Description
Claims (1)
- 【特許請求の範囲】 1. 液体生物試料を受け入れるための試料セル、該試料セルは試料室および 該液体生物試料を該試料室内へ加えるための試料入り口を持っている;および 乾燥核酸増幅試薬および乾燥均質核酸アッセイ試薬、該試薬は該液体生 物試料と反応させるため該試料室の内部に接着されている: を含む液体生物試料に対して均質核酸増幅および核酸アッセイを実施するための 装置。 2. 該乾燥核酸増幅試薬および該均質核酸アッセイ試薬が該試料室の内部表 面に接着された単一スポットの形で提供される請求項1に記載の装置。 3. 該液体生物試料が該試料室に加えられた後に該試料入り口を封じるため に該試料室に付着可能なシーリング部材をさらに含む請求項1に記載の装置。 4. 該均質核酸アッセイ試薬が蛍光偏光アッセイ試薬を含み、および該試料 セルの少なくとも一部が該試料室の液体生物試料中で起こっている蛍光偏光反応 の外部検出を可能にするように十分に光透過性である請求項1に記載の装置。 5. 該均質核酸アッセイ試薬が蛍光偏光アッセイ試薬を含み; 該試料セルの少なくとも一部が該試料室の液体生物試料中で起こってい る蛍光偏光反応の外部検出を可能にするように十分に光透過性であり;および 該シーリング部材が光透過性物質で作られておりおよび該試料室の光透 過性部分に付着可能である請求項1に記載の装置。 6. 該シーリング部材の少なくとも一つおよび該試料室の該光透過性部分が 光偏光物質で作られている請求項5に記載の装置。 7. 該試料室がさらに該試料室内への該液体生物試料の添加の間に該試料室 から置き換えられるべき空気のための通気孔を含む請求項1に記載の装置。 8. 該試料セルが該試料室の底壁を形成する下層、該試料室の側壁を形成す る隙間を持つ中間層および該試料室の上壁を形成する上層を持つ実質的に平坦な 積層カード様部材に形成され、該試料入り口は該上層に形成されており;および 該試料セルが該カード様部材の長さおよび幅に渡って二次元的配列で配 置された多数の実質的に同一の試料セルの一つである請求項1に記載の装置。 9. 液体生物試料が該試料室へ加えられた後、該試料入り口を封じるための シーリング部材をさらに含み、該シーリング部材が柔軟な物質の層の下面に保持 された感圧接着剤により該薄板の上層へ付着できる柔軟な層を含む請求項8に記 載の装置。 10. 液体生物試料を受け入れるための少なくとも一つの試料セルを持つ実質 的に平坦なカード様部材、該試料セルは試料室、該試料室へ該液体生物試料を加 えるための試料入り口、該試料室内への該液体生物試料の添加の間に該試料室か ら置き換えられるべき空気のための通気孔および該液体生物試料と反応させるた め該試料室の内部表面に付着された乾燥試薬を持っており;および 該試料室へ該液体生物試料が加えられた後、該試料入り口および該通気 孔を封じるための該カード様部材に付着可能なシーリング部材から成る液体生物 試料に生物学的過程を実施するための装置。 11. 該カード様部材が向かい合う上および下表面を持ち、および該試料入り 口および該通気孔が両方とも該カード様部材の上表面に形成されている請求項1 0に記載の装置。 12. 該カード様部材が該試料室の底壁を形成する下層、該試料室の側壁を形 成する隙間を持つ中間層および該試料室の上壁を形成する上層を持つ積層を含み 、該試料入り口および該通気孔が該上層に形成されている請求項10に記載の装 置。 13. 該シーリング部材が柔軟な物質の層の下面に保持された感圧接着剤によ り該薄板の上層へ付着できる柔軟な層を含む請求項12に記載の装置。 14. 該乾燥試薬が該液体生物試料と反応して光学指標を産生し、および該光 学指標の外部的検出を可能にするために該試料室上の領域で該シーリング部材お よび該上層が十分に光透過性である請求項13に記載の装置。 