Abstract

(57)【要約】 【課題】 メラニンモノマーをトラップすることによっ
て、ＤＯＰＡｃｈｒｏｍｅのポリマー化を抑制する全く
新しいタイプの色白剤を提供する。 【解決手段】 メラニンモノマーと複合体を形成する性
質をもつ物質群、例えばホウ素含有化合物および／また
は動・植物あるいは微生物由来の天然物質、を有効成分
として含有する色白剤。ホウ素含有化合物としては、ボ
ロノフェニルアラニン、ボログリシン、ボロジメチルグ
リシン、ほう酒石酸カリウム、ほう酸、ジヒドロキシフ
ェニルボラン、ボロベタインおよび四ほう酸ナトリウム
からなる群より選ばれた１種または２種以上のものが例
示される。

【発明の詳細な説明】
【０００１】
【産業上の利用分野】本発明は、メラニンモノマーをト
ラップすることによって、ＤＯＰＡｃｈｒｏｍｅのポリ
マー化を抑制する全く新しいタイプの色白剤に関するも
のであって、より詳しくは、メラニンモノマーと複合体
を形成する性質を持つ物質群を有効成分とする色白剤に
関する。
【０００２】
【従来の技術】ヒトの皮膚に現れる、シミやそばかす等
の斑点を消失させ、更に皮膚全体の美白作用を高めるた
めの色白剤について様々な研究が重ねられている。正常
のヒト皮膚の色調に関する因子としては、メラニン、カ
ロチン、血流量、皮膚の厚み及び皮膚透明度が関与して
いるとされている。これらの中で主としてメラニンがヒ
ト皮膚の色調に影響を及ぼし、特に褐色や黒色のユウメ
ラニンの量が色素沈着の要因となっていることが知られ
ている。このような、シミやソバカスの原因となるメラ
ニン生成を抑制し、更に皮膚全体の美白を目的とした各
種の色白化粧料の研究も長年にわたり行われている。
【０００３】最近、動物が生成するメラニンポリマーの
生成過程においては、従来から知られているｔｙｒｏｓ
ｉｎａｓｅのみならず、２種のメラニンモノマー、ＤＨ
ＩＣＡ（５，６−ｄｉｈｙｄｒｏｘｙｉｎｄｏｌｅ−２
−ｃａｒｂｏｘｙｌｉｃ ａｃｉｄ）およびＤＨＩ（ｄ
ｉｈｙｄｒｏｘｙ−ｉｎｄｏｌｅ）を経由する経路や、
それらモノマー生成に関与するＤＯＰＡｃｈｒｏｍｅ
ｔａｕｔｏｍｅｒａｓｅ，ＤＨＩＣＡ−ｏｘｉｄａｓｅ
等の酵素群の存在とその役割が解明されつつある。
【０００４】ところで、コウジ酸をはじめとする種々の
ｔｙｒｏｓｉｎａｓｅ活性抑制化合物が、メラニン生成
を抑制する手段のひとつとして用いられてきたが、効果
が弱かったり、あるいは速効性が期待できないなどの問
題点があったため、新規な作用機序を持った確実な効果
をあげ得る色白剤の開発が待ち望まれていた。
【０００５】
【本発明が解決しようとする課題】そこで、本発明の目
的は、これまでの作用機序とは異なるまったく新しいタ
イプの色白剤を提供することにある。
【０００６】
【課題を解決するための手段】本発明者らは、有効な治
療として確立した悪性黒色腫に対するホウ素熱中性子捕
捉療法において、使用薬剤であるＢｏｒｏｎｏｐｈｅｎ
ｙｌ ａｌａｎｉｎｅ（以下、「ＢＰＡ」ということが
ある）が悪性黒色腫に選択的に蓄積されることにまず着
目した。更に本発明者らは、この知見を元に鋭意研究を
重ねた結果、ＢＰＡを初めとする一連のホウ素化合物群
ならびにＣＶ（ｃｏａｔｅｄ ｖｅｓｉｃｌｅｓ）含有
の天然物質が、細胞内でメラニンモノマー（ＤＨＩＣ
Ａ，ＤＨＩおよびＤＯＰＡ）と錯体を形成し、悪性黒色
腫内でのメラニンポリマーの生成を抑制することによっ
て色素沈着を抑制するという全く新たな作用メカニズム
が認められ、これを元に本発明を完成させた。
【０００７】すなわち、本発明によれば、メラニンモノ
マーと複合体を形成する性質をもつ物質群を有効成分と
して含有することを特徴とする色白剤が提供される。
【０００８】また、本発明によれば、上記物質群が、ホ
ウ素含有化合物および／または動・植物あるいは微生物
由来の天然物質である上記色白剤が提供される。
【０００９】また、本発明によれば、ホウ素含有化合物
が、ボロノフェニルアラニン、ボログリシン、ボロジメ
チルグリシン、ほう酒石酸カリウム、ほう酸、ジヒドロ
キシフェニルボラン、ボロベタインおよび四ほう酸ナト
リウムからなる群より選ばれた１種または２種以上のも
のである上記色白剤が提供される。
【００１０】また、本発明によれば、動・植物あるいは
微生物由来の天然物質が、オルガネラであるメラノソー
ム、ＣＶおよびライソソームまたはそれらのオルガネラ
抽出物である上記色白剤が提供される。

