発明の背景
を椎動物の免疫系は、宿主に異物であると認識される、病原体、細菌およびウィ
ルスのような物質を認識し排除するように機能する。さらに、免疫系はまた、細
胞表面上に改変されたタンパク質を発現するために異物であると見なされる悪性
細胞を、排除する監視系としての役割も果たす。この抗原と呼ばれる、異物物質
に対する免疫応答には、体液性応答および細胞性応答が包含される。体液性応答
には、抗原を認識して抗原に結合する、特異的な抗体または免疫グロブリンの生
産が含まれる。細胞性応答には、抗原の排除を助ける細胞の増殖が含まれる。
免疫応答は、抗原または抗原に類似した分子を含むワクチン(または免疫原)に
よる能動免疫によって、しばしば誘導または増強され得る。しばしば、免疫原に
加えて、アジュバントと呼ばれる免疫増強剤を提供することが必要とされる。
本発明は、能動免疫の分野に関し、より詳細には、リンパ腫に対する免疫に有用
なワクチン、および免疫応答の程度および特性を変化させるのに有用なアジュバ
ントに関する。
Bリンパ系腫瘍の大部分は、細胞膜上の免疫グロブリン(Ig)の発現により特
徴づけられる。表面1gのイディオタイプ(id)は、腫瘍特異的抗原またはマ
ーカーと見なし得、これは免疫療法の標的として使用されてきた。モノクローナ
ル抗id抗体は、これらの腫瘍の免疫生物学の研究に用いられ、油膜にも使用さ
れた。しかし、抗idモノクローナル抗体(Mabs)、特にマウス起源のもの
の受身投与は、多数の問題によって妨害されており、そのため、これらの抗体の
臨床上の応用性が限定されてきた。これらの問題としては、(i)循環している
フリーの抗原、(ii)マウス■gに対する免疫応答の生起、および(iii)
Ill瘍細胞の不均一性がある。いくつかの研究により、イディオタイプタンパ
ク質またはサブフラグメントによる免疫が、形質細胞腫またはIgを表面に有す
るリンノス腫の過成長から動物を防護し得ることが報告されている。誘導される
免疫応答には、抗イデイオタイプ抗体およびT細胞依存性免疫が含まれる。腫瘍
細胞のチャレンジに対してこのような抗体および免疫を引き起こすためには、イ
ディオタイプタンパク質を強力な免疫原性キャリヤに結合させ、この複合体を強
力なアジュバントの存在下に提供する必要があることが、繰り返し示されている
。
イディオタイプの不均一性が、抗イデイオタイプ抗体を用いた臨床油膜において
開示されている。モノクローナル抗体によって誘導される初期の部分的応答の後
、オリジナルの腸瘍部位からイディオタイプ変異腫瘍細胞が出現した。このよう
なイディオタイプ変異腫瘍細胞は、モノクローナル抗体処置前から既に存在して
いたが、これらはイディオタイプ陽性腫瘍細胞の選択的な除去の後、増殖可能に
なったものと思われる。
同系腫瘍由来のIgまたはそのサブフラグメントによる動物の能動免疫は、抗i
d抗体の生産を生じ、これは引続き腫瘍細胞に曝されて防御を誘導する。しかし
、大抵の場合、腫瘍は、あまりにも弱いか、または治療上有利に間に合うには遅
すぎる応答しか引き起こさない。従って、修飾されたid−1gまたは強力な非
生理的アジュバントのいずれかが必要であった。非生理的免疫原または非特異的
アクチベーターは、ヒトの患者への使用には非常に望ましくないものである。自
己免疫疾患の発生を導き得るポリクローナルβ細胞応答の誘導の可能性を含む、
副作用のためである。
従って、ワクチンの免疫原性を高める方法が長年型まれてきた。好ましくは、こ
のような免疫と一緒に使用される全てのアジュバントは、治療上有用であるのに
十分な強さの免疫応答を提供すべきである。特に、有効なポリクローナル応答が
引き起こされるように、同系1gで腫瘍宿主を能動免疫する手段が、必要とされ
ている。本発明は、この必要性を満足し、それに関連した利点をも提供する。
発明の要旨
本発明は、目的のイディオタイプタンパク質を予めノイルスした樹枝状細胞を投
与することにより、病原性リンパ球に対する有効な免疫応答を誘導する方法を提
供する。1つの実施態様においては、リンパ腫に対する哺乳類の能動免疫の方法
