次に、マウスメラノーマB16−F10細胞、或いはマウス4T1乳癌細胞を足踵に注射し肺転移を起こすモデルマウスに抗Exon−17ポリクローナル抗体を週に一度投与し続けたところ、モデル作製後3〜5週間で原発巣の増殖のみならず原発巣から肺への転移率・転移コロニー数を有意に抑制した(後述実施例14、実施例15)。また、マウス4T1乳癌細胞においては原発巣からの乳癌細胞の骨浸潤及び癌の骨浸潤による骨破壊を有意に抑制した(後述実施例15)。これにより、抗Exon−17ポリクローナル抗体が癌病態の進行とともに起こる原発巣の増殖を抑制する活性、骨浸潤を抑制する活性及び癌の骨浸潤による骨破壊を抑制する活性ならびに原発巣から肺への転移を抑制する活性を持つ抗体であることが明らかとなった。このことから、PN−1に対する中和抗体はPN−1が有する接着阻害作用を抑制し、悪性黒色腫或いは乳癌細胞の増殖および肺転移並びに乳癌細胞の骨浸潤、骨破壊を抑制する新規治療薬として有用であることが示された。以上の知見から、癌病態において、PN−1が原発巣の増殖ならびに原発巣からの転移をおこす機能を有していることが明らかとなり、PN−1に特異的に存在するExon−17によりコードされるペプチド領域に対する抗体により、これらの癌病態の進行を抑制することができることを見出した。

本発明は、抗細胞接着作用を有するペリオスチンのスプライシングバリアント(PN−1タンパク質)を認識する抗ペリオスチン抗体を含む、癌治療用医薬組成物である。
2.抗体
本発明に用いられる抗体は、PN−1タンパク質に特異的に存在する領域に対する抗体である。
3.抗体の調製
本発明に用いる抗体は、癌病態時等の癌細胞に特異的なスプライシングバリアントが形成されるC-末端ドメインのExon−17領域によりコードされるアミノ酸配列からなるペプチドを化学合成したものを抗原として作製できるが、係るペプチドは、ペリオスチンタンパク質の酵素消化や遺伝子工学的手法などによっても得られ、その由来は問わない。
4.ヒト型抗体の作製
免疫グロブリンG(以下、単に「IgG」という。)は、分子量約23000の軽ポリペプチド鎖(以下「軽鎖」という)、分子量約50000の重ポリペプチド鎖(以下「重鎖」という)の各2本ずつから構成される。重鎖、軽鎖とも約110残基からなる、アミノ酸配列が保存されている領域の繰り返し構造を持ち、これらはIgGの3次元構造の基本単位(以下、「ドメイン」という。)を構成する。重鎖および軽鎖は、それぞれ連続した4個、および2個のドメインから構成されている。重鎖、軽鎖いずれにおいても、アミノ末端のドメインは他のドメインに比べ各抗体分子間でのアミノ酸配列の変異が大きく、このドメインは可変ドメイン(variable domain:以下、「Vドメイン」という。)と呼ばれる。IgGのアミノ末端においては、重鎖、軽鎖のVドメインが相補的に会合し可変領域を形成している。これに対し、残余のドメインは、全体として定常領域を形成する。定常領域は、各動物種に特徴的な配列を有し、例えば、マウスIgGの定常領域はヒトIgGの定常領域とは異なっているので、マウスIgGはヒトの免疫系によって異物として認識され、その結果、ヒト抗マウス抗体(Human Anti Mouse Antibody:以下「HAMA」という。)応答が起こる(Schroff RW. et al Cancer Res. (1985)45,879-85)。従って、マウス抗体はヒトに繰返し投与することはできない。このような抗体をヒトに投与するためには、抗体の特異性を保持したままHAMA応答を起こさないように抗体分子を修飾する必要がある。X線結晶構造解析の結果によれば、一般に、このようなドメインは3本から5本のβ鎖からなる逆平行βシートが二層重なり合った長円筒状の構造をとる。可変領域では、重鎖、軽鎖のVドメインそれぞれにつき各3個のループが集合し、抗原結合部位を形成する。この各ループは相補性決定領域(complementarity determining region:以下、「CDR」という。)と呼ばれ、アミノ酸配列の変異が最も著しい。可変領域のCDR以外の部分は、一般に、CDRの構造を保持する役割を有し、「フレームワーク」と呼ばれる。カバトらは、重鎖、軽鎖の可変領域の一次配列を多数収集し、配列の保存性に基づき、それぞれの一次配列をCDRおよびフレームワークに分類した表を作成した(Kabatt et al. SEQUENCES OF IMMUNOLOGICAL INTEREST, 5th edition, NIH publication, No.91-3242, E.A.)。また、各フレームワークは、アミノ酸配列が共通の特徴を有する複数のサブグループに分類された。さらに、ヒトとマウスの間で対応するフレームワークが存在することも見いだされた。このようなIgGの構造的特徴に関する研究から以下のヒト化抗体の作製法が考案された。研究初期の段階では、マウス由来抗体の可変領域をヒト由来の定常領域に接合したキメラ抗体が提案された(Morrison SL. et al Proc Natl Acad Sci U S A. (1984)81,6851-5)。しかし、そのようなキメラ抗体は、依然として、多くの非ヒトアミノ酸残基を含むので、特に長期間投与した場合にはHAMA応答を誘導しうる(Begent et al., Br. J. Cancer, (1990)62, 487)。
(a)アクセプターのフレームワーク領域中のアミノ酸がその位置において稀であり、ドナーの対応するアミノ酸がアクセプターの前記位置において普通であること
(b)該アミノ酸がCDRのひとつのすぐ近くであること
(c)該アミノ酸が三次元免疫グロブリンモデルにおいてCDRの約3Å以内に側鎖原子を有し、そして抗原とまたはヒト化抗体のCDRと相互作用することができると予想されること。
5.ヒト抗体の作製
