Abstract
Description
また、複数回の投与によってマラリアに特異的な抗体の産生を得ること、それによりマラリアの感染を抑制し得ることを見出した。更に、癌抗原として悪性黒色腫の表面抗原をコードしたpDNAを用いた場合に悪性黒色腫に特異的な細胞性免疫を誘導し、悪性黒色腫の増殖と転移を抑制し得ることを見出した。
また、抗原もしくは薬物とカチオン性分子との複合体にホスファチジルセリン、N−ラウロイルザルコシンおよびグリチルリチンから選択されるアニオン性分子を添加することにより、それぞれ、脾臓、肺または肝臓への選択性(臓器選択性)を付与した、抗原もしくは薬物送達複合体の作製にも成功した。
さらに、アニオン性分子としてγ−ポリグルタミン酸を使用することで、樹状細胞へ指向性を有する複合体を作製できることを発見した。
これらの複合体は、抗原もしくは薬物とカチオン性分子との複合体、そしてアニオン性分子が静電的に結合しているので安全性が極めて高く、細胞障害性がないことも明らかになった。
さらに、抗原もしくは薬物とカチオン性分子との複合体に、さらにアプタマー、miRNAおよび抗体から選択されるアニオン性分子を添加することで、より特異的な細胞もしくは臓器選択性を付与した複合体の作製にも成功した。

[28’]薬物がDNA、siRNA、miRNA、アンチセンスDNAなど負電荷の核酸である、[15’]又は[25’]に記載の薬物送達複合体。
特に本発明の複合体は、標的となる細胞や臓器への高い指向性を有することを特徴とするので、新たなドラッグデリバリーシステムの有効成分となり得る。
以下、本発明の抗原送達複合体と、本発明の薬物送達複合体のそれぞれについて説明する。
そのような病原体抗原としては、病原体の表面抗原(例えば、タンパク質(抗原)、ペプチド(抗原)など)及びそれらをコードする核酸(例えば、DNA)抗原等から選択される。
より大きなサイズの核酸については、通常用いられる方法で適宜調製することができる。
本発明において用いられる核酸は、特に限定されないが、当業者が通常用いる方法により精製されていることが好ましい。
不活化ウイルスとしては、通常行われている手法により得られたものであればいかなるものでも使用することができるが、例えば、ホルマリン処理、紫外線照射、β−プロピオラクトン等により得たものを使用することができる。
また、RNAの種類は、使用の目的に応じて適宜選択することができ、特に限定されないが、例えばsiRNA、miRNA、shRNA、アンチセンスRNA、メッセンジャーRNA、一本鎖RNAゲノム、二本鎖RNAゲノム、RNAレプリコン、トランスファーRNA、リボゾーマルRNA等が挙げられ、好ましくはsiRNA、miRNA、shRNA、mRNA、アンチセンスRNA、RNAレプリコンである。
本発明の薬物送達複合体は、赤血球凝集を引き起こさず、また細胞毒性も低く、しかも細胞内取り込み効率に優れ、また細胞や臓器への指向性を有するので、安全かつ有効に用いられる分子プローブであるといえる。
カチオン性デンドリマーが有するカチオン性の基(例、アミノ基)には、共有結合を介して色素や蛍光色素分子などの薬物を導入することができ、またキレート剤にも、放射性核種やMRI造影剤、ランタノイドなどの薬物を導入することが可能である。
カチオン性デンドリマーにキレート剤を結合させる方法や、本発明の薬物とカチオン性デンドリマーおよびキレート剤との複合体の製造方法は、当業者に公知である。例えば、薬物複合体の製造方法としては、カチオン性デンドリマーとキレート剤を混合させてカチオン性デンドリマーとキレート剤との複合体を形成し、これをイメージング用核種で標識する方法が挙げられる。
