Abstract
Description
本発明者らの一部は、オリゴヌクレオチドであっても、それをポリ(エチレンオキシド)と共有結合させてコンジュゲートとした後、ポリカチオンとPICミセルを形成させることによりPICミセル内にオリゴヌクレオチドを比較的安定に保持できることを見出した。さらに、該コンジュゲートを形成する際に、連結基部分に生理学的環境下、特に、エンドソームのpH(=5.5)において開裂できるエステル結合を組み込むことにも成功し、公表した(例えば、下記の非特許文献2参照。)。また、こうしてPICミセル内に内包されたオリゴヌクレオチドは、in vitroでのヌクレアーゼの作用に抵抗性も示す。
他方、オリゴヌクレオチド誘導体それ自体がヌクレアーゼに抵抗性を示すものとして、ホスホロチオエートオリゴヌクレオチド(例えば、下記の非特許文献3参照。)、ホスホロアミデートオリゴヌクレオチド(例えば、下記の非特許文献4参照。)および2′−O−メチルホスホエートオリゴヌクレオチド(例えば、下記の非特許文献5参照。)も合成され、一部は遺伝子治療法の確立を目差して臨床試験に供されているものもある。また、遺伝子治療の標的遺伝子の発現を確実に抑制することを目差し、標的遺伝子と特異的に共有結合により架橋し得るようにオリゴヌクレオチド内の2−アミノプリン環の6位に−CH2CH2S(O)nPh(ここで、nは0〜2の整数を表す。)で表される基(ビニル基の前駆基)を導入した機能性オリゴヌクレオチドも提案されている(例えば、下記の非特許文献6または特許文献1参照)。具体的には、このような機能性オリゴヌクレオチドは、該ビニル基の前駆基を利用してin vitroで相補的な配列のオリゴヌクレオチドのシトシンのアミノ基と共有結合により架橋することも示されている。

本発明は、ある一定のオリゴヌクレオチド(非特許文献2参照)とポリ(エチレンオキシド)のコンジュゲートを内包するPICミセルが動物細胞内に効果的に取り込まれ、しかも標的遺伝子の発現を強く抑制し得ることを見出した。また、非特許文献3および非特許文献4に記載された機能性オリゴヌクレオチドもポリ(エチレンオキシド)とのコンジュゲートを形成させ、ポリカチオンとのPICミセルにすると動物細胞内へ安定かつ、効果的に送達でき、しかも標的遺伝子の発現を抑制しうることも見出した。
さらに、これらのコンジュゲートは、水性媒体中で極めて可動性が高く、親水性の高いポリ(エチレンオキシド)鎖がオリゴヌクレオチド鎖に共有結合しているにもかかわらず、標的遺伝子と特異的に結合し(水素結合等を介して)、さらに機能性オリゴヌクレオチド(非特許文献4参照)とポリ(エチレンオキシド)のコンジュゲートにあっては、標的遺伝子の発現が非機能性オリゴヌクレオチドを用いたコンジュゲートよりも有意に強く抑制されるとの、驚くべき作用効果を奏することも見出された。これは、理論に拘束されるものでないがポリ(エチレンオキシド)またはその鎖が近傍に存在するにもかかわらず、機能性オリゴヌクレオチドの標的遺伝子への特異的な接近を可能にし、しかも機能性オリゴヌクレオチド上のビニル基と標的遺伝子中のシチジン上のアミノ基とが共有結合を形成することによると考えられる。
したがって、本発明によれば、ある特定の遺伝子の発現を抑制する系で有用なオリゴヌクレオチドのコンジュゲートが提供される。具体的には、本発明によれば、一般式(A):
FuNT−L1−L2−PEG (A)
式中、FuNTはリボースの3′もしくは5′ヒドロキシル基末端においてリン酸エステル結合を介してL1−L2−PEGに結合した機能性オリゴヌクレオチドの残基を表し、ここで機能性オリゴヌクレオチドは、
