Description
本発明は良姜(リヨウキヨウ)、忍冬(ニンド
ウ)、何首烏(カシユウ)、クロモジ、南天実(ナ
ンテンジツ)、山査子(サンザシ)、牡丹皮(ボタ
ンヒ)、香〓(コウジユ)、遠志(オンジ)、杏仁
(キヨウニン)、桃仁(トウニン)、紫蘇(シソ)
及び柿葉(シヨウ)より選ばれる植物の有機溶媒
抽出物(粗エキス)を配合することにより、歯垢
の形成を抑制してう蝕及び歯周疾患を予防する口
腔用組成物に関する。
歯の表面に付着する歯垢は、約70%の細菌、細
菌により形成された約20%の多糖、及び約10%の
食物残渣よりなり、固く歯面にこびりついてい
る。そして、その内部に貯えられた酸がエナメル
質を脱灰し、菌及び産生された毒素が歯肉炎、歯
周炎、更には歯槽膿漏をもたらすといわれてお
り、口腔の二大疾患であるう蝕、歯槽膿漏の原因
として注目されている。
この歯垢はストレプトコツカス・ミユータンス
を中心とする口腔内細菌によつて口腔内に存在す
るシヨ糖を利用して形成される。即ち、ストレプ
トコツカス・ミユータンスはGTF(グルコシルト
ランスフエラーゼ、デキストラン合成酵素)を産
生し、これによりシヨ糖からデキストラン、ムタ
ン等の粘着性多糖を合成する。そして、この合成
された多糖はストレプトコツカス・ミユータンス
をはじめ、他の菌(病原菌)を巻き込み、一定の
菌叢を有する歯垢を形成する。また、ストレプト
コツカス・ミユータンス等の菌は種々の糖を利用
して酸を産生し、この酸は多糖及び菌の壁の中に
滞留することにより、エナメル表面を脱灰してい
く。更に、菌の死骸や産生された種々の物質、毒
素は歯肉に対して悪影響を及ぼし、歯槽膿漏の原
因として作用する。
従つて、歯垢の形成を抑制、阻止することは、
う蝕、歯周疾患を予防するという点から非常に有
効なことである。
本発明者らは、このような点を考慮し、う蝕、
歯周疾患の予防に好適な口腔用組成物につき鋭意
研究を行ない、天然物添加剤として主に漢方処方
に用いられる種々の植物をスクリーニングした結
果、前記13種の植物の有機溶媒抽出物がストレプ
トコツカス・ミユータンス由来のGTFによる歯
垢の形成を抑制し、従つて前記有機溶媒抽出物が
口腔用組成物の有効成分としてう蝕、歯周疾患の
予防に好適であることを知見し、本発明をなすに
至つた。
以下、本発明につき詳しく説明する。
本発明に係る口腔用組成物は練歯磨、粉歯磨、
水歯磨などの歯磨類、マウスウオツシユ、トロー
チ、パスタ、塗布剤、チユーインガム、うがい用
錠剤等として使用されるもので、有効成分として
良姜、忍冬、何首烏、クロモジ、南天実、山査
子、牡丹皮、香〓、遠志、杏仁、桃仁、紫蘇及び
柿葉の有機溶媒抽出物の1種又は2種以上を配合
してなるものである。
前記植物の抽出に用いる有機溶媒としては、メ
タノール、エタノール等の低級アルコール、アセ
トン、エチルエーテル、酢酸エチルなどの極性溶
媒、ベンゼン、ヘキサン、石油エーテル等の非極
性溶媒が使用でき、また含水メタノール、含水エ
タノール等の混合溶媒も使用し得る。本発明にお
いて有効成分として用いる有機溶媒抽出物は、上
述した植物をこれら有機溶媒の1種もしくは2種
以上の混合物、又はこれらの有機溶媒と水との混
合物で抽出し、その抽出残渣を除去し、必要に応
じて抽出液から溶媒を留去することによつて製造
できる。なお、抽出方法としては公知の方法が採
用でき、例えば有機溶媒中に前記植物の乾燥粉末
を温浸する等の方法が採用し得る。
前記有機溶媒抽出物の口腔用組成物中への配合
量は、全体の0.001〜5%(重量%、以下同じ)、
特に0.01〜2%とすることが好ましい。