15. 該乾燥試薬が核酸蛍光偏光アッセイ試薬を含んでいる請求項14に記載 の装置。 16. 該試料セルが一般的に該カード様部材の面に平行な方向に伸びた形を持 ち、該試料入り口が該試料セルの一つの末端に位置しおよび該通気孔がその反対 の末端に位置している請求項10に記載の装置。 17. 該試料セルが該カード様部材の長さおよび幅に渡って二次元的配列で配 置された多数の実質的に同一の試料セルの一つである請求項10に記載の装置。 18. 液体生物試料を受け入れるための試料セル、該試料セルは試料室および 該液体生物試料を該試料室内へ加えるための試料入り口を持っている;および 該液体生物試料と反応させるため該試料室の内部表面に接着されている 乾燥核酸蛍光偏光アッセイ試薬から成る液体生物試料に対して核酸蛍光偏光アッ セイを実施するための装置であって; ここで該試料セルの少なくとも一部は該試料室の液体生物試料中で起こ っている蛍光偏光反応の外部検出を容易にするために十分に光透過性の光偏光物 質から作られている。 19. 該試料セルの該部分が該液体生物試料が該試料室に加えられた後に該試 料入り口を封じるために該試料室に付着可能なシーリング部材を含む請求項18 に記載の装置。 20. 該試料セルが該試料室の底壁を形成する下層、該試料室の側壁を形成す る隙間を持つ中間層および該試料室の上壁を形成する上層を持つ実質的に平坦な 積層カード様部材に形成され、該試料入り口は該上層に形成されており;および 該試料セルの該部分が該積層カード様部材の上層を含む請求項18に記 載の装置。 21. 該試料セルが該カード様部材の長さおよび幅に渡って二次元的配列で配 置された多数の実質的に同一の試料セルの一つである請求項20に記載の装置。 22. 該液体生物試料と反応させるために該試料室の内部表面に接着された乾 燥核酸増幅試薬をさらに含む請求項18に記載の装置。 23. 液体生物試料を光透過性部分を持つ試料セル内へ導入し; 該液体生物試料を乾燥核酸増幅試薬および均質核酸蛍光偏光アッセイ試 薬と該試料セル内で接触させ; 該試料セルを封じ; 該液体生物試料と乾燥核酸増幅試薬および均質核酸蛍光偏光アッセイ試 薬を反応させるために該試料セルをインキュベートし;そして 該試料セルの該光透過性部分を通して該液体生物試料中の蛍光偏光を検 出する工程からなる、液体生物試料に対する統合された核酸増幅および均質核酸 蛍光偏光アッセイの実施法。 24. 該液体生物試料中の蛍光偏光を検出する工程が該試料セルの該光透過性 部分を通して偏光光を向けることを含む請求項23に記載の方法。 25. 該試料セルの該光透過性部分が光偏光物質で作られ、および該液体生物 試料中の蛍光偏光を検出する工程が該光透過性部分を通して非偏光光を向けるこ とを含む請求項23に記載の方法。 26. 乾燥核酸増幅試薬を含む試料セルを核酸増幅に適した温度まで前もって 加熱し; 該前もって加熱した試料セル内へ液体生物試料を導入して該液体生物試 料と該乾燥核酸増幅試薬を接触させ; 該液体生物試料の温度を該前もって加熱した試料セルの温度と平衡化し ;そして 該温度平衡が実質的に完了した後、該試料セルで該核酸増幅反応を開始 させる工程からなる、液体生物試料に対する核酸増幅反応の実施法。 27. 該液体生物試料の温度の該前もって加熱した試料セルの温度への平衡化 工程が該試料セルおよび該液体生物試料間の熱輸送を促進するため該試料セル中 で該液体生物試料の薄層を形成させることを含む請求項26に記載の方法。 28. 該温度平衡化が起こるのに必要とされる時間が該液体生物試料が該乾燥 核酸増幅試薬を溶解させるのに必要な時間と実質的に同じである請求項26に記 載の方法。
