タンパク質のメチル化を阻害する化合物発明の背景
本発明は、腫瘍細胞の増殖の制御に関する。
活性化ラス遺伝子は、複数のヒトの癌に関連してきた。活性化ラス遺伝子、H−
ラス−1は、初めて発見された非ウィルス性の癌遺伝子である。H−ラス−2(
H−ris−2) 、K−ラス−1、K−ラス−2、N−ラス等のその他のいく
つかのヒトラス原癌遺伝子が後に同定されてきた。これらのラス遺伝子について
、いくつかの活性化突然変異体型が同定されてきた。活性化に一ラス遺伝子は、
ヒト結腸の前悪性腫瘍及びヒト前白血病において検出されてきた。
ラスタンパク質及びラス様タンパク質、あるいはシグナル導入Gタンパク質のよ
うなその他のタンパク質は、保持されたカルボキシル末端の−CAAXモチーフ
を有している(C−システィン、A−脂肪族アミノ酸、X−任意のアミノ酸)。
このモチーフは、ポリイソプレニル化、カルボキシル末端のタンパク分解、カル
ボキシル基のメチル化などの一連の翻訳後修飾に関与している。
R−ラス、ラス2、rap−2、pho B等のいくつかのラス関連の低分子G
TP結合タンパク質も、カルボキシ末端の−CAAXモチーフを有し、これらの

メビノリンは、メバロン酸の細胞合成を阻害し、これによってポリイソプレノイ
ドを除去し、ポリイソプレニル化反応を妨害するものと考えられる。メビノリン
は、ラスタンパク質の翻訳後処理に影響を及ぼし、ラスの局在化を妨害するCH

妨害することが観察されたことから、メビノリンまたはメビノリンの誘導体は、
新規細胞傷害性/抑制剤であり、さらに、抗ラス剤の開発の開始点になるのでは
ないかということが示唆されるに至った()Iancock et al、、

発明の概要
概して、本発明は、カルボキシル末端の−CAAXモチーフ(C−システィン、
八−脂肪族アミノ酸、X−任意のアミノ酸)を有するタンパク質のメチル化を阻
害するある種の新規化合物を特徴とする。
概して、本発明は、W−Y−Q−Zまたはw−y−zを有する化合物を特徴とす
る。ここで、Wはファルネシル基か、ゲラニルゲラニル基か、置換ファルネシル
基か、置換ゲラニルゲラニル基であり、YはN0
−5−か、−〇−か、−5e−か、−3−か、−8−か、−8−か、一3e−か
、−3e−であり、

炭素を表し、2個以上の炭素が存在する場合、これらは共有結合によって直鎖状

その塩またはエステル(好適にはメチルが、エチルが、プロピル)が、−CON
H2か、−N02か、−po3が、これらの塩またはエステル(好適にはメチル
か、エチルか、プロピル)が、−CNが、−803が、これらの塩またはエステ
ル(好適にはメチルが、エチルが、プロピル)である。−COOHが、−PO3
か、−803のエステルは、遊離の酸であることが望ましい。これは、遊離の酸
の方が、細胞によってより容品に取り込まれるからである。多くの細胞は遊離の
酸を再生することのできるエステラーゼを有し、メチル化の阻害には好ましい場
合がある。
本発明に係る化合物は、カルボキシル末端の−CAAXモチーフを有するタンパ
ク質のメチル化を阻害することができる。ここで、C−システィン、八−任意の
脂肪族アミノ酸、X−任意のアミノ酸である。
ファルネシル部分及びゲラニルゲラニル部分に関しては、一般に水素がフッ素に
よって置換され、一般にメチル基は臭素によって置換されている。したがって、
°置換ファルネシル基”とは、1個以上の水素がフッ素によって置換され、1個
以上のメチル基が臭素によって置換されたファルネシル部分を意味し、“置換ゲ
ラニルゲラニル基°とは、1個以上の水素がフッ素によって置換され、1個以上
のメチル基力嗅素によって置換されたゲラニルゲラニル部分を意味する。本発明
の第1の態様には、下記に記載のAFC化合物を除くよう予告がある。
種々の好適な実施態様において、Yはスルホキシドで、n−1または2、n−1
で、化合物は、表1に示された化合物で、ペプチドはトランスデユーシンのγサ
ブユニットである。
関連の態様において、本発明は、カルボキシル末端の一〇AAXモチーフを有す
るタンパク質のメチル化を阻害する能力のある前述の化合物を含む治療用の組成
を特徴とする。
関連の態様において、本発明は、患者において腫瘍細胞の増殖を制御する方法を
特徴とする。本方法は、前述の(最終の警告はない)化合物を含む治療用組成の
患者への投与を含む。
下記に説明するように、−CAAXタンパク質のメチル化は可逆的である。メチ
ル化反応はl”I)Vl’VOで可逆的であり、ラスが効率良く膜に結合するに
は重要であるため、ラスタンパク質の病原作用を阻害するためには有用なターゲ
ットとなる。
本発明のその他の特徴及び利点は、以下の好適な実施態様の説明及び請求項がら
明らかになるであろう。
詳細な説明
まず図面について簡単に説明する。
図1は、N−アセチル−5−ファルネシルシスティンの構造の略図である。2番