が提供される。この実施態様は、樹枝状細胞をイディオタイプIgにさらして、
イディオタイプをパルスされた樹枝状細胞状細胞を哺乳類に戻す注射を行う工程
とを包含し、これにより、リンパ腫細胞に対する免疫が誘導される。もう1つの
実施態様では、本発明は、イディオタイプ細胞およびパルスされた樹枝状細胞の
両方を投与することに関する。

発明の詳細な説明
本発明は、免疫の効果を高める方法に関する。このような方法は、多(の腫瘍マ
ーカーのように、有効な免疫応答を生じさせるのが困難な状況での免疫に対して
、特に有用である。
しかし、この方法はまた、様々な病原に対する免疫(こ組み合わせても適用され
る。
近年、インビトロでウィルスまたはポリクローナル抗イディオタイプ抗体(Ab
2)をフィルスされたマウスの樹枝状細胞力(、劇的に抗ウイルス免疫応答を増
大させること力(、FrancotteおよびUrbain PNAS 82:
8149 (1985)によって報告された。樹枝状細胞(DC)は、その細胞
表面上に付着した抗原(こ対する免疫応答の強力な刺激者であることが示されて
(入る。抗原を、<ルスされたDCは、休眠Tリン、<球を始動させ、この1ノ
ンノ<球は次に、抗原特異的Bリン、f球に、必要な介助を与える。
本発明は、インビトロにおいて病原性リンノく球力)らのイディオタイプタンパ
ク質をフィルスされたマウスDC力(、これまでは病原性リンパ球に対して有効
な免疫応答を生起させるの(こ必要と考えられていた、免疫原性キャリヤおよび
非生理的アジュバントの代わりをし得る、という予期されなかった結果を含んで
いる。本願で使用される用語「病原性リンパ球」とは、制御されない悪性リンパ
球、または自己免疫応答に関連するリンパ球のいずれかを指す。用語「病原性リ
ンパ球からのイディオタイプタンパク質」とは、免疫グロブリン、またはイディ
オタイプエピトープを有する免疫グロブリンのフラグメント(FAB)を指す。
引続く腫瘍細胞投与に対する防御は、免疫原性キャリヤに結合させ、フロインド
アジユバントに乳化させた同系イディオタイプを用いて、またはインビトロでイ
ディオタイプをパルスされたDCを用いて、予め免疫することによって得ること
ができる。無関連なIgMをパルスされた同数のDCで処理され、または可溶性
同系38C131gMの同一の投与量で免疫されたコントロール群は、長期にわ
たる生存を示さなかった。これらのデータは、樹枝状細胞による増大効果および
腫瘍増殖のイディオタイプ特異的抑制を、明確に示している。
C3H/Heマウスを、同系311C13リンパ腫由来のイディオタイプ免疫グ
ロブリンM(IgM)で免疫した。イディオタイプ特異的な体液性および細胞性
の免疫、および致死の腫瘍細胞チャレンジに対する防御を得るためには、免疫原
性キャリヤタンパク質(キーホールリンベットヘモシアニン;にLH)および強
力な非生理的免疫刺激剤(完全な70インドアジュバント;CFディオタイブ提
示のために用いられる場合には、免疫原キャリヤもアジユバントも必要ではなか
った。インビトロで非修飾イディオタイプタンパク質をフィルスされ、動物に再
注射された樹枝状細胞は、続く腫瘍接種に対して有意な抵抗性を誘導し得た。あ
るいは、FABフラグメントまたはイディオタイプエピトープを有する合成ペプ
チドを、接種に使用し得た。天然の38C13イデイオタイプ(id)に対する
応答として増殖した、イディオタイプ特異的Tリンノく球および細胞障害性TI
Jン/f球が、両群において観察された。細胞免疫応答は、38C13−にLH
および完全フロインドアシュIインドで処理した場合よりも、樹枝状細胞で処理
した動物においてより強かった。両免疫方法の結果は、イディオタイプ変異体の
出現に起因する腫瘍細胞のエスケープ、または腫瘍細胞の休眠を伴うことなく、
長期間の生存を得た。注目すべきことには、−年後で1よ、80%のマウスがま
だ生存していた。
免疫した動物の血清分析によると、防御効果は、血清抗イデイオタイプ力価と単
純には相関していなかった。フロインドアジユバントを用いた過免疫によって誘
