- 【特許請求の範囲】 1.プロテアーゼネキシン−1の変異体であって、P4、P3、P2、P1、P 1’、P2’、P3’、P4’よりなる群から選ばれる位置のアミノ酸残基が、 その位置に天然に存在するアミノ酸残基とは異なる天然アミノ酸残基で置換され ている変異体。 2.変異体が、プロテアーゼネキシン−1と比較して異なるプロテアーゼ特異性 を、および/または特定のプロテアーゼについてプロテアーゼネキシン−1と比 較して上昇した率の会合定数を有する、請求の範囲1記載のプロテアーゼネキシ ン−1の変異体。 3.プロテアーゼネキシン−1の変異体であって、プロテアーゼネキシン−1の 活性部位アミノ酸残基が、プロテアーゼネキシン−1以外のセリンプロテアーゼ 阻害剤の同数の活性部位アミノ酸残基で置換されている変異体。 4.セリンプロテアーゼ阻害剤が、抗トロンビンIII、ヘパリン補因子II、α− 1抗トリプシン、α−1プロテアーゼ阻害剤、プラスミノーゲン活性化因子阻害 剤I、IIおよびIII、α−2抗プラスミン、カリクレイン結合タンパク質ならび にC1阻害剤よりなる群から選ばれる、請求の範囲3記載の変異体。 5.プロテアーゼネキシン−1の活性部位のP4、P3、P2、P1、P1’、 P2’、P3’、P4’位のアミノ酸残基が、下記: よりなる群から選ばれるアミノ酸配列で置換されている請求の範囲3記載の変異 体。 6.プロテアーゼネキシン−1の変異体であって、プロテアーゼネキシン−1の 活性部位の3個またはそれ以上のアミノ酸残基が、与えられたプロテアーゼに特 異的な基質配列を含む異なるアミノ酸残基で置換されている変異体。 7.与えられたプロテアーゼが、エラスターゼ、カテプシンG、C1エステラー ゼ、トロンビン、カリクレイン、およびXa因子、IXa因子、XIa因子、XIIa因子、 VIIIa因子、V’因子、活性化プロテインC、トリプシン、キモトリプシンより なる群から選ばれる請求の範囲6記載の変異体。 8.少なくとも1個がシステインである3個もしくはそれ以上のアミノ酸を含む タンパク質またはその一部であって、ポリエチレングリコールが該システインの チオ基に共有結合しているタンパク質またはその一部。 9.少なくとも1システイン残基を有する天然に存在するタンパク質のアミノ酸 配列を含む修飾されたタンパク質であって、該修飾が、ポリエチレングリコール と該タンパク質のシステイン残基とを結合させることを含む修飾されたタンパク 質。 10.ポリエチレングリコールとタンパク質とを結合させる方法であって、 所望のタンパク質を同定し、ポリエチレングリコールとタンパク質とを結合さ せる部位を決定する段階と、 ポリエチレングリコールを該タンパク質のシステイン残基のチオ基に共有結合 させ、ポリエチレングリコールと該タンパク質とを該部位で結合させる段階と、 を含む方法。 11.システイン残基がタンパク質中に天然の状態で存在する請求の範囲10記 載の方法。 12.通常は存在しないシステイン残基を含むようにタンパク質を変化させ、該 追加したシステイン残基にポリエチレングリコールを共有結合させる請求の範囲 10記載の方法。 13.ポリエチレングリコールを結合させる部位が、タンパク質の溶媒接近可能 領域内にある請求の範囲12記載の方法。 14.グリコシル化される部位でシステイン残基を有するようにタンパク質を変 化させる請求の範囲12記載の方法。 15.多数のシステイン残基を有するようにタンパク質を変化させる請求の範囲 12記載の方法。 16.ポリエチレングリコールが少なくとも2個のタンパク質反応性部分を含む 請求の範囲10記載の方法。 17.ポリエチレングリコールが200〜10,000の分子量である請求の範 囲10記載の方法。 18.所望のタンパク質がプロテアーゼネキシン−1である請求の範囲10記載 の方法。 19.