Claims (1)
- 【特許請求の範囲】 1.IgG、IgM、IgA、IgDおよびIgEのクラスの一つまたはいくつ かからの免疫グロブリンのH鎖のFd部分に対して特異的であり、少なくとも実 質的に、これらの免疫グロブリンの抗原結合能を遮蔽する、いくつかの異なる抗 体および/または抗体フラグメントで構成される組成物。 2.免疫グロブリンGのH鎖のFd部分に対して特異的である、請求項1に記載 の組成物。 3.遮蔽すべき抗体分子に対して5倍モル過剰で、抗体分子の抗原結合能を少な くとも50%阻害する、請求項1または2に記載の組成物。 4.遮蔽すべき抗体分子に対して5倍モル過剰で、抗体分子の抗原結合能を少な くとも90%阻害する、請求項3に記載の組成物。 5.第一の免疫グロブリンクラスに対して特異的であり、他の免疫グロブリンク ラスとの最大交差反応性が、第一の免疫グロブリンクラスに対する反応性に対し て10-2である、請求項1〜4のいずれか一項に記載の組成物。 6.IgGに対して特異的であり、IgMとの最大交差反応性がIgGに対する 反応性に対して10-2である、請求項5に記載の組成物。 7.最大交差反応性が10-3である、請求項6に記載の組成物。 8.一価の抗体フラグメントで構成される、請求項1〜7のいずれか一項に記載 の組成物。 9.Fabフラグメントで構成される、請求項1〜8のいずれか一項に記載の組 成物。 10.ヒト免疫グロブリンに対して特異的であり、ヒト以外の免疫グロブリンとの 最大交差反応性がヒト免疫グロブリンに対する反応性に対して10-2である、請求 項1〜9のいずれか一項に記載の組成物。 11.ポリクローナル抗体および/または抗体フラグメントで構成される、請求項 1〜10のいずれか一項に記載の組成物。 12.(a)実験動物を第一の免疫グロブリンクラスからの免疫グロブリンのH鎖 のFd部分を含む免疫原で免疫感作し; (b)ポリクローナル抗血清を実験動物から得て; (c)ポリクローナル抗血清を必要により、分解によって一価の抗体フラグメ ントに変換し; (d)抗血清または一価の抗体フラグメントを、第一の免疫グロブリンクラス のH鎖のFd部分に対して特異的である抗体および/または抗体フラグメントの 選択を可能にする一工程または数工程の免疫収着に供し;そして (e)第一の免疫グロブリンクラスのFd部分に対する特異性を有する抗体ま たは抗体フラグメント組成物を単離する、 請求項1〜11のいずれか一項に記載の組成物の作製方法。 13.免疫グロブリンのFc成分を含まない免疫原を使用する、請求項12に記載の 方法。 14.免疫収着工程が、(i)第一の免疫グロブリンクラスのH鎖のFd部分を含 む抗原に対する少なくとも一つの正の免疫収着、および組成物の結合成分の単離 ;ならびに (ii)第一の免疫グロブリンクラスとは異なる免疫グロブリンクラスから選択 される抗原またはその成分に対する少なくとも一つの負の免疫収着、および組成 物の非結合成分の単離; を含む、請求項12または13に記載の方法。 15.免疫収着工程がさらに、(iii)正の免疫収着工程(i)における免疫グロ ブリンの種とは異なる種の第一の免疫グロブリンクラスから選択される抗原また はその成分に対する少なくとも一つの負の免疫収着、および組成物の非結合成分 の単離を含む、請求項14に記載の方法。 16.正の免疫収着(i)を第一の免疫グロブリンクラスのヒト免疫グロブリンに 対して行い、負の免疫収着(iii)を第一の免疫グロブリンクラスのヒト以外の種 の免疫グロブリンに対して行う、請求項15に記載の方法。 17.免疫収着工程を固定化抗原を使用して行う、請求項12〜16のいずれか一項に 記載の方法。 18.免疫原は、免疫グロブリンGのH鎖のFd部分を含むものを使用し、正の免 疫収着(i)は、免疫グロブリンGまたはH鎖のFd部分を含むそのフラグメ ントに対して行い、結合成分を単離する、請求項12〜17のいずれか一項に記載の 方法。 