本発明で使用される「ヒト抗体」あるいは「ヒト免疫グロブリン」とは、免疫グロブリンを構成するH鎖の可変領域(VH)及びH鎖の定常領域(CH)並びにL鎖の可変領域(VL)及びL鎖の定常領域(CL)を含む全ての領域がヒトイムノグロブリンをコードする遺伝子に由来するイムノグロブリンである。換言すれば、H鎖がヒト免疫グロブリン重鎖遺伝子に由来し、軽鎖がヒト免疫グロブリン軽鎖遺伝子に由来するものである抗体を意味する。ヒト抗体は、常法に従って、例えば、少なくともヒトイムノグロブリン遺伝子をマウス等のヒト以外の哺乳動物の遺伝子座中に組込むことにより作製されたトランスジェニック動物を、抗原で免疫感作することにより、前述したモノクローナル抗体の作製法と同様にして製造することができる。例えば、ヒト抗体を産生するトランスジェニックマウスは、既報(Mendez MJ et al.Nature Genetics(1997)15, 146-56, Green LL et al. Nature Genetics(1994)7, 13-21, 表平4-504365号公報;国際出願公開WO94/25585号公報;日経サイエンス、6月号、第40〜第50頁、1995年;Nils Lonberg et al. Nature(1994) 368, 856-9, 及び特表平6-500233号公報)に記載の方法に従って作製することができる。
6.抗体断片
抗体断片としては、例えば、Fab、F(ab’)2、Fv、一本鎖抗体(scFv)、ジスルフィド安定化抗体(dsFv)、CDRを含有するペプチド等を挙げることができる。
7.診断薬
本発明はまた、上記の抗体をマーカー標識することにより作製した癌の診断薬を提供する。ここでマーカーとしては、酵素、放射性同位元素、蛍光色素等を用いることができる。ここで用いる酵素としては、ターンオーバー数(turn over number)が大きく、かつ酵素と結合しても安定であること、基質と特異的に反応して発色させることができる等の条件を満たす物であれば特に制限されるものではなく、通常の酵素免疫アッセイ(EIA)に用いられる酵素を使用することができる。好ましい酵素の例としては、ペルオキシダーゼ、β−ガラクトシダーゼ、アルカリフォスファターゼ、グルコースオキシダーゼ、アセチルコリンエステラーゼ、グルコース−6−リン酸脱水素酵素、リンゴ酸脱水素酵素等を用いることができる。また、酵素阻害物質や補酵素等を用いることもできる。
8.診断方法
本発明はまた、本発明に使用される抗体を用いることによる生体試料、例えばヒト又は動物の血液から調製した血清中におけるExon−17でコードされるペプチド領域をもつペリオスチン量を測定することによる癌の診断方法である。本方法において、いわゆるサンドイッチELISA法(Enzyme-linked immunosorbent assay:酵素免疫測定法)によってExon−17でコードされるペプチド領域をもつペリオスチンを検出することができる。診断キットを用いる場合には、まず抗一次抗体を固相化したプレートに試料を接触させて両者を結合させ、この結合体にマーカー標識した二次抗体を結合させ、この三者の結合体におけるマーカーのシグナル強度を測定することにより、Exon−17でコードされるペプチド領域をもつペリオスチンを検出または定量することができる。特にExon−17でコードされるペプチド領域を持つペリオスチンは、癌等の病態時に高発現し、原発巣の増殖および癌の転移に関するスプライシングバリアントであるので、その産生をモニターすることにより癌等における病態の診断をすることができる。
<実施例>
[製造例1]サブトラクション法によるペリオスチンの検索
1−1 心不全病態モデルラットの作製及び左心室サンプルの採取
雄性ダール食塩感受性ラット(Dahl−S)(清水実験材料)を6週齢より8%高食塩含有食で飼育し、心肥大期(11週齢)および心不全期(14週齢)に各3匹の左心室を採取した。
総RNAは上記左心室約500mgからISOGEN(ニッポンジーン社)を用いて説明書に記載の方法にしたがって調整した。次に心肥大期、心不全期それぞれ3匹分を合わせた総RNA約400μgよりFast Track 2.0 Kit(インビトロジェン社)を用いて説明書に記載の方法にしたがってmRNAを精製し、それぞれ約3μgのmRNAを回収した。
cDNAサブトラクションは、PCR−Select cDNA サブトラクションキット(クローンテック社)により、説明書に記載の方法にしたがって行った。すなわち、上記1−2で得られたmRNAそれぞれ2μgよりcDNAを合成し、制限酵素Rsalで消化した。次に、14週齢から合成されたcDNAをテスターcDNA、11週齢から合成されたcDNAをドライバーcDNAとし、テスターcDNAにkit添付の2種類のアダプターを別々に連結させた後、サブトラクトハイブリダイゼーションを行った。次に、アダプターに相補的なプライマーを用いてPCRを行い、発現量に違いのあるcDNA断片を特異的に増幅させ、増幅産物1を得た。
A.ドットブロットの作製
増幅産物1をPCR IIベクター(インビトロジェン社)にTAクローニングし、挿入断片の入ったクローンを選択した。各クローンの挿入断片を増幅したPCR反応後、それぞれ1μLを熱処理後、2枚のナイロンメンブレンフィルター(ベーリンガー社)にドットブロットし、UVクロスリンカー(ストラタジーン社)により固定した。
増幅産物1を制限酵素Rsal及びEael、Smalで消化し、アダプターを除去し、DIGハイプライムDNAラベリング/検出キットII(ベーリンガー社)を用いて説明書に記載の方法に従ってDIG−dUTPでランダムプライム標識を行い、cDNAプローブ1を作製した。増幅産物2より同様にして、cDNAプローブ2を作製した。
上記Aで作製したドットブロットメンブランの1枚をcDNAプローブ1、他の1枚をcDNAプローブ2とハイブリダイゼーションを行った。具体的には、ベーリンガー社のDIGハイプライムDNAラベリング/検出キットIIを用いて、説明書に記載の方法にしたがい、DIGイージーハイブ液中42℃で一晩ハイブリダイセーションを行った。2×SSC、0.1%SDSで室温にて5分間2回、0.1×SSC、0.1%SDSで68℃にて15分間2回洗浄した後、キットに添付のブロッキングバッファー中でアルカリホスファターゼ標識抗DIG抗体と反応後、CSPD ready−to useを加えて化学発光を進行させ、X線フィルムを露光させた。cDNAプローブ1でのシグナルがcDNAプローブ2でのシグナルより強いクローンをポジティブクローンとして選択し、塩基配列の決定を行った。