本発明に係る化合物の特徴は、カルボキシル末端の−CAAXモチーフ(たとえ
ばラス遺伝子生成物)を有するタンパク質のメチル化を低減する能力である。
阻害されるメチル化反応は、−CAAXモチーフに関与した一連の翻訳後修飾の
一部である。これらの修飾には、−CAAXモチーフのシスティンのポリイソプ
レニル化(イオウ上)、カルボキシル末端の3個のアミノ酸(−AAX)のタン
パク分解、ンステインのカルボキシル基のメチル化がある。
本発明の化合物は、古典的な競合的阻害剤か、あまり好ましくはないが、それに
代るものとしては、通常のタンパク基質(たとえばラス遺伝子生成物)のメチル
化を効果的に低減するのに十分な低いKmを有する酵素基質である。標準的なア
ッセイが用いられ、基質のKmと阻害剤のKiが決定される。一般に、好適な化
合物は小さいKiまたはKmの値を有する。Ki及びKmの値は、従来法による

を説明するため実施例を下記に示すが、これは、本発明の限定を意図するもので
はない。
メチル化作用の適切なアッセイの1つでは、ウシ網膜外杆体分節(boνine
re【Inal rod outer sagseno (RO3)メチルト
ランスフェラーゼと天然基質トランスデユーシンを使用する。興味深いことに、
この系は、主要な関心事であるラス遺伝子生成物のメチル化の系の代りとして有
用である。したがって、本発明に係る化合物の好適な種類の1つには、トランス
デユーシンのメチル化の阻害剤がある。その他の適切なアッセイでは、N−アセ
チル−8−ファルネシルシスティンをROSメチルトランスフェラーゼの代りの
基質として使用する。
さらに他の適切なアッセイでは、活性化ラス遺伝子を産生ずる細胞の培養を使用
し、細胞増殖を阻害する候補化合物の能力を測定する。
N−アセチル−5−ファルネシルシスティン(AFC)を示す図1について説明
する。次の構造ガイドラインが、本発明の化合物を設計するのに有用である。
1、AFC及びその誘導体の3位の炭素を置換すると、阻害剤が生成する。
2、AFCの2位の炭素または3位の炭素を脱離すると競合的阻害剤が生成する
。
3.2位の炭素と3位の炭素の間の結合を二重結合に変えると、阻害剤が生成す
る。
4、ファルネシル基を還元すると阻害剤が生成する。
5、C2炭素は、基質活性と適合するいくつもの異なる置換基を受け入れること
ができる。ペプチド結合は、この位置には必要ない。
本発明の化合物の設計は、メチルトランスフェラーゼによる認識に必要な構造の
情報に基づいている。認識は基質である化合物に対する酵素の作用または酵素阻
害によって示される。基質の構造は、下記に説明するように、阻害剤の設計に対
する基準となる。
単純な修飾アミノ酸N−アセチル−5−ファルネシル−し−システィン(AFC
)は、カルボキシル末端の−CAAXモチーフを存するタンパク質をメチル化す
ることができるメチルトランスフェラーゼ、Rosメチルトランスフェラーゼの
基質となる。これは、天然のタンパク基質のポリペプチド配列は、基質認識に必
須ではないことを示している。事実、タンパク結合は基質認識には必要ではなく
、AFCのN−アセチル部分の水素による置換によってS−ファルネシルチオプ
ロピオン酸(FTP)を生成するが、本化合物もメチルトランスフェラーゼの基
質である。
AFC及びFTP誘導体を解析することにより、メチルトランスフェラーゼ阻害
剤の設計に対する指針が得られる。たとえば、図1について説明すると、AFC
の炭素2のN−アセチル基を他の基で置換すると、一般に基質を生成するが、3
位の炭素上で置換すると、一般に競合的阻害剤を生成する。AFCのカルボキシ
ル基を他の基で置換すると、一般に基質活性を妨害する。カルボキシル基の炭素
とイオウとの間の距離が重要で、3位の炭素の脱離、または2位の炭素と3位の
