【発明の詳細な説明】
本出願は、1992年7月22日出願の出願第07/918,292号の一部継
続出願であり、その出願を本出願の一部を構成するものとしてここに引用する。
本発明は癌を治療するためのワクチンに関するものである。そのワクチンは、p
53タンパクを免疫原として自存する。
ヒトゲノムの染色体17p上に見い出されるp53遺伝子は、野性型の状態では
腫瘍抑制遺伝子である。レビン(Levine)らによる“p53腫瘍抑制遺伝
子“と題された研究記事がネイチャー(以、453〜456.1991年)に掲
載されている。
ヒトの腫瘍の50%以上は、p53遺伝子の突然変異型を発現する細胞を含有し
ている。多くの腫瘍では、り53遺伝子の1つの対立遺伝子がタンパクの突然変
異型をコードする点突然変異を自存しているが、他の対立遺伝子は部分的または
完全に失われる。このようなパターンは、例えば、結腸癌の約70〜80%、乳
癌の約50%、肺癌の約50%で観察され、小細胞肺癌の100%で観察される
。野性型p53の消失を検出することによって癌の診断を行なう提案がなされて
いる(ボゲルスタイン(Voge I s t e in)らのヨーロッパ特許

p53遺伝子中の点突然変異の位置あるいは場所は腫瘍の種類によって異なる。
例えば、B型肝炎とアフロトキシンに暴露されたヒトの肝細胞カルシノーマの5
0%はコドン249にp53突然変異を有し、肺腫瘍では、コドン154と27
3に優先的に突然変異を存するようであり、結腸腫瘍では、コドン175.24
8および273に複数の独立した突然変異を有している。癌および前癌状態にお
ける重症度と性質を含む種々の表現型は、p53突然変異の場所を決定すること
によって区別できることを示す証拠が提出されている。レビン(Levine)

癌を有するヒトの約10〜20%は、p53タンパクに対する抗体を産生ずる腫
瘍を持っている(デ・フロメンチル(de Fromentel)ら、Inte
rnational Journal of Cancer 39.185〜I

B (1982年))。これらの抗体の存在は、ヒトHLAまたはネズミH−2
遺伝子座のクラスII受容体が、CD−4ヘルパーT細胞およびB細胞システム
に対してp53のペプチド抗原を提供でき、その結果免疫応答が生じることを示
唆する。しかし、抗体は、効果的な抗腫瘍剤ではないと考えられている。したが
って、癌を有するヒトにおける抗p53抗体の存在は、癌患者が効果的な抗腫瘍
免疫応答をしている可能性を示唆するものでない。
腫瘍抗原によって免疫処理された動物は同じ抗原からは保護されると報告されて
いる。したがって、シミアンウィルス40(SV40)ラージT抗原を用いて動
物を免疫処理すると、生きた腫瘍形成SV40によって形質転換された細胞によ
るその後の攻撃から保護できる(テベチア(Tevethia)ら、ColdS
pring Harbor Symp、Quant、Biol、土工、235〜
242.1980年参照)。
同様に、フレイ(F r e y)とレビン(Levine)は、p53付加r
as−転換の、B3と命名された照射されたフィッシャーラット細胞株で免疫さ
れたラットは、同じ生細胞によるその後の腫瘍攻撃から保護できたことを報告し
た。
そのp53付加ras−転換ラット細胞株は、別個に得たp53付加ras−転
換細胞株の85%と共通する腫瘍特異的な移植拒絶抗原を発現することが報告さ
れた。フレイとレビンは、p53タンパクが腫瘍特異的な移植拒絶抗原でなく、
B3細胞による攻撃に対して保護できないことを示す証拠を提出した(Jour
nal of Virology 63,5440〜5444.1989年を参
照)。
現在の癌治療では、化学物質や放射線など、腫瘍細胞に対する特異性が不十分な
細胞毒性剤を使用している。正常な細胞に影響を与えることがない、より特異的
な治療が要求されている。特に、患者自身の免疫系を刺激することによる癌治療
が必要とされている。
発明の概要
上記の目的および当業者に明らかとなるであろう他の目的は、哺乳類の免疫系に
投入されたとき効果的な免疫応答を誘発するような形の突然変異型または野性型
のp53タンパクを含むワクチン組成物を提供することによって達成される。
さらに、本発明は、免疫学的に効果のある量の突然変異型または野性型のp53
タンパクを用いて哺乳類を治療することを含む、ヒトの腫瘍の成長を阻止する方
法を提供する。
発明の詳細な説明
本発明は、哺乳類の免疫系に投入されたとき効果的な免疫応答を誘発するような
形の突然変異型または野性型のp53タンパクを含むワクチン組成物を提供する
。例えば、突然変異型または野性型のp53タンパクは、抗原表現細胞の表面、
または薬学的に許容される補助剤と結合して提供される。
本明細書では、“野性型のp53タンパク“という語は、マトラシェウスキ−(
Mat Iashewski)ら、EMBOJ、+3.3257〜3262.1
984年、サカット・ホウリ(Zaku t−Hour i)ら、EMBOJ。