塩基配列は、THERMO SequenaseTM II dye terminator cycle sequencing kit(アマシャムファルマシア社)を用いて自動DNA配列読み取り装置モデル373A(PE Applied Biosystems社)で解析することにより決定した。得られた遺伝子配列をGenBanKのデータバンクに照会した結果、クローンの1つ(SF014)がマウスペリオスチン(GenBank アクセッションNo.D13664)と86%ホモロジーのある遺伝子であることが判明した。
ラットペリオスチンcDNAの単離はλgtIIベクターに挿入されたrat aorta cDNAライブラリー(クローンテック社)より作製した約4000クローンのファージサブプール10個(計約4万クローン)をSF014の塩基配列を基に設計したプライマー(1)5’−GTTCATTGAAGGTGGCGATGGTC−3’(配列番号15)、(2)5’−GAGATAAAATCCCTGCATGGTCCT−3’(配列番号16)を用いてPCRスクリーニングを行い、3個のポジティブなサブプールを得た。そのうち1個のサブプールを上記PCRによって増幅された断片をAlkPhos DirectTM(アマシャムファルマシア社)を用いてアルカリフォスファターゼ標識したプローブを用いてハイブリダイゼーションによるスクリーニングを行い、1個のポジティブクローンラットペリオスチン#1を得た。その挿入断片をpBluescript II(ストラタジーン社)のEcoRI部位に組み込み、製造例1−5の方法に従って全塩基配列を決定した。
製造例2で得られたラットペリオスチン遺伝子のコード領域から翻訳されるタンパク質のカルボキシル末端にMycエピトープと6個のヒスチジンタグを有し、CMVプロモーターを有する発現ベクターを作製した。
製造例3で得られたプラスミドpcDNA4/Myc−His/ラットペリオスチンを、制限酵素SacI及びPmelで消化し、ペプチド断片rat PN−1/Myc−Hisを切り出した。これを、pFastBacHTc(インビトロジェン社)を制限酵素SacIとKpnI(blunting)で消化して得られたベクター断片に、ライゲーションキット(宝酒造(株))を用いて連結し、得られた発現ベクターをpFastBac/ラットペリオスチン−1/Myc−Hisと命名した。挿入部分の塩基配列は製造例1−5に記載の方法により確認した。
Escherichia coliのDH10BAC細胞を、製造例5で得られたpFastBac/ラットペリオスチン−1/Myc−Hisで形質転換し、組換えバキュロウイルスを調製した。得られたバキュロウイルスは、電気泳動およびPCRにより、目的のものが挿入されていることを確認した。
製造例5で得られた培養上清2000mLを平衡化バッファー(50mM酢酸ナトリウムバッファー pH6.0 0.1M塩化ナトリウム)で平衡化したSPsepharose Fast Flow 10mL bedに供し、得られたFlow Through(通過分画)をSPセファロース通過分画とした。
<実施例1>ラットExon−17ペプチド鎖の合成およびポリクローナル抗体の作製
ノーマルのSDラットの心臓にPN−1遺伝子を高発現させると心拡大が惹起され、またダール心不全モデルラットの心臓にラットペリオスチンのアンチセンスオリゴヌクレオチドを投与すると生存率が改善されたことに加え、報告されてきたPN−2とは異なり、ラットPN−1タンパク質には細胞接着作用が無いことが明らかとなったことを受け、ラットPN−1タンパク質に特異的な構造は夫々の配列比較からExon−17配列であると同定した(図1参照)。このExon−17によりコードされるアミノ酸配列のN末端にCys残基を付加したペプチドを、純度80%以上にて10mg、化学合成した。キャリアタンパク質としてKLH6mgを結合させたものをウサギ(Kbl:JW)に免疫させた。免疫方法は初回免疫にはFCA(フロイント完全アジュバント)を使用し、2次免疫以降はFIA (フロイント不完全アジュバント)を使用した。また、投与部位は背部皮下20ヶ所、投与週 は0、2、4、6週、投与量は初回免疫では800μgペプチド/匹、2次免疫以降では400μgペプチド/匹にて免疫した。抗体力価はELISA法にて測定し 投与週7週目に全血清を集めた。その後、合成ペプチドを用いたアフィニティーカラムを作製し、Exon−17ペプチドに特異的に反応する抗体のみを回収した。以降、上記の、ラットペリオスチンのExon−17によりコードされるペプチドに対するポリクローナル抗体を、抗ラットExon−17ポリクローナル抗体と称する。
<実施例2>in vitroにおけるラットPN−1タンパク質の非細胞接着活性の有無の検討
細胞培養用96穴マルチウェルプレートに10mg/mlフィブロネクチン、100μg/ml BSA、 10mg/mlPN−1タンパク質をそれぞれ添加し4℃にて一晩コーティングした。タンパク質液を除去したウェルに、DMEM(10% BSA,PC/SM)に懸濁したマウスメラノーマB16−F10細胞(ATCC番号:CRL−6475)若しくはマウス4T1乳癌細胞(ATCC番号:CRL-2539)104個/ウェルを加え、37℃インキュベーターにて3時間培養した。細胞の接着量測定は、培養上清を除去した後、2.5%グルタルアルデヒドにて30分間固定し、0.02%クリスタルバイオレッドにて染色した後プレートリーダー(BIO−RAD、 Model 680 マイクロプレートリーダー)にてOD 550 nmの吸光度を測定した。バックグラウンドとして、無処理のウェルを染色したものを用い、その吸光度値にて補正した値を比較した。データの解析はFisherのPLSD検定により行った(図2A、B)。その結果、陽性コントロールであるフィブロネクチンでは細胞が接着し、陰性コントロールであるBSAでは細胞が接着しなかった。ラットPN−1タンパク質を添加した群では細胞の接着が認められなかったことから、ラットPN−1タンパク質には細胞を接着させない作用、すなわち非細胞接着作用を有していることが明らかとなった。