炭素との間に二重結合があると競合的阻害剤を生成する。ファルネシル基も重要
で、ファルネシル基を還元したり、ゲラニルゲラニル基を還元すると、一般に基
質としては活性でなくなる。この解析により、−CAAXモチーフを有するタン
パク質のメチル化を妨害する能力のある分子の設計に対する指針が得られる。
これらの指針は、本発明を特定の前述の化合物に限定するものではない。
メチル化を妨害することによってラス活性を妨害する化合物は明らがであるが、
これらの化合物の他の効果も生理学的に重要である。たとえば、本発明の阻害剤
は膜に局在化したラス受容体に結合するか、若しくはこれを妨害する。阻害剤の
作用の正確な機序に関らず、概説した構造は、ラス機能を妨害し、ラス依存性の
腫瘍の増殖を阻害するのに必要且つ十分なものである。さらに、本発明の化合物
は、たとえば、ラミンBまたは種々のラス関連タンパク質といった、カルボキシ
ル末端の−CAAXモチーフを有する他のタンパク質のメチル化を阻害すること
ができる。このようなタンパク質は、ラスタンパク質のように、細胞増殖に関与

我々は、ウシ網膜トランスデユーシンγサブユニットのファルネシル化システィ
ンの遊離カルボキシル基をメチル化する、ウシ網膜外杆体分節(RO3)のS−
アデノシル−し−メチオニン依存性メチルトランスフェラーゼ活性を同定した。
ウシ網膜トランスデューンンγサブユニットは、カルボキシル末端の−CAAX
モチーフを有し、ラスにおいて観察されるのと同様のファルネシル化反応を受け
る(Lai et al、、 Proc、 Natl、^cad、 Sc1.
US^87:7B73.1990; Fukada et ≠煤A。
Nature 346:65g、 1990 ) 、ウシ網膜外杆体分節S−ア
デノシル−L−メチオニン−依存性メチルトランスフェラーゼ(ROSメチルト
ランスフェラーゼ)により、カルボキシル末端の−CAAXモチーフを有するタ
ンパク質のメチル化を妨害する能力のある化合物を同定する手段が得られる。
RO3,トランスデユーシン、洗浄RO3膜の調製冷凍ウシ網膜(vanda
Lawson Co、、 Lincoln、 NE )を使用し、ウェスリング
−レスニック(Wessl lng−Resnick)らの方法(、/、 11
10/、 r&v262:3697.19+17)の修正法を用い、周囲のタン
パク質を除去したRO3膜を調製した。簡単に説明すると、RO3膜を低イオン
強度の緩衝液及び100μMのGTPで洗浄し、50mM Hepes−Na
(pH7,4) 、100mM NaC1,5mM MgC12,0,1mMフ
ッ化フェニルメチルスルフォニル、0゜1mMジチオスレイトール(緩衝液A)
中に再懸濁し、小分割し、使用するまで一80’Cに保存した。トランスデユー
シンは、RO3膜を細いニードルに通して破壊し、wess I ing−Re
snickら()、 ll゛o/、 r&v262:3G97.19117)に
よって記載されたように、破壊した膜を一連の遠心分離ステップで洗浄して単離
した。最後に、トランスデユーシンは、100μM GTPで膜から溶出した。
トランスデユーシンが豊富な上清を、Centrtprep (Amicon)
を使用して濃縮した。
基質類似体の合成
N−アセチル−s−トランス、トランス−ファルネシル−し−システィン(AF
C)は、カミャ(Kai+ya)ら(,1znc、 Jシ’o/、 1Jei、
43:363.1979 ) lこ記載されたように、N−アセチル−し−シ
スティンと臭化トランス、トランス−ファルネシルより合成した。AFCをメタ
ノール及びHCIで処理し、AFCのメチルエステルを生成した。
In Vitrθ メチル化反応
最初の検討用に、反応混合液は、S−アデノシル−[メチル−3H]メチオニン

