Claims (1)
- 【特許請求の範囲】 1.マンヌロナンC-5エピメラーゼをコードする配列を含むDNA化合物であ って、該配列が天然供給源から単離されたかまたは合成的に誘導されたものであ る、DNA化合物。 2.マンヌロナンC-5エピメラーゼをコードする単離されたDNA化合物であ って、DNAブロックAおよび/またはDNAブロックSおよび/またはDNA ブロックRを含む、単離されたDNA化合物。 3.少なくとも1つのDNAブロックA、1つのDNAブロックS、および0か ら少なくとも5つのDNAブロックRを含む、請求項2に記載のマンヌロナンC -5エピメラーゼをコードする単離されたDNA化合物。 4.図6に記載のDNA配列の少なくとも1部分を含む、請求項1に記載の単離 されたDNA。 5.図6に記載のタンパク質アミノ酸配列の少なくとも1部分を含む、請求項1 に記載の単離されたDNA。 6.請求項1に記載のDNA配列を含む、組換えDNAベクター。 7.マンヌロナンC-5エピメラーゼ活性を有する酵素の生産および/またはア ルギン酸塩の微生物学的生産のための組換え宿主細胞の構築に用いる、請求項1 〜6のいずれか1つに記載のDNA配列の使用。 8.75〜98%の高Gブロック含有率を有するアルギン酸塩の微生物学的生産 のための、DNA配列の請求項7による使用。 9.アルギン酸塩の微生物学的生産のためにDNA配列が所望のM/Gブロック 含有率を有するアルギン酸塩を生産させるように選択されることからなる、該D NA配列の請求項7に記載の使用。 10.エピメラーゼ活性を発現し得る組換え宿主細胞の構築による、マンヌロナ ンC-5エピメラーゼを有する酵素の生産方法であって、 (a)記宿主細胞内で機能するプロモーターおよび翻訳活性化配列;および (b)DNAブロックAおよび/またはDNAブロックSおよび/またはDNA ブロックRを含み、前記プロモーターおよび翻訳活性化配列により発現される位 置にある、マンヌロナンC-5エピメラーゼをコードするDNA配列、 を含む組換えDNA発現ベクターにより前記宿主細胞を形質転換させることを特 徴とする、酵素の生産方法。 11.生来アルギン酸塩を産生する宿主細胞内のマンヌロナンC-5エピメラー ゼをコードするDNA配列の1つ、幾つかまたは全てを選択的に不活性化するこ とを特徴とする、アルギン酸塩の微生物学的生産方法。 12.純粋なポリMアルギン酸塩または0〜25%のGブロック含有率を有する アルギン酸塩を生産するための、請求項11に記載の方法。 13.アルギン酸塩を生産するための請求項11に記載の方法であって、前記D NA配列を選択的に不活性化して所望のM/Gブロック含有率およびGブロック 分布を有するアルギン酸塩を生産する、方法。 14.図6に開示された配列の少なくとも1部分を包含する、マンヌロナンC- 5エピメラーゼ活性を有する酵素のアミノ酸配列。 15.DNA化合物がアルギン酸塩産生宿主内で生来的に生じるものではなく、 かつ天然供給源から単離されたかまたは合成的に誘導された少なくとも1つのA ブロックおよび少なくとも1つのRブロックを含む、請求項1に記載のDNA化 合物。 16.DNA化合物が少なくとも1つのSブロックをさらに含む、請求項15に 記載のDNA化合物。 17.マンヌロナンC-5エピメラーゼ活性を有するポリペプチドであって、該 ポリペプチドがアルギン酸塩産生宿主内で生来的に生じるものではなく、かつ天 然供給源から単離されたかまたは合成的に誘導された少なくとも1つのAブロッ クおよび少なくとも1つのRブロックを含む、ポリペプチド。 18.前記ポリペプチドが少なくとも1つのSブロックをさらに含む、請求項1 7に記載のポリペプチド。