<実施例3>in vitroにおけるラットPN−1タンパク質の抗細胞接着活性の有無の検討
細胞培養用96穴マルチウェルプレートに、DMEM(10% BSA,PC/SM)に懸濁したマウスメラノーマB16−F10細胞若しくはマウス4T1乳癌細胞104個/ウェルを添加し、37℃インキュベーターにて一晩培養した。培養上清を除去した後、10μg/mlフィブロネクチン(SIGMA)、100μg/ml BSA (SIGMA)、PN−1タンパク質を添加し37℃インキュベーターにて1〜3時間培養した。細胞の接着量測定には、培養上清を除去した後、2.5% グルタルアルデヒドにて30分間固定し、0.02%クリスタルバイオレッドで染色した後、プレートリーダー(BIO−RAD、 Model 680 マイクロプレートリーダー)によりOD 550nmの吸光度を測定した。バックグラウンドとして、無処理のウェルを染色したものを用い、その吸光度値にて補正した値を比較した。データの解析はFisherのPLSD検定により行った(図3A、B)。細胞は、実体顕微鏡LEICA MZ16付属のNikon COOLPIX4500にて撮影した。その結果、コントロールであるフィブロネクチンおよびBSAを投与した群ならびに無添加群では接着していた細胞の剥離は起こらず、抗細胞接着作用が見られなかったが、ラットPN−1タンパク質を添加した群では細胞の剥離が認められたことから、ラットPN−1タンパク質は接着細胞の剥離作用、すなわち抗細胞接着作用を有していることが明らかとなった。
<実施例4>in vitroにおける抗ラットExon−17ポリクローナル抗体の中和活性の検討
実施例3と同様に、細胞培養用96穴マルチウェルプレートに、DMEM(10% BSA,PC/SM)に懸濁したマウスメラノーマB16−F10細胞若しくはマウス4T1乳癌細胞104個/ウェルを添加し、37℃インキュベーターにて一晩培養した。培養上清を除去後、培養液を10μg/mlのシクロヘキシミドを加えた10%FBS添加DMEM培地に変更し、37℃、1時間培養した。その後、予め37℃に温めたDMEM培地(無血清)で細胞を2度洗い、終濃度10μg/mlのラットペリオスチンタンパク質と抗ラットExon−17ポリクローナル抗体を終濃度100 μg/mlになるようにDMEM培地(無血清)に添加した。陽性コントロールとしてラットペリオスチンタンパク質のみを、陰性コントロールとしてBSAを用いた。37℃で1時間培養後の顕微鏡観察においてラットペリオスチンタンパク質のみを添加した群はほぼ完全に細胞が剥がれたため、PBS(−)にて2度洗った後、10%中性緩衝ホルマリン液にて30分間細胞を固定した。その後、PBS(−)にて3度洗った後、クリスタルバイオレットを用いて30分間細胞を染色した。その後、550nmのプレートリーダー(BIO−RAD、 Model 680 マイクロプレートリーダー)を用いて細胞の染色度を測定した(図4A、B)。その結果、抗ラットExon−17ポリクローナル抗体は、PN−1タンパク質による接着細胞の剥離を抑制する、つまりPN−1タンパク質の抗細胞接着作用を抑制する、すなわちラットPN−1タンパク質の抗細胞接着作用を中和する活性を有した抗体であることが判明した。
<実施例5>in vitroにおける抗ラットExon−17ポリクローナル抗体の細胞増殖に与える影響の検討
実施例3と同様に、細胞培養用96穴マルチウェルプレートに、DMEM(無血清,PC/SM)に懸濁したマウスメラノーマB16−F10細胞104個/ウェルを添加し、37℃インキュベーターにて一晩培養した。培養上清を除去した後、抗ラットExon−17ポリクローナル抗体およびウサギIgG抗体が各々100mg/ml、10μg/ml、1μg/mlの終濃度になるように添加したDMEM(無血清,PC/SM)培地を加えて、再度一晩培養した。翌日、全てのウェルの培地をDMEM(10% BSA,PC/SM)に交換し、Cell Titer 96 AQueous One Solution Cell Proliferation Assay kit(プロメガ)を用いて、Cell Titer液を100μlの培地に対し20μlずつ加え、37℃で1時間培養した。その後、490nmのプレートリーダー(BIO−RAD、 Model 680 マイクロプレートリーダー)を用いて細胞の染色度を測定した(図5A)。同様に、マウス4T1乳癌細胞においては抗ラットExon−17ポリクローナル抗体およびウサギIgG抗体が各々200μg/ml、100μg/ml、50μg/mlの終濃度になるように添加し測定した(図5B)。その結果、抗ラットExon−17ポリクローナル抗体は、高濃度にて細胞増殖を抑制する活性を有した抗体であることが判明した。
<実施例6>ヒトペリオスチンのExon−17ペプチド鎖に対するモノクローナル抗体の作製
(1) 抗原の作製
ヒトペリオスチンExon−17によりコードされるアミノ酸配列(配列番号4)のN末端にCys残基を付加したペプチド(抗原ペプチド;配列番号25)を、Fmoc法にて化学合成し、純度90%以上の抗原ペプチド10mgを得た。この抗原ペプチド5mgにキャリアタンパク質としてKLH(CALBIOCHEM社製)5mgを結合させ、抗原溶液を得た。すなわち、KLHをPBS(0.01M)に溶解して3.3mg/mLに調整し、0.2524mg/mLのMBS溶液(GEヘルスケアバイオサイエンス社製)を滴下して室温で60分間攪拌し反応させた。ジクロロメタンを用いてフリーのMBSを除き、KLH−MBを得た。このKLH−MB5mgと、0.01Mリン酸ナトリウム緩衝液(pH7.2)に溶解した抗原ペプチド5mgとを混合し、4℃で12時間攪拌して反応させ、抗原溶液を得た。
(2) 免疫