Daポリペプチドは、SDS/ポリアクリルアミドゲルのTγと一致する。さら
に、精製トランスデユーシンを洗浄RO3及び[3HI SAMのみとインキュ
ベートすると、6kDaのポリペプチドと膜関連の23〜29kDaタンパク質
を標識化する。これらの後者のタンパク質は、網膜の培養細胞中に見られるプレ
ニル化及びカルボキシル基がメチル化されたスモールGタンパク質(Malte
se et all、〕、 11.’o/、 (’/lex、 2G5:214
8.1990 )及び脳から精製された23kDaのGタンパク質(Yaman

989 )の類似体であると見られる。この結論は、これらがプレニル化されて
いるとの我々の以前の観察(Lai et at、、 Proc Jyt/、
jcid、 Sr/、 43”j87:7673.1990 )と、これらがメ
チル化されるという事実と、これらの分子量と、GTPをニトロセルロース膜上
に結合させるというこれらの能力に基づいている。
放射活性標識した6kDaのタンパク質をHPLCで分析すると、放射活性はT
γと共に溶出する。放射活性ポリペプチドをv8プロテアーゼで分解してHPL
Cで分析すると、放射活性の2つの主ピークが検出される。1つ目は放射活性メ
タノールであると予測される位置の3分に、2番目のピークは42分に検出され
る。エドマン分解によってこのタンパク質を解析すると、Tγのカルボキシル末
端フラグメントに対応する配列Leu−Lys−Gly−Gly−Xaaが得ら
れ、最近報告されたように(Fukuda et al、、 ri’、y1we
346:65B、 1990 ) 、メチル化がシスティン残基に起こっている
ことを裏づけた。
メチル化のプロセスの性質は、下記に説明する。RO3膜に存在するメチルトラ
ンスフェラーゼ活性は、熱によって失活し、2つの特徴の良くわかっている阻害
剤、SAM−依存性メチルトランスフェラーゼ、すなわち、SAH(Barbe
r et al、+)’、 l/10/、 l:’1lei、 259ニア11
5.1984)及びs inefungin (Pughetall、)’、
l/10/、 (’/m、 253:4075.197g)によって阻害される
。
AFCは[3H] SAMによるタンパク質への標識の取り込みを低減した。A
FCも、メチルトランスフェラーゼに対する基質として作用し、AFC[3H:
1メチルエステルを生成する。この生成物の正体は、TLC及びHPLCの標準
による標品と共に溶出することから確認された。AFCに対する酵素メチルトラ
ンスフェラーゼ活性は、予想通り、熱によって失活し、SAH及びシネファンギ
ン(s inefungin)によって阻害された。基質活性に対してファルネ
シル部分が必要であることは、N−アセチルシスティンがメチルトランスフェラ
ーゼの基質でないのを示すことにより、例証された。
メチル化は可逆的なプロセスである
非放射活性メチルエステルを[3HlSAM及び洗浄RO5膜と共にインキュベ
ートすると、少なくとも2時間の間、放射活性が、直線的に、時間依存的にAF
Cメチルエステルに取り込まれる。この結果は、メチル化反応が可逆であること
を示している。脱メチル化プロセスを直接検討するため、AFC[3Hコメチル