好ましいアニオン性分子としては、アニオン性高分子、アニオン性脂質(例、ラウロイルサルコシン(以下、LSと略記する場合がある)、ホスファチジルセリン(以下、DOPSと略記する場合がある)など)、全体として負電荷を帯びた遺伝子(例、アプタマーなど)、全体として負電荷を帯びた抗体などが挙げられる。なかでも、例えばラウロイルサルコシン、γ−ポリグルタミン酸(以下、γ−PGAと略記する場合がある)、グリチルリチン(以下、GLYと略記する場合がある)、ホスファチジルセリン、コンドロイチン硫酸、アルギン酸などが特に好ましい。
これらのアニオン性分子は、種々の核種(例えば、天然放射性核種、人工放射性核種、陽電子放出核種など)を含むものであってもよいし、核種によって放射ラベルされたものであってもよい。あるいは、種々の化合物(例えば、公知の蛍光色素、酵素など)で標識されたものであってもよい。
たとえば、脾臓、肺または肝臓への指向性を与える場合、アニオン性分子としては、ホスファチジルセリン、ラウロイルサルコシンおよびグリチルリチンから選択することが望ましい。特に脾臓への指向性を与える場合はホスファチジルセリンを選択し、肺への指向性を与える場合はラウロイルサルコシンを選択し、そして肝臓への指向性を与える場合はグリチルリチンをそれぞれ選択することが望ましい。
各臓器において本発明の複合体が取り込まれる細胞としては特に限定されず、各臓器を形成する何れの細胞であってもよい。例えば、肝臓への指向性を有する本発明の複合体は、肝実質細胞に取り込まれる。
(a)抗原もしくは薬物とカチオン性分子(カチオン性高分子又はカチオン性脂質により構成されるリポソームやミセルを含む)との混合により自己組織化して得られる正の表面電荷を有する複合体(以下、自己組織化複合体という場合がある);
(b)抗原もしくは薬物を内包したカチオン性ミセル(マイクロスフェア);
(c)抗原もしくは薬物を内包したカチオン性リポソーム;
(d)カチオン性デンドリマーおよびキレート剤との複合体を形成する薬物と、カチオン性分子との混合により自己組織化して得られる正の表面電荷を有する複合体
などが挙げられるが、それらに限定されない。リポソームの場合、単層型、多重層型のいずれであってもよい。
特に本発明の薬物送達複合体において、薬物がsiRNAである場合、本発明の複合体はジオレイルホスファチジルエタノールアミンを含有したリポソームと自己組織化されることが好ましい。
このような形態のカチオン性複合体としては、抗原もしくは薬物と、カチオン性脂質ならびにDOPEおよびコレステロールから選択される脂質からなるリポソームあるいはミセルとから構成されるカチオン性複合体が挙げられる。この実施形態において、カチオン性脂質は、DOTMA、DOTAPおよびDC−Cholから選択されることが特に好ましい。

すなわち「配合比」は、本発明の複合体を形成する各物質における、カチオン性の官能基(例えばアミノ基)またはアニオン性の官能基(例えばカルボキシル基やスルホニル基)の「電荷比」によっても定義することができる。
すなわち本発明の好ましい実施態様によれば、アニオン性分子が、ホスファチジルセリンであり;抗原もしくは薬物、カチオン性分子、ホスファチジルセリンの配合比が1:2:4〜1:8:8であり;かつ脾臓への指向性を有することを特徴とする、複合体を提供する。配合比をこの範囲内にすることで、脾臓指向性に最適な表面荷電を有する本発明の複合体が得られる。
この場合のカチオン性高分子およびカチオン性脂質としては特に限定されないが、ポリエチレンイミンやポリリジン、ポリアルギニンであることが好ましい。
この場合のカチオン性高分子およびカチオン性脂質としては特に限定されないが、カチオン性高分子がポリエチレンイミンやポリリジン、ポリアルギニン、カチオン性脂質がDOTMAまたはDOTAPであることが好ましい。
この場合の抗原もしくは薬物として好ましくは、核酸である。カチオン性分子として好ましくは、ポリエチレンイミンやポリリジン、ポリアルギニンである。
特に抗原もしくは薬物として蛋白抗原を用いる場合、カチオン性分子として塩化ベンザルコニウムを用い、かつアニオン性分子としてγ−ポリグルタミン酸を用いると、樹状細胞への指向性が一層強力となる。