6週齢のBALB/c雌性マウス3匹の両足に、(1)で得られたKLH結合抗原ペプチド 100μg を含む抗原溶液 50μlと、FCA(フロイント完全アジュバント) 50μlとの混合乳濁液全量を皮下注射した。その後2週間間隔で2回、用時調製した上記抗原溶液とFIA (フロイント不完全アジュバント) との混合乳濁液を両足に投与した。その後、そのマウスを頸椎脱臼により致死させ、無菌的に足部リンパ節を採取した。RPMI培地(コージンバイオ株式会社製)を供給しながら上記リンパ節を破砕し、孔径約10μmのメッシュを通過させて RPMI培地に懸濁状態のリンパ節細胞を得た。これを1000rpm、10分間の遠心分離にかけて、リンパ節細胞を沈殿画分として得た。この沈殿画分に、0.84 %の塩化アンモニウム溶液に20mMのHEPES緩衝液(pH 7.4) を加えた溶液1mlを入れて溶血させ、赤血球を除いた後、1,000rpm、5分間の遠心分離にかけた。得られた沈殿画分(細胞画分)をRPMI培地で数回洗浄し、細胞融合に用いた。
(3) ミエローマ細胞の調製
8-アザグアニン耐性でかつイムノグロブリン非分泌型のマウスミエローマ細胞株P3X63Ag8U.1(P3U1 株)を、20%のウシ胎児血清(FCS) を含む RPMI培地で、10%CO2 ・37℃インキュベーター内で培養し、対数増殖期にある細胞を集め、1,000rpm、5分間の遠心分離にかけて沈殿画分として細胞のみを取得し、RPMI培地に懸濁させた。
(4) 細胞の融合
(2)で得た免疫化リンパ節細胞108 〜3×108 個を含む RPMI培地と、(3)で得たミエローマ細胞108 個を含むRPMI培地とを混合した後、1,000rpm、10分間の遠心分離にかけた。上清を静かに除いて沈殿画分として細胞を取得し、これに25%(w/v)のポリエチレングリコール1500(PEG 1500 、ベーリンガー社製)1mlを加えた後、更にRPMI培地をゆっくりと加えて総量を10mlとした。これに20% FCSを含む RPMI培地10mlを加えて、少し静置させた後、1,000rpm、5分間の遠心分離にかけ、得られた沈殿画分(細胞画分)に20%FCSを含む RPMIを加えて細胞濃度が106 個/mlになるように調整した細胞懸濁液を、コーニング社製の96穴培養プレートに200μL/ウェルずつ分注した。5%CO2 ・37℃インキュベーター中で24時間培養後、HAT 溶液(インビトロジェン社製)を添加した後、更に2週間培養した。
(5)ELISA法によるスクリーニング
培養上清が抗原ペプチドと反応する陽性ウェルのスクリーニングを行った。
(6) 抗体産生細胞のクローニング
(5)のELISA法で抗原ペプチドとの反応性が確認された陽性ウェル内の細胞から、限界希釈法により、抗体産生細胞株のクローニングを行った。すなわち、陽性ウェル内の細胞を96穴培養プレートの各ウェルに撒き込み、5%CO2 ・37℃インキュベーター内で2週間培養した。各ウェルの培養上清について、(5)の方法と同様に、ELISA法にて、抗原ペプチドとの反応性を確認し、陽性ウェルについて、再度、限界希釈法によるクローニングを行い、抗原ペプチドとの反応性が高く、細胞コロニーの発育が良好な30個の細胞を得た。これら細胞を24穴培養プレートに移し、5%CO2 ・37℃インキュベーター内で2週間培養した。培養上清について、再び、(5)の方法と同様に、ELISA法にて、抗原ペプチドとの反応性(抗体価)を確認した。OD490nmの吸光度が高かった10ウェル内の細胞、すなわち10個のハイブリドーマ細胞株を、抗体産生細胞として有用であると判断し、選別した。
(7)ヒトペリオスチンタンパク質(PN−1)との結合性の確認
(6)で得られた抗体産生細胞10個が産生する抗体と、ヒトペリオスチンタンパク質(PN−1)との結合性について、ドットブロット法にて確認した。すなわち、実施例8で得られた合成タンパク質(30μg/ml)を5μlずつHybond-ECLニトロセルロースメンブレン(GEヘルスケアバイオサイエンス株式会社製)にスポットし、TBS溶液(10mM Tris−HCl(pH8.0)、150mM NaCl)で1回洗浄した。ブロッキングバッファー(ブロックエース、雪印乳業株式会社製)を加えて、室温で1時間振盪した。メンブレンに(6)で得られたモノクローナル抗体(一次抗体)の1μg/ml濃度溶液を加えて3時間振盪した後、TBS溶液で10分間振盪洗浄を4回行った。メンブレンにHRP標識抗マウスIgG抗体(プロメガ社製)(二次抗体)の0.4mg/ml濃度溶液を加えて室温にて1時間振盪した後、メンブレンをTBS溶液で10分間振盪洗浄を4回行った。検出用試薬(ECL plus western blotting detection system、GEヘルスケアバイオサイエンス株式会社製)を加えて1分間反応させ、化学発光法により検出した。その結果、(6)でクローニングした抗体産生細胞10個全てが、ヒトペリオスチンPN−1と結合することが確認された。
(8)モノクローナル抗体の大量調製と精製
BALB/cマウスの腹腔内にプリスタン[2,6,10,14-テトラメチルペンタデカン(和光純薬製)]0.5mlを投与し、2〜3週間飼育した。予め、対数増殖期に維持しておいたモノクローナル抗体産生ハイブリドーマ No.1およびNo.3を回収し、培養上清を除いた沈殿画分の細胞にFCS不含のRPMI培地を加え、細胞数が1×107個/mlになるように細胞液を調製した。この細胞液を、プリスタン前投与したBALB/cマウスの腹腔中に注入し、3週間後頃から漏出した腹水を腹部より注射器で回収した。採取した腹水を、孔径0.22μmφのフィルターを用いて濾過した後、濾液をプロテインG-セファロースカラム(Millipore、11511324)によるアフィニティークロマトグラフィーによって常法に従い精製し、抗ヒトExon−17モノクローナル抗体2種を調製した。
<実施例7>抗ヒトExon−17モノクローナル抗体のヒトペリオスチンExon−17ペプチド鎖における認識部位解析
得られたモノクローナル抗体2種(No.1およびNo.3)についてヒトペリオスチンExon−17ペプチド鎖における認識部位の解析(エピトープの同定)を行った。すなわち、セルロースメンブレン上に、ヒトペリオスチンExon−17ペプチド鎖(配列番号4;1位のスレオニンから27位のグルタミン酸まで)のN末端から−9番目のフェニルアラニンから、C末端から9番目のイソロイシンまでの計45個のアミノ酸からなるアミノ酸配列において、下記のアミノ酸10個からなるペプチド36種を膜結合型ペプチドアレイを作製した(シグマアルドリッチジャパン株式会社カスタムSpots サービス)。
