Description
Claims (14)
- 1.細胞内成分および該成分を抽出するために用いた物質を含有する溶液を供給 することにより細胞内成分の抽出物を調製する方法において、該溶液を抽出用物 質の中和のために適切な種類および適切な量のサイクロデキストリンまたはサイ クロデキストリン誘導体と接触させることを特徴とする方法。
- 2.溶液が細胞を抽出用物質と接触させることにより供給される、請求の範囲第 1項に記載の方法。
- 3.サイクロデキストリンが溶液状で使用される、請求の範囲第1項または第2 項に記載の方法。
- 4.サイクロデキストリンが抽出用物質より化学量論的に過剰に使用される、請 求の範囲第1項ないし第3項のいずれかに記載の方法。
- 5.サイクロデキストリンがa−サイクロデキストリン、β−サイクロデキスト リンまたはγ−サイクロデキストリンである、請求の範囲第1項ないし第4項の いずれかに記載の方法。
- 6.抽出用物質がノニオン、アニオン、カチオンまたはツビッターイオン界面活 性剤、またはそれらの界面活性剤の混合物である、請求の範囲第1項ないし第5 項のいずれかに記載の方法。
- 7.細胞内成分がATPである、請求の範囲第1項ないし第6項のいずれかに記 載の方法。
- 8.抽出物中の細胞内成分が酵素アッセイされる、請求の範囲第1項ないし第7 項のいずれかに記載の方法。
- 9.酵素アッセイがホタルルシフェラーゼ反応に基づくものである、請求の範囲 第1項ないし第8項のいずれかに記載の方法。
- 10.細胞内成分がDNAまたはRNAである、請求の範囲第1項ないし第6項 のいずれかに記載の方法。
- 11.抽出されたDNAまたはRNAが増幅または酵素による修飾もしくは制限 処理により後続処理される、請求の範囲第10項に記載の方法。
- 12.酵素アッセイまたは後続処理がサイクロデキストリンまたは誘導体の存在 下で実施される、請求の範囲第8項、第9項または第11項に記載の方法。
- 13.請求の範囲第1項ないし第9項のいずれかに記載の方法により生物検体中 のATPを抽出およびアッセイするためのキットにおいて、下記の構成成分:a )抽出用物質(他の成分と分離して保存)b)サイクロデキストリン、 c)ホタルルシフェラーゼ試薬、 d)アッセイ用緩衝液 を含むキット。
- 14.−抽出用物質が、流体検体と接触した際に適切なサイズの試料を捕獲する キャリヤー上または中において乾燥されており、−サイクロデキストリンがアッ セイ用緩衝液に溶解されており、−ホタルルシフェラーゼ試薬が、該試薬をアッ セイ用緩衝液中へ放出しうるキャリヤー上または中において乾燥されている、請 求の範囲第13項に記載のキット。
