Description
［産業上の利用分野］
本発明は尿素水溶液配合製剤において長期間尿
素の安定性を保持させる方法に関するものであ
る。 ［従来の技術］ 尿素は皮膚の角層の吸湿性を高める作用や、角
質溶解作用があるので、魚鱗癬、老人性乾皮症、
アトピー性皮膚炎等の疾患用の外用製剤中に配合
されたり、保湿効果を高めるために化粧料中に配
合されてきた。また、単純水溶液を利尿剤として
内服する場合もある。 しかし、尿素は水の共存下で除々に分解するこ
とが知られており、この分解は酸、アルカリの共
存下ではさらに促進される。尿素が分解するとア
ンモニアガスが発生し、このものは、周知の通
り、強い刺激臭を有するので、大きな欠点となつ
ていた。 ［発明が解決しようとする問題点］ 本発明者らはかかる事情に鑑み尿素の分解反応
を抑制して安定な尿素水溶液配合製剤を開発すべ
く鋭意検討を重ねた結果、本発明を完成した。 ［問題点を解決するための手段］ すなわち、本発明は、尿素水溶液に、塩基性ア
ミノ酸塩を配合することを特徴とする尿素の安定
化法である。 以下、本発明について詳述する。 本発明において用いられる塩基性アミノ酸塩を
構成する塩基性アミノ酸としてはヒスチジン、リ
ジン、オルニチン、アルギニンなどがあり、塩と
しては塩酸塩、臭化水素酸塩をはじめ、ピロリド
ンカルボン酸塩、ウロカニン酸塩などの有機酸塩
も用いることができる。 塩基性アミノ酸塩の添加量は、配合される尿素
の量あるいは保存される条件、期間などや希望す
る分解抑制の度合によつてそれぞれ適量を選ぶこ
とができるが、尿素量に対する塩基性アミノ酸塩
量が増すほど抑制効果は大きくなる。安定性を向
上させる目的で配合される塩基性アミノ酸塩の量
は、尿素に対して重量で1/10倍以上、３倍以下が
望ましい。但し、過剰の塩基性アミノ酸塩の添加
は塩基性アミノ酸塩自身が変質し、変色、変臭な
どを生じるために注意が必要である。また本発明
で用いられる尿素水溶液の濃度は任意である。さ
らに、尿素水溶液が油相等と乳化されて乳状製剤
の形等になつていても本発明の効果には全く差し
支えない。 尿素の微量の分解を追跡するためにはPH変化を
観察するのが簡便でしかも正確である。すなわ
ち、尿素が分解するとPHが上昇する。以下に尿素
と塩基性アミノ酸塩を併用した場合の系のPH変化
などを示す。表１は尿素および塩基性アミノ酸塩
をそれぞれの量、精製水に溶解し、全量を100と
した水溶液のPH値変化とアンモニア臭の評価結果
である。塩基性アミノ酸塩を添加しない尿素のみ
の水溶液の場合はPH値が上昇し、それに伴つてア
ンモニア臭が発生する。しかし本発明の塩基性ア
ミノ酸塩を添加したものについてはPH変化は若干
あるもののかなり低くおさえられ、アンモニア臭
も認められない。

次に本発明を実施例および比較例によりさらに
詳細に説明するが、本発明はこれにより限定され
るものではない。 実施例 １ Ａ セタノール 3.0％ 固型パラフイン 5.0 ワセリン 15.0 流動パラフイン 20.0 ステアリン酸モノグリセリンエステル 2.0 P.O.E.(20)ソルビタンモノステアレート 2.0 Ｂ 尿素 2.0 アルギニン塩酸塩 5.0 リジン酢酸塩 1.0 精製水 全体を100とする量 Ａに含まれる成分を混合し70℃に加熱し、同様
に加熱したＢの成分中に加える。乳化処理後冷却
処理を行い、外用尿素配合剤を得た。 比較例 １ 実施例１処方で塩基性アミノ酸塩を添加しない
もの。 実施例１および比較例１の試料を40℃にて２ケ
月保存した後、尿素の定量を行つた結果、実施例
１では96〜99％の尿素が残存していたのに対し、
比較例１では80〜85％に減少していた。 実施例 ２ 尿素 5.0％ 精製糖 2.0 Ｌ−オルニチン塩酸塩 3.0 精製水 90.0 各成分を精製水に混合溶解し、内用利尿剤とす
る。 比較例 ２ 実施例２処方でＬ−オルニチン塩酸塩を添加し
ないもの。 実施例２および比較例２の試料を40℃にて２ケ
月保存したのち尿素の定量を行つた結果、実施例
２では97〜99％の尿素が残存していたのに対し、
比較例２では76〜83％に減少していた。