配列を意味する。これらの配列はアンバンク(GenBank)から入手可能で
ある。
野性型のp53には、アミノ酸72におけるプロリン/アルギニン多型および対
[5口するヌクレオチド多型が含まれる。
p53タンパクは突然変異されてもよい。腫瘍細胞において突然変異が起きるこ
とか頻繁に見い出される位置か、または野性型p53遺伝子の不活化を導く位置
において突然変異が行なわれることが好ましい。突然変異は、単一の置換または
複数(即ち、2〜20、好ましくは2〜IO1より好ましくは2〜5)の置換で
ある。
好ましい突然変異体ヒトp53遺伝子は、レビ〉・・ニー・ジエー(Levin
e A、J、))ら、“p53腫瘍抑制遺伝子”、Nature、351.45
3〜456(1991年)に記載されている。p53遺伝子で生じる点突然変異
の殆どは、ミスセンス突然変異であり、変換されたp53タンパクを生しさせる
。突然変異の多くは、アミノ酸残基130から290の間に密集しており、その
タンパクの、4個の“ホットスポット”領域に最も集中している。
このホントスポット領域は、p53遺伝子の4つの最も保存された領域に一致し
ているにクロ(Nigro)ら、Nature、342.705〜708(19
89年)参照)。4個の“ホットスポット“突然変異領域は、132〜143.
174〜179.236〜248、および272〜281のコドンに存在する。
これらのホットスポットの生成頻度と分布は癌ができた組織の種類によって異な
る。
野性型p53遺伝子とタンパクは知られており、公知の方法により自然にまたは
りコンビナンドとして得られる。その方法には、細胞からタンパクを直接分離す
る方法、当該タンパクをコードするDNAを分離または合成し、このDNAを使
用してリコンビナントタンパクを産生する方法、個々のアミノ酸から当該タンパ
クを化学的に合成する方法が含まれる。野性型p53遺伝子とタンパクを得る方
法は、マトラシエウスキ−(Mat Iashewski)ら、EMBOJ。
ラム(Lamb)とクローフォード(Crawford)、Mo1.Ce1l。
Biol、5.1379〜1385.1986年)に記載されている。突然変異
株は、位置特異的突然変異生成により野性型p53遺伝子から調製してもよい(
例えば、1ラー(Zoller)とスミス(Smi th) 、Nuc I。
完全なp53遺伝子またはp53遺伝子のフラグメントは、例えば、オリゴヌク
レオチド・プローブを横築するための公知のDNA配列を用いて分離してもよい
。こうするために、公知の配列から得た探識化オリゴヌクレオチドを使用し、サ
ザン・ハイブリダイゼーションによってDNA制限フラグメントを同定する。
あるいは、p53をコードするDNAを個々のヌクレオチドから合成してもよい
。DNAを合成する公知の方法には、重複する二重鎖オリゴヌクレオチドを調製
し、空隙に充填し、終端を相互に結合する方法が含まれる。
上記のようにして調製されたDNAを複製連鎖反応(PCR)によって増殖して
もよい。あるいは、そのDNAをクローニングベクターに挿入して増殖し、それ
を適切な宿主細胞に形質移入し、その宿主細胞から953DNAを回収するよう

