Claims (1)
- 【特許請求の範囲】 1. アルファウイルスRNAゲノムの少なくとも一部、および翻訳増強活性 をもつアルファウイルス塩基配列の下流に位置に生物活性をもつ物質をコードす る異種RNAを含む、自己複製および転写能をもつ組み換えアルファウイルスR NA分子であって、該翻訳増強塩基配列が、完全なアルファウイルス・キャプシ ド遺伝子または該遺伝子の5’部分を含んでなる組み換えRNA分子。 2. 異種RNAが、該翻訳増強塩基配列の後のアルファウイルスサブゲノム 領域中に挿入される、請求の範囲1の組み換えRNA分子であって、細胞中に導 入された後のRNA分子が、その分子の5’領域によってコードされたアルファ ウイルス・レプリカーゼを用いて複製され、そしてアルファウイルスサブゲノム プロモーターによってmRNAに転写され、そのmRNAから、異種遺伝子産物 が、該遺伝子の5’部分によってコードされる完全なキャプシドタンパク質また はキャプシドタンパク質のアミノ末端部分をもつ融合産物として、その細胞で発 現される、組み換えRNA分子。 3. アルファウイルスがセムリキ森林熱ウイルスである、請求の範囲1また は2の組み換えRNA分子。 4. 翻訳増強塩基配列が、配列 をもつ、キャプシド遺伝子の最初の81個、好ましくは、最初の102 個の塩基を含んでなる、請求の範囲3の組み換えRNA分子。 5. 異種RNAが、該融合産物として発現され、それが、キャプシドタンパ ク質の自己分解的プロテアーゼ活性によって、その融合部位で切断される、請求 の範囲2,3または4の組み換えRNA分子。 6. 異種RNA配列が、抗原決定基を含むタンパク質またはポリペプチドを コードしており、該タンパク質またはポリペプチドが、免疫原性または抗原性で ある、請求の範囲1〜5のいずれかの組み換えRNA分子。 7. 異種RNAが、治療活性をもつタンパク質またはポリペプチドをコード している、請求の範囲1〜5のいずれかの組み換えRNA分子。 8. 請求の範囲1〜7のいずれかの組み換えRNA分子に相補的なDNA配 列を含む、DNA分子。 9. さらに、細胞中で組み換えRNA分子を転写するためのプロモーターを コードするDNA配列、そしてさらに、E.コリ(E.coli)におけるプラ スミド増殖に必要な特性をコードするDNA配列を含む、請求の範囲8のDNA 分子。 10.プロモーターがSP6である、請求の範囲9のDNA分子。 11.前記請求の範囲のいずれかの組み換えRNAまたはDNA分子を含む形 質転換細胞。 12.動物またはヒトの細胞である、請求の範囲11の細胞。 13.該RNAまたはDNA分子により安定に形質転換される、請求の範囲1 1または12の細胞を含む細胞系。 14.生物活性物質をコードする異種RNA配列を発現する方法であって、請 求の範囲1〜7のいずれかの組み換えRNA、または請求の範囲 8、9または10の組み換えDNAによる細胞の形質転換、そして続く、異種R NA配列の発現を可能にする条件下での形質転換細胞の増殖を含む方法。 15.形質転換がトランスフェクションによって達成される請求の範囲14の 方法。 16.RNAが、治療活性、または免疫原性もしくは抗原性のような予防活性 をもつタンパク質、ポリペプチドまたはペプチドをコードする、請求の範囲14 または15の方法。 17.発現産物が、細胞から分離され、場合によって精製される、請求の範囲 14〜16のいずれかの方法。 18.生物活性物質をコードする異種RNA配列を発現する方法であって、請 求の範囲1〜7のいずれかの組み換えRNA、または請求の範囲8、9または1 0の組み換えDNAにより、動物またはヒト個体において、イン・ビボで細胞を 形質転換、例えばトランスフェクションすることを含む方法。 19.野生型アルファウイルス・コートによって包まれた組み換えアルファウ イルスRNAゲノムを含む組み換えアルファウイルスを生産する方法であって、 該RNAゲノムが、該組み換えRNAゲノムと、アルファウイルス構造タンパク 質の発現能を含有するヘルパーRNAとによる細胞の同時トランスフェクション のような同時形質転換によって、請求の範囲1〜7のいずれかの組み換えRNA 分子を含み、そしてシスに働く複製シグナルであるが包膜シグナルではないアル ファウイルス構造タンパク質のコーディング配列を含み、細胞を培養し、そして 感染性組み換えアルファウイルス粒子を含む培地を回収する方法。 20.請求の範囲1〜7のいずれかの組み換えRNA分子と、アルファウイル ス構造タンパク質の発現能を含有するヘルパーRNAとを含み、そしてシスに働 く複製シグナルであるが包膜シグナルではないアルファウイルス構造タンパク質 のコーディング配列を含む形質転換細胞であって、野生型アルファウイルス・コ ートによって包まれた該組み換えRNAを含む感染性組み換えアルファウイルス 粒子を生産することができる細胞。 21.野生型アルファウイルス・コートによって包まれた請求の範囲1〜7の いずれかの組み換えRNA分子からなるゲノムを含む感染性組み換えアルファウ イルス粒子。 22.請求の範囲21の組み換えアルファウイルス粒子による、細胞培養また は動物もしくはヒト個体における細胞の感染を含む、生物活性物質をコードする 異種RNA配列を発現する方法。 23.RNAが、治療活性、または免疫原性もしくは抗原性のような予防活性 をもつタンパク質、ポリペプチドまたはペプチドをコードする、請求の範囲22 の方法。 24.発現が細胞培養において達成され、そして発現産物が、細胞から分離さ れ、場合によって精製される、請求の範囲22または23のいずれかの方法。 25.発現が動物において達成され、そして発現産物が、該動物からの体液の 収集によって回収される、請求の範囲22または23の方法。 26.動物またはヒト個体において治療または予防活性を達するために、 該個体において生物活性物質をコードする異種RNA配列を発現する方 法であって、該個体への請求の範囲20記載の細胞の投与を含む方法。 27.異種RNA配列が、それが発現される個体の免疫を引き起こす免疫原ま たは抗原をコードする、請求の範囲26の方法。 28.生理的に投与しうる形状における請求の範囲1〜7のいずれかの組み換 えRNAを含む医薬調製物。 29.請求の範囲20記載の形質転換細胞を含む請求の範囲28の調製物。 30.請求の範囲21記載の感染性粒子を含む請求の範囲28の調製物。 31.動物もしくはヒト個体において組み換えRNAの発現を達成するための 使用に関する、請求の範囲28記載の医薬調製物の調製における、請求の範囲1 〜7のいずれかの組み換えRNAの使用。
