Abstract
PURPOSE: To obtain a high-purity D-penicillamine, by reacting crude D-penicillamine with acetone in the absence or presence of an organic acid to form D-isopropylidenepenicillamine, which is then hydrolyzed.
CONSTITUTION: Crude D-penicillamine is added to acetone and heated in the absence or presence of an organic acid, e.g. acetic or formic acid at 30°C to reflux temperature to dissolve the D-penicillamine. The solution is filtered, and the filtrate is cooled and concentrated to give D-isopropylidenepenicillamine, which is hydrolyzed with water in the atmosphere of N2 under refluxing for 30 min, and water is vacuum distilled to afford D-penicillamine crystals. The presence of the organic acid promotes the dissolving of D-penicillamine into acetone, thus reducing the amount of acetone.