当該タンパクを調製するためには、当技術分野でよく知られたりコンビナンド法
を使用してもよい。簡単に述へると、p53をコードするDNAを発現ベクター
に挿入し、それを好適な宿主細胞に形質移入する。DNAの発現後、当該タンパ
クを収穫する。サンプルツク(Sambrook)ら参照(前述)。
突然変異型または野性型p53タンパクの同等物を本発明のワクチンに使用して
もよい。そのような同等物には、突然変異型または野性型p53タンパクに匹敵
する免疫応答を誘発する類似体が含まれる。さらに、当該同等物は、それらに対
応する突然変異型または野性型p53タンパクと免疫学的に交差反応性(cro
ss−react 1ve)を存する。その同等物は、例えば、タンパクのフラ
グメント、あるいは突然変異型または野性型p53タンパクの置換型、付加型ま
たは削除型突然変異株でもよい。
突然変異型または野性型p53タンパクは、抗原のエピトープを定義するための
十分なアミノ酸残基を含有していることが好ましい。例えば、そのフラグメント
は、エピトープのみをコートするミニ遺伝子であってもよい。公知の免疫タンパ
クから免疫原性フラグメントを分離して同定する方法は、サルフェルト(Sal
feld)らによって、J、Virol、63,798〜808(1989年)
に、およびイソラ(Isola)らによって、J、Virol。
63.2325〜2334 (1989年)に記載されている。
フラグメントがエピトープを定義しているものの、免疫原としては短すぎる場合
には、キャリヤーとなる分子に結合してもよい。好ましいキャリヤー分子には、
キーホール・リンペット・ヘモシアニン、Ig配列、TrpE、およびヒトまた
はウシ血清アルブミンが含まれる。結合は、当技術分野で知られている方法で行
なうことができる。その一つの方法は、当該フラグメントのシスティン残基をキ
ャリヤー分子上のシスティン残基と結合させることである。
同等のタンパクは、同等のアミノ酸配列を有する。他の配列と実質的に同一であ
るが、1つ以上の置換、付加または削除によって当該能の配列と相違するアミノ
酸配列は、同等な配列と考えることができる。好ましくは、配列内のアミノ酸残
基の数の25%以下、より好ましくは10%以下、最も好ましくは5%以下の数
のアミノ酸残基が、本発明のタンパクに対して、置換、付加、または削除される
。
例えば、配列内のアミノ酸を同等なアミノ酸を用いて置換することは知られてい
る。一般に同等と考えられているアミノ酸のグループは下記の通りである。

パクは、免疫系に適切に与えた場合、予想を越えた効果的な免疫応答を誘発する
。本明細書では、効果的な免疫応答とは、癌細胞の成長を禁止、すなわち阻止、
遅延または停止するか、癌細胞を無くすことを意味する。その効果的な免疫応答
は、キラーT細胞反応であることが好ましい。哺乳類は、ヒト、またはマウス、
ラットまたはウサギのような実験に通常使用される動物である。
突然変異型または野性型p53タンパクは、ベヒクルによりワクチンとして免疫
系に与えられる。例えば、突然変異型または野性型のp53タンパクは、抗原表
現細胞の表面、または薬学的に受け入れられる補助剤と結合して与えられる。
抗原表現細胞は、一般に、主要組織適合性遺伝子複合体(MHC)を有する真核
性細胞であり、好ましべは、それらの細胞表面にあるクラスIIの遺伝子産物で
ある。本明細書の目的の範囲では、抗原表現細胞には、抹消血細胞なとのりコン
ビナンド真核細胞、およびリコンビナント細菌細胞も含まれる。本明細書によっ
て定義された抗原表現細胞の例には、樹枝状細胞、MHCクラスIi陽性である
ことが好ましいマクロファージ、MHCクラスII陽性であることが好ましい単
核細胞、およびリンパ球が含まれる。
本発明の一実施例においては、抗原表現細胞は、突然変異型または野性型p53
タンパクをコードする外因性DNAを発現するりコンビナンド真核細胞である。
リコンビナント真核細胞は、インビボ、またはインビトロで調製できる。
本発明の一実施例においては、突然変異型または野性型p53をコードするDN
Aを、リコンビナント・ウィルスベクターを用い、インビボで真核細胞に挿入す
る。ベクターには、ワクシニアウィルスのような弱毒化したりコンビナンド・ポ
ックスウィルスが含まれ、国際出願PCT/US 92101906(1992
年3月2日)に記載されたように、必須でないウィルスコード化遺伝子機能が不
活化されたオウムボックスが好ましい。ワクシニアゲノムに外部のDNAを挿入
するだめの技術は、当技術分野で知られている。リコンビナントを構築するため
のプラスミドベクターは、例えば、チャクラパーティ(Chakrabar t

