Claims (1)
- 【特許請求の範囲】 1. (i)ブタ核酸の試料を採取すること、および (ii)KIT遺伝子の全部または一部の複製が存在するか否かを決定するために (i)で得られた核酸を分析すること、 を含むブタ毛色ゲノタイプの決定方法。 2.ブタ核酸はゲノムDNAである請求項1記載の方法。 3.段階(ii)は、PCRサイクルの一またはそれ以上および一対の適切なプラ イマーにより、少なくともKIT遺伝子の一部を増幅することを含むものである 請求項2記載の方法。 4.PCR生産物を定量するものである請求項3記載の方法。 5.一対のプライマーは、 GAATATTGTTGCTATGGTGATCTCC CCGCTTCTGCGTGATCTTCCTG、 GG(C/T)AATCACATGAATATTGTGAA TCACCATAGCAACAATATTCTGT、 TC(A/G)TACATAGAAAGAGA(C/T)GTGACTC CCTTT(A/G)ACCAC(A/G)TAATT(A/C)GAATC、または GTGATG(A/G)T(G/T)CT(C/G)ACCTACAAATA GTCTATGTAAACATAATTGTTTCC である請求項3または4記載の方法。 6.一対のプライマーは、KIT遺伝子の境界に存在する特異領域および複製領 域をハイブリッド形成するものである請求項3または4記載の方法。 7.決定は、内部標準との比較を含むものである請求項3〜6のいずれ か一に記載の方法。 8.内部標準は、一対の適切なプライマーを用いたPCRサイクルの一またはそ れ以上により、少なくとも別のヌタレオチド配列の一部を増幅することによって 生成されるものである請求項7記載の方法。 9.内部標準PCR生産物を定量するものである請求項8記載の方法。 10.内部標準PCR生産物は、少なくとも筋カルシウム放出チャンネル(CR C)遺伝子の配列の一部を増幅することにより生じるものである請求項8または 9記載の方法。 11.下記一対のプライマーは、内部標準PCR反応のために用いられるもので ある請求項10記載の方法。 CTGGATGTCCTGTGTTCCCTGT および AGGTTTGTCTGCAGCAGAAGCTC 12.下記一対のプライマーは、内部標準PCR反応のために用いられるもので ある請求項10記載の方法。 GATGAACTTTGAGGTCCCTGAGGAG および TTTCTTCTGAGAATGCCGAAGATCTG 13.内部標準PCR反応は、KIT配列の増幅と同時に進行するものである請 求項8〜12のいずれか一に記載の方法。 14.KIT領域のコピー:内部標準領域のコピー、の比率を決定するものであ る請求項8〜13のいずれか一に記載の方法。 15.さらに下記段階を含む請求項1〜14のいずれか一に記載の方法。 (iii)任意の存在する複製がIまたはIPいずれの存在によるものであるかを決 定すること 16.少なくとも複製領域の1つにおいて欠失が存在するか否かを決定するもの である請求項14記載の方法。 17.非欠失領域のコピー:欠失領域のコピー、の比率を決定するもの である請求項16記載の方法。 18. (i)ブタゲノムDNAの試料を採取すること、 (ii)(i)で得られたゲノムDNAを一またはそれ以上の適切なプライマーで ハイブリッド形成すること、 (iii)(ii)で得られたハイブリッド形成された核酸を用いて一またはそれ以 上のPCRサイクルを行わせること、および (iv)PCR反応生産物の量を決定すること、 を含むブタ毛色ゲノタイプの決定方法。 19.さらに下記段階を含む請求項18記載の方法。 (v)任意の存在する複製がIまたはIPのいずれの存在によるものかを決定す ること 20.少なくとも複製領域の一において欠失が存在するか否かを決定するもので ある請求項19記載の方法。 21.クレーム6〜14の特徴の一またはそれ以上により修正された請求項18 〜20のいずれか一に記載の方法。 22. (i)ブタKIT蛋白質の試料を採取すること、および (ii)KIT遺伝子の全部または一部の複製が存在するか否かを決定するために (i)で得られた蛋白質を分析すること、 を含むブタ毛色ゲノタイプの決定方法。 