Claims (1)
- 【特許請求の範囲】 1. セファロスポリンCまたはその誘導体を、7−アミノセファロスポラン 酸またはその対応する誘導体に変換する一段階変換方法であって、前記セファロ スポリンCまたはその誘導体を、Pseudomonas vesicularis B965、または、何 れかのセファロスポリンCアミドヒドロラーゼを産生するか、或いは潜在的に産 生するこれらの継代物から誘導されるセファロスポリンCアミドヒドロラーゼで 処理することを具備した方法。 2. セファロスポリンCまたはその誘導体を、7−アミノセファロスポラン 酸またはその対応する誘導体に変換する一段階変換方法であって、前記セファロ スポリンCまたはその誘導体を、Pseudomonas vesicularis B965、または、何 れかのセファロスポリンCアミドヒドロラーゼを産生するか、或いは潜在的に産 生するこれらの継代物のDNAの発現から誘導されるセファロスポリンCアミド ヒドロラーゼで処理することを具備した方法。 3. セファロスポリンCまたはその誘導体を、7−アミノセファロスポラン 酸またはその対応する誘導体に変換する一段階変換方法であって、前記セファロ スポリンCまたはその誘導体を、配列識別番号1のDNA配列の一部または全部 を含有するDNA;またはこれらの遺伝コードまたは対立または機能性変異の結 果として生じるDNA配列変性体の発現から誘導されるセファロスポリンCアミ ドヒドロラーゼで処理することを具備した方法。 4. 請求の範囲第3項または4項に記載の方法であって、セファロスポリン Cアミドヒドロラーゼが、配列識別番号1の配列の一部または全部を含有する組 換えDNA分子;またはこれらの遺伝コードまたは対立または機能性変異の結果 として生じるDNA配列変性体の発現から誘導される方法。 5. 請求の範囲第1項から第4項の何れかで調製された7−アミノセファロ スポラン酸を用いた、セファロチン、セファロリジンおよびセフロキシンを含有 する群から選択される半合成セファロスポリンの製造方法。 6. 請求の範囲第1項から第4項の何れかに記載の方法であって、セファロ スポリンCアミドヒドロラーゼが固定化されている方法。 7. 真核生物または原核生物ホスト内でセファロスポリンCアミドヒドロラ ーゼを発現する方法であって、 a)ホスト細胞を、 (i)ホスト細胞内で機能する発現コントロール配列、および (ii)発現コントロール配列に外科的に結合されたセファロスポリンCアミ ドヒドロラーゼをコードする組換えDNA配列 を含有する組換えDNA発現ベクターで形質転換すること、 b)ステップa)で形質転換されたホスト細胞を、セファロスポリンCアミドヒ ドロラーゼ活性の発現を可能にする条件下で培養すること、 を具備した方法。 8. 真核生物または原核生物ホスト内でセファロスポリンCアミドヒドロラ ーゼを発現する方法であって、組換えDNA配列が、配列識別番号1またはこれ らの対立変異若しくは他の機能的に等価なこれらの変異のアミノ酸配列を有する セファロスポリンCアミドヒドロラーゼポリペプチドをコードする方法。 9. 実質的に精製された形態で、Pseudomonas vesicularis B965 から誘 導されるセファロスポリンCアミドヒドロラーゼ。 10. 固定化された請求の範囲第9項に記載のセファロスポリンCアミドヒ ドロラーゼ。 11. 7−アミノセファロスポラン酸の製造方法における請求の範囲第9項 または第10項に記載のセファロスポリンCアミドヒドロラーゼの使用。
