Abstract
Description
単純ヘルペスウイルス1型(HSV−1)とHHV−6またはHHV−7は、進化的には同じ祖先から発生しているが、遺伝子の構造が全く異なり、互いの遺伝子配列の相同性が低く、ベクターの作成や遺伝子治療を行なう際に重要である細胞指向性(cellular tropism)が全く異なる。このため、HHV−6またはHHV−7由来ベクターの作成には、単純ヘルペスウイルス1型(HSV−1)ベクターとは異なった技術開発が必要である。
本発明の課題は、外因性ヌクレオチド配列を挿入することができ、哺乳類の宿主細胞に容易に導入することができ、外因性ヌクレオチド配列にコードされた遺伝子を宿主細胞内で発現させることができ、病原性を有する危険性が低く、哺乳類の遺伝子治療に好適に用いることができる、ウイルスベクターを提供することである。
本明細書において、HHV−6とは、ヒトヘルペスウイルス6(Human herpesvirus6)のvariant AおよびBを意味するものとする。
本発明の組換ウイルスおよび組換ウイルスベクターは、HHV−6由来の組換ウイルスおよび組換ウイルスベクターであって、HHV−6のU2、U3、U4、U5、U6、U7、U8、U24およびU25領域からなる群より選ばれる少なくとも1領域に相当する部位に、外因性ヌクレオチド配列を備える、組換ウイルスおよび組換ウイルスベクターである。
本発明の組換ウイルスおよび組換ウイルスベクターは、HHV−7由来の組換ウイルスおよび組換ウイルスベクターであって、HHV−7のU2、U3、U4、U7、U8、U24、U24aおよびU25領域からなる群より選ばれる少なくとも1領域に相当する部位に、外因性ヌクレオチド配列を備える、組換ウイルスおよび組換ウイルスベクターであってもよい。
また、HHV−7の上記の部位は、配列番号2で表されるHHV−7のDNA配列におけるヌクレオチド番号11631〜17221または34744〜36118の範囲に存在する部位であってもよい。この範囲については、後述する実施例において前者は欠失可能である事が、後者は組み換え可能であることが実験的に確認されているからである。
上記の外因性ヌクレオチド配列は、DNA配列および/またはRNA配列であってもよい。DNA配列はゲノムDNA配列であってもよく、cDNA配列であってもよい。
本発明の組換ウイルスおよび組換ウイルスベクターの製造方法は、HHV−6由来の組換ウイルスおよび組換ウイルスベクターの製造方法であって、HHV−6のU2、U3、U4、U5、U6、U7、U8、U24およびU25領域からなる群より選ばれる少なくとも1領域に相当する部位に、外因性ヌクレオチド配列を挿入するステップを備える、組換ウイルスおよび組換ウイルスベクターの製造方法である。
本発明の宿主細胞の形質転換方法は、上記の組換ウイルスおよび組換ウイルスベクターによる哺乳類の宿主細胞の形質転換方法であって、上記の組換ウイルスおよび組換ウイルスベクターにより上記の宿主細胞を0.01〜20の感染多重度(MOI)で形質転換するステップを備える、宿主細胞の形質転換方法である。
ここで、これらの細胞は、従来のベクターでは導入効率や発現などに問題があったが、本発明の組換ウイルスベクターを用いることにより、これらの細胞にも外来遺伝子を導入して発現させることが可能になる利点がある。
本発明の形質転換宿主細胞は、上記の宿主細胞の形質転換方法により得られる形質転換宿主細胞である。
本発明の遺伝子治療方法は、ヒト以外の哺乳類の遺伝子治療方法であって、上記の形質転換細胞をこの哺乳類へ投与するステップを備える、遺伝子治療方法である。
ヒトヘルペスウイルス6(HHV−6)のU3−U7遺伝子クラスターを、ヒトサイトメガロウイルスの主要な即時型エンハンサー−プロモーター(MIEP)の制御下にある増強された緑色蛍光タンパク質(enhanced green fluorescent protein:EGFP)と、SV40初期プロモーターの制御下にあるピューロマイシン耐性遺伝子と、を含む遺伝子カセット(EGFP−puroカセット)に置換した、組換えウイルスH6R28を構築した。相同組換によってEGFP−puroカセットをHHV−6ゲノムに挿入するために、ウイルスのゲノムの約1キロベース(kb)の部分がカセット(図1)のそれぞれの末端に挿入された。
