Claims (8)
- 1.最初のヌクレオシドをシリルエーテル結合を介して支持体に結合させること を特徴とするオリゴヌクレオチド合成用の支持体系。
- 2.式 ▲数式、化学式、表等があります▼ 〔式中、 Bはヌクレオシドまたはデオキシヌクレオシド塩基であり、 R1は保護基であり、 R2はH、OHまたはOR5(式中、R5は保護基である)であり、 R3、R4およびXはそれぞれ、アルキル、アリール、シクロアルキル、アルケ ニル、アラールキル、シクロアルキルアルキル、アルキルオキシ、アリールオキ シ、シクロアルキルオキシ、アルケニルオキシ、アラールキルオキシであり、 Sは固体支持体である。 ただし、R3、R4およびXの1個だけが、アルキルオキシ、アリールオキシ、 シクロアルキルオキシ、アルケニルオキシ、アラールキルオキシまたはシクロア ルキルアルキルオキシである〕を有することを特徴とする請求項1記載の支持体 系。
- 3.R1がトリチル、モノメトキシトリチル、ジメトキシトリチルまたはピキシ ルであり、基Bはアデニン、グアニン、シトシン、ウラシル、チミンまたはイノ シンであることを特徴とする請求項2記載の支持体系。
- 4.R3はアリール基であり、Xはアルキルまたはアルキルオキシ鎖であり、R 4は三級アルキルまたは三級アルキルオキシであることを特徴とする請求項2ま たは3に記載の支持体系。
- 5.固体支持体(S)上でオリゴヌクレオチドを合成する方法において、最初の ヌクレオシドをシリルエーテル結合を介して支持体(S)に結合させ、ついで以 下の工程 (i)最初のヌクレオシド上へのヌクレオチドの縮合、 (ii)オリゴヌクレオチド上の5′−保護基を除くすべての保養基の除去およ び酸触媒脱保護時に形成された非プリン部位の切断、 (iii)支持体からのオリゴヌクレオチドの選択的切断、 (iv)5′−保護基をアフィニティーハンドルとして用いた、逆相クロマトグ ラフィーによるオリゴヌクレオチドの精製 を行うことを特徴とする方法。
- 6.5′−保護基はジメトキシトリチルであることを特徴とする請求項5記載の 方法。
- 7.切断反応(iii)はテトラアルキルアンモニウムフルオリドまたはヒドロ キシドを用いて行うことを特徴とする請求項6記載の方法。
- 8.請求項2〜4のいずれかに記載の支持体系を利用することを特徴とする請求 項5〜7のいずれかに記載の方法。