導された、高レベルの抗イデイオタイプ抗体は、腫瘍細胞の表面1gを改変し得
、次いでこの腫瘍細胞は、id特異的T細胞による破壊を免れることができたの
であろう。
イディオタイプタンパク質の存在下に増殖したT細胞(孟、免疫の3ケ月以降で
も実証することができた。これらの細胞Cよ、その表現型は知られていないが、
同系膿瘍細胞1こ細胞障害性効果を有していた。
以下の実施例は、本発明を説明することを巨的としており、本発明を限定するも
のではない。
実施例1

樹枝状細胞(DC)は5teirvanおよびCohen、 J、 Exp、
Med。
139:380−397 (1974)に記載の方法を用いて単離した。簡単に
述べると、凝集物またはクラスターを含まない肺臓細胞の懸濁液をウシ血清アル
ブミン(BSA) (P 111.082 g/cm3) ;画分V (Sig
ma Chemical Co、、 St、 Louis、 MO)の溶液に、
11Il当りlx 10’細胞の濃度で懸濁した。低密度のBSA溶液(p−1
,060g/am3)を、この上に積層した。試験管を、4℃、10.000g
で30分間回転させて平衡化した。浮遊細胞を回収し、RPM! 1640培地
で2回洗浄した。細胞を完全培地(12PMr 1640.5%FCS、 5
x 10−5M 2−メルカプト−エタノール、ペニシリン、ストレプトマイシ
ン、最小必須アミノ酸、およびピルビン酸ナトリウム)に再懸濁し、1ml当り
1 x 10’細胞の濃度でプラスチックベトリ皿に移して、5%の002中で
37℃で3時間放置した。非付着細胞を、ゆるやかにピペットで除去し、付着細
胞を完全培地にさらに16時間維持した。低密度の非付着細胞を含む上清み液を
、DC源として使用した。マクロファージはプラスチック表面に再度付着する傾
向があるが、DCは付着しないままであったことから、夾雑しているマクロファ
ージを抗原をパルスする間に除去した。最終細胞調製物の純度は、走査型電子顕
微鏡、透過型電子顕微鏡、免疫蛍光検査、およびアクリジンオレンジ染色により
調べた。FITCで標識したヤギ抗マウスIg、およびビオチンで標識した抗T
hy−1,2(BectonDickinson)を用いて、それぞれBおよび
T細胞を特徴づけた。
樹枝状細胞は、24ウエルの平底プレート(1ウエル当す1m1)に1ml当り
5 x 105細胞濃度で、完全培地に再懸濁した。50マイクログラムの精製
した38C131gM (に)イディオタイプタンパク質、または無関連のマウ
スIgM (x ) (ABPC−SigmaChe+*1cal Co、、
St、 Louts、 MO)を加えた。プレートを、5%のco2中に37℃
で、4から5時間放置した。細胞を無菌PBSで数回洗浄し、1ml当り2.5
X 105細胞に再懸濁した。
実施例II
イディオ イブ1を いたEXIsの
C3H/Heマウスは、Catholic University of Le
uvenのLaboratory Animal Centerより入手した。
Ba1b/cおよびFl (C3H/He x Ba1b/c)マウスは、Fr
ee University of Brussels (VUB)のLabo
ratory Animal Facilityで飼育されたo 3aC13は
、C3H起源の、発がん物質(f)MBA)により誘導されたB細胞腫瘍である
。これらの腫瘍細胞およびこの研究に使用したインビトロ適合された細胞系は、
細胞膜上にIgM(に)を発現するが、大量のIgは分泌しない。イディオタイ
プIgM(に)を分泌する細胞系は、腫瘍細胞を非分泌骨髄腫細胞系(P3 x
63 Ag 8.653)と融合させることにより得られた。
各々10匹のマウスからなる4つの群を、表1に示すスケジュールに従って、K
LHに架橋し、フロイント完全アジュバント(CFA) 、不完全アジュバント
(ICFA)またはPBSに乳化した50μgのイディオタイプ38C131g
M、または無関連のARPCIgMで免疫した。最後の注射から一週間後、3a