予想される領域の中にEGFP−puroカセットが挿入されたことを確認するために、相同ヒンジ領域の外側の領域に設計されたプライマー(外側のプライマーセット:U2R2−U8F2、内側のプライマーセット:U2R1−U8F1、図1参照)を用いて、二重ネストPCRによりウイルスのDNAを増幅した。PCRにはKODプラスDNAポリメラーゼ(東洋紡)を用いた。
本発明者は、独立した3つの個別のエレクトロポレーションにより3つの個別のH6R28を作出して、CBMCsで複製の動態を調べた。前述のように、ウイルス力価の測定はCBMCsを用いてAsadaらの方法(H.Asada,et.al,J.Clin.Microbiol.27:2204−2207,1989)により行われた。CBMCsにMOI0.05の条件で感染させると、3つのH6R28クローンとwtウイルスとはウイルスの感染の拡大(図3(A))およびウイルス生産(図3(B))が、長い時間にわたって類似の水準であった。
HHV−6のU24とU25を組み換え部位に使用した組み換えウイルスH6R24−25を構築した。
本発明者は次に潜伏感染を確立する能力、再活性化の効率に関しH6R28を用いて実験した。
図6(A)は、HHV−6のDNA陽性細胞の存在割合を示す。HHV−6のDNAが検出される細胞の存在割合は、感染後4週目と6週目に調べられた。wtウイルスとH6R28の3つのクローンの3回の繰返し培養の平均値と標準偏差が示される。白色柱は、野生型ウイルスを示し;灰色柱は、H6R28を示す。
(1)マクロファージ、CBMCs、Molt−3、HeLa
興味深いことに、HHV−6の潜伏感染の間に、本発明者はHCMVの主要な即時型エンハンサーとプロモーター遺伝子(MIEP)によって促進されるEGFPの発現を認めることができなかった(図7(A))。他方、EGFPの発現が、図1に示したプラスミドpU2−U8EGFP−puroをトランスフェクトされた、潜伏感染しているマクロファージ(図7(B))、再活性化−誘導マクロファージ(図7(C))、生産的感染したCBMC細胞とMolt−3細胞(図7(D)と7(E))、流産感染したHela細胞(図7(F))で観察された。
H6R28のfree virusを、interleukin−2(IL−2)存在下で培養した成人末梢血由来単核球にmultiplicity of infection(MOI)1で感染させた。CD56陽性細胞(NK細胞)への遺伝子導入を、感染3日後にFACSを用いてEGFP発現で確認した。
H6R28のfree virusを、basic fibroblast growth factor(bFGF)存在下で培養したヒト初代培養アストロサイトにmultiplcity of infection(MOI)1で感染させた。遺伝子導入は2日後に蛍光顕微鏡を用いてEGFPの発現で確認した。
H6R28のfree virusを、phytohemagglutinin(PHA)存在下で培養した成人末梢血由来単核球にmultiplicity of infection(MOI)1で感染させた。CD4陽性T細胞への遺伝子導入を、感染1日後にFACSを用いてEGFP発現で確認した。
H6R28のfree virusを、phytohemagglutinin(PHA)存在下で培養した成人末梢血由来単核球にmultiplicity of infection(MOI)1で感染させた。CD8陽性T細胞への遺伝子導入を、感染3日後にFACSを用いてEGFP発現で確認した。
H6R28のfree virusを、interleukin−4(IL−4)とgranulocyte−macrophage colony−stimulating factor(GM−CSF)存在下で培養した成人末梢血由来単球にmultiplicity of infection(MOI)1で感染させた。CD83陽性細胞(樹状細胞)への遺伝子導入を、感染3日後にFACSを用いてEGFP発現で確認した。
なお、実施例には記載しないが、血液幹細胞、肝細胞、血管内皮細胞、繊維芽細胞においても、野生株のHHV−6が感染することは報告されており(参考文献1〜5参照)、H6R28のfree virusが上記細胞に感染することが示唆される。

<潜伏感染HHV−6におけるHCMVプロモーターの機能>
