Claims (1)
- 【特許請求の範囲】 1.形質転換された宿主細胞であって、 リプレッサーによる結合に感受性のオペレーターを含むプラスミド、 該リプレッサーをコードする第1の染色体遺伝子、および 該オペレーターに機能的に結合しており、そして細胞増殖に必須な第2の染色体 遺伝子を含み、 ここで、該プラスミドは、該細胞中に該リプレッサーを滴定するのに充分な数で 存在し、その結果該必須な遺伝子が発現され、これにより細胞増殖を可能にする 、形質転換された宿主細胞。 2.前記リプレッサーが、E.coli lacリプレッサー、γリプレッサー、E.coli trpリプレッサー、E.coli galRリプレッサー、E.coli araCリプレッサー、E .coli tetリプレッサー、およびE.coli ArgRNVリプレッサーからなるの群から 選択される、請求項1に記載の宿主細胞。 3.前記宿主細胞が、哺乳動物細胞または昆虫細胞のような動物細胞、植物細胞 、酵母細胞のような菌類あるいは細菌細胞である、請求項1または2に記載の宿 主細胞。 4.前記細胞が細菌細胞である、請求項1または2に記載の宿主細胞。 5.前記細胞がグラム陰性細菌細胞である、請求項4に記載の宿主細胞。 6.前記細胞がE.coliである、請求項5に記載の宿主細胞。 7.前記細胞がSalmonellaである、請求項5に記載の宿主細胞。 8.前記細胞がグラム陽性細菌細胞である、請求項4に記載の宿主細胞。 9.前記細胞がBacillusである、請求項8に記載の宿主細胞。 10.前記細胞が酵母細胞である、請求項1または2に記載の宿主細胞。 11.前記細胞が哺乳動物細胞である、請求項1または2に記載の宿主細胞。 12.前記プラスミドが、宿主細胞あたり該プラスミドの約10〜50コピーの複製 を可能にする複製起点を含む、前記請求項のいずれか1項に記載の宿主細胞。 13.前記プラスミドが、宿主細胞あたり該プラスミドの約100〜200コピーの複 製を可能にする複製起点を含む、請求項1〜11のいずれか1項に記載の宿主細 胞。 14.前記プラスミドがpBR322である、請求項12に記載の宿主細胞。 15.前記プラスミドがpUCである、請求項13に記載の宿主細胞。 16.前記プラスミドが、目的の遺伝子の挿入のためのクローニング部位を含む 、前記請求項のいずれか1項に記載の宿主細胞。 17.前記プラスミドが、哺乳動物細胞における発現のための制御配列に作動可 能に結合した目的の遺伝子をさらに含む、請求項16に記載の宿主細胞。 18.前記プラスミドが、本質的に、リプレッサーによる結合に感受性のオペレ ーター、複製起点、および目的の遺伝子の挿入のためのクローニング部位からな る、前記請求項のいずれか1項に記載の宿主細胞。 19.前記プラスミドが、約1000bpの長さである、請求項18に記載の宿主細胞 。 20.宿主細胞中のプラスミドを維持する方法であって、以下の工程: 請求項1〜19のいずれか1項に記載の形質転換細胞を、該細胞が増殖し得る のに充分な時間および条件下で培養する工程、 を包含する、方法。 21.プラスミドDNAを産生する方法であって、以下の工程: 請求項1〜19のいずれか1項に記載の形質転換細胞を、該細胞が増殖し得る のに充分な時間および条件下で培養する工程、ならびに プラスミドDNAを該培養細胞から単離する工程、 を包含する、方法。 22.組換えタンパク質を産生する方法であって、以下の工程: 請求項17〜19のいずれか1項に記載の形質転換細胞を、該細胞が増殖し得 るのに充分な時間および条件下で培養する工程、ならびに 該組換えタンパク質を該培養細胞から単離する工程、 を包含する、方法。