上記培養時間が短すぎると、抗原もしくは薬物が十分細胞内へ導入されず、培養時間が長すぎると、細胞が弱ることがある。
更なる培養の時間は、抗原もしくは薬物に期待される機能等を考慮して、適宜設定することが可能であるが、抗原もしくは薬物が発現ベクター等のプラスミドDNAである場合は、通常8〜72時間、好ましくは8〜60時間、より好ましくは8〜48時間、更に好ましくは8〜36時間、最も好ましくは12〜32時間の範囲である。
即ち、本発明の複合体を対象に投与することにより、該複合体が標的細胞または標的臓器へ到達・接触し、生体内で該複合体に含まれる抗原もしくは薬物が細胞または臓器へと導入される。
具体的には、抗原もしくは薬物として病原体抗原又はそれをコードするDNAを用いた場合に感染症ワクチンとして使用することができ、抗原もしくは薬物として癌抗原を用いた場合に癌ワクチンとして使用することができる(以下、本発明の感染症ワクチン及び癌ワクチンを総称して本発明のワクチンともいう)。
本発明の複合体は、自身が有する指向性に基づいて特定の細胞または臓器に送達されるので、上記輸送体(以下、本発明の輸送体と記載する場合がある)は、プローブのみを用いる従来の診断方法・イメージング方法に比べて、目的部位をより正確に診断することが可能である。
本発明の輸送体は、本発明の複合体そのものであってもよいし、本発明の複合体を含んでなるものであってもよい。
しかしながら、特にリンパ節のイメージングを行う場合、カチオン性分子としてポリエチレンイミンを選択し、アニオン性分子としてγ−ポリグルタミン酸を選択することが好ましい。
この薬物複合体/ポリエチレンイミン/γ−ポリグルタミン酸の組合せにより、本発明の複合体がリンパ節への指向性を有するようになるため、薬物複合体に含まれるイメージング用核種によって、様々な用途に適したリンパ節イメージングが可能となる。
センチネルリンパ節は、腫瘍からリンパ流に入った癌細胞が最初に到達し、領域リンパ節の中で最初に転移が生ずるリンパ節のことをいう。そのため、悪性腫瘍から他組織への転移が起こっているか否かを判別する上でセンチネルリンパ節の生検を行う必要があるが、センチネルリンパ節は肉眼で区別することができないため、確実に採取する上でイメージングは重要である。
本発明によれば、様々なイメージング法に適用可能であり、かつリンパ節への指向性を有し、特にセンチネルリンパ節に滞留可能なイメージング用核種の輸送体が提供される。すなわち、抗原もしくは薬物が、イメージング用核種、カチオン性デンドリマーおよびキレート剤からなる複合体であり;カチオン性分子が、ポリエチレンイミンであり;アニオン性分子が、γ−ポリグルタミン酸であり;そしてリンパ節(特にセンチネルリンパ節)への指向性を有することを特徴とする複合体を含む、イメージング用核種の輸送体が提供される。
この場合、本発明の複合体における、抗原もしくは薬物、カチオン性分子、アニオン性分子のいずれかが診断装置(方法)やイメージング装置(方法)に適した修飾をうけていることが望ましく、そのような修飾としては、種々の核種(例えば、PET用放射性核種、SPECT用放射性核種など)による標識、MRI造影剤(MRI造影効果を有する元素)による標識、蛍光色素(例えば、FITC、Alexa Fluorなど)による標識、酵素(例えば、ペルオキシダーゼなど)による標識などが挙げられる。
方法
マラリア抗原MSP−1をコードしたpDNAであるpMSP−1とポリエチレンイミン(PEI)、γ−ポリグルタミン酸(γ−PGA)の電荷比が1:8:6になるようにpMSP−1/PEI/γ−PGA複合体を作製した。また、薬効を持たないホタルルシフェラーゼをコードしたpCMV−Lucを用いたpCMV−Luc/PEI/γ−PGA複合体をコントロールのために用いた。pCMV−Luc/PEI/γ−PGA複合体をDNA量として40μg(A)、pMSP−1/PEI/γ−PGA複合体をDNA量として0.4(B)、4(C)、40(D)、80(E)μgになるようにマウスに三週間おきに計三回静脈内投与した。最終免疫から10日後にマウス血清を採取し、ELISAを用いてマラリアに対する抗体価を測定した。また、最終免疫から三週間後にマウスにマラリア原虫を感染させ、感染後のマウスの生存を経日的に観察した。