a) 標的遺伝子の発現に関与するヌクレオチド配列に対して、相補的であるかまたはストリンジェントな条件下でハイブリダイズするオリゴヌクレオチド配列からなり、かつ、該オリゴヌクレオチド配列中に存在する少なくとも1個の2−アミノプリン環の6位に、式−CH2CH2S(O)nR(ここで、nは0〜2の整数を表し、Rはオキシ(=O)、シアノもしくはカルボキシで置換されていてもよいC1−C6アルキルまたはオキシ(=O)、シアノ、ニトロ、カルボキシ、C1−C6アルキルもしくはハロで置換されていてもよいフェニルを表す。)を有するか、あるいは
b) 標的遺伝子の発現に関与するヌクレオチド配列に対して、相補的であり、かつ、各ヌクレオチドのホスフェート部がホスホロチオエート、ホスホロアミデートおよび2′−O−メチルホスホエートの形態にあるものからなる群より選ばれるものであり、
L1は酸素原子もしくは硫黄原子により1もしくは2以上の箇所で中断されていてもよい総原子数3〜30のアルキレン基を表し、
L2は生理学的条件下で開裂しうる連結基を表し、そして
PEGはL2との末結合末端に、連結基を場合により介して、水素原子、アルキル基、アラルキル基、官能基もしくはリガンドの残基を担持するポリ(エチレンオキシド)基を表す、
で表されるオリゴヌクレオチドとポリエチレンオキシドのコンジュゲートが、提供される。
また、本発明によれば、該コンジュゲートとポリカチオン性セグメントを含むポリマーとのポリイオン複合体(PIC)も提供される。
以下、本発明をより具体的に記述する。
本発明に従う、コンジュゲートおよびポリイオン複合体は、ある特定の遺伝子の発現を抑制することを主たる目的とするものである。したがって、機能性オリゴヌクレオチドは特定の遺伝子(または標的遺伝子)の発現に関与するヌクレオチド配列に対して、相補的であるか、またはストリンジェントな条件下でハイブリダイズすることができる配列を含むものである。本発明に関連して、標的遺伝子の発現に関与するヌクレオチド配列とは、特定のコーディング配列およびそれらの転写や翻訳に関与する遺伝子またはその遺伝子領域における配列をも包含する。したがって、これらの配列に相補的である配列(一般に、アンチセンスオリゴヌクレオチド)には、限定されるものでないが、標的遺伝子の転写の阻害、RNAプロセシング阻害、mRNAの核からの移行阻害、翻訳阻害をもたらすものが挙げられる。
特に、上記のa)に分類できる機能性オリゴヌクレオチドは、オリゴヌクレオチド配列中に、式(B):
(式中、nは0〜2の整数を表し、Rはオキシ(=O)、シアノ(−CN)、ニトロ、カルボキシで置換されていてもよいC1〜C6アルキル(例えば、メチル、エチル、n−プロピル、iso−プロピル等)またはオキシ、ニトロ、カルボキシ、C1〜C6アルキル(例えば、メチル)もしくはハロ(例えば、フッ素、塩素)で置換されていてもよいフェニルを表す。)
の機能性ヌクレオチド単位を少なくとも1個有し、このような単位における−CH2CH2S(O)nR基を利用して、標的遺伝子の相補的なオリゴヌクレオチド配列におけるシトシンのアミノ基(−NH2)との間で共有結合を形成しうる。また、この機能性ヌクレオチド単位は、機能性オリゴヌクレオチド鎖中のいかなる位置に存在していてもよい。このような共有結合は、標的遺伝子と機能性オリゴヌクレオチドとが水性媒体または生理学的環境下で接触した場合に、−CH2CH2S(O)mRが−CH=CH2に変換され、次いでアミノ基と反応することにより形成されるものと理解されている。