エステルを、メチル化阻害剤の存在下、RO8膜とインキュベートした。メチル
エステルは、膜のデメチラーゼ活性によって急速に加水分解された。反応を加熱
すると、酵素活性について予測されるように、急激に活性が低下した。同様の条
件下で、Tγの脱メチル化とスモールGタンパク質(23〜29kDa)である
と考えられるものも観察された。
ROSメチルトランスフェラーゼ基質
前述のウシ網膜外杆体分節のS−アデノシル−し−メチオニン依存性メチルトラ
ンスフェラーゼは、カルボキシル末端の−CAAXモチーフを有するラスタンパ
ク質のようなタンパク質を修飾するメチルトランスフェラーゼのモデルを提供す
る。AFCはRO8I−ランスフェラーゼに対する基質として作用するため、酵
素相互作用に対して局所12ペプチド構造は一般に必要ではない。したがって、
メチルトランスフェラーゼ活性を妨害する化合物は、比較的小さい。
まずAFCについて説明すると、変更されて基質及び阻害剤を生成するAFCの
構造態様には、複数のものがある。たとえば、変更は、N−アセチル基、ファル
ネシル基、カルボキシル基においてなされる場合がある。イオウはスルホキシド
に変更され、イオウとカルボキシル基の炭素との間の距離は、炭素原子の付加ま
たは除去、または二重結合の形成によって変更され、イオウとカルボキシル基と
の間の1つ以上の炭素に置換基が付加される場合がある。
メチルトランスフェラーゼ基質及び阻害剤の合成N−アセチルホモシスティンを
、無水酢酸によってN−アセチル化し、次いで塩基性で加水分解することにより
、ホモシスティンチオールアセトンから合成した。ファルネシル化合物、AFC
は臭化トランス、トランス−ファルネシル及びN−アセチル−L−システィンか
ら、N−アセチル−8−ファルネシル−ホモシスティン(AFHC)はN−アセ
チルホモシスティンから、3−ファルネシルチオプロピオン酸(FTP)は3−
メルカプトプロピオン酸から、S−ファルネシルチオ酢酸(PTA)はメルカプ
ト酢酸から、それぞれ、AFCの合成について記載された一般的な手順を用いて
合成した。この手順は、Kaaiyaらの方法(Agric、 Biol、 C
hes、 43:363.1979 )の変法である。N−アセチル−L−シス
ティン(1,0g、6mM)と、グアニジノ炭酸塩(1,3g、7mM)と、臭
化トランス、トランスーファルネシル(1,7g、6mM)をアセトン75m1
に溶かし、その結果生成した溶液を室温にて終夜撹拌した。この溶媒を減圧留去
し、残渣を酢酸エチル中に取り、10%HCI及び水で連続して洗浄し、乾燥し
くNa2S04)、小容量に濃縮した。この物質のこの物質をンリカカラムの酢
酸エチル/メタノール(4:1〜1:2)で溶出するクロマトグラフィーにより

。蒸発後、生成物を分取用薄層クロマトグラフィープレートに添加し、ヘキサン
/酢酸エチルで展開した。シス及びトランス生成物を1(PLOによって分離し
た。メタノール中でシスとトランスの比は約7:]であった。より大量のトラン
ス異性体をシス異性体から、光化学的に、あるいは、テトラヒドロフラン中でト
リメチルアミンを塩基として反応することによって合成することが可能である。

