Claims (1)
- 【特許請求の範囲】 1.脂肪族ケトンドナーとアルデヒドアクセプター間のアルドール付加反応を触 媒する抗体分子又は抗体結合部位部分を含む分子であって、ε−アミノ基を有す るリシンをもち、1,3−ジケトンハプテンと前記触媒抗体のリシンのε−アミノ 基間の複合体の形成によって1,3−ジケトンハプテンによる阻害を受け、前記複 合体が安定な共有結合ビニローグアミド、複合エナミン及びシッフ塩基からなる 群より選ばれることを特徴とする、前記抗体分子又は抗体結合部位部分を含む分 子。 2.クラム・フェルキン方向及びアンチクラム・フェルキン方向のアルドール付 加反応のジアステレオ面の選択性を制御する、請求項1記載の抗体分子又は抗体 結合部位部分を含む分子。 3.前記脂肪族ケトンドナーが下記構造によって表される群より選ばれる、請求 項1記載の抗体分子又は抗体結合部位部分を含む分子。 4.前記アルデヒドアクセプターが下記構造によって表される群より選ばれる、 請求項1記載の抗体分子又は抗体結合部位部分を含む分子。 5.ATCC受託番号 HB12005を有するハイブリドーマ38C2によって分泌される、請 求項1記載の分子。 6.ATCC受託番号 HB12004を有するハイブリドーマ 33F12によって分泌される、 請求項1記載の分子。 7.培養液中で培養した場合、脂肪族ケトンドナーとアルデヒドアクセプター間 のアルドール付加反応を触媒し、かつε−アミノ基を有するリシンをもち、1,3 −ジケトンハプテンと前記触媒抗体のリシンのε−アミノ基間の複合体の形成に よって1,3−ジケトンハプテンによる阻害を受け、前記複合体が安定な共有結合 ビニローグアミド、複合エナミン及びシッフ塩基からなる群より選ばれることを 特徴とする前記抗体分子又は抗体結合部位部分を含む分子を産生する細胞。 8.該培養液中に前記モノクローナル抗体分子又は抗体結合部位部分を含む分子 を更に分泌する、請求項7記載の細胞。 9.ハイブリドーマ細胞である、請求項8記載の細胞。 10.ATCC受託番号 HB12005を有するハイブリドーマ38C2である、請求項9記載の ハイブリドーマ細胞。 11.ATCC受託番号 HB12004を有するハイブリドーマ 33F12である、請求項9記載 のハイブリドーマ細胞。 12.脂肪族ケトンドナーとアルデヒドアクセプター間のアルドール付加反応を触 媒する方法であって、 工程A:請求項1記載のモノクローナル抗体分子又は抗体結合部位部分を含む 分子の触媒的に有効な量と、前記脂肪族ケトンドナーと前記アルデヒドアクセプ ターとを水性培養液中で混合して反応混合液を形成する工程;及び 工程B:前記工程Aの前記反応混合液を、前記抗体分子又は抗体結合部位部分 を含む分子が前記脂肪族ケトンドナーと前記アルデヒドアクセプター間の前記ア ルドール付加反応を触媒するのに十分な時間維持する工程 を含む、前記方法。 13.前記抗体分子又はその抗体結合部位部分を含む分子がATCC受託番号 HB12005 を有するハイブリドーマ38C2によって分泌される、請求項12記載の方法。 14.前記抗体分子又はその抗体結合部位部分を含む分子がATCC受託番号 HB12004 を有するハイブリドーマ 33F12によって分泌される、請求項12記載の方法。 15.アルドール付加反応を行う方法であって、 工程A:脂肪族ケトンドナー、アルデヒドアクセプター、及び該脂肪族ケトン ドナーと反応するε−アミノ基を有するリシンを含むモノクローナル抗体又は前 記モノクローナル抗体のパラトープ含有部分の触媒的に有効な量を混合すること により反応混合液を形成してエナミン中間体を形成する工程;及び 工程B:前記反応混合液を生物学的反応条件下で前記工程Aのエナミン中間体 が該アルデヒドアクセプターと反応するのに十分な時間維持してアルドール付加 生成物を形成する工程 を含む、前記方法。 16.培養液中で培養した場合、脂肪族ドナーとアルデヒドアクセプター間のアル ドール付加反応を触媒する抗体分子又は抗体結合部位部分を含む分子を産生する 細胞の調製方法であって、 工程A:1,3−ジケトンハプテンを含む免疫原を動物に免疫する工程; 工程B:前記動物が前記ハプテンリガンドと免疫反応する抗体を分泌するのに 十分な時間前記動物を維持する工程; 工程C:前記工程(B)の前記維持し免疫した動物の抗体産生細胞からの抗体 分子又は抗体結合部位部分を含む分子をコード化する遺伝子をホスト細胞に移入 して少なくとも2種の供給源から遺伝子を含むハイブリッド細胞を形成し、形成 したハイブリッド細胞が、(i)培養した場合に前記移入した遺伝子から抗体分 子又は抗体結合部位部分を含む分子を産生しかつ(ii)ほぼ無制限に培養される 工程; 工程D:該ハイブリッド細胞を適切な培養液中で該ハイブリッド細胞が抗体分 子又は抗体結合部位部分を含む分子を産生するのに十分な時間培養する工程; 工程E:該培養したハイブリッド細胞から抗体分子又は抗体結合部位部分を含 む分子を回収する工程; 工程F:該アルドール付加反応を触媒する得られた抗体分子又は抗体結合部位 部分を含む分子をスクリーニングする工程;及び 工程G:該脂肪族ドナーと該アルデヒドアクセプター間のアルドール付加反応 を触媒する抗体分子又は抗体結合部位部分を含む分子を産生する前記同定ハイ ブリッド細胞のクローンを増殖する工程 を含む、前記方法。 17.前記工程Cで形成された細胞がハイブリドーマ細胞である、請求項12記載の 方法。