ct[癌細胞(102細胞/マウス)を、腹腔内注射した。この投与量は、30
0日目でに、非処理動物の100%を致死させる。腫瘍の存在および死亡日を記
録した。
図1は、生存率に対する免疫処置の防御効果を、典型的実験について示す。最も
効果的な免疫スケジュールは、イディオタイプIgMをCFAを用いて1回、I
CFAを用いて1回の2回投与するものであった(群I+)。これらの結果には
再現性があった。無関連のIgM−1[LH複合体(群mまたは複合体にしない
イディオタイプIgM(群V[[)を用いた場合には、長期にわたる生存例はな
かった。

ブタンノでり質のダースター投与を行った結果、38C13−KLH複合体にC
FAを用0た実験と同等な、腫瘍投与後の生存を得た。コントロールの免疫動物
(無関連なIgMをパルスしたDCまたはイディオタイプタンノ(り質単独)は
、回答防御を示さなかった。
実施例IV
゛ および の一
体液性免疫の役割を評価するために、同系の抗イデイオタイプ抗体のレベルを、
以下のように調製したIgG特異的EL[SAで測定した。
Maloneyら、Hybridoma 4:191−209 (1985)の
方法に従って、グルタルアルデヒドを用いてキーホールリンペットヘモシアニン
(KLH) (Calbfochem−Behring、 Hoechst)と
架橋した精製イディオタイプタンパク質で、Ba1b/cマウスを免疫した。
上記文献はここに援用される。膵臓細胞を、P3 x 63 Ag 8.553
骨髄腫細胞系にハイブリダイズした。モノクローナル抗体E4および1IE3は
、38C131gM (に)と強く反応し、正常なC3111血清によって阻害
されず、正常な肺臓細胞にも精製された1gM骨髄腫タンパク質にも結合しなか
った。C3H起源のモノクローナル抗イデイオタイプ抗体55A8およびラット
モノクローナル抗体R7D7を、硫酸アンモニウム(40%)を使用して2回沈
澱によって、腹水液から精製した。当該技術分野で周知の方法に従って、抗体の
ビオチン標識を行った。
腫瘍細胞を注射する前に、眼窩静脈叢の穿刺により、マウスから採血した。個々
のマウスからの血清を同一の実験群についてプールした。同系の抗イデイオタイ
プ抗体を、上記のELISAアッセイによって検出した。
図3の結果は、38C13−にLHとCFAにより免疫したマウスで抗id抗体
レベルが高いことを示している。DCで免疫した動物の血清では、これよりはる
かに低レベルの抗id抗体が検出され、このことは、抗体レベルと生存との間に
は明確な相関関係がなかったことを示している。CFA中のIgM−KLHを3
回注射することにより、血清の抗id抗体のレベルは高いが、生存率は低い結果
となった(図1)。
両免疫方法における長期生存者の肺臓にイディオタイプ特異的1978球を検出
することによって、細胞性免疫を測定した。T細胞について富化した膵臓細胞を
、可溶性イディオタイプタンパク質またはセファロースビーズに結合させたイデ
ィオタイプIgMの存在下で3日間培養した。コントロールのウェルは、4μg
/itのフンカナバリンAを含有させて24時間培養したラット牌細胞培養物の
培養上清を10%(v/v)含むIL2含有培地中に、肺細胞を含むか、または
無関連の1gMタンパク質を含んでいた。生存する動物の肺臓細胞の懸濁液を0
.2%のBSAでプレコートしたプラスチックベトリ皿(80mm)に、3ml
の完全培地に107細胞/mlで移し、37℃で1時間(27)維持した。
非付着細胞を、ウサギ抗マウス(に)(10μg/+*1) テプレコードシた
プレートに移し、4℃で1時間放置した。2回目のブレーティングを、ヤギ抗マ
ウスIg (Tago、 Burlinga+we、 CA)でプレコートした
プレートで行った。この2回にわたるプレーティング工程後の回収率は、一般に
、有核細胞の25から30%であった。後の実験では、T細胞は、当該技術分野
に周知の方法により、ナイロンクールに結合させることにより富化された。B細
胞の混入は、蛍光ヤギ抗マウスIg抗体(Tago)を用いた免疫蛍光検査によ