真核細胞にDNAを挿入するための好適なりローニング/発現ベクターには、5
V−40、アデノウィルス、サイトメガロウィルス(CMV)の公知の誘導体、
およびレトロウィルス由来のDNA配列が含まれる。これらのベクターは何れも
、プラスミドと、ファージDNA、即ちシャトルベクターとの組合せから得られ
るベクターと結合することにより、原核細胞および真核細胞の両方においてタン

突然変異型または野性型p53タンパクは、リコンビナント細菌細胞の表面上で

クローニングベクターは、染色体、非染色体、および合成りNA配列のセグメン
トを含んでいてもよい。幾つかの好適な原核クローニングベクターは、coIE
l、pcRI%pBR322、pMB9、pUCSpKsM、およびRP4など
の大腸菌からのプラスミドを含有する。また原核ベクターは、M13なとのファ
ージDNAの誘導体、および他の線状−重鎖DNAファージを含有する。
細菌、特に大腸菌のタンパクを発現するためのベクターも知られている。そのよ

挿入する。有用な発現コントロール配列の例は、Iacシステム、mシステム、
tacシステム、↓rcシステム、ファージλの主要なオペレータおよびプロモ
ータ領域、fd外皮タンパクのコントロール領域、および、ポリオーマ、アデノ
ウィルス、レトロウィルス、およびシミアンウィルスから得たプロモータ、例え
ば、5V−40の早期および晩期プロモータ、および原核細胞または真核細胞内
での遺伝子の発現をコントロールするとして知られている他の配列、およびそれ
らのウィルスまたはその組合せである。
本ワクチンは、ムラミルペプチドなどの薬学的に許容される補助剤、インターフ
ェロン、インターロイキン−1およびインターロイキン6などのリンフ才力イン
、または細菌的補助剤を更に含有していてもよい。その補助剤は、突然変異型ま
たは野性型p53タンパクを吸着する、酸化アルミニウムなどの適切な粒子を含
有していてもよい。補助剤を含むこれらのワクチンは、本技術分野で知られた方
法で調製できる。
細菌性補助剤の例はBCGである。上記のように抗原表現細胞として使用した場
合、リコンビナントBCGはそれ自身の補助剤としても作用するであろう。この
場合、追加の補助剤は必要てないが、1種類以上の補助剤を任意的に存在させて
もよい。天然(リコンビナントでない)の状態で使用する場合、BCGは突然変
異型または野性Wp53と組合せられることによって単独で補助剤として作用し
、効果的な免疫応答を誘発する形になる。
本ワクチンは、適切な媒体を含んでいてもよい。好ましい媒体には、燐酸緩衝生
理食塩水、リボゾームおよびエマルジョンなどの薬学的に許容されるキャリアー
が挙げられる。
さらに、本発明は、突然変異型または野性型のp53を含むワクチンを免疫的に
効果のある量だけ用い、腫瘍を有するか、腫瘍形成の可能性に切迫した哺乳類を
治療することを含む、哺乳類における腫瘍の成長を阻止する方法を包含する。
哺乳類が異常な前癌状態にあると診断された場合、その哺乳類は腫瘍形成の危険
に切迫している。
突然変異型または野性型p53は、効果的な免疫応答を誘発するような形で、即
ち、抗原表現細胞の表面で、または薬学的に許容される補助剤と結合して哺乳類
の免疫系に提供される。突然変異型または野性型p53は、薬学的に許容される
キャリアーなとの媒体中にあるのが好ましい。
本ワクチンは、本技術分野で知られた方法によって哺乳類に投与される。そのよ
うな方法には、静脈内、腹腔内、皮下、筋肉内、局所、または皮肉の投与が含ま
れる。

- 1.哺乳類の免疫系に投入されたとき効果的な免疫応答を誘発する形態の突然変 異体p53タンパクを含有するワクチン組成物。
- 2.薬学的に許容される媒体をさらに含有する請求項1に記載のワクチン組成物 。
- 3.上記形態が、突然変異体p53タンパクが抗原表現細胞の表面にある形態か 、または突然変異体p53タンパクが秦学的に許容される補助剤と結合した形態 である請求項1に記載のワクチン組成物。