<実施例8>抗ラットExon−17ポリクローナル抗体のヒトペリオスチンExon−17ペプチド鎖における認識部位解析
実施例1で作製したポリクローナル抗体について、実施例7と同様に、ヒトペリオスチンExon−17ペプチド鎖における認識部位の解析(エピトープの同定)を行った。その結果、実施例7のモノクローナル抗体と同様に、該ポリクローナル抗体は合成ペプチドNo.9とのみ反応し、モノクローナル抗体と同様の部位を特異的に認識していることが明らかとなった。従って、ラットペリオスチンExon−17ペプチドを抗原としてポリクローナル抗体を作製しても、ヒトペリオスチンExon−17ペプチドを抗原としてモノクローナル抗体を作製しても、同様な特異性をもつ抗体が得られることが示唆された。
<実施例9>ラットペリオスチンタンパク質(PN−1)との結合性の確認
得られたモノクローナル抗体2種(No.1およびNo.3)について、ラットペリオスチンタンパク質(PN−1)との結合性について、ドットブロット法にて確認した。すなわち、製造例6で得られた精製タンパク質(30μg/ml)を5μlずつHybond-ECLニトロセルロースメンブレン(GEヘルスケアバイオサイエンス株式会社製)にスポットし、TBS溶液(10mM Tris−HCl(pH8.0)、150mM NaCl)で1回洗浄した。ブロッキングバッファー(ブロックエース、雪印乳業株式会社製)を加えて、室温で1時間振盪した。メンブレンにモノクローナル抗体(一次抗体)の1μg/ml濃度溶液を加えて3時間振盪した後、TBS溶液で10分間振盪洗浄を4回行った。メンブレンにHRP標識抗マウスIgG抗体(プロメガ社製)(二次抗体)の0.4μg/ml濃度溶液を加えて室温にて1時間振盪した後、メンブレンをTBS溶液で10分間振盪洗浄を4回行った。検出用試薬(ECL plus western blotting detection system、GEヘルスケアバイオサイエンス株式会社製)を加えて1分間反応させ、化学発光法により検出した。その結果、得られたモノクローナル抗体2種はラットペリオスチンPN−1とも結合することが確認された。
<実施例10>抗ヒトExon−17モノクローナル抗体のエピトープ解析
実施例7の結果から、抗ヒトExon−17モノクローナル抗体のエピトープ部分はヒトペリオスチンExon−17ペプチド鎖(配列番号4)のN末端のスレオニンから9番目のバリンまでのアミノ酸配列(TTKIITKVV;配列番号22)を認識していることが明らかとなり、また実施例9の結果において、抗ヒトExon−17モノクローナル抗体がラットペリオスチンタンパク質(PN−1)とも結合することが確認された。このことから、抗ヒトExon−17モノクローナル抗体のエピトープ部分はヒトペリオスチンExon−17ペプチド鎖(配列番号4)のN末端のスレオニンから9番目のバリンまでのアミノ酸配列(TTKIITKVV;配列番号22)のうち、ヒトとラットの種間においてアミノ酸に相違がない部分、すなわち、ヒトペリオスチンExon−17ペプチド鎖(配列番号4)またはラットペリオスチンExon−17ペプチド鎖(配列番号3)のN末端のスレオニンから7番目のリジンまでのアミノ酸配列の全部またはその一部分を認識していることが示唆された。そこで、さらに詳細にエピトープ部分を解析するために、アラニンスキャンの手法にて解析を行った。
<実施例11>抗ラットExon−17ポリクローナル抗体のヒトペリオスチンExon−17ペプチド鎖における認識部位解析
実施例1で作製したポリクローナル抗体について、実施例10と同様に、ヒトペリオスチンExon−17ペプチド鎖における認識部位の解析(エピトープの同定)を行った。その結果、実施例10のモノクローナル抗体と同様に、該ポリクローナル抗体は合成ペプチドNo.#7と強く反応し、#1および#2とは弱く反応し、#3および#8とは更に弱く反応し、モノクローナル抗体と同様の部位を特異的に認識していることが明らかとなった。従って、ラットペリオスチンExon−17ペプチドを抗原としてポリクローナル抗体を作製しても、ヒトペリオスチンExon−17ペプチドを抗原としてモノクローナル抗体を作製しても、同様な特異性をもつ抗体が得られることが示唆された。
<実施例12>マウスメラノーマB16−F10細胞肺転移モデルマウスを用いた原発巣でのExon−17によりコードされるペプチド領域を持つペリオスチンの発現
マウスメラノーマB16−F10細胞注射後2週目にマウス原発巣を採取し、ホモジナイズした組織をRIPAバッファー (RIPA Lyses Buffer 10×; Upstate)、180mM Na3VO4、プロテアーゼ阻害剤カクテル (ナカライテスク)、200mM NaF を添加し、氷上にて15分静置した。その後、15000rpm、4℃で20分間遠心し、その上清を採取した。抽出したタンパク質はDCプロテインアッセイ試薬(BIO−RAD) により濃度を測定した。その後、2×サンプルバッファー (Laemmli Sample Buffer (BIO−RAD)、5% 2−メルカプトエタノール) に混合し、98℃、5分加熱処理を加えた。調製したタンパク質のサンプルをマルチゲルIIミニ7.5 (第一化学) を用いて 10mAで180分間泳動した後、Immobilon−P Transfer Membranes (MILLIPORE) に30V、4℃で一晩転写させた。その後、メンブレンは5% スキムミルクin PBS−Tに1時間、または ブロッキングワンP (ナカライテスク) に20分間浸し、室温で振盪させブロッキングした。その後一次抗体(抗ヒトExon−17モノクローナル抗体(No.3)、1:500に希釈、コントロールとして抗ニワトリα−tublinモノクローナルIgG抗体(Sigma)、1:5000に希釈)を添加し4℃にて一晩反応させた後、PBS−Tで5分×3回の洗浄を行った。その後、二次抗体(Anti mouse IgG HRP (PROMEGA)、1:10000に希釈)を添加し1時間室温で反応させた。PBS−Tにて10分×1回、30分×2回洗浄した後、ECL−Plus (Amersiam Biosciences)を用いて化学発光法によりバンドの検出を行なった。その結果、正常下肢ではExon−17によりコードされるペプチド領域を持つペリオスチンの発現は見られなかったのに対し、腫瘍組織では発現が亢進していることが明らかとなった(図6A)。