本発明の他の成分としては、口腔用組成物の種
類に応じた適宜な成分が用いられる。例えば練歯
磨の場合であれば、第2リン酸カルシウム、炭酸
カルシウム、ピロリン酸カルシウム、不溶性メタ
リン酸ナトリウム、非晶質シリカ、結晶質シリ
カ、アルミノシリケート、酸化アルミニウム、水
酸化アルミニウム、レジン等の研磨剤、カルボキ
シメチルセルロース、ヒドロキシエチルセルロー
ス、アルギン酸塩、カラゲナン、アラビアゴム、
ポリビニルアルコール等の粘結剤、ポリエチレン
グリコール、ソルビトール、グリセリン、プロピ
レングリコール等の粘稠剤、ラウリル硫酸ナトリ
ウム、ドデシルベンゼンスルホン酸ナトリウム、
水素添加ココナツツ脂肪酸モノグリセリドモノ硫
酸ナトリウム、ラウリルスルホ酢酸ナトリウム、
N−ラウロイルザルコシン酸ナトリウム、N−ア
シルグリタミン酸塩、シヨ糖脂肪酸エステル等の
発泡剤、それにペパーミント、スペアミント等の
精油、l−メントール、カルボン、オイゲノー
ル、アネトール等の香料素材などの香料、サツカ
リンナトリウム、ステビオサイド、ネオヘスペリ
ジルジヒドロカルコン、グリチルリチン、ペリラ
ルチン、p−メトキシシンナミツクアルデヒドな
どの甘味剤、防腐剤などの成分を水と混和し、常
法に従つて製造する。また、マウスウオツシユ等
の口腔洗浄剤その他においても、製品の性状に応
じた成分が適宜配合される。
なお、本発明においては、上述した植物の有機
溶媒抽出物に加えて塩化リゾチーム、デキストラ
ナーゼ、溶菌酵素、ムタナーゼ、クロルヘキシジ
ン、ソルビン酸、アレキシジン、ヒノキチオー
ル、セチルピリジニウムクロライド、アルキルグ
リシン、アルキルジアミノエチルグリシン塩、ア
ラントイン、ε−アミノカプロン酸、トラネキサ
ム酸、アズレン、ビタミンE、モノフルオロリン
酸ナトリウム、フツ化ナトリウム、フツ化第1
錫、水溶性第一もしくは第二リン酸塩、第四級ア
ンモニウム化合物、塩化ナトリウムなどの有効成
分を配合することもできる。
本発明に係る口腔用組成物は、前記13種の植物
の有機溶媒抽出物の1種又は2種以上を配合した
ことにより、ストレプトコツカス・ミユータンス
由来のGTFによる歯垢の形成を効果的に抑制し、
従つてう蝕の発生、歯周疾患を良好に防止する。
次に実験例を示し、本発明の効果を具体的に説
明する。
実験例
良姜の乾燥粉末80gにメタノール300mlを加え、
70〜80℃の水浴上で冷却管を付けて1時間還流し
た。次いでメタノール画分を別し、残渣に新し
いメタノールを加え、同様の還流を2回繰り返
し、計3回の抽出操作を行なつた。メタノール抽
出液はまとめて減圧濃縮し、暗褐色のペースト
5.7gを得た。
第1表に示す他の植物についても上記と同様に
操作し、各植物のメタノール抽出物を得た。
次に、上記抽出物を試験試料とし、下記反応付
着法によりその歯垢形成抑制の度合を評価した。
反応付着法
1%のシヨ糖、0.01%の殺菌剤(NaN3)及び
熱殺菌したストレプトコツカス・ミユータンス
6715株の菌体を含む0.05Mリン酸緩衝液(PH7.0)
に上記試験試料を0.1%添加し、これにあらかじ
め調製しておいたストレプトコツカス・ミユータ
ンス6715株のGTFを含む菌体外酵素を加えて37
℃、16時間反応させた。反応後、水で2回洗浄
し、次いで反応系と同量の水を加え、約20秒間の
超音波処理を行なつて歯垢を均一に懸濁させた
後、フオトメーターを用いて550nmで吸光度
(濁度)を測定し、付着歯垢量を求めた。
結果を第1表に示す。なお、結果は試験試料を
添加しないコントロールの付着歯垢量を100%と
した場合の百分率で示した。