しかし、機能性オリゴヌクレオチドのヌクレオチド配列が標的遺伝子の特定のヌクレオチド配列に、必ずしも完全に相補的でない場合であっても、ストリンジェントな条件下(例えば、Molecular Cloning,2nd.ed.,Cold Spring Harbor Laboratory 1989,New Yorkの1.101〜1.104頁参照)で該遺伝子にハイブリダイズしうる限り、配列特異的な結合がもたらされるので、このような機能性オリゴヌクレオチドも本発明の範囲内にある。
他方、上記のb)に分類される機能性オリゴヌクレオチドは、ヌクレオチドのホスフェート結合が、それぞれ式(C)〜(E):
で表されるようなホスホロチオエート、ホスホロアミデートおよび2′−O−メチルホスホエート結合に誘導されたものである。
本発明にいうオリゴヌクレオチドは、所謂、「オリゴ」が意味する数個から10数個のヌクレオチドからなるものに限定されず、最大、200個までのヌクレオチドからなるものも包含する。しかし、オリゴデオキシヌクレオチドにあっては、10〜50のヌクレオチドを含むものが好ましい。
また、各ヌクレオチド間のホスホジエステル結合の向きは、通常、3′→5′もしくは5′→3′であるが、ヌクレオチドの種類によっては、2′→5′もしくは5′→2′であってもよく、また、これらの結合様式が混在していてもよい。さらに、オリゴヌクレオチドを構成するヌクレオチドはDNAまたはRNAを構成するいずれのものであってもよい。
このようなオリゴヌクレオチドには、通常のDNAおよびRNAの方向付けに基づいて、3’末端側(すなわち、リボースの5’位はリン酸ジエステル結合に関与している。)の3’ヒドロキシル基か、または5’末端側(すなわち、リボースの3’位はリン酸ジエステル結合に関与している。)の5’ヒドロキシル基を介するリン酸エステル結合を形成するように式−L1−L2−PEGで表される基が共有結合される。
該式中のL1は、酸素もしくは硫黄原子、または−NHCO−、好ましくは酸素原子により1もしくは2以上の箇所で中断されていてもよい総原子数3〜30のアルキレン基であることができる。このようなアルキレン基は、分岐していてもよく、したがって、限定されるものでないが、
−CH2CH2CH2−、−CH2(CH3)CH−、−CH2(CH2)2CH2−、
−CH2CH2−O−CH2CH2−、−CH2CH2−S−CH2−CH2−、
−CH2CH(CH2OH)CH2CH2CH2CH2NHCOCH2CH2−等が例示される。
L2は連結基を表し、本発明の目的に沿う限り、FuNT−L1部とPEG部とを連結しうるいかなる基、例えば、アルキレン基(上記の例示を参照。)、アリレン基、−O−、−S−、−NH−、−COO−、−SS−、−NHCO−、−NHCO−NH−、−SO−から選ばれる1種以上からなるものであってもよい。限定されるものでないが、L2の例としては、−SS−、−SCH2CH2−COO−、−OCH2CH2COO−、−NHCH2CH2COO−、−SCH2CH2−COOCH2−、−SOCH2CH2COO−が挙げられる。このような連結基のうち、生理学的条件下(動物の組織、細胞、体液等に含まれるペプチダーゼ、エステラーゼ、還元性物質等の存在下)で開裂されうる基、例えば−SS−基、−COO−もしくは−OCO−基を連結基内に少なくとも1個含むものが好ましい。特に、エンドソームのpH(=5.5)で開裂しうる連結基を1個含むものが好ましい。
−PEGは未結合末端(L2との結合末端のもう一方の末端)に連結基を場合により介して、水素原子、アルキル基、アラルキル基、官能基もしくはリガンドの残基を担持するポリ(エチレンオキシド)基を表す。具体的には、−PEGは、式(F):
−(CH2CH2O)n−L3−X (F)
式中、L3は単結合または上記のL1もしくはL2について定義したのと同義のアルキレン基もしくは連結基から選ばれる連結基であり、