1.5当量を10分間に渡って撹拌しながらメタノールに加えた。反応混合物を
さらに1時間撹拌し、希塩酸で中和した。

酢酸エチルて展開した。純粋な生成物をプレートから取り、メタノールで溶出し

解した。塩化ベンゾイル1.1当量を冷却して加え、懸濁液を1時間撹拌した。
この溶液を希塩酸で中和した。蒸発後、生成物を分取用薄層クロマトグラフィー
プレートに添加し、ヘキサン/酢酸エチルで展開した。純粋な生成物をプレート
から取り、メタノールで溶出した。収率は50〜75%の間であった。生成物は
予想通りのnmr及び赤外スペクトルを示した。
親の酸は、メタノール中で水酸化ナトリウム1当量を加え、エステルから合成し
た。生成物は予想通りのnmr及び赤外スペクトルを示した。
N−ブロモアセチルファルネシルシスティンは同一の方法で合成した。
3−ファルネシルオキシプロピオン酸:全一トランス−ファルネソールを、0.
1当量の1当量のtertブトキシドカリウム/テトラヒドロフラン/l−ブタ
ノールの存在下、アクリル酸メチルと室温で3時間反応させた。この溶液を0.
IN塩酸で酸性化し、蒸発乾固し、薄層クロマトグラフィーによってシリカ上で
精製した。遊離酸は、メタノール中で水酸化カリウムによってエステルを処理し
て合成した。生成物は予想通りのnm「及び赤外スペクトルを示した。
N−アセチル−3e−ファルネシル−D、 L−システィン:N−アセチルセレ
ノシスティンメチルエステルを、AFCの生成と同様に臭化ファルネシルと反応
させ、セレノ誘導体を生成した。
3−ファルネシルセレノプロピオン酸:1−セレノファルネサレ(1−sele
nofarnesare) は1臭化全一トランス−ファルネシル(al 1−
trans−farnesylbroiide) をセレノ尿素と反応し、次い
で水酸化ナトリウムで処理して合成した。サリノファルネサン(sallnof
arnesane )をテトラヒドロフラン及びトリエチルアミン中でアクリル
酸メチルと反応させ、メチルエステル生成物を生成した。この溶液を0.IN塩
酸で酸性化し、蒸発乾固し、薄層クロマトグラフィーによってシリカ上で精製し
た。遊離酸は、メタノール中で水酸化カリウムによってエステルを処理して合成
した。生成物は予想通りのnmr及び赤外スペクトルを示した。
本発明に従う他の化合物は、前述のものと同様の方法で、有機化学の標準的な技
法を用いて合成される。本発明の化合物の合成に必要な化合物は、Pierce
、Aldrich、Fluka、Avant i (Blrminghas、^
L)から入手した。
類似体の解析
前述のメチルトランスフエラーザアッセイを用いて前述の類似体を解析した。
ROSメチルに対する基質としてのAFC(表1)に関して、RO8膜をメチル
トランスフェラーゼ酵素源として使用し、メチル化のKM23μMを測定した。
この系におけるAdoMetの見かけのKMは2μMであり、酵素の至適pHは
約8. 0である。基質としてのAFCのKM +iミーラスタンパクから誘導
された合成ペプチドのメチル化について別の系で決定されたものの範囲内にある
(Stephenson et at、、)、〃θ/、(”/let 265:
1624g、 1990 ) 、このことは、タンパク質のペプチド部分は、一
般に、メチルトランスフェラーゼによる認識に重要でないことを裏付けている。

