Abstract
Description
(i) キレート剤を含有する培地中にて、0.008 MPa以上かつ0.08 MPa以下の平均圧力条件下にて該微生物を培養する工程、及び
(ii) 培養物より、生成されたゼアキサンチンを回収する工程、
を含む、上記方法。
本発明におけるゼアキサンチン生産微生物は、パラコッカス(Paracoccus)属に属する微生物であり、生産される総カロテノイド量に対するゼアキサンチンの比率が高い微生物である。
本発明にて用いられる培地は、キレート剤を含有する。「キレート剤」としては、鉄元素をキレート化し、培地中に可溶化させる作用を有する物質であれば特に限定されない。このようなキレート剤としては公知のものを利用することができ、例えば、クエン酸、リンゴ酸、コハク酸、フマル酸、酒石酸、酢酸、グルコン酸、フィチン酸、ポリアミノリン酸、乳酸、アルキルジホスホン酸、エチレンジアミン四酢酸(EDTA)、ニトリロ三酢酸、1,2−ジアミノシクロヘキサン四酢酸、ヒドロキシエチルエチレンジアミン三酢酸、ヒドロキシエチルイミノ二酢酸、イミノ二酢酸、ジエチレントリアミン五酢酸、ジヒドロキシエチルグリシン、ジアミノプロパン四酢酸、ニトリロトリス(メチレンホスホン酸)、エチレンジアミンテトラキス(メチレンホスホン酸)及びそれらの塩が挙げられる。塩としては水溶性塩が挙げられ、例えばナトリウム塩やカリウム塩などのアルカリ金属塩やアンモニウム塩等が挙げられる。キレート剤として好ましくはクエン酸又はクエン酸塩である。このようなキレート剤を培地に含めることによって、ゼアキサンチン生産微生物によるゼアキサンチン生産及び当該微生物の増殖を阻害する圧力条件下においても、ゼアキサンチン生産微生物のゼアキサンチン生産能及び増殖能を向上させることができる。これらキレート剤の培地に加える添加量は、用いるキレート剤の種類や鉄元素をキレート化する能力によって変更されるが、ゼアキサンチン生産微生物によるゼアキサンチン生産及び当該微生物の増殖を阻害する圧力条件下においても、ゼアキサンチン生産微生物のゼアキサンチン生産能及び増殖能を向上させる範囲にて適宜調節することができる。用いるキレート剤がクエン酸又はその塩であれば、培地1L当たり14 g未満、好ましくは10.0g以下、より好ましくは8.4 g以下である。クエン酸又はその塩の添加量の下限は特に限定されないが、少なすぎれば十分なキレート効果が得られないことから、培地1L当たり0.5 g以上、好ましくは1.0 g以上、より好ましくは1.5 g以上、さらに好ましくは2.8 g以上である。したがって、クエン酸又はその塩の添加量として、培地1L当たり好ましくは0.5 g以上かつ10.0g以下、より好ましくは2.8 g以上かつ8.4 g以下、の範囲から選択される量を用いることができる。
本発明はゼアキサンチン生産微生物の培養において、当該ゼアキサンチン生産微生物に対して0.008 MPa以上かつ0.08 MPa以下、または0.008 MPa以上かつ0.08 MPa未満、好ましくは0.02 MPa以上かつ0.08 MPa以下、または0.02 MPa以上かつ0.08 MPa未満、さらに好ましくは0.05 MPaの圧力が付加される状況を有する培養方法に適用することができる。ただし、大型の培養槽を用いた場合、液相部分の圧力(水圧)は深度によって異なり、培地上部の圧力条件と培地下部の圧力条件に差が生じる。したがって、大型の培養槽を用いた場合の上記圧力値は培養系(液相部分)にかかる平均圧力値とすることができる。例えば、大型培養槽(高さ10 M)を用いた一般的な培養方法について例示すると、当該培養槽に対して深さ6 Mまで培地を満たし、気相に0.02 MPaの圧力を付加した場合、当該培養系にかかる平均圧力値は、0.02 MPa(気相部)+0.03 MPa(培地水深中央値3 Mの水圧)として求めることができる。
本発明にて生産されたゼアキサンチンは、公知の手法に基づいて回収することができる。すなわち、培養終了後、培養液からろ過法、遠心分離法などにより培地成分や水分を除去することによって、ゼアキサンチンを含む培養沈殿物を得ることができる。必要であればさらに、当該培養沈殿物を噴霧乾燥、ドラム乾燥、凍結乾燥等により乾燥し、ゼアキサンチンを含む沈殿乾燥物を得ることができる。
シード用フラスコ培地は以下の組成を有するものを使用した。
基本培地は以下の組成を有するものを使用した。