Xは水素原子もしくは、アルキル基(例、メチル、エチル、プロピル、ブチル、またはヘキシル等)、アラルキル基(例、ベンジル、フェネチル等)、ヒドロキシル基、アルデヒド(もしくはホルミル基:−CHO)、アセタール化ホルミル基、アミノ基、保護されたアミノ基、マレイミド基、カルボキシル基および保護されたカルボキシル基(なお、「保護された」と称する場合の保護基は、限定されるものでないが、ペプチド合成で慣用されるアミノ基およびカルボキシル基に対する保護基を意味する。)からなる群より選ばれる官能基または該官能基を介して結合したリガンド(例、糖、ビオチン、ハプテン、ホルモン、または酵素の基質等)の残基であり、そして
nは5〜500の整数である、
で表されるポリ(エチレンオキシド)基である。
式(F)または−L2−(CH2CH2O)n−L3−Xをもたらすポリ(エチレンオキシド)誘導体は、本発明者らの一部によって提案されたヘテロテレケリック親−疎水性ブロックコポリマーを製造する際の、中間段階である、ポリ(エチレンオキシド)セグメントの形成に従って(例えば、WO 96/33233、WO 96/32434、WO 97/06202、前述の特許文献1および2参照。)、または必要により、それらに当業技術分野で周知の技法を加えて製造することができる。
限定されるものでないが、典型的なポリ(エチレンオキシド)誘導体の製造法は次に示す反応スキームに従って実施できる。

以上の反応スキームに示す各反応段階の条件は、前述の特許文献1を参照できる。
次に、一般式(A)におけるL2に−SS−結合または−COO−結合を有するオリゴヌクレオチドとポリエチレンオキシドのコンジュゲートは、例えば、次に示す反応スキームに従って製造できる。

上式中、oligo−FuNTは、上記の機能性オリゴヌクレオチドについて定義したa)およびb)に分類されるものであることができる。
a)に分類されるものは、非特許文献4または特許文献1に記載された方法に基づいて取得でき、またb)に分類されるものは、非特許文献3に記載された方法に基づいて取得できる。また、oligo−FuNT−CH2CH2CH2SHの取得には、非特許文献2に記載の方法を参考にすることができる。
本発明の機能性オリゴヌクレオチドが標的とする遺伝子は、本発明の目的に沿う限り、如何なる遺伝子であってもよいが、好ましくは、動物の病原遺伝子およびそれらの発現に関与するDNAまたはRNA部分であることができる。限定されるものでないが、標的遺伝子には、非小細胞肺癌などに関係のあるPKCα、悪性黒色腫などに関係のあるBCL−2、クローン病に関係のあるICAM−1、C型肝炎に関係のあるHCV、関節リウマチ、乾鮮に関係のあるTNFα、喘息に関係のあるアデノシンA1受容体、卵巣癌などに関係のあるc−raf kinase、膵臓癌などに関係のあるH−ras、冠動脈疾患癌に関係のあるc−myc、大腸癌に関係のあるPKA RIα、エイズに関係のあるHIV、固形癌に関係のあるDNAメチル−トランスフェラーゼ、癌に関係のあるVEGF受容体、腎臓癌に関係のあるリボヌクレオチド還元酵素、CMV性網膜炎に関係のあるCMV IE2、前立腺癌に関係のあるMMP−9、悪性グリオーマに関係のあるTGFβ2、多発性硬化症に関係のあるCD49d、糖尿病に関係のあるPTP−1B、癌に関係のあるc−myb、乳癌などに関係のあるEGFR、癌に関係のあるmdr1およびGLUT−1の遺伝子を挙げることができる。そしてこれらの遺伝子の発現を効果的に抑制することのできるアンチセンス配列も、それ自体公知の配列のものを利用できる。