って調べた。B細胞画分は、通常、5から7%であった。T細胞が富化された細
胞懸濁液を、丸底ウェル(200μm/ウェル、5 x 105細胞/ml)に
置いた。20μlの刺激剤(50μg/ml)をウェルに加えた。3日間培養の
後、1.5μCiの[メチル3H1−チミジンを各ウェルに加えた。こうして1
8時間のパルスを行った後、細胞をガラスファイバーフィルターに収穫し、組み
込まれた放射能をシンチレーシゴンカウンティングによって測定した。測定はす
べて4試行で行い、データは、平均cpmtsEMで表した。
インビトロにおける刺激の結果を表I+に示す。DC−38C13で処理された
マウスからのT細胞iよ、CFA中のイディオタイプKLH複合体で処理された
マウスまたはコントロールマウスと比較して、イディオタイプタンパク質により
良(反応した。

長期生存動物“における増殖応答
[3H]チミジン取り込み(cpm+sEM)表2.゛富化された膵臓子細胞を
、38C131gM、セファロースビーズに結合した38C131gM、 IL
2含宵培地または可溶性無関連IgMの存在下で、3日間インビトロで培養した
。培養の最後の18時間に、3トチミジン取り込みの程度により、刺激を測定し

T細胞を富化した膵臓細胞はまた、以下のように、通常の細胞で媒介性の細胞障
害性アッセイにも使用した。
腫瘍細胞をL−[4,5−”[1]ロイシン(Amersham)で標識し、U
底のマイクロウェルに1 x 10’個の標的細胞(100μm)で、入れた。
エフェクター細胞(生存動物からの富化された膵臓T細胞)を、100/1.5
o/1.25/1.6.25/1.3/1.1.5/1の割合で3試行ずつ、最
終容量を200μlとなるように加えた。細胞を、ゆるやかな遠心分離により沈
降させ、プレートを5%co2を含む湿潤雰囲気中に37℃で16時間インキュ
ベートした。50マイクロリツトルの上清み液を用いて、遊離した放射能をカウ
ントした。自発遊離および最大i!1Mは、T細胞の代わりに100μmの完全
培地または1ooμlの0.1%NP40を加えることによって決定した。特異
的な細胞障害性を、以下の式に従って計夏した。

免疫リンパ球による細胞障害性を、T細胞を単離した日、およびインビトロにお
いて38C13が結合したセファロースビーズで刺激した3日後に、測定した。
刺激しないT細胞は、38C13標的細胞を溶解できなかった。しかし、インビ
トロにおける刺激後は、(100/lのエフェクター/標的比で16時間インキ
コベーションしたアッセイにおいて) 38C13−KLHで刺激された細胞に
よる38C13履瘍細胞の特異的溶解が11%であったのに対して、DC−38
C13で刺激されたT細胞による溶解は21%であった。
実施例V
1昆l簾皿思
長期生存動物の肺臓における残存腫瘍細胞の存在を、免疫組織学およびインビト
ロにおける培養により追跡した。クリオスタット切片をアセトン中に固定し、モ
ノクローナル抗体、次いでストレブトアビジンーホースラディッシコベルオ牛シ
ダーゼおよびジアミノベンジジンテトラヒドロクロリドで染色した。長期生存動
物からの肺臓の凍結切片を、ビオチン化モノクローナル抗イデイオタイプ抗体で
染色した。これらの抗体は、腫瘍が優麗した肺臓切片で予めテストした。この肺
臓切片において、これらの抗体は表面と細胞質イディオタイプIgMとの両方を
染色する。長期生存動物の肺臓の連続切片で、残存膿瘍細胞は検出され得なかっ
た。肺臓から単離された8973球を、富化された馴化培地においてインビトロ
で培養した。この研究で使用された3gCl3腫瘍細胞は、インビトロでの増殖
に適応しているが、いずれの培養物も腫瘍細胞の発生を示さなかった。最後に、
B細胞を、放射線照射した同系の未処置動物に移入した。6ケ月間観察した結果
、腫瘍の増殖は見られなかった。これらのデータは、長期生存動物の肺臓に、残
存腫瘍細胞が残っていなかったことを示す。
実施例v■
のその の ゛
10X/%7クス平衡塩顕溶液(IIBss) (10x HBSSから希釈;
Gibco Cat、 Nr、 310−4065A/J、)中の、100ユニ