23.段階(ii)は、蛋白質と一またはそれ以上の抗体とを反応させることによ り行われるものである請求項22記載の方法。 24.方法は、ELISAである請求項23記載の方法。 25.KIT配列の全部または一部の複製の存在を指示することのできる一また はそれ以上の試薬を含むブタ毛色ゲノタイプ決定用キット。 26.キットは、ブタゲノムDNAの試料とともに使用されるのに適合したもの である請求項25記載のキット。 27.少なくとも一対のプライマーとともにPCRを行うための試薬を含む請求 項25または26記載のキット。 28.一対のプライマーは、 GAATATTGTTGCTATGGTGATCTCC CCGCTTCTGCGTGATCTTCCTG、 GG(C/T)AATCACATGAATATTGTGAA TCACCATAGCAACAATATTCTGT、 TC(A/G)TACATAGAAAGAGA(C/T)GTGACTC CCTTT(A/G)ACCAC(A/G)TAATT(A/C)GAATC、または GTGATG(A/G)T(G/T)CT(C/G)ACCTACAAATA GTCTATGTAAACATAATTGTTTCC である請求項27記載のキット。 29.一対のプライマーは、KIT遺伝子の境界に存在する特異領域および複製 領域をハイブリッド形成するものである請求項27記載の方法。 30.さらに、一またはそれ以上のPCRサイクルにより、少なくとも別のヌク レオチド配列の一部を増幅することのできる、第二の一対のプライマーを含む請 求項26〜29のいずれか一に記載のキット。 31.第二の一対のプライマーは、少なくとも筋カルシウム放出チャンネル(C RC)遺伝子の配列の一部をハイブリッド形成するものである請求項30記載の キット。 32.第二の一対のプライマーは下記配列を有するものである請求項31記載の キット。 CTGGATGTCCTGTGTTCCCTGT および AGGTTTGTCTGCAGCAGAAGCTC 33.第二の一対のプライマーは、少なくともブタインターフェロン−β遺伝子 の配列の一部をハイブリッド形成するものである請求項30記載のキット 34.第二の一対のプライマーは下記配列を有するものである請求項33記載の キット。 GATGAACTTTGAGGTCCCTGAGGAG および TTTCTTCTGAGAATGCCGAAGATCTG 35.KIT蛋白質と反応することのできる一またはそれ以上の抗体を含む請求 項25記載のキット。 36. (i)ブタ核酸の試料を採取すること、および (ii)KIT遺伝子配列において欠失が存在するか否かを同定するために(i) で得られた核酸を分析すること を含むブタ毛色ゲノタイプの決定方法。 37.ブタ核酸はゲノムDNAである請求項36記載の方法。 38.段階(ii)は、一またはそれ以上のPCRサイクルおよび一対の適切なプ ライマーにより、少なくともKIT遺伝子の一部を増殖することを含むものであ る請求項36または37記載の方法。 39.一対のプライマーは、 TGTGGGAGCTCTTCTCTTTAGG CCAGCAGGACAATGGGAACATCT、または GAAAGTGA(C/T)GTCTGGTCCTAT(C/G)GGAT AGCCTTCCTTGATCATCTTGTAG である請求項38記載の方法。 40.方法はIとiの識別に用いられるものである請求項36〜39のいずれか 一に記載の方法。 41.KIT配列において欠失が存在するか否かを指示することのできる一また はそれ以上の試薬を含むブタ毛色ゲノタイプの決定用キット。 42.キットは、ブタゲノムDNAの試料とともに使用するのに適合したもので ある請求項41記載のキット。 43.少なくとも一対のプライマーとともにPCRを行うための試薬を含む請求 項41または42記載のキット。 44.一対のプライマーは、 TGTGGGAGCTCTTCTCTTTAGG CCAGCAGGACAATGGGAACATCT、または GAAAGTGA(C/T)GTCTGGTCCTAT(C/G)GGAT AGCCTTCCTTGATCATCTTGTAG である請求項43記載のキット。