ワクチン抗体価を図1に示す。血清希釈が320倍以上である場合には抗体価に差異はないが、希釈倍率がそれより低い場合には複合体に含まれるpDNA量の多いものの方がより高い抗体価であった。また、コントロール群では抗体価の上昇は認められなかった。
マラリア感染後の生存率を図2に示す。pCMV−Luc/PEI/γ−PGA複合体投与群、又はpMSP−1/PEI/γ−PGA複合体投与群のpDNA量が〜4μgの場合は感染後9日で生存率がほぼ0%になったのに対して、pDNAが40μgと80μgの場合は9日経過後も生存率が高く保たれていた。
方法
不活化インフルエンザウイルス(Vaccine)を塩化ベンザルコニウム(BAC)とγ−PGAを用いて微粒子化したVaccine/BAC/γ−PGA複合体を作製した。Vaccine単体、VaccineにBAC又はγ−PGAのいずれかを混合したもの、あるいはVaccine/BAC/γ−PGA複合体をマウスに投与し、4週間後に採血し、インフルエンザウイルスに対する抗体価をELISAを用いて測定した。
図3には各ワクチンの抗体産生効果(ワクチン抗体価)を示す。VaccineにBAC又はγ−PGAのいずれかを混合しても、抗体価はVaccine単体と有意な差異は得られなかったが、Vaccine/BAC/γ−PGA複合体はVaccine単体と比較して有意に高い抗体価が得られた。
方法
モデルタンパク質として牛血清アルブミン(BSA)をローダミンBイソチオシアネートを用いて蛍光標識したものを用いた。BSA又はBSAに塩化ベンザルコニウム(BAC)とγ−PGAを用いて作製したBSA/BAC/γ−PGA複合体を樹状細胞株であるDC2.4細胞に添加し、2時間インキュベートした。細胞を洗浄し、22時間後に細胞の蛍光像を蛍光顕微鏡を用いて測定した。
細胞の蛍光顕微鏡観察像を図4に示す。BSAのみでは細胞内取り込みは認められなかったが、BSA/BAC/γ−PGA複合体を添加した細胞においてはBSAの蛍光が強く観察され、BSA/BAC/γ−PGA複合体が樹状細胞に高い細胞内取り込みを示すことが明らかとなった。
方法
メラノーマ抗原遺伝子としてgp100とTRP−2のユビキチン化エピトープをコードしたpDNAであるpUb−MとPEI、γ−PGAの配合比(電荷比)が1:8:6になるようにpUb−M/PEI/γ−PGA複合体を作製した。C57BLマウスに5%糖液(control)又はpUb−Mのみ、薬効をもたないpCMV−LucとPEI、γ−PGAを用いて作製したpCMV−Luc/PEI/γ−PGA複合体、pUb−M/PEI/γ−PGA複合体を二週間おきに計四回投与し、免疫を惹起した。最終免疫から二週間後にメラノーマ細胞株B16−F10細胞、又はルシフェラーゼを恒常発現したB16−F10−Luc細胞を皮内又は静脈内に投与し、それぞれ皮内移植モデルと肺転移モデルを作製した。皮内移植モデルの腫瘍径を経日的に測定した。また、肺転移モデル作製から21日目に肺を摘出し、観察した後に、肺のルシフェラーゼ活性を測定し、肺転移細胞数を算出した。
皮内移植された悪性黒色腫の腫瘍重量の変化を図5に示す。pUb−Mのみ、又はpCMV−Luc/PEI/γ−PGA複合体を投与したマウスの腫瘍増殖はコントロール群と有意な差異は認められなかったが、pUb−M/PEI/γ−PGA複合体を投与したマウスの腫瘍増殖はコントロール群と比較して有意に低値を示した。