<実施例13>マウス4T1乳癌細胞肺転移モデルマウスを用いた原発巣でのExon−17によりコードされるペプチド領域を持つペリオスチンの発現
マウス4T1乳癌細胞注射後2週目にマウス原発巣を採取し、ホモジナイズした組織をRIPAバッファー (RIPA Lyses Buffer 10×; Upstate)、180mM Na3VO4、プロテアーゼ阻害剤カクテル (ナカライテスク)、200mM NaF を添加し、氷上にて15分静置した。その後、15000rpm、4℃で20分間遠心し、その上清を採取した。抽出したタンパク質はDCプロテインアッセイ試薬(BIO−RAD) により濃度を測定した。その後、2×サンプルバッファー (Laemmli Sample Buffer (BIO−RAD)、5% 2−メルカプトエタノール) に混合し、98℃、5分加熱処理を加えた。調製したタンパク質のサンプルをマルチゲルIIミニ7.5 (第一化学) を用いて 10mAで180分間泳動した後、Immobilon-P Transfer Membranes (MILLIPORE) に30V、4℃で一晩転写させた。その後、メンブレンは5% スキムミルク(PBS−T中)に1時間、または ブロッキングワンP (ナカライテスク) に20分間浸し、室温で振盪させブロッキングした。その後一次抗体(抗ヒトExon−17モノクローナル抗体(No.3)、1:500に希釈、コントロールとして抗ニワトリα−tublinモノクローナルIgG抗体(Sigma)、1:5000に希釈)を添加し4℃にて一晩反応させた後、PBS−Tで5分×3回の洗浄を行った。その後、二次抗体(Anti mouse IgG HRP (PROMEGA)、1:10000に希釈)を添加し1時間室温で反応させた。PBS−Tにて10分×1回、30分×2回洗浄した後、ECL−Plus (Amersiam Biosciences)を用いて化学発光法によりバンドの検出を行なった。その結果、正常下肢でのペリオスチンの発現は見られなかったのに対し、腫瘍組織では発現が亢進していることが明らかとなった(図6B)。
<実施例14>マウスメラノーマB16−F10細胞肺転移モデルマウスを用いた抗ラットExon−17ポリクローナル抗体および抗ヒトExon−17モノクローナル抗体の効果
マウスメラノーマB16−F10細胞を37℃インキュベーターにて培養し、PBSで洗浄した後、トリプシン/EDTAにて細胞を浮遊させ回収した。その後1500rpm、3分間遠心を行い回収した細胞を5×105個/匹になるようにカウントし100μlのPBSに懸濁した。調整した細胞は、29Gマイジェクター注射針付インスリン用シリンジ (TERMO) を用いマウスC57BL/6Nオス8週齢の足底部に注入した。先ず初めに実施例6で得られた2種のモノクローナル抗体(No.1およびNo.3)の効果を検討するために細胞の接種と同時にマウス頸静脈より29Gマイジェクター注射針付インスリン用シリンジにて抗体2μg/匹を投与した。コントロールとしてNormal Rabbit IgG(R&D Systems)を用いた。細胞および抗体の投与後1週間での下肢腫大部の径をノギスにて計測し、原発巣の増加率を評価した。その結果、コントロール群で74.5±6.3%(n=11)、抗体(No.1)群で17.8±4.4%(n=10)、抗体(No.3)群で19.5±9.9% (n=10)であり、両抗体共に癌増殖抑制活性を同程度に持つことが明らかとなった(図7)。
<実施例15>マウス4T1乳癌細胞肺転移モデルマウスを用いた抗ラットExon−17ポリクローナル抗体の効果
マウス4T1乳癌細胞を10%牛血清アルブミン (FBS)(Bio west)、Penicillin-streptomycin Mixed solution (ナカライテスク)、含有RPMI1640(Gibco) を用いて10cm Tissue Culture Dishes (Greiner)に播種し、37℃インキュベーターにて24時間培養した。その後、培養上清を除去し、PBSにて洗浄した後にトリプシン/EDTAにより浮遊させた。細胞を回収し1500rpm、3分間遠心した後、1.5×105個の細胞を継代し37℃インキュベーターにて72時間培養後、対数増殖期にある細胞を1×106個/匹になるようにカウントし100μlのPBSに懸濁した。調整した細胞は、29Gマイジェクター注射針付インスリン用シリンジ(TERMO)を用いマウスBALB/cメス8週齢の足底部に注入した。また、マウス頸静脈より29Gマイジェクター注射針付インスリン用シリンジにて抗体20μg/匹を投与した。抗ラットExon−17ポリクローナル抗体 (1mg/ml)を用いた実験では、細胞注射と同時に抗体を投与し、さらに細胞注射を行なった1週間後と2週間後に抗体を投与した。コントロールとしてNormal Rabbit IgG (R&D Systems)を用いた。細胞注射を行なった後、1週間毎に体重測定と下肢腫大部の径をノギスにて計測し、原発巣の体積を評価した。評価方法はDethlefsen LA. et al. J. Natl. Cancer Inst., 40, 389(1968)に記載方法に従い、(足裏の長さ)×(足裏の幅の2乗)÷2にて求めた。細胞注射後3週目に剖検を行い、体重測定、原発巣から肺への転移の有無と肺への転移コロニー数のカウントを行なった。それぞれ、Studentのt検定によりデータ解析を行なった。結果は、体重変化において細胞注射後2週目までは両群間に有意な差は見られなかったが3週間後において、コントロール群で17.50±0.703g(n=6)、中和抗体群で21.24±0.517g(n=6)で有意に原発巣の増殖を抑制していた (P<0.05)(図10)。