所望のタンパク質がプロテアーゼネキシン−1の変異体であって、該変異 体は、P4、P3、P2、P1、P1’、P2’、P3’およびP4’よりなる 群から選ばれる位置のアミノ酸残基が、その位置に天然に存在するアミノ酸残基 とは異なる天然アミノ酸残基で置換されている請求の範囲10記載の方法。 20.所望のタンパク質が、プロテアーゼネキシン−1の活性部位の少なくとも 1アミノ酸残基がプロテアーゼネキシン−1以外のセリンプロテアーゼ阻害剤の 同数の活性部位アミノ酸残基で置換されている、プロテアーゼネキシン−1の変 異体である請求の範囲10記載の方法。 21.所望のタンパク質が、プロテアーゼネキシン−1の活性部位の3個または それ以上のアミノ酸残基が、与えられたプロテアーゼに特異的な基質配列を含む 異なるアミノ酸残基で置換されている、プロテアーゼネキシン−1の変異体であ る、請求の範囲10記載の方法。 22.少なくとも1システイン残基を有する天然に存在するプロテアーゼネキシ ン−1タンパク質のアミノ酸配列を含む、修飾されたプロテアーゼネキシン−1 であるタンパク質であって、該修飾が、ポリエチレングリコールと該タンパク質 のシステイン残基とを結合させることを含む修飾されたタンパク質。 23.プロテアーゼネキシン−1のアミノ酸配列を含む、プロテアーゼネキシン −1の修飾された変異体であって、P4、P3、P2、P1、P1’、P2’、 P3’およびP4’よりなる群から選ばれる位置のアミノ酸残基が、その位置に 天然に存在するアミノ酸残基とは異なる天然アミノ酸残基で置換されていて、か つ該配列は少なくとも1システイン残基を有し、該修飾が、ポリエチレングリコ ールと該タンパク質のシステイン残基とを結合させることを含む修飾された変異 体。 24.変異体が、プロテアーゼネキシン−1と比較して異なるプロテアーゼ特異 性を、および/または特定のプロテアーゼについてプロテアーゼネキシン−1と 比較して上昇した率の会合定数を有する、請求の範囲23記載のプロテアーゼネ キシン−1の修飾された変異体タンパク質。 25.プロテアーゼネキシン−1の修飾された変異体であって、プロテアーゼネ キシン−1の活性部位のアミノ酸残基がプロテアーゼネキシン−1以外のセリン プロテアーゼ阻害剤の同数の活性部位アミノ酸残基で置換されていて、かつ該変 異体タンパク質のアミノ酸配列は少なくとも1システイン残基を有し、該修飾が 、ポリエチレングリコールと該タンパク質のシステイン残基とを結合させること を含む修飾された変異体。 26.セリンプロテアーゼ阻害剤が、抗トロンビンIII、ヘパリン補因子II、α −1プロテアーゼ阻害剤、プラスミノーゲン活性化因子阻害剤I、IIおよびIII 、 α−2抗プラスミン、カリクレイン結合タンパク質ならびにC1阻害剤よりなる 群から選ばれる請求の範囲25記載の修飾された変異体。 27.プロテアーゼネキシン−1の活性部位のP4、P3、P2、P1、P1’ 、P2’、P3’およびP4’位のアミノ酸残基が、下記: よりなる群から選ばれるアミノ酸配列で置換されていて、かつ変異体タンパク質 は少なくとも1システイン残基を有し、該修飾が、ポリエチレングリコールと該 タンパク質のシステイン残基とを結合させることを含む請求の範囲25記載の修 飾された変異体。 28.プロテアーゼネキシン−1の修飾された変異体であって、プロテアーゼネ キシン−1の活性部位の3個またはそれ以上のアミノ酸残基が、与えられたプロ テアーゼに特異的な基質配列を含む異なるアミノ酸残基で置換されていて、かつ 該変異体タンパク質のアミノ酸配列は少なくとも1システイン残基を有し、該修 飾が、ポリエチレングリコールと該タンパク質のシステイン残基とを結合させる ことを含む修飾された変異体。 29.