ゼアキサンチン生産微生物は、パラコッカス(Paracoccus)属に属するアスタキサンチン生産微生物より上記手法により得られた変異株を使用した。
微生物を、上記シード用フラスコ培地に一白金耳植菌し、28℃で2日間、100rpmで回転振とう培養を行った後、その培養液80mLを基本培地を入れた発酵槽に植菌した。
圧力の制御は手動で背圧弁を調整して制御した。
ゼアキサンチンの定量は、高速液体クロマトグラフィー(HPLC)を用いて以下のとおり行った。
[菌体濃度の測定]
波長610 nmにて、培養終了後の培養液の吸光度(OD610)を、分光光度計を用いて計測しこの値を培養液の菌体濃度とした。
ゼアキサンチン生産微生物を0.003MPa、0.025MPa、0.05 MPa、0.075MPa、又は0.1 MPaの圧力条件下、クエン酸ナトリウム未添加の基本培地を用いて上記培養方法にて培養を行った。
ゼアキサンチン生産微生物を0.05 MPaの圧力条件下、クエン酸ナトリウムを0 g/L、2.8 g/L、8.4 g/L、14g/L、又は21g/Lの濃度となるように添加して調製した基本培地を用いて、上記培養方法にて培養を行った。
ゼアキサンチン生産微生物を0.003 MPa、0.02 MPa、0.04 MPa、0.06 MPa、0.08 MPa、又は0.1 MPaの圧力条件下、クエン酸ナトリウムを2.8 g/Lの濃度となるように添加して調製した基本培地を用いて、上記培養方法にて培養を行った。
クエン酸ナトリウムを0 g/L、2.8 g/L、8.4 g/L、又は14 g/Lの濃度となるようにそれぞれ添加して調製した基本培地を遠心分離して上清を回収した。次いで、得られた各上清中のカルシウムイオン、鉄イオン、カリウムイオン、マグネシウムイオン、及びリン酸イオンの濃度をそれぞれ測定した。各陽イオン濃度はICP発光分析法により測定し、リン酸イオンについてはモリブデンブルー吸光度法で測定した。
Claims (17)
- パラコッカス(Paracoccus)属に属するゼアキサンチン生産微生物を用いたゼアキサンチンの製造方法であって、
(i) キレート剤を含有する培地中にて、0.008 MPa以上かつ0.08 MPa以下の平均圧力条件下にて該微生物を培養し、ゼアキサンチンを生成する工程、及び
(ii) 培養物より、生成されたゼアキサンチンを回収する工程、
を含む、上記方法。 - 工程(ii)が、培養物より培地成分及び水分を除去して乾燥させることによりゼアキサンチンを含む組成物を得る工程を含む、請求項1に記載の方法。
- さらに、ゼアキサンチンを含む組成物よりゼアキサンチンを精製する工程を含む、請求項2に記載の方法。
- キレート剤がクエン酸又はその塩である、請求項1〜3のいずれか1項に記載の方法。
- 培地がクエン酸又はその塩を14 g/L未満の濃度で含有する、請求項4に記載の方法。
- 培地がクエン酸又はその塩を0.5 g/L以上かつ10 g/L以下の濃度で含有する、請求項5に記載の方法。
- 0.02 MPa以上かつ0.08 MPa以下の平均圧力条件下にて前記微生物を培養する、請求項1〜6のいずれか1項に記載の方法。
- キレート剤を含有する培地中にて、パラコッカス(Paracoccus)属に属するゼアキサンチン生産微生物を培養することを含む、0.008 MPa以上かつ0.08 MPa以下の圧力条件下における該微生物のゼアキサンチン生産能を向上させる方法。
- キレート剤がクエン酸又はその塩である、請求項8に記載の方法。
- 培地がクエン酸又はその塩を14 g/L未満の濃度で含有する、請求項9に記載の方法。
- 圧力条件が0.02 MPa以上かつ0.08 MPa以下である、請求項8〜10のいずれか1項に記載の方法。
- キレート剤を含有する培地中にて、パラコッカス(Paracoccus)属に属するゼアキサンチン生産微生物を培養することを含む、0.008 MPa以上かつ0.08 MPa以下の圧力条件下における該微生物を培養する方法。
- キレート剤がクエン酸又はその塩である、請求項12に記載の方法。
- 培地がクエン酸又はその塩を14 g/L未満の濃度で含有する、請求項13に記載の方法。
- 圧力条件が0.02 MPa以上かつ0.08 MPa以下である、請求項12〜14のいずれか1項に記載の方法。
- 請求項2に記載の方法により製造された、ゼアキサンチンを含む組成物。
- 請求項3に記載の方法により製造された、ゼアキサンチン。