静脈注入された悪性黒色腫の肺転移の様子を図6に示す。また、肺転移した細胞数を図7に示す。5%糖液投与群(control)に比べ、pUb−M/PEI/γ−PGA複合体投与群では肺転移が顕著に抑制された。
方法
pDNA(pCMV−Luc)とPEIを混合し、pDNA/PEI複合体を調製した。また、このpDNA/PEI複合体にN−[1−(2,3−ジオレイルオキシ)プロピル]−N,N,N−トリメチルアンモニウム(DOTMA)を添加し、さらにN−ラウロイルザルコシン(LS)を添加することで新規薬物送達複合体を調製した。pDNAとPEIとDOTMAとLSの配合比(電荷比)が1:2:0:0、1:2:2:0、1:2:2:2、1:2:2:4になるように各複合体を作製し、粒子径と表面電荷を測定した。更に、配合比(電荷比)を1:2:2:0、1:2:2:2、1:2:2:4とした複合体をそれぞれマウスに尾静脈内投与し、6時間後に肝臓、脾臓、及び肺の遺伝子発現を調べた。また、1:8:0:0の複合体を対照として用いた。
配合比(電荷比)が1:8:0:0と1:2:2:2の複合体をマウスに投与し、in vivoイメージング装置を用いて遺伝子発現を観察した。
また、様々な配合比(電荷比)の複合体を作製し、多変量解析を用いて各成分の肺における遺伝子発現量に及ぼす寄与率を測定した。
粒子サイズと表面電荷を図8に示す。粒子サイズはLSを添加してもほぼ一定であり、表面電荷はLS添加により負電荷に近づくことが確認された。
ルシフェラーゼ活性を図9に示す。LSの添加によって肺における遺伝子発現量が著しく上昇した。
ルシフェラーゼでイメージングされたマウスを図10に示す。LSを添加した複合体では、肺でルシフェラーゼが高発現していることが確認された。
多変数のスプライン補間法及び寄与指数を図11に示す。これより肺への送達にはLSが最も寄与することが明らかになった。
方法
ホスファチジルセリン(DOPS)のみからなるリポソームを作製した。pCMV−Lucを用いて作製したpDNA/PEI複合体にDOPSリポソームをpDNAとPEI、DOPSの配合比(電荷比)が1:8:6になるように添加して、pDNA/PEI/DOPS複合体を作製した。pDNA/PEI複合体とpDNA/PEI/DOPS複合体をマウスに静脈内投与し、6時間後に各臓器の遺伝子発現量を測定した。
各臓器における遺伝子発現(ルシフェラーゼ活性)を図12に示す。pDNA/PEI/DOPS複合体はpDNA/PEI複合体に比べ、特に脾臓に高い遺伝子発現効果を示すことが明らかにされた。
方法
癌表面抗原MUC1を認識するアプタマーに加え、非特異配列のアプタマーを用いた。pCMV−Lucとポリエチレンイミンの電荷比が1:8になるようにpDNA/PEI複合体を作製した。pDNA/PEI複合体にpDNAとPEI、アプタマーの配合比(電荷比)が1:8:1.25、1:8:2.5、1:8:5、1:8:10になるようにアプタマーをそれぞれ添加することで、pDNA/PEI/アプタマー複合体を作製し、粒子径と表面電荷を測定した。また、電荷比が1:8:10の複合体のA549細胞(MUC1+)における遺伝子発現量を測定した。
pDNAとPEIの電荷比が1:8になるようにpDNA/PEI複合体を作製した。STEAP−4(six−transmembrane epithelial antigen of the prostate−4:脂肪細胞の膜タンパク質)に対する抗体を10μgのpDNAを含んだpDNA/PEI複合体に200、300、400μgそれぞれ添加することで、pDNA/PEI/抗体複合体を作製し、粒子径と表面電荷とを測定した。
アプタマーで被覆した複合体の粒子サイズ及び表面電荷を図13に示す。pDNA/PEI/アプタマー複合体の配合比(電荷比)が1:8:10であるとき粒子サイズが小さく表面電荷が負になり、最も好ましい複合体が得られた。