IE1/IE2プロモーター遺伝子からの遺伝子発現を検討するために、5’RACEを行なった(J.Virol.77:2258−2264,2003、J.Virol.76:4145−4151,2002、Proc.Natl.Acad.Sci.USA,93:11137−11142,1996)。手短かに言えば、cDNAの5′末端はdA−tailが付加されて、そしてアンカープライマー、RL−1と一緒にアニーリングされた。PCRの最初の10のサイクルは、Taqポリメラーゼ(Roche Diagnostics)を用いて以下の条件で行なった。変性は94°Cにおいて1分間、アニーリングは55°Cにおいて1分間、伸長は72°Cにおいて1分間の条件で行なった。
MIEPがHHV−6潜伏感染条件で潜伏感染と関係のある性質を示したので、HHV−6潜伏感染の転写調節がHCMV潜伏感染と若干の共通の機構を共有するかもしれないことが示唆される。これらの知見は、潜伏感染の部位の場合のように、HHV−6とHCMVとが比較的共通した性質を示すという事実と関係があるかもしれない。
ヒトヘルペスウイルス7(HHV−7)のU2−U8遺伝子クラスターを、ヒトサイトメガロウイルスの主要な即時型エンハンサー−プロモーター(MIEP)の制御下にある増強された緑色蛍光タンパク質(enhanced green fluorescent protein:EGFP)と、SV40初期プロモーターの制御下にあるピューロマイシン耐性遺伝子と、を含む遺伝子カセット(EGFP−puroカセット)に置換した組換えウイルスH7R28を構築した。相同組換によってEGFP−puroカセットをHHV−7ゲノムに挿入するために、ウイルスのゲノムの約1キロベース(kb)の部分がカセット(図14)のそれぞれの末端に挿入された。
HHV−7の組換えウイルスベクター構築の確認は、HHV−6の組換えウイルスベクターH6R28構築の確認の方法と同様の方法により行われ、同様の結果を得た。なお、重複記載を避けるため、説明を繰返さない。
H7R28の細胞から細胞への広がりの効率および感染細胞でのウイルス産生の効率を検討した。なお、実験方法は、抗HHV−6モノクローナル抗体の代わりに、抗HHV−7モノクローナル抗体(KR4)を使用したこと以外は、前記H6R28と同じである。
HHV−7のU24とU25を組み換え部位に使用した組み換えウイルスH7R24−25を構築した。
HHV−7の組換えウイルスベクターの潜伏感染能力および再活性化効率は、HHV−6の組換えウイルスベクターの潜伏感染能力および再活性化効率の確認の方法と同様の方法により行われ、同様の結果を得た。なお、重複記載を避けるため、説明を繰返さない。
(1)マクロファージ
H7R28のfree virusを、コラーゲンコートディッシュで培養した成人末梢血由来単球にmultiplicity of infection(MOI)1で感染させた。接着細胞でかつCD11c陽性の細胞(マクロファージ)への遺伝子導入を、感染3日後にFACSを用いてEGFP発現で確認した。
H7R28のfree virusを、phytohemagglutinin(PHA)存在下で培養した成人末梢血由来単核球にmultiplicity of infection(MOI)1で感染させた。CD4陽性T細胞への遺伝子導入を、感染3日後にFACSを用いてEGFP発現で確認した。
H7R28のfree virusを、interleukin−4(IL−4)とgranulocyte−macrophage colony−stimulating factor(GM−CSF)存在下で培養した成人末梢血由来単球にmultiplicity of infection(MOI)1で感染させた。CD83陽性細胞(樹状細胞)への遺伝子導入を、感染3日後にFACSを用いてEGFP発現で確認した。
HHV−6、HHV−7ともに、U2〜U8の外来遺伝子挿入可能部位が大きいので、比較的大きな遺伝子の導入が可能である。特にBacterial artificial chromosome(BAC)の複製開始点を挿入できれば、所謂BACシステムによるベクター産生が可能となる。BACシステムによる組み換えウイルス作製は、他のヘルペスウイルスなど、多くのウイルスで確立しているが、HHV−6およびHHV−7においては未だ確立されていない。
図21は、H6R28 BACの構造を表す模式図である。

