Journal of Lipid Research 2002;43:598-603 Circulation 2004;109:2980-2985 厚生労働省特定疾患原発性高脂血症調査研究班平成12年度研究報告書 2000:37-41
動脈硬化性疾患を発症していない男性および女性健常人360名、および本人、家族歴からFCHLに羅患していることが確認されている9例を用いてsmall,dense LDLコレステロールの測定を行った。small,dense LDLコレステロールの測定はsd LDL-C「生研」(デンカ生研社製)を使用した。
実施例1で用いたFCHL患者群において検体中の各種脂質パラメーターの比較を行った。small,dense LDLコレステロールの測定は実施例1と同様の試薬を使用した。中性脂肪、LDL-コレステロール、HDL-コレステロールの測定は酵素法を使用した。
糖尿病、冠動脈疾患のない健常人1345例のsd LDL-Cをsmall,dense LDLコレステロールの測定はsd LDL-C「生研」(デンカ生研社製)を用いて測定した。
実施例3で求めたようにsd LDL-C値40mg/dLをカットオフ値として用いてFCHLを検出する方法と、既存のFCHLを検出する方法であるLDLサイズによる方法、アポ蛋白BとLDLコレステロールの比による方法と比較した。LDLサイズによる方法とは粒子径25.5nm未満LDLの存在すれば陽性とする方法であり、アポ蛋白BとLDLコレステロールの比による方法とはApoB/LDL-C>1.0を陽性とする方法(動脈硬化性疾患診療ガイドライン2002年版、日本動脈硬化学会編)である。
Claims (9)
- 被験者から採取した被検体試料中のsmall,dense LDLコレステロール濃度を測定することを含む、家族性複合型高脂血症を検出する方法。
- 被検体試料中のTGまたはLDL-Cのレベルが正常レベルである患者から採取した被検体試料中のsmall,dense LDLコレステロール濃度を測定することを含む、請求項1記載の家族性複合型高脂血症を検出する方法。
- small,dense LDLコレステロール濃度の測定が、被検体試料中のsmall,dense LDLをそれ以外のLDLと分離する、またはそれ以外のLDLを反応させコレステロールを消去する第1工程、ならびにsmall,dense LDL以外のLDLを分離または消去した後でsmall,dense LDL中のコレステロールを測定する第2工程からなる請求項1または2に記載の家族性複合型高脂血症を検出する方法。
- 被検体試料が血清または血漿であり、血清または血漿中のsmall,dense LDLコレステロール定量値が40mg/dLを超えた場合に家族性複合型高脂血症に羅患していると判断する、請求項1〜3のいずれか1項に記載の方法。
- 被験者から採取した被検体試料中のsmall,dense LDLコレステロール濃度を測定することを含む、家族性複合型高脂血症患者において動脈硬化性疾患を起こす危険性を判定する方法。
- 被験者から採取した被検体試料中のsmall,dense LDLコレステロール濃度の増加が、前記被験者の動脈硬化性疾患を起こす危険性の増加を示す、請求項5記載の家族性複合型高脂血症において動脈硬化性疾患を起こす危険性を判定する方法。
- small,dense LDLコレステロール濃度の測定が、被検体試料中のsmall,dense LDLをそれ以外のLDLと分離する、またはそれ以外のLDLを反応させコレステロールを消去する第1工程、ならびにsmall,dense LDL以外のLDLを分離または消去した後でsmall,dense LDL中のコレステロールを測定する第2工程からなる請求項5または6に記載の家族性複合型高脂血症において動脈硬化性疾患を起こす危険性を判定する方法。
- 被検体試料が血清または血漿であり、血清または血漿中のsmall,dense LDLコレステロール定量値が40mg/dLを超えた場合に家族性複合型高脂血症患者において動脈硬化性疾患を起こす危険性があると判定する、請求項5〜7のいずれか1項に記載の方法。
- small,dense LDLコレステロールの測定用試薬を含む、家族性複合型高脂血症検出用キット。