ツト/mlのコラゲナーゼ(CLS目L Worthington、 Cat、
Nr 4182. Ig/バイアル)溶液(無菌GIBCO7,5%重炭酸イ
オンでpH7,2に調節)を用いて、肺臓を還流した。細胞を10%のFCSを
含むRPMI培地を含む試験管に収集した。残った部分を、インキニベータ中で
、400ユニツト/厘1のコラゲナーゼ溶液中で15分間インキュベートした。
この溶液を約50回ピペッティングして、大半の細胞を収集した。すべての細胞
をプールし、ベレット化した。
ウシ血清アルブミンを以下のように調製した: B(IVUMINARCOHN
FRACTION V粉末(Armour Pharmaceutical
Company、にankakee、1llinois)を、デシケータ中に4
℃で保存した。ガラ゛ スピーカー(アルミニウム箔で封鎖した)中に、65m
1の蒸留水、1116m1のPBS、29*lのLM NaOHおよび107g
のBSAを混合し、攪拌せずl二4℃で24時間放置した。屈折率測定器で濃度
を調べて、1.3855−1.3865の間に維持した。溶液の濃度が高すぎる
場合には、ゆっくりと攪拌して、冷PBSを加えた。溶液を濾過し、4℃で放置
した。ベレットを冷えたBSA溶液に再懸濁し、約2mlの血清を含まない培地
を勾配の一番上に加えて10.000gで遠心分離した。低密度の細胞を収集し
たところ、膵臓細胞の約10%であった。
100■■の培養皿を用いて、10%のFCSを含む培地で、細胞を2時間培養
した。激しくピペッティング(1皿当り15分間)することによって、非付着の
細胞を洗浄除去した。細胞を再び血清を含まない培地で1時間インキュベートし
、ゆるやかにピペットすることによって非付着の細胞を除去した。残った付着細
胞を、10%のFCSを含むRPMI中で、−晩培養した。次ぎの日、非付着の
細胞を、ゆるやかにピペットすることによって収集した。n臓1個当り、2 x
105から5 x 10’個の樹枝状細胞が得られた。
本発明は、好ましい上記の実施態様を参照して説明したが、本発明の精神を逸脱
しない限り、様々な改変が可能であることを理解されたい。従って、本発明は、
以下の請求の範囲によってのみ限定される。

本発明は、目的のイディオタイプタンパク質を予めパルスした樹枝状細胞を投与
することにより、病原性リンパ球に対して有効な免疫応答を誘導する方法を提供
する。1つの実施態様において、哺乳類をリンパ腫に対して能動免疫する方法が
提供される。この実施態様は、樹枝状細胞をイディオタイプ□Igにさらして、
イディオタイプをパルスした樹枝状細胞を作成する工程と、このイディオタイプ
をパルスした樹枝状細胞を哺乳類に注射する工程とを包含し、これにより、リン
パ腫細胞に対する免疫が誘導される。もう1つの実施態様では、本発明は、イデ
ィオタイプ細胞およびパルスした樹枝状細胞国際調査報告
Claims (8)
- 1.細胞膜上にイディオタイプタンパク質を発現する病原性リンパ球に対して有 効な、体液性および細胞性免疫応答を、哺乳類に誘導する方法であって、 a)インビトロにおいて、樹枝状細胞を該病原性リンパ球のイディオタイプタン パク質にさらして、イディオタイプをパルスした樹枝状細胞を得る工程;および b)該イディオタイプをパルスした樹枝状細胞を該哺乳類に注射する工程、 を包含する方法。
- 2.前記病原性リンパ球が腫瘍細胞である、請求項1に記載の方法。
- 3.前記腫瘍細胞が、リンパ腫細胞である、請求項2に記載の方法。
- 4.前記リンパ腫細胞が、Bリンパ球様細胞である、請求項2に記載の方法。
- 5.前記イディオタイプタンパク質が、免疫グロブリンである、請求項1に記載 の方法。
- 6.前記イディオタイプタンパク質が、イディオタイプを有する免疫グロブリン のフラグメントである、請求項1に記載の方法。
- 7.前記哺乳類が、ヒトである、請求項1に記載の方法。
- 8.インビトロにおいてイディオタイプ免疫グロブリンにさらされた樹枝状細胞 を含む、イディオタイプをパルスした樹枝状細胞。