MUC1発現細胞におけるルシフェラーゼ活性を図14に示す。MUC1を発現したA549細胞では、MUC1に対するアプタマーで被覆した複合体が非特異的なアプタマーに比べて顕著にルシフェラーゼ活性が高かった。
抗体で被覆した複合体の粒子サイズと表面電荷を図15に示す。粒子サイズは抗体が300μgであるときに最も小さくなり、表面電荷は抗体で被覆した複合体いずれでも差異は見られなかった。したがって、pDNA/PEI/抗体300μgの複合体が最も適していると考えられた。
方法
pCMV−Lucを用いて作製したpDNA/PEI複合体にグリチルリチンをpDNAとPEI、GLYの配合比(電荷比)が1:8:12になるように添加して、pDNA/PEI/GLY複合体を作製した。pDNA/PEI複合体(市販の遺伝子ベクター)とpDNA/PEI/GLY複合体(新規遺伝子ベクター)をマウスに静脈内投与し、6時間後に各臓器の遺伝子発現量を測定した。
各臓器における遺伝子発現(ルシフェラーゼ活性)を図16に示す。pDNA/PEI/GLY複合体はpDNA/PEI複合体に比べ、特に肝臓に高い遺伝子発現効果を示すことが明らかにされた。
方法
蛍光色素を用いて蛍光標識した不活化インフルエンザウイルス(以下Vaccine)に塩化ベンザルコニウム(BK)とγ―PGAを自己組織化したVaccine/BAC/γ−PGA複合体を作成し、樹状細胞株(DC2.4)に添加した。
不活化ウイルスの取り込みおよびマウスの抗体価を図17に示す。ウイルス単体では樹状細胞への取り込みが認められなかったが、Vaccine/BAC/γ−PGA複合体にする事によって、取り込み量が大きく増加した。
方法
pCMV−Lucを用いて作製したpDNA/PEI複合体にγ−PGAをpDNAとPEI、γ−PGAの配合比(電荷比)が1:8:6になるように添加して、pDNA/PEI/γ−PGA複合体を作製した。樹状細胞株DC2.4細胞にpDNAのみ、pDNA+γ−PGA、pDNA/PEI/γ−PGA複合体、市販の遺伝子ベクターとしてLipofectinとpDNAとの複合体を添加し、22時間後のルシフェラーゼ活性を測定した。
樹状細胞株における遺伝子発現(ルシフェラーゼ活性)を図18に示す。pDNAのみ、またはpDNAとγ-PGAを添加しても遺伝子発現は得られなかったが、pDNA/PEI/γ-PGA複合体は市販の遺伝子ベクターと比較して顕著に高い遺伝子発現効果を示した。
方法
siRNAに対し、どのような複合体が最も機能するのかを試験するために、ルシフェラーゼ活性を指標とした遺伝子抑制試験を行った。ルシフェラーゼを恒常発現した大腸癌細胞株colon26細胞を用いた。siRNAとしてルシフェラーゼに対するsiRNAに、市販の遺伝子ベクターであるLipofectamine RNAimaxまたはPEI、ポリアルギニン(PLA)、DOTMA−Cholリポソーム、DOTMA−DOPEリポソーム、DC−Chol−EggPCリポソーム、DC−Chol−DOPEリポソームと混合し、カチオン性の複合体を作成した。さらに、γ−PGAを添加し、それぞれ、アニオン性の複合体を構築した。各複合体を細胞に添加し、22時間後のルシフェラーゼ活性を測定した。値は未処理の細胞のルシフェラーゼ活性を100%として算出した。
ルシフェラーゼ活性を指標とした遺伝子抑制効率を図19に示す。様々なカチオン性の高分子やリポソームを検討した結果、siRNAにはカチオン性分子として、高分子よりリポソームが適していた。さらに、膜融合脂質であるDOPEを含有したリポソームとsiRNAとの複合体をγ−PGAを用いて被膜することで、市販のsiRNAベクターと匹敵する効果を示しながら、標的化が可能なベクターを開発できた。
方法
複合体の投与量(DNA量として)を100μgに増加した以外は実施例1と同様にして、マウスにマラリアDNAワクチン(pMSP−1)のみまたはpMSP−1/PEI/γ−PGA複合体を3週間おきに計三回投与し、最終免疫から10日後にマラリア原虫を感染させた。