こうして取得される本発明に従うコンジュゲートは、必要により、さらにリガンドを付する反応に供してもよい。例えば、上記の(f)または(g)で表されるコンジュゲートのアセタール化ホルミル基[(CH3CH2O)CH−]を酸加水分解に供してホルミル基(−CHO)に転化し、次いで、必要によりアミノ基を導入するかもしくはアミノ基を保持するリガンドとの反応により、リガンドをコンジュゲートに導入することもできる。
これらのコンジュゲートは、ポリカチオン性セグメントを含むポリマーと水性媒体(例えば、緩衝化された生理食塩水、等)中でPICを形成し、自動的に会合(または自己集成)してPICミセルを形成し得る。このようなPICまたはPICミセルも本発明の一態様である。本発明によれば、ポリカチオン性セグメントを含むポリマーとしては、ポリカチオン性セグメントがポリリジン、ポリエチレンイミン、ポリメタクリル酸ジ−C1−3アルキルアミノエチル、オリゴアルギニン、tatおよびKALAからなる群より選ばれるホモポリマー、さらには、該ポリカチオン性セグメントとポリ(エチレンオキシド)セグメントを含有するブロックコポリマーを挙げることができる。
これらのポリマーは、一部は市販されており、それらをそのまま用いるか、また、非特許文献1等を参考に取得したものを用いることができる。
上記のPICミセルは、希薄な水溶液でも安定に機能性オリゴヌクレオチドとポリ(エチレンオキシド)のコンジュゲートをミセル内に保持することができる。また、該PICミセルは動物細胞に効果的に取り込まれ、エンドソームのpH(=5.5)の環境下で該コンジュゲートから機能性オリゴヌクレオチドを開裂して放出でき、しかも上記のa)に分類される機能性オリゴヌクレオチドは配列特異的に標的遺伝子に共有結合の形成を介して架橋しうるので、該遺伝子の発現をより確実に抑制できる。したがって、上記のPICミセルはin vitro、ex−vivoおよびin vivoの系での遺伝子の発現を制御するのに使用できるとともに、遺伝子治療の分野でも有用である。
図2は、本発明に従うコンジュゲートおよび該コンジュゲートとPICミセルのアンチセンス効果の結果を示すグラフである。縦軸は、ホタルルシフェラーゼの発現量/ウミシイタケルシフェラーゼの発現量(%)を表し、横軸に示す(1)〜(12)は、実施例2を参照されたい。
図3は、本発明に従う、リガンド(ラクトース)を有するコンジュゲートとポリリジンからなるPICミセルのアンチセンス効果を示すグラフである。縦軸は、ホタルルシフェラーゼの発現量/ウミシイタケルシフェラーゼの発現量(%)を表し、横軸は、PEG−機能性オリゴヌクレオチドコンジュゲートを表す。黒塗りバーは、ラクトースありSPhコンジュゲートのPICミセルに関し、そして白抜きバーは、ラクトースなしSPhコンジュゲートのPICミセルに関する結果である。
以下、具体的な態様を挙げてさらに本発明を説明する。
参考製造例1:アセタール−PEO−アクリレートの製造
アルゴン下、ナスフラスコ中、室温において、開始剤3,3−ジエトキシ−1−プロパノール1.0mmol(0.16mL)を溶媒テトラヒドロフラン(THF)25mlにマイクロシリンジで加え、K−ナフタレン1.0mmol(0.325mol/1−THF溶液、3.1mL)を加えて10分間メタル化を施した。次いで、エチレンオキシド110mmol(5.5mL)を加えて水冷下で2日間撹拌し、アニオン開環重合を行った。その後、トリエチルアミンを3倍モル量3.0mmol(0.42mL)と停止剤としてアクリロイルクロライドを3倍モル量3.0mmol(0.24mL)を順次加え、室温において1日停止反応を行った。その後、2−プロパノール沈澱(2l)、遠心分離(5000×g、45分間)、減圧乾燥、ベンゼン凍結乾燥により精製を行った。この生成物の収量は5.0g(98%)であった。