また、細胞注射後1週目の原発巣の腫瘍体積は、コントロール群で301.3±11.49mm3(n=10)、中和抗体群で235.9±7.842mm3(n=10)で有意に原発巣の増殖を抑制していた(P<0.05)、また、細胞注射後2週目の原発巣の腫瘍体積は、コントロール群で842.4±34.71mm3(n=10)、中和抗体群で613.9±45.17mm3(n=10)で有意に原発巣の増殖を抑制していた (P<0.05) (図11)、細胞注射後3週目ではコントロール群の足が壊死による脱落が起こったが、中和抗体群では脱落は見られなかった。また、原発巣からの乳癌細胞の骨浸潤による骨破壊の評価は下肢距骨に対してHE染色、TRAP染色を行った後、骨面積および破骨細胞数を測定した。骨への直接浸潤による残存骨面積はコントロール群では326656±53628.7(n=5)であったのに対し、中和抗体群では545756.8±65928.8(n=5)で有意に骨の残存が見られた(p<0.05)(図12)。さらに、破骨細胞数も残存骨面積に逆相関し、コントロール群では49±9.576個(n=5)であったのに対し、中和抗体群では20±5.167個(n=5)と有意に減少していた(p<0.05)(図13)。一方、原発巣から肺への転移率は、何れの群でも転移が見られたが、転移コロニー数はコントロール群では89±28.9個であったのに対し、中和抗体群では30.5±6.30個で有意に転移コロニー数を抑制していた(P<0.05)(図14)。以上の結果より、抗ラットExon−17ポリクローナル抗体は乳癌細胞の原発巣の増殖を抑制すると共に、肺への転移抑制効果を有することが明らかとなった。
<実施例16>ヒト癌細胞および正常組織におけるExon−17領域を持ったペリオスチンの発現の検討
先ず初めにExon−17領域に特異的なプライマー(sense鎖:5’−TAAAATTATAACCAAAGTTGTGGAACC−3’(配列番号35)、antisense鎖:5’− AGTGTGGGTCCTTCAGTTTTGAT−3’(配列番号36))を作製した。
次に、下記1)〜6)のヒト癌細胞株をATCC(American Type Culture Collection)およびECACC(European Collection of Cell Cultures)より購入し、当該細胞株の説明書に記載の方法に従って培養後、ISOGEN(ニッポンジーン社)を用い説明書に記載の方法に従ってtotal RNAを抽出した。

これらのtotal RNAに対しSuperscript IIファーストストランドシステム(Invitrogen(社))を用いて 42℃ 50min、70℃ 10minにてcDNAを作製した。その後、配列番号35,36のプライマーとKOD plus Taq DNA polymerase(東洋紡(株))を用いたPCR法 (熱変性94℃ 30sec、アニーリング56℃ 30秒、伸長反応68℃ 30秒×40サイクル) を行った後、3%アガロースゲルを用い電気泳動を行ない、Exon−17領域を持ったペリオスチンの発現の検討を行った。その結果、検討した全ての癌種(食道癌、肺癌、胸腺癌、膵臓癌、甲状腺癌、胃癌、小腸癌、結腸(大腸)癌、直腸癌、肝癌、膀胱癌、腎臓癌、乳癌、乳管癌、乳腺癌、子宮癌、子宮頚癌、卵巣癌、精巣癌、リンパ腫、副腎癌、前立腺癌、骨肉腫、悪性黒色腫(メラノーマ)、滑膜腫、白血病)で発現していることを確認した(図15、図16、図17、図18)。一方、それに対応した正常組織において発現を検討した結果、乳腺で僅かな発現を認めたが癌組織と比べると発現が非常に弱かった。また、他の正常組織では殆ど認められなかった(図19)。以上の結果から、Exon−17領域を持ったペリオスチンは癌組織に特異的に発現していることが示され、その発現量を測定することによって癌の診断が可能であることが示された。ペリオスチンのExon−17部位によりコードされるペプチドに対する抗体は、メラノーマ或いは乳癌のみならず上記癌をもターゲットとして良いことが示された。
Claims (12)
- 配列番号3、配列番号4および配列番号21からなる群から選択されるいずれか一つのアミノ酸配列からなるペプチドに対する抗体を含有する、癌治療用医薬組成物。
- 前記抗体が配列番号26のアミノ酸配列からなる部位と特異的に結合する抗体である、請求項1記載の癌治療用医薬組成物。
- 前記抗体が配列番号34のアミノ酸配列からなる部位を特異的に認識する抗体である、請求項1または2記載の癌治療用医薬組成物。
- 癌治療が癌の転移の抑制である、請求項1〜3のいずれか一項記載の癌治療用医薬組成物。
- 癌治療が癌の原発巣の増殖の抑制である、請求項1〜3のいずれか一項記載の癌治療用医薬組成物。
- 癌治療が癌の骨浸潤および/または癌の骨浸潤による骨破壊の抑制である、請求項1〜3のいずれか一項記載の癌治療用医薬組成物。
- 前記癌が食道癌、肺癌、胸腺癌、膵臓癌、甲状腺癌、胃癌、小腸癌、結腸(大腸)癌、直腸癌、肝癌、膀胱癌、腎臓癌、乳癌、乳管癌、乳腺癌、子宮癌、子宮頚癌、卵巣癌、精巣癌、リンパ腫、副腎癌、前立腺癌、骨肉腫、悪性黒色腫、滑膜腫、または白血病である、前請求項1〜6のいずれか一項記載の癌治療用医薬組成物。
- 前記癌が悪性黒色腫または乳癌である、請求項1〜6のいずれか一項記載の癌治療用医薬組成物。
- 配列番号3、配列番号4および配列番号21からなる群から選択されるいずれか一つのアミノ酸配列からなるペプチドに対する抗体を用いて、生体試料中のExon−17によりコードされるペプチド領域をもつペリオスチン量を測定する工程を含む、癌の診断方法。
- 配列番号3,配列番号4および配列番号21からなる群から選択されるいずれか一つのアミノ酸配列からなるペプチドに対する抗体を含む、癌の診断薬。
- 配列番号35のヌクレオチドおよび配列番号36のヌクレオチドからなるPCRプライマーにより生体試料中のExon−17によりコードされるペプチド領域をもつペリオスチン量を測定する工程を含む、癌の診断方法。
- 配列番号35のヌクレオチドおよび配列番号36のヌクレオチドからなるPCRプライマーを含む癌の診断薬。
