Abstract

NEW MATERIAL:Mandelic 3-hydroxy-1,2-diazin-6-yl-thiol ester of formula I.
USE: An acylating agent for amines and hydrazines. Especially, they are very useful as an active ester in the synthesis of cephalosporins, for example, enabling easy synthesis of 7-mandelamide-3-(3-hydroxy-1,2-diazin-6-yl)tiomethyl-3-cephem-4-carboxylic acid, which is used as an antibiotic having excellent antibacterial activity against gram-positive and -negative bacteria.
PREPARATION: The reaction of 3-hydroxy-1,2-diazine-6-thiol of formula II or its reactive derivative with mandelic acid or ts derivative is effected in a solvent such as acetone at -50W100°C, preferably -20W50°C to give the compound of formula I.