ゲルパーミエーションクロマトグラフィーの測定により、得られたポリマーは単峰性であり、その数平均分子量は4830であり、仕込み分子量5080とほぼ一致していることが確認できた。
同様にMALDI−TOF−MS(マトリックス支援レーザー脱離イオン化飛行時間型質量分析計)の測定においても、得られたポリマーは単峰性であり、その数平均分子量は4460であった。また、これらのピークの測定値と計算値を比較した結果、このポリマーはエチレンオキシド骨格を主鎖に有し、α−末端にアセタール基、ω−末端にアクリレート基を有するヘテロテレケリックポリエチレンオキシドであることが確認された。
さらに、得られたポリマーの重DMSO中での1H−NMR(プロトン核磁気共鳴)スペクトルより、このポリマーの平均分子量は4870と計算された。またこのポリマーはエチレンオキシド骨格を主鎖に有し、α−末端にアセタール基、ω−末端にアクリレート基を有するヘテロテレケリックポリエチレンオキシドであることが確認された。
式中、Xは、下記式のいずれかで表されるデオキシヌクレオチド誘導体残基である。
2−アミノ−6−ビニルプリンのアミダイト前駆体を用いて2−アミノ−6−ビニルプリンを含む3′末端アミノオリゴマーを合成した。このオリゴマー0.5μmolを1.5μmolのマグネシウムモノパーフタレート(MMPP)を用いて酸化し、引き続き反応液をアルカリ性(pH10)にすることでビニル基を有する3′末端アミノオリゴマーを効率よく合成した。このオリゴマー0.5μmolにチオフェノール10μmolおよび2−カルボキシチオフェノール25μmolを反応させビニル体の安定前駆体であるスルフィド誘導体を合成した。さらに、保護したチオール基を有する活性エステル3−(2−ピリジルチオ)プロピオン酸N−スクシイミジル(SPDP)を1mMのエチレンジアミン四酢酸(EDTA)を含むリン酸緩衝液中(pH7.5)で反応させ、次いでジチオスレイトールで還元することで、3′末端にチオール基を有する機能性オリゴヌクレオチドを合成した。得られた3′末端にチオール基を有する修飾オリゴヌクレオチド0.2μmolをPEG誘導体とリン酸緩衝液(pH8.0)中、5時間反応させた。目的のPEGと機能性オリゴヌクレオチドがコンジュゲーションしたPEG−機能性オリゴヌクレオチドコンジュゲートが収率よく得られた(図1参照)なお、反応スキームを参照されたい。
実施例2: PEG−機能性オリゴヌクレオチドコンジュゲートとポリリジンからなるポリイオンコンプレックス(PIC)ミセルの製造
PEG−機能性オリゴヌクレオチドコンジュゲート60nmolおよびポリリジン(平均重合度=100)144nmolを、それぞれ10mMトリス−塩酸緩衝溶液(pH7.4)に溶解させた後、0.1μmのフィルターで濾過しゴミを取り除いた。次に、これらの溶液をポリリジンの正電荷とPEG−機能性オリゴヌクレオチドコンジュゲートの負電荷の比が1(N/P=1)となるように混合した。さらに、10mMトリス−塩酸緩衝溶液(pH7.4、0.3M NaCl)を混合液と同量加え、12時間静置しPICミセルを形成させた。
実施例3: PEG−機能性オリゴヌクレオチドコンジュゲートとポリリジンからなるPICミセルのアンチセンス効果