19.IgM、IgD、IgEおよび/またはIgAあるいはそれらの成分に対す る負の免疫収着(ii)を行い、非結合成分を単離する、請求項18に記載の方法。 20.負の免疫収着をIgMまたはその成分に対して行う、請求項19に記載の方法 。 21.ヒト以外の免疫グロブリンGに対する負の免疫収着(iii)を行い、非結合 成分を単離する、請求項18〜20のいずれか一項に記載の方法。 22.ポリクローナル抗血清を、パパインによる分解によりFabフラグメントに 変換する、請求項12〜21のいずれか一項に記載の方法。 23.IgG、IgM、IgA、IgDおよび/またはIgEの抗体の選択的クラ ス特異的測定に対するイムノアッセイにおける、他のクラスの抗体によって引き 起こされる干渉を抑制するための、請求項1〜11のいずれか一項に記載の組成物 の使用。 24.一工程のイムノアッセイにおける請求項23に記載の使用。 25.他の免疫グロブリンクラスの抗体も含むサンプル液体における、一つまたは いくつかの選択した免疫グロブリンクラスからの特異的抗体の選択的免疫学的検 出方法であって、サンプル液体を請求項1〜11のいずれか一項に記載の組成物と 前インキュベートして、他の免疫グロブリンクラスによって引き起こされる干渉 を抑制し、次いで、サンプル液体における選択した検出すべきクラスの抗体の有 無および/または量を測定する方法。 26.組成物を、サンプル液体に存在する他の免疫グロブリンクラスの抗体に対し て活性成分が少なくとも5倍モル過剰となる量で添加する、請求項25に記載の方 法。 27.組成物を10倍〜1000倍モル過剰で添加する、請求項26に記載の方法。 28.前インキュベーションを、5〜60分間行う、請求項25〜27のいずれか一項に 記載の方法。 29.測定を、反応性固相ならびに検出すべき抗体に結合可能な2つのレセプター R1およびR2(R1は、固相に結合しているか、固相に結合可能であり、R2は、 直接または間接的に標識されている。)の存在下で、不均一イムノアッセイの原 理に従って行い、検出すべき抗体を、固相および/またはインキュベーション液 体中の標識を測定することにより測定する、請求項25〜28のいずれか一項に記載 の方法。 30.検出すべき抗体と特異的に反応する抗原および固相結合基のコンジュゲート をレセプターR1として使用し、検出すべき抗体と特異的に反応する抗原を、直 接または間接的に標識されているレセプターR2として使用する、請求項29に記 載の方法。 31.検出すべき抗体と特異的に反応する抗原および固相結合基のコンジュゲート をレセプターR1として使用し、選択した抗体クラスを認識し、直接または間接 的に標識されている抗体または対応する抗体フラグメントをレセプターR2とし て使用する、請求項29に記載の方法。 32.選択した抗体クラスを認識する抗体または対応する抗体フラグメントおよび 固相結合基のコンジュゲートをレセプターR1として使用し、直接または間接的 に標識され、検出すべき抗体と特異的に反応する抗原をレセプターR2として使 用する、請求項29に記載の方法。 33.ストレプトアビジンまたはアビジンで被覆した反応性固相およびビオチニル 化レセプターR1を使用する、請求項29〜32のいずれか一項に記載の方法。 34.発光金属複合体を標識として使用し、標識を電気化学発光によって測定する 、請求項29〜33のいずれか一項に記載の方法。 35.酵素を標識として使用し、標識を酵素反応の検出によって測定する、請求項 29〜33のいずれか一項に記載の方法。 36.請求項1〜11のいずれか一項に記載の組成物を含む、一つまたはいくつかの 選択した免疫グロブリンクラスから特異的抗体を選択的免疫学的に検出するため の試薬。