双方の例とも、本は図示した分子の残りの部分に結合している炭素原子を示す。
単純化のため、はとんどの場合において、炭素は実線の末端及び有機化合物を表
す通常の方式における角度で交差する2つの実線の交差点によって示した。した
かって、ファル不ンル基は15個の炭素を含み、ファルネシル基は10個の炭素
を含んでいる。通常の方式に従い、概して水素も特には示していない。Kmは基
質に対して示し、Kiは通常の阻害剤に対して示した。基質または通常の阻害剤
としての化合物の状態を、KmまたはKiの値の位置に示した場合かある。
AFCのジアステレオマーのスルホキシドであるAFC3は、500μMてしメ
チル化されなかった(表1)。AFC5に対して活性か欠如していることは、本
質的な特異性は、基質のイオウ原子にあることを示している。興味深いことに、
S−ファルネシルホモシスティン類似体(AFHC)もメチルトランスフェラー
ゼの基質とはならなかった(表1)。このことは、イオウ(あるいは、他の誘導
体では酸素またはセレン)原子とカルボキシル基の距離が基質活性に対して重要
であることを示している。
ファルネシル部分が重要であることは、N−アセチル−8−ゲラニル−し−シス
ティン(AGC)が、酵素に対して非常に弱い基質であることによって示される
(表1)。さらに、完全に飽和されたファルネシル(3,7,11−トリメチル
ドデシル)誘導体(ATC)は、基質として不活性である。これらの実験は、本
質的な特異性は、ファルネシル側鎖及びチオプロピオン酸部分にあることを示し
ている。
アセチルアミド部分がないS−ファルネシル−3−チオプロピオン酸(FTP)
は、活性な基質であることが明らかとなり、KMは13.7μM(表1)で、こ
れは実際にはAFCより低い。したがって、メチルトランスフェラーゼ活性には
ペプチド結合は必要なく、酵素活性は本質的にファルネシルチオプロピオン酸部
分にある。さらに、S−ファルネシル−2−チオ酢酸(PTA)のようにイオウ
原子とカルボキシル基との間の距離が縮められると、基質活性は消失した(表1
)。この結果は、AFHCについて観察された結果と一致する。
FTAとAFHCとAFC5が基質ではなく、これらがすべてAFCメチル化の
強力な競合的阻害剤であることから、これらはメチルトランスフェラーゼ活性を
阻害し、ラスのようなタンパク質のメチル化を阻害するのに使用することができ
る。PTAによるAFCのメチル化の阻害に対しては、Ki 4.6μMが算出
された。同様の解析を用い、AFHCに対してはKi 30.2μMが決定され
た。
AFC5に対しては、算出Kiは11.6μMであった。PTAの存在下または
PTAがない状態でS−アデノシル−[メチル−3H]メチオニン(2,3μM
、85Ci/mmo 1)とインキュベートしたRO3試料のSDSゲルを蛍光
撮影に露出し、濃度測定によって定二した結果、FTAのマイクロモルの濃度(
10μM)もl’l) Vl’tfOでトランスセユーシンγサブユニットのカ
ルボキシル基のメチル化を70%阻害した。
前述の峙果は、天然のメチルトランスフェラーゼ基質トランスデユーシンの構造
的なff1Thlさが大きく低減されても、メチル化酵素によって認識されるこ
とのできる分子を生成することを示している。
メチルトランスフェラーゼによる認識に重要な2つの特異的な特徴を下記に説明
する。1つは、カルボキシル基から特定の距離にあるIIでないイオウ原子が好
ましいことである。未変化のファルネシルが、本質的な基質活性には好ましい。
これは、ファルネサン誘導体(ATC)の活性の欠如、ゲラニル誘導体(AGC
)の非常に低い活性によって示されている(表1)。
細胞増殖阻害のアッセイ
細胞増殖を阻害する能力につき、PTAとFTPとAFHCとAFCとを次のよ
うに試験した。