24穴ポリスチレン製細胞培養プレート(ファルコン社製)に、人肝癌細胞(Huh7 cell)を5×104cells/well播種し、24時間培養した後、市販の遺伝子導入試薬であるLipofectAMINE(1.22μL/well,インビィトロジェン社製)を用い、ホタルルシフェラーゼプラスミドDNA(pGL3,0.084μg/well,プロメガ社製)とウミシイタケルシフェラーゼプラスミドDNA(pRL−TK,0.75μg/well,プロメガ社製)のレポーター遺伝子を細胞にトランスフェクションした。次に、ホタルルシフェラーゼ遺伝子に対するアンチセンス配列を有するPICミセル溶液(実施例2で調製した)、PEG−機能性オリゴヌクレオチドコンジュゲート単独および機能性オリゴヌクレオチド単独の溶液を所定量加え(培地濃度換算で5μMのコンジュゲートとなるように加え)、24時間細胞と接触させた。培地交換後、さらに24時間培養した後、細胞を回収し両レポーター遺伝子の発現量をDual Luciferase Reporter Assay System(プロメガ社製)により測定しアンチセンス効果を評価した(n=3)。
比較のために、実施例3と同様の条件で、PEG−アンチセンスDNAコンジュゲートとポリリジンからなるPICミセル、PEG−アンチセンスDNAコンジュゲート単独およびアンチセンスDNA単独のアンチセンス効果も評価した。
結果を図2に示す。図中、コントロール(1)、アンチセンスDNA単独(2)、SPh機能性オリゴヌクレオチド単独(3)、PEG−アンチセンスDNAコンジュゲート単独(4)およびPEG−Sph機能性オリゴヌクレオチドコンジュゲート単独(5)では、アンチセンス効果を全く示さないことが確認された。一方、PICミセルでは、アンチセンス効果を示すことが確認された。具体的には、SMe構造を有するPEG−機能性オリゴヌクレオチドコンジュゲートのPICミセル(8)(約80%のアンチセンス効果)およびSPh構造を有するPEG−機能性オリゴヌクレオチドコンジュゲートのPICミセル(9)(約80%のアンチセンス効果)は、PEG−アンチセンスDNAコンジュゲートPICミセル(6)(約55%のアンチセンス効果)よりも、高いアンチセンス効果を示すことが確認された。
また、ランダムな塩基配列(ホタルルシフェラーゼ遺伝子に関係ない配列)を有するSPh構造を有するPEG−機能性オリゴヌクレオチドコンジュゲートのPICミセル(10)では、アンチセンス効果を全く示さなかったことから、以上のアンチセンス効果が標的遺伝子特異的なものであることがわかる。
さらに、一塩基ミスマッチ配列有するPEG−アンチセンスDNAコンジュゲートPICミセル(11)のアンチセンス効果は、一塩基ミスマッチを有さないPICミセルとほぼ同等であったのに対し、一塩基ミスマッチ配列有するSPh構造を有するPEG−機能性オリゴヌクレオチドコンジュゲートのPICミセル(12)のアンチセンス効果は、一塩基ミスマッチを有さないPICミセルに比べ著しくアンチセンス効果が低下することが確認されたことから、このSPh構造を有するPEG−機能性オリゴヌクレオチドコンジュゲートのPICミセル(9)が高い配列認識を持っていることが明らかとなった。
実施例3と同様の条件で、ラクトース(リガンド)を有するSPh構造を有するPEG−機能性オリゴヌクレオチドコンジュゲートとポリリジンからなるPICミセルのアンチセンス効果も評価した。
結果を図3に示す。1μMおよび5μMのいずれの濃度においても、ラクトースを有するSPh構造を有するPEG−機能性オリゴヌクレオチドコンジュゲートPICミセル(黒塗りバー)のほうが高いアンチセンス効果を示すことが確認された。
Claims (10)
- 一般式(A):
FuNT−L1−L2−PEG (A)
式中、FuNTはリボースの3′もしくは5′ヒドロキシル基末端においてリン酸エステル結合を介してL1−L2−PEGに結合した機能性オリゴヌクレオチドの残基を表し、ここで機能性オリゴヌクレオチドは、