【発明の実施の形態】本発明によって解明された、上記
有効成分とメラニンモノマーとの錯体形成のメカニズム
を、ＢＰＡの場合を例にとって示すと、下記のようにな
る。

【００１２】本発明の有効成分として配合されるホウ素
含有化合物は、例えば、ボロノフェニルアラニン、ボロ
グリシン、ボロジメチルグリシン、ほう酒石酸カリウ
ム、ほう酸、ジヒドロキシフェニルボラン、ボロベタイ
ンおよび四ほう酸ナトリウムからなる群より選ばれた１
種または２種以上から選ばれたものが好適なものとして
挙げられる。また、動・植物あるいは微生物由来の天然
物質としては、Ａ．Ｗｉｌｃｚｅｋｓの論文に記載され
ているような手法により調製されるオルガネラであるメ
ラノソーム、ＣＶおよびライソソーム等、さらにはそれ
らのオルガネラ抽出物を例示できる。
【００１３】本発明の色白剤の剤型は、外用施用上適す
るものであれば特に制限はなく、パップ剤、プラスター
剤、ペースト剤、クリーム、軟膏、チンキ剤、エアゾー
ル剤、乳剤、ローション、乳液、エッセンス、ゲル剤、
パック、パウダー、ファンデーション、サンケア、バス
ソルト等の医薬品、医薬部外品ならびに化粧料として許
容しうる公知の形態で幅広く使用に供されるものであ
る。
【００１４】さらに、本発明の色白剤を調製する場合、
特にアスコルビン酸、アルブチン、コウジ酸等の本発明
の有効成分とは作用機序の異なる公知の色白剤とのｄｕ
ａｌ作用によって色白効果を増強あるいは相乗的に高め
ることができ、難治な色素沈着症に対しても極めて有効
に作用するものとなる。
【００１５】またこのような色白剤以外にも、通常に用
いられる種々の公知の有効成分、例えば、塩化カルプロ
ニウム、セファランチン、ビタミンＥ、ビタミンＥニコ
チネート、ニコチン酸、ニコチン酸アミド、ニコチン酸
ベンジル、ショウキョウチンキ、トウガラシチンキ等の
末梢血管拡張剤、カンフル、メントール、ハッカ油等の
清涼剤、ヒノキチオール、塩化ベンザルコニウム、ウン
デシレン酸等の抗菌剤、副腎皮質ホルモン、ε−アミノ
カプロン酸、塩化リゾチーム、グリチルリチン、アラン
トイン等の消炎剤、胎盤抽出物、甘草抽出物、紫根エキ
ス、乳酸菌培養抽出物等の動物・植物・微生物由来の各
種抽出物等を本発明の目的を損なわない範囲で、その時
々の目的に応じて適宜添加して使用することができる。
【００１６】さらに、本発明の色白剤には前述した公知
の有効成分に加え、油脂類等の基剤成分のほか、必要に
応じて公知の保湿剤、防腐剤、酸化防止剤、キレート
剤、ｐＨ調整剤、香料、着色剤、紫外線吸収剤、散乱剤
等の種々の添加剤を本発明の目的を損なわない範囲で併
用することができる。