上記のように、全体的に、組換ウイルスH6R28により、HHV−6のかなり大きい遺伝子クラスターU3−U7がウイルスの複製、潜伏感染と再活性化のために欠失可能であることが明らかとなった。削除された遺伝子について、U4とU6の機能は報告されていない。
また、H7R28によりHHV−7のかなり大きい遺伝子クラスターU3−U7がウイルスの複製、潜伏感染と再活性化のために欠失可能であることが明らかとなった。
さらに、本発明のウイルスベクターの製造方法で使用されたウイルスの組み換え方法は、他のベクター開発技術であるBACシステムやアンプリコンシステムをHHV−6およびHHV−7に応用しようとする際に、必要不可欠な技術である。
これらは、5’RACE法によって分析された。使われたRACE法としては通常に用いられる方法を採用した。転写物の5′末端はdAテイルであって、そしてアンカープライマーRL−1とともにアニーリングした(図13(A))。そして最初にプライマーN2−EGFP R2と共に、それからプライマーN1−EGFP R1と共に増幅した。PSS(〜360bp)、LSS1(〜720bp)、そして/あるいはLSS2(〜650bp)で始まる転写物の5′末端が検出された。Hae IIIによって切断されたΦ X174 DNA断片がサイズマーカー(ΦX)として用いられた。

- HHV−6由来の組換ウイルスベクターであって、HHV−6のU2、U3、U4、U5、U6、U7、U8、U24およびU25領域からなる群より選ばれる少なくとも1領域に相当する部位に、外因性ヌクレオチド配列を備える、組換ウイルスベクター。
- 前記部位は、配列番号1で表されるHHV−6のDNA配列におけるヌクレオチド番号9041〜17446または36250〜37775の範囲に存在する、請求の範囲1に記載の組換ウイルスベクター。

- 前記外因性ヌクレオチド配列を挿入するステップは、それぞれ配列番号30−31および34−35に示す配列のプライマーの組合せまたは配列番号40−41および42−43に示す配列のプライマーの組合せを用いて増幅されるDNA配列と、HHV−7のDNA配列との相同組換を行なうステップを含む、請求の範囲14に記載の組換ウイルスベクターの製造方法。
- 前記外因性ヌクレオチド配列を挿入するステップは、正常かつ/または臍帯血由来の細胞内において前記外因性ヌクレオチド配列を挿入するステップを含む、請求の範囲11または14に記載の組換ウイルスベクターの製造方法。
- 請求の範囲1または4に記載の組換ウイルスベクターによる哺乳類の宿主細胞の形質転換方法であって、前記組換ウイルスベクターにより、ヒト、ヒト以外の霊長類およびHHV−6またはHHV−7の感染可能な宿主からなる群より選ばれる1種以上の哺乳類由来の前記宿主細胞を形質転換するステップを備える、宿主細胞の形質転換方法。
- 前記形質転換するステップは、前記組換ウイルスベクターにより、T細胞、マクロファージ、末梢血単核球細胞、血液幹細胞、肝細胞、血管内皮細胞、繊維芽細胞、グリア細胞、アストロサイト、CD4陽性T細胞、CD8陽性T細胞、樹状細胞およびナチュラルキラー細胞からなる群より選ばれる1種以上の前記宿主細胞を形質転換するステップを含む、請求の範囲18に記載の宿主細胞の形質転換方法。
- 請求の範囲18または19に記載の宿主細胞の形質転換方法により得られる、形質転換宿主細胞。
- 哺乳類の遺伝子治療方法に用いられる、請求の範囲20に記載の形質転換宿主細胞。
- 前記遺伝子治療は、ヒト免疫不全ウイルス(HIV)に対する易感染性細胞内におけるHIVウイルス感染を予防するための遺伝子治療および/または癌の免疫治療のための遺伝子治療である、請求の範囲21に記載の形質転換宿主細胞。
- 前記宿主細胞は、前記遺伝子治療を行なう対象となる種類の哺乳類と同種の哺乳類由来の宿主細胞である、請求の範囲21に記載の形質転換宿主細胞。
- ヒト以外の哺乳類の遺伝子治療方法であって、請求の範囲20〜23のいずれかに記載の形質転換細胞を前記哺乳類へ投与するステップを備える、遺伝子治療方法。
- ヒト以外の哺乳類の遺伝子治療方法であって、請求の範囲1または4に記載の組換ウイルスベクターにより、前記哺乳類の体内の宿主細胞を0.01〜20の感染多重度(MOI)で形質転換するステップを備える、遺伝子治療方法。
- さらに前記組換ウイルスベクターに備わる外因性ヌクレオチド配列にコードされる遺伝子を発現させるステップを備える、請求の範囲24または25に記載の遺伝子治療方法。