マラリア感染後のマウスの生存率を図12に示す。DNAワクチンのみではマラリアの感染抑制効果はほとんど得られなかったが、pMSP−1/PEI/γ−PGA複合体を投与することで、マラリアの感染を100%抑制することが可能であった。
方法
卵白アルブミン(OVA)と塩化ベンザルコニウム(BK)、γ−PGAを用いてOVA/BK/γ−PGA複合体を構築した。マウスにOVAのみ、塩化ベンザルコニウムとγ−PGA(ベクターのみ)、OVA/BK/γ−PGA複合体をそれぞれ一週間おきに計4回投与して免疫感作させ、最終免疫から2週間後に血清を採取し、ELISAでOVAに対する抗体価を測定した。抗体価の測定には二次抗体としてHRP標識ヤギ由来抗マウスIgG抗体を用いた。
マウス血清における抗体価を図21(A:血清を希釈した場合の抗体価の変動、B:希釈率 10−4 における血清の抗体価)に示す。OVAのみやBKとγ−PGAの混合物では免疫誘導効果が得られなかったが、塩化ベンザルコニウムとγ−PGAを用いて作成したOVA/BK/γ−PGA複合体は高い液性免疫誘導効果を示した。
方法
卵白アルブミン(OVA)と塩化ベンザルコニウム(BK)、γ−PGAを用いてOVA/BK/γ−PGA複合体を構築した。マウスにOVAのみ、塩化ベンザルコニウムとγ−PGA(ベクターのみ)、OVA/BK/γ−PGA複合体を一週間おきに計4回投与し、最終免疫から2週間後にOVA発現癌細胞(EG7)を皮内に移植し、移植後の腫瘍増殖を観察した。
EG7の腫瘍移植からの腫瘍増殖の観察結果を図22に示す。OVAのみやBKとγ−PGAの混合物では腫瘍増殖抑制効果が得られなかったが、OVA/BK/γ−PGA複合体を投与したマウスでは、高い免疫誘導効果によって、OVA発現癌細胞を完全に抑制することが可能であった。
方法
ポリアミドアミンデンドリマー(G4)とp−SCN benzyl diethylenetriamine pentaacetic acid(DTPA)を反応させることでG4の1分子に対し約53個のDTPAを導入したG4−DTPAを合成した。これをPEIと混合させることでPEI/G4−DTPAとした後、γ−PGAと混合し、γ−PGA/PEI/G4−DTPAを作製した。一方、111In標識体はG4−DTPA−111Inを用いて同様の方法で作製した。粒子径とゼータ電位を測定したところ、膜電位47.7±6.0mV、粒子径30.2±3.2nmのPEI/G4−DTPAと膜電位−48.9±1.6mV、粒子径24.8±3.7nmのγ−PGA/PEI/G4−DTPAの生成を確認した。比較対照として別のアニオン性高分子であるアルギン酸(ALG)を含有するALG/PEI/G4−DTPA およびその111In標識体も同様の操作で作製した。
RAW264にPEI/G4−DTPA、γ−PGA/PEI/G4−DTPA、ALG/PEI/G4−DTPAを添加し、細胞障害性を評価した。さらに、各複合体を雄性ラットのfoot padから投与し、経時的に各臓器での放射能を測定することにより各粒子の生体内分布を評価した。
PEI/G4−DTPAでは非常に強い細胞障害性を示したが、γ−PGA/PEI/G4−DTPA、ALG/PEI/G4−DTPAでは細胞障害性が認められなかった(図23)。また、マウス赤血球を用いて各複合体の赤血球凝集能、溶血性を評価した結果、PEI/G4−DTPAでは赤血球の著しい凝集とわずかな溶血が確認されたが、γ−PGA/PEI/G4−DTPA、ALG/PEI/G4−DTPAでは赤血球の凝集、溶血は認められなかった。 γ−PGA/PEI/G4−DTPA−111Inの投与24時間後におけるセンチネルリンパ節への集積は、他の複合体よりも有意に高いものであった。また、いずれの複合体でもセンチネルリンパ節、投与部位以外の臓器への取り込みは非常に低い値を示し、各複合体のリンパ管から血管への漏出は起こりにくいことが示された(図24)。またSPECT/CT撮像においても、γ−PGA/PEI/G4−DTPA−111Inは他の複合体に比べてセンチネルリンパ節へ高く集積し、明瞭なイメージングに成功した(図25)。