【実施例】次に実験及び処方例を開示して本発明を説明
するが、これらの開示は本発明の好適な態様を示すもの
であって、本発明を何ら限定するものではない。
【００１８】≪試験例１≫ ドーパクロムからのメラノ
クロムの生成阻害試験 試験 ａ）試験方法 ＜試薬の調製：５ｍＭ ドーパクロム溶液の調製＞ Ａ溶液：１０ｍＭ ドーパ Ｂ溶液：２０ｍＭ メタ過よう素酸ナトリウム水溶液 Ａ溶液とＢ溶液を１：１等容量混合する（朱色の溶
液）。但し、調製後すぐさま実験に供する。
【００１９】＜試験手順＞１０００ｍＭ燐酸Ｂｕｆｆｅ
ｒ ｌｍｌに、表１から表７に示す濃度になるようにボ
ロノフェニルアラニン、ほう酒石酸カリウム、ジヒドロ
キシフェニルボラン、ボログリシン、四ほう酸ナトリウ
ム、ほう酸を加え、さらに純水を加え１．８ｍｌとし
た。次いで３７℃のウォーターバスにつけ、調製直後の
ドーパクロム溶液（５ｍＭ／ｍｌ）０．２ｍｌを加え、
すばやく混合撹拌し反応を開始させた。３０分間ウォー
ターバスで反応させた後、直ちに５００ｎｍにおける吸
光度を測定し、色調を目視観察した。
【００２０】ｂ）試験結果 下記表１ないし６に結果を示す。３０分間の反応後にお
いて、本発明の有効成分を含まない溶液は、黒くメラノ
クロムが生成し、非常に微細な沈殿が生成し始めてい
た。本発明の有効成分を含む反応溶液は、褐色を示し、
肉眼的には沈殿は観察されなかった。この結果から、本
発明の有効成分は濃度依存的にメラノクロム生成を抑制
していることがわかる。また、これらの溶液の５００ｎ
ｍでの吸光度を測定したところ、目視観察と同様の結果
が得られた。また、これらを一晩室温に放置したものに
おいて、本発明の有効成分を含まない溶液は、青みがか
った黒色の明白な沈殿が観察されたのに対して、本発明
の有効成分を含む溶液では、透明な褐色がかった溶液状
態のままで沈殿は観察されなかった。
【００２１】ｃ）考察 本発明の有効成分を含む反応溶液は、メラノクロムの生
成が抑制される。この機構は本発明の有効成分がドーパ
クロムより自発的に生成されるメラニンモノマーと錯体
を形成し、ドーパクロムを安定化してドーパクロムの重
合を阻害するためと考えられる。

【００２８】≪試験例２≫ 培養メラノーマ細胞に対す
る白色化作用 ａ）試験方法 ＜試験手順＞１×１０5 細胞のＢ１６Ｆ１０細胞をｓｅ
ｅｄｉｎｇし、８時間後ボロノフェニルアラニン、ほう
酒石酸カリウム、ジヒドロキシフェニルボラン、ボログ
リシン、四ほう酸ナトリウム、ほう酸を表７から表１２
に示す濃度で含む培地に交換し培養を継続した。培養開
始３日後に培地を交換し、６日後細胞を回収し、目視観
察を行った。
【００２９】ｂ）実験結果 下記表７ないし１２にその結果を示す。本発明の有効成
分を含む培地で培養したＢ１６Ｆ１０細胞において、本
発明の有効成分を含まない培地で培養した細胞に比べ、
本発明の有効成分を含む培地で培養した細胞は白色化が
認められた。

【発明の効果】本発明によれば、メラニンモノマーと複
合体を形成する性質を持つ物質群を有効成分として含有
する新規な美白剤が提供され、この美白剤はヒトに対す
る安全性が高く、また優れた色白効果を有する色白剤で
ある。

Claims (4)

    【特許請求の範囲】
  1. 【請求項１】 メラニンモノマーと複合体を形成する性
    質をもつ物質群を有効成分として含有することを特徴と
    する色白剤。
  2. 【請求項２】 上記物質群が、ホウ素含有化合物および
    ／または動・植物あるいは微生物由来の天然物質である
    請求項１記載の色白剤。
  3. 【請求項３】 ホウ素含有化合物が、ボロノフェニルア
    ラニン、ボログリシン、ボロジメチルグリシン、ほう酒
    石酸カリウム、ほう酸、ジヒドロキシフェニルボラン、
    ボロベタインおよび四ほう酸ナトリウムからなる群より
    選ばれた１種または２種以上のものである請求項１また
    は２記載の色白剤。
  4. 【請求項４】 動・植物あるいは微生物由来の天然物質
    が、オルガネラであるメラノソーム、ＣＶおよびライソ
    ソームまたはそれらのオルガネラ抽出物である請求項１
    ないし３のいずれか１項記載の色白剤。