与えられたプロテアーゼが、エラスターゼ、カテプシンG、C1エステラ ーゼ、トロンビン、カリクレイン、およびXa因子、IXa因子、XIa因子、XXIa因子 、VIIIa因子、V’因子、活性化プロテインC、トリプシンおよびキモトリプシ ンよりなる群から選ばれる請求の範囲28記載のプロテアーゼネキシン−1の修 飾された変異体。 30.下記の一般構造式: R1−S−PEG−S−R2 [式中、R1およびR2は、それぞれ独立に、アミノ酸配列であり、Sは、それぞ れ、R1およびR2のそれぞれのシステイン残基のチオ基であり、PEGはポリエ チレングリコールである] を有する化合物。 31.R1およびR2が、それぞれ独立に、約6〜約1,000個のアミノ酸を含 む請求の範囲30記載の化合物。 32.ポリエチレングリコールが200〜10,000の分子量である請求の範 囲30記載の化合物。 33.R1およびR2が同一である請求の範囲30記載の化合物。 34.R1がヘモグロビンa鎖であり、R2がヘモグロビンb鎖である請求の範囲 30記載の化合物。 35.下記の一般構造式: R1−(S−PEG−S−R2)n [式中、R1およびR2は、それぞれ独立に、アミノ酸配列であり、Sは、R1お よびR2のそれぞれのシステイン残基のチオ基であり、PEGはポリエチレング リコールである] を有する化合物。 36.ポリエチレングリコールが200〜10,000の分子量である請求の範 囲35記載の化合物。 37.R1およびR2が同一である請求の範囲35記載の化合物。 38.R1がヘモグロビンa鎖であり、R2がヘモグロビンb鎖である請求の範囲 37記載の化合物。 39.請求の範囲1記載のプロテアーゼネキシン−1の変異体をコードしている DNA配列。 40.請求の範囲2記載のプロテアーゼネキシン−1の変異体をコードしている DNA配列。 41.請求の範囲4記載のプロテアーゼネキシン−1の変異体をコードしている DNA配列。 42.請求の範囲5記載のプロテアーゼネキシン−1の変異体をコードしている DNA配列。 43.請求の範囲8記載のキメラプロテアーゼをコードしているDNA配列。 44.薬剤学上許容され得る担体と、請求の範囲1記載のプロテアーゼネキシン −1の変異体とを含む薬剤学的組成物。 45.薬剤学上許容され得る担体と、請求の範囲2記載のプロテアーゼネキシン −1の変異体とを含む薬剤学的組成物。 46.薬剤学上許容され得る担体と、請求の範囲4記載のプロテアーゼネキシン −1の変異体とを含む薬剤学的組成物。 47.薬剤学上許容され得る担体と、請求の範囲5記載のプロテアーゼネキシン −1の変異体とを含む薬剤学的組成物。 48.変異体タンパク質を製造する方法であって、 第一の天然に存在するタンパク質の受容体結合領域のアミノ酸をコードしてい るDNAを、該第一のタンパク質とは異なる第二のタンパク質またはその生体活 性部分のアミノ酸をコードしているDNAに接続する段階と; 該DNAを適切な宿主中で発現させる段階と; を含む方法。 49.第二のタンパク質が、P4、P3、P2、P1、P1’、P2’、P3’ 、P4’よりなる群から選ばれる位置のアミノ酸残基が、その位置に天然に存在 するアミノ酸残基とは異なる天然アミノ酸残基で置換されているPN−1の変異 体である、請求の範囲48記載の変異体。 50.受容体結合領域が、ウロキナーゼ、tPA、IX因子、X因子、プロテイン C、表皮増殖因子およびEGP様ドメインよりなる群から選ばれる第一のタンパ ク質に由来する、請求の範囲48記載の方法。 51.受容体結合領域がウロキナーゼのアミノ末端断片である、請求の範囲50 記載の方法。 52.受容体結合領域が、ウロキナーゼの第1〜135番または第1〜87番ア ミノ酸を含むアミノ末端断片である請求の範囲51記載の方法。 53.薬剤学上許容され得る担体、および請求の範囲8記載のタンパク質または その一部を含む薬剤学的組成物。
