保持される。したがって、流通路54と疎水性溶媒供給部56との境界は互いを区画する部材を必要としない。通常、水中において安定な油滴の大きさは100μm程度であるので、内部の高さAは、100μm以下であることが好ましい。
次に上記のように構成された検出装置50の動作及び効果について説明する。まず、図11Bに示すように、蓋部60を取り外し、一方の貫通穴58から生体試料と試薬65を含む親水性溶媒42を流通路54内に導入する。これにより、試薬65は各収容部8に収容される。
次いで、疎水性溶媒供給部56の変形部62に対し外部から力を付加し、変形部62を弾性変形させる。この力により疎水性溶媒供給部56に収容されていた疎水性溶媒44が流通路54内に流入し、各収容部8に疎水性溶媒44により覆われた液滴を形成し、収容部8内に生体試料を封入することができる。
本実施形態に係る検出装置50は、生体試料と試薬65を含む親水性溶媒42を導入した後、1回的な動作で収容部8内に生体試料を封入することができる。したがって疎水性溶媒44を流入させる準備を省略することができるので、検査作業をより簡略化することができる。
なお、本実施形態では、流通路54と疎水性溶媒供給部56との境界は互いを区画する部材がない場合について説明したが、本発明はこれに限られない。例えば、流通路54と疎水性溶媒供給部56との境界に隔壁を設けることとしてもよい。当該隔壁は、変形部62に付加された力によって破断可能に形成される。
これにより、生体試料と試薬65とを含む親水性溶媒42を流通路54内に導入した段階では疎水性溶媒44は隔壁によって疎水性溶媒供給部56に確実に収容されている。そして、変形部62に力を付加することによって、当該力が疎水性溶媒供給部56に収容されている疎水性溶媒44を介して隔壁に伝達される。伝達された力によって隔壁が破断すると、疎水性溶媒供給部56に収容されていた疎水性溶媒44が流通路54内に流入し、各収容部8に疎水性溶媒44により覆われた液滴を形成し、収容部8内に生体試料を封入することができる。
このように流通路54と疎水性溶媒供給部56との境界に隔壁を設けることにより、生体試料と試薬65とを含む親水性溶媒42を流通路54内に導入した段階、すなわち試薬65が各収容部8に収容される段階において疎水性溶媒44が流通路54内に進入することをより確実に防ぐことができるので、作業の確実性を向上することができる。

本発明は上記実施形態に限定されるものではなく、本発明の趣旨の範囲内で適宜変更することが可能である。

Claims (7)
- 生体試料を含む親水性溶媒を充填する複数の収容部を有するマイクロチャンバーアレイと、
前記収容部に対応して画素が設けられているイメージセンサと
を備える検出装置において、
前記マイクロチャンバーアレイは、
前記収容部の開口部に連通している流通路と、
前記流通路に連設されている疎水性溶媒供給部と、
前記流通路に対し前記親水性溶媒が出入り可能な貫通穴と
を有し、
前記疎水性溶媒供給部は外部から加えられた力によって疎水性溶媒を前記流通路に流入させる
ことを特徴とする検出装置。 - 前記疎水性溶媒供給部と前記流通路の間に前記力によって破断可能な隔壁が設けられていることを特徴とする請求項1に記載の検出装置。
- 前記疎水性溶媒供給部は外部から加えられた力によって弾性変形可能な変形部を有することを特徴とする請求項1又は2に記載の検出装置。
- 前記流通路に前記試料を含まない前記親水性溶媒が充填されていることを特徴とする請求項1~3のいずれか1項に記載の検出装置。
- 前記貫通穴を閉塞する開閉可能な蓋部が設けられていることを特徴とする請求項1~4のいずれか1項に記載の検出装置。
- 複数の収容部に生体試料を含む親水性溶媒を充填する工程と、
前記収容部に励起光を照射する工程と、
前記試料の蛍光反応を前記収容部毎にイメージセンサで検出する工程と
を備え、
前記親水性溶媒を充填する工程は、
前記収容部の開口部から前記親水性溶媒を流し込む工程と、
前記開口部を疎水性溶媒で閉塞する工程と
を有することを特徴とする検出方法。 - 前記励起光を前記開口部に対し斜め上方から照射することを特徴とする請求項6に記載の検出方法。