特に本発明の複合体は、標的となる細胞や臓器への高い指向性を有することを特徴とするので、新たなドラッグデリバリーシステムの有効成分となり得る。
さらに本発明の複合体は、イメージングプローブの輸送体として用いることも可能である。
Claims (15)
- 抗原もしくは薬物とカチオン性分子との複合体、ならびにそれを内包するアニオン性分子を含有し、細胞または臓器への指向性を有することを特徴とする、抗原もしくは薬物送達複合体。
- 抗原もしくは薬物が、核酸、ペプチド、タンパク質または不活化ウイルスである、請求項1に記載の複合体。
- アニオン性分子が、ホスファチジルセリン、ラウロイルザルコシンおよびグリチルリチンから選択され、脾臓、肺または肝臓への指向性を有することを特徴とする、請求項1または2に記載の複合体。
- アニオン性分子が、ホスファチジルセリンであり;
抗原もしくは薬物、カチオン性分子、ホスファチジルセリンの配合比が1:2:4〜1:8:8であり;かつ
脾臓への指向性を有することを特徴とする、請求項3に記載の複合体。 - アニオン性分子が、ラウロイルザルコシンであり;
カチオン性分子が、カチオン性高分子およびカチオン性脂質からなり;
抗原もしくは薬物、カチオン性高分子、カチオン性脂質、ラウロイルザルコシンの配合比が1:2:2:1〜1:2:2:4であり;かつ
肺への指向性を有することを特徴とする、請求項3に記載の複合体。 - アニオン性分子が、グリチルリチンであり;
抗原もしくは薬物、カチオン性分子、グリチルリチンの配合比が1:2:8〜1:8:16であり;かつ
肝臓への指向性を有することを特徴とする、請求項3に記載の複合体。 - アニオン性分子が、γ−ポリグルタミン酸であり、樹状細胞または脾臓への指向性を有することを特徴とする、請求項1または2に記載の複合体。
- 抗原もしくは薬物、カチオン性分子、γ−ポリグルタミン酸の配合比が1:2:8〜1:8:16であり;かつ
樹状細胞または脾臓への指向性を有することを特徴とする、請求項7に記載の複合体。 - アニオン性分子が、アプタマー、miRNAおよび抗体から選択される、請求項1または2に記載の複合体。
- 抗原もしくは薬物、カチオン性分子、アニオン性分子の配合比が1:2:8〜1:8:16である、請求項9に記載の複合体。
- 複合体が、
抗原もしくは薬物;
ポリリジン、ポリアルギニン、ポリヒスチジン、ポリエチレンイミンおよびカチオン性デンドリマーから選択されるカチオン性高分子、あるいはDOTMA、DOTAP、DC−Cholおよび塩化ベンザルコニウムから選択されるカチオン性脂質;
から構成される、請求項1〜10のいずれか一項に記載の複合体。 - 複合体が、
抗原もしくは薬物;そして
DOTMA、DOTAPおよびDC−Cholから選択されるカチオン性脂質ならびにDOPEおよびコレステロールから選択される脂質からなるリポソームあるいはミセル;
から構成される、請求項1〜10のいずれか一項に記載の複合体。 - イメージングに使用するための請求項1〜12のいずれか一項に記載の複合体を含む、イメージング用核種の輸送体。
- 抗原もしくは薬物が、イメージング用核種、カチオン性デンドリマーおよびキレート剤からなる複合体であり;
カチオン性分子が、ポリエチレンイミンであり;
アニオン性分子が、γ−ポリグルタミン酸であり;そして
リンパ節への指向性を有することを特徴とする、請求項14に記載の輸送体。 - イメージング用核種、カチオン性デンドリマーおよびキレート剤からなる薬物複合体;ポリエチレンイミン;ならびにγ−ポリグルタミン酸から構成される複合体を用いることを特徴とする、センチネルリンパ節のイメージング方法。