、 Betl+esda、 HD) にて増殖させた。これらの細胞を、ジメチ
ルスルホキシド(DP、1SO)に溶解した様々な濃度のPTAか、FTPか、
AFHCか、AFCか、またはDMSOのみで処理した(すべての実験において
、最終のDMSOi11度は1%以下であった)。処理した細胞及び未処理の細
胞を標準的な培養条件で数日間インキュベートした。細胞は、生きている完全な
細胞のみを数えるトリバンブルー染料排除アッセイを用い、手作業で24時間間
隔で数えた。
図2に示したように、FTP (図A) 、PTA (図B) 、AFC(図C
)、AFHC(図D)は、HL−60細胞の増殖を阻害した。
本アッセイは、新たに創製された化合物のスクリーニングに使用することができ
る。
HL−60細胞におけるメチルトレンスフエラーゼ活性ロス膜について前述した
のと同様のity Vitro標識化反応において、23kDaのラスタンパク
質は、崩壊したHL−60細胞の[3H] SAM標識によってメチル化された
主要なタンパク質だった。本アッセイにおいて、50μMのFTAは、23kD
aのタンパク質のメチル化を61%阻害した。これは、FTAが特異的にラスの
メチル化を阻害できることを示している。
AFCはHL−60細胞の膜に存在する活性によってメチル化することができ、
この活性は10倍過剰のPTAによって90%阻害することができる。AFCメ
チルエステルはHL−60細胞の抽出物によって分解され、メチル化反応はHL
−60細胞抽出物において可逆的であることを示している。最後に、トランスデ
ユーシンのγサブユニットは、HL−60細胞抽出物に存在するメチルトランス
フェラーゼによってメチル化される。
用途
本発明の化合物は、単独または薬剤学的に許容される担体または賦形剤との配合
において効果的な量で投与することができる。本化合物または組成は、単独また
は他の治療剤との配合で投与することができる。
本発明の化合物は、好都合な手段、たとえば、静脈投与、経口投与、筋肉投与、
経鼻投与によって投与する。特に腫瘍部位においては、延長放出システムを使用
する。

Claims (8)
- 1.構造式W−Y−Q−ZまたはW−Y−Zの化合物であり、ここで、Wはファ ルネシル基か、ゲラニルゲラニル基か、置換ファルネシル基か、置換ゲラニルゲ ラニル基であり、 Yは−S−か、−O−か、−Se−か、▲数式、化学式、表等があります▼か、 ▲数式、化学式、表等があります▼か、▲数式、化学式、表等があります▼か、 ▲数式、化学式、表等があります▼か、▲数式、化学式、表等があります▼であ り、Qは、▲数式、化学式、表等があります▼・・・・▲数式、化学式、表等が あります▼で、n−1、2、3、4、5、または6であり、T1′・・・・Tn ′及びT1′′・・・Tn′′のそれぞれは、独立してF1か、Brか、−NH COCH3か、−NH2か、ペプチドか、アルカン基か、アルケン基か、ポリエ チレングリコール基か、飽和脂肪酸か、不飽和脂肪酸か、単糖類か、二糖類であ り、 Zは−COOHか、その塩またはエステルか、−CONH2か、−NO2か、− PO3か、これらの塩またはエステルか、−CNか、−SO3か、これらの塩ま たはエステルであり、 ただし、Wがファルネシルの場合、Yは−S−であり、n−2で、T2′または T2′′のいずれかが−NHCOCH3であり、Zは−COOHではない。 前記化合物は、カルボキシル末端モチーフ−CAAXを有するタンパク質の酵素 的メチル化を妨害することを特徴とし、ここで、C−システイン、A−脂肪族ア ミノ酸、X−任意のアミノ酸である。
- 2.請求の範囲第1項の阻害剤において、Yが−S−で、n−1または2である 阻害剤。
- 3.請求の範囲第1項の阻害剤において、n−1である阻害剤。
- 4.請求の範囲第1項の阻害剤において、前記化合物が表1に掲載された化合物 である阻害剤。
- 5.請求の範囲第1項の阻害剤を薬剤学的に許容される担体中に含む治療組成。
- 6.患者において腫瘍細胞の増殖を制御する方法において、前記方法が、請求の 範囲第5項の治療組成の前記患者への投与を含む方法。
- 7.患者において腫瘍細胞の増殖を制御する方法において、前記方法が、薬理学 的に許容される化合物の前記患者への投与を含み、前記化合物が、カルボキシル 末端モチーフ−CAAXを有するタンパク質の酵素的メチル化を妨害することを 特徴とし、ここで、C−システイン、A−脂肪族アミノ酸、X−任意のアミノ酸 である方法。
- 8.請求の範囲第1項の化合物において、前記ペプチドがトランスデューシンの 7サブユニットである化合物。表1:メチルトランスフェラーゼの基質及び阻害 剤