a) 標的遺伝子の発現に関与するヌクレオチド配列に対して、相補的であるかまたはストリンジェントな条件下でハイブリダイズするオリゴヌクレオチド配列からなり、かつ、該オリゴヌクレオチド配列中に存在する少なくとも1個の2−アミノプリン環の6位に、式−CH2CH2S(O)nR(ここで、nは0〜2の整数を表し、Rはオキシ(=O)、シアノもしくはカルボキシで置換されていてもよいC1−C6アルキルまたはオキシ(=O)、シアノ、ニトロ、カルボキシ、C1−C6アルキルもしくはハロで置換されていてもよいフェニルを表す)を有するか、あるいは
b) 標的遺伝子の発現に関与するヌクレオチド配列に対して、相補的であり、かつ、各ヌクレオチドのホスフェート部がホスホロチオエート、ホスホロアミデートおよび2′−O−メチルホスホエートの形態にあるものからなる群より選ばれるものであり、
L1は酸素原子もしくは硫黄原子により1もしくは2以上の箇所で中断されていてもよい総原子数3〜30のアルキレン基を表し、
L2は生理学的条件下で開裂しうる連結基を表し、そして
PEGはL2との末結合末端に、連結基を場合により介して、水素原子、アルキル基、アラルキル基、官能基もしくはリガンドの残基を担持するポリ(エチレンオキシド)基を表す、
で表されるオリゴヌクレオチドとポリエチレンオキシドのコンジュゲート。 - 機能性オリゴヌクレオチドがアンチセンスデオキシオリゴヌクレオチドであって、該デオキシオリゴヌクレオチドにおける少なくとも1個の2−デオキシグアノシンの6位に、式−CH2CH2S(O)nR′(ここで、nは0〜2の整数を表し、R′はメチルまたはフェニルを表す)を有し、そしてポリ(エチレンオキシド)の数平均分子量が200〜30000である請求項1に記載のコンジュゲート。
- L2の生理学的条件下で開裂しうる連結基が主鎖にエステル結合(−COO−)またはジスルフィド結合(−SS−)を有する請求項1に記載のコンジュゲート。
- −PEGが式:
−(CH2CH2O)n−L3−X
式中、L3は単結合またはL1もしくはL2について定義したのと同義のアルキレン基もしくは連結基から選ばれる連結基であり、
Xは水素原子もしくは、アルキル基、アラルキル基、ヒドロキシル基、ホルミル基、アセタール化されたホルミル基、アミノ基、保護されたアミノ基、マレイミド基、カルボキシル基および保護されたカルボキシル基からなる群より選ばれる官能基または該官能基を介して結合したリガンドを表し、そしてnが5〜500の整数である請求項1〜3のいずれかに記載のコンジュゲート。 - オリゴヌクレオチドが10〜50のヌクレオチドを含むオリゴデオキシヌクレオチドである請求項1に記載のコンジュゲート。
- ポリカチオン性セグメントを含むポリマーと請求項1に記載のコンジュゲートを含有する複合体。
- ポリカチオン性セグメントを含むポリマーがホモポリマーまたは該セグメントとポリ(エチレンオキシド)セグメントとを含有するブロックコポリマーである請求項6に記載の複合体。
- ポリカチオン性セグメントがポリリジン、ポリエチレンイミン、ポリメタクリル酸 2−(N,N−ジ−C1−3アルキルアミノ)エチル、オリゴアルギニン、tatおよびKALAからなる群より選ばれるホモポリマーである請求項6に記載の複合体。
- 複合体が水性媒体中でポリ(エチレンオキシド)鎖から主として構成されるシエル部と機能性オリゴヌクレオチドから主として構成されるコア部とからなるポリイオン複合体ミセルを形成する請求項6に記載の複合体。
- 機能性オリゴヌクレオチドがアンチセンスデオキシオリゴヌクレオチドであって、該デオキシオリゴヌクレオチドにおける少なくとも1個の2−デオキシグアノシンの6位に、式−CH2CH2S(O)nR′(ここで、nは0〜2の整数を表し、R′はメチルまたはフェニルを表す。)を有する請求項6に記載の複合